Study of Effect and Mechanism of Curcumin Protect Myocardial Cells Against Ischemia and Reperfusion Injury

Abstract:[Objective] To investigate the protective effect of curcumin on myocardial ischemia and reperfusion (IR) injury and the possible mechanism. [Methods] Rabbits were randomly divided into four groups: simple IR group, curcumin co-treatment group, curcumin pre-treatment group, and curcumin+Zinc protoporphrinⅨ (ZnPPⅨ) pre-treatment group. Each group was given respectively the simple IR, curcumin (30 mg/kg) injecting via ear marginal vein with IR injury, curcumin (30 mg/kg) injecting via ear marginal vein 8hrs before IR injury, and curcumin (30mg/kg)+ZnPPⅨ (7.5 mg/kg) injecting via ear marginal vein 8hrs before IR injury. Myocardial IR was carried out by ligation of left anterior descending coronary artery for 40min followed by 6hrs of relaxtion. The myocardial HO-1 protein expression and its activity before IR and the neutrophil infiltration, superoxide dismutase (SOD), malodiadehyde (MDA), myocardial infarction range, and myocardial cells apoptosis index (AI) after IR were detected. [Result] campared with simple IR group, curcumin only had slight antioxidative effect and reduced myocardial infarction range gently (P 0.05 respectively), but exerted no effect on AI (P 0.05). Pretreatment with curcumin resulted in an enhanced expression and activity of HO-1 protein, which prevented the neutrophil infiltration and oxidative stress and reduced the myocardial infarction range and AI significantly (P 0.01 respectively). This protective effect was considerably attenuated by ZnPPⅨ, an inhibitor of HO-1. [Conclusion] Curcumin could protect against myocardial IR injury by antioxidative effect and it up-regulatis the expression and activity of HO-1 protein, which was a main component in curcumin-mediated protective effect.
