Abstract:beta-glucosidase produced by Aspergillus niger AS 3.4309 was carried out under Solid State Fermentation (SSF). The effects of medium composition and incubation conditions on the enzyme activity were investigated and suitable solid medium components and optimum incubation conditions for fermentation process were therefore established. The maximum beta-glucosidase production (508 U g(-1) dry matter) occurred after 72 h of incubation at 28 degrees C in optimized solid medium containing wheat bran 100%, ammonium chloride 2%, MgSO4 center dot 7H(2)O 0.5% and KH2PO4, 1.5% at the optimal conditions including moisture content of solid substrate 70% (w/w) initial pH 6.0 and inoculum level at 1 x 10(7) spores g(-1) of solid substrate.
High Expression of Thermostable Xylanase XYNB from Aspergillus Niger in Pichia Pastoris

Abstract:The high expression of xylanase with high specific activity is the effective way to further improve the fermentation potency of xylanase and cut down the production costs. In this study, a recombinant plasmid pPIC9K-XynB was constructed by inserting gene XynB (without signal peptide) from Aspergillus niger into Pichia pastoris secretary vector pPIC9K. Linearized pPIC9K-XynB was transformed into P. pastoris GS115 by electroporation. The positive recombinant strain identified by G418 selection and confirmed by PCR analysis was induced by 0.5% methanol at 28 ℃ to express the recombinant xylanase. The SDS-PAGE analysis showed that the protein molecular weight was about 20 000. The enzyme production was high and stable (4 757 U/mg) when the methanol induced time interval was 12 h and the methanol concentration was 0.5%. The optimal temperature and pH of the enzyme activity was 55 ℃ and 5.0, respectively. After treated for 30 min at 80 ℃, there was still 74% residual activity.

Abstract:The components of a natural medium were optimized to produce cellulase from a marine Aspergillus niger under solid state fermentation conditions by response surface methodology. Eichhornia crassipes and natural seawater were used as a major substrate and a source of mineral salts, respectively. Mineral salts of natural seawater could increase cellulase production. Raw corn cob and raw rice straw showed a significant positive effect on cellulase production. The optimum natural medium consisted of 76.9 % E. crassipes (w/w), 8.9 % raw corn cob (w/w), 3.5 % raw rice straw (w/w), 10.7 % raw wheat bran (w/w), and natural seawater (2.33 times the weight of the dry substrates). Incubation for 96 h in the natural medium increased the biomass to the maximum. The cellulase production was 17.80 U/g the dry weight of substrates after incubation for 144 h. The natural medium avoided supplying chemicals and pretreating substrates. It is promising for future practical fermentation of environment-friendly producing cellulase.

Abstract:Aspergillus niger NRRL3 was cultivated in a moist wheat bran and ground corncob solid medium supplemented with inorganic minerals for the production of cellobiase (β-1,4-glucosidase, EC 3.2.1.21). With this method, A. niger NRRL3 was able to produce a high concentration of cellobiase (215 IU/gofsolid substrate) after 96 h of incubation. Temperature and moisture content affected final cellobiase titers. The best conditions for cell obiase production from solid substrate by A. niger NRRL3 were determined to be 70% moisture and 35°C.

Abstract:The optimal fermentation conditions for production of xylanase by engineered Pichia pastoris GS115-ATx in shake-flask cultivation was investigated. The results showed that the best carbon and nitrogen sources for xylanase production were bran and beef extract, respectively. Judging from their effect on xylanase production efficiency, organic nitrogen was better than inorganic nitrogen. The best methanol concentration was 0.5% for inducing xylanase expression. The xylanase activity was increased when 0.5% glycerin was added into the culture every 24 h. In addition, Toween 20, Toween 80, and betaine were found to increase the yield of xylanase, and among them betaine was the optimal stimulator. SDS-PAGE analysis revealed that the molecular weight of xylanase expressed by GS115-ATx was about 19.0 kDa. The xylanase activity of the culture supernatant reached its peak after 96-h methanol induction.

Abstract:The mature peptide of Aspergillus niger xylanase A (AnxA) was successfully expressed in Pichia pastoris at high levels under the control of AOX1 promoter. The recombinant AnxA (reAnxA) was secreted into culture medium. After 96-h 0.25% methanol induction, the activity of reAnxA in the culture supernatant reached the peak, 175 U/mg, which was 1.9 times as high as that of the native AnxA (92 U/mg). Studies on enzymatic properties showed that the optimum temperature and optimum pH of reAnxA were 50 degrees C and 5.0, respectively. The reAnxA was very stable in a wide pH range of 3.0-8.0. After incubation at the pH 3.0-8.0, 25 degrees C for 1h, all the residual activities of reAnxA were over 80%. The K(m) and k(cat) values for reAnxA were 4.8 mg/ml and 123.2s(-1), respectively. HPLC analysis showed that xylotriose was the main hydrolysis product of birchwood xylan and bran insoluble xylan by reAnxA.
Optimization by Response Surface Methodology of Solid-state Fermentation Medium for α-Galactosidase Production by Aspergillus niger ZJUQH

Abstract:In order to enhance the production of α-galactosidase by Aspergillus niger ZJUQH,response surface methodology was applied to optimize the solid culture medium.A fractional factorial design was used to investigate the main factors affecting α-galactosidase yield.The results showed that two factors played important roles in the production of α-galactosidase.Central composite design and response surface analysis were applied to establish a second-order regression equation model for the yield of α-galactosidase.At last,the optimal fementation condition were obtained by response surface analysis as follows: peptone 0.6 g,Na2HPO4.12H2O 0.08 g,ratio of solid to water 0.41 and inoculum quantity 1.5 mL/5g.Under the optimal conditions,the activity of α-galactosidase reached(77.21±2.01) U/g,which was increase by 57.41% compared with(49.05 ± 2.11) U/g before the oprimization,that is,peptone 0.4 g,Na2HPO4.12H2O 0.08 g,ratio of solid to water 0.33 and inoculation quantity 1.5 mL/5 g.
Effect of Culture Conditions on the Growth and Hydrolytic Performance of Aspergillus niger

Abstract:The effects of different culture conditions,including different types and concentrations of nitrogen sources and surfactants,fermentation time,temperature,and rotation speed,on the biomass and hydrolytic activity of Aspergillus niger (A.niger) cells were studied in this paper.The results showed that different nitrogen sources enhanced the production of the induced enzymes with hydrolytic activities in A.niger cells,and the rate of enhancement by the different nitrogen sources were different.Among the tested nitrogen sources,yeast extract showed the best inducing effect on the cells.For yeast extract concentration of 1.5 g/L,the biomass of A.niger corresponds 1.35 g/L.Among the surfactants examined,the optimal hydrolytic activity was found in the A.niger cells cultured using Triton X-100.In the culture media with 0.5％ Triton X-100,the hydrolysis efficiency ofA.niger reached 58.12％.In addition,the effects of inorganic salts,culture temperature,and rotation speed were also investigated and the optimal conditions to cultivate Aspergillus niger cells with hydrolytic activities were as follows:yeast extract 1.5 g/L,Triton X-100 0.5％,magnesium sulfate heptahydrate 1 g/L,monopotassium phosphate 1 g/L,potassium chloride 0.5 g/L,ammonium sulfate 1.5 g/L,and anhydrous calcium chloride 0.1 g/L.The ideal culture temperature and rotation speed were confirmed as 35℃and 140 r/min,respectively.Under the optimal culture conditions,biomass and hydrolysis efficiency of A.niger cells were 1.73 g/L and 90.23％,respectively.
Optimized Solid-State Fermentation of Aspergillus Niger for Production of Feed-Purpose Β-Glucanase and Xylanase Using Rice Residues from Mass Production of Fungal Biocontrol Agents

Abstract:Rice residues from mass production of fungal biocontrol agents against insect pests were explored for use in the production of feed-purpose β-glucanase and xylanase,which is usually achieved by solid-state fermentation (SSF) of Aspergillus niger with wheat bran. A mixture of 70% wheat bran and 30% milled rice was selected as an acceptable substrate in the trials of 10 ratios of the two components and yielded 353.8 and 343.8 U·g-1 substrate for β-glucanase and xylanase after 3-day SSF,respectively. Factors found to significantly affect the enzyme yields in the mixture during SSF were the type of metal ion as trace nutrition,the content of (NH4)2SO4 as supplementary nitrogen,and the concentration of Tween-80 and pH in the water used to stir the mixture for SSF. Combinations of the selected factors were further optimized for enzyme production in triple four-factor,three-level orthogonal experiments. The optimized combination of the rice-bran mixture consisted of 0.1% KH2PO4,1.0% (NH4)2SO4 and the water containing 0.3% Tween-80 with pH 5. When this optimized mixture was used as substrate in three repeated trials of 54-h SSF at 25 ℃ for confirmation,the mean (±SD) yields of β-glucanase and xylanase reached (716.1±9.2) and (453.0±62.5) U·g-1,which were greatly enhanced compared to those before optimization. The results highlight an alternative approach to utilizing the rice residues for the enzyme production at the reduced cost of 30% wheat bran and would facilitate to establish a rice-based,environment-friendly technology for bulk production of fungal biocontrol agents.

Abstract:To obtain the optimum fermentation medium and circumstances for extracellular lipase construction (as an important biocatalyst and promising industrial enzyme) by Aspergillus Niger, the fermentation conditions of Aspergillus Niger were optimized by single factor and response surface design, the enzymatic properties of the crude enzyme were studied. The results displayed that the optimum fermentation medium was soluble starch 4%, (NH4)2SO4 0.1%, K2HPO4 0.1%, MgSO4·7H2O 0.05%, peptone 3%, olive oil 1.05% and initial pH 7. The optimal fermentation conditions were 30℃, the sample size was 26 mL/250 mL and the shaking speed was 213 r/min. The optimized lipase activity was 1.55 U/mL, which was 7.75 times of the pre-optimized lipase. It was found that when the pH value of lipase was 7.0, the activity of lipase reached its maximum value of 79.3±6.82%. When pH value was between 6.0 and 8.0, the activity of lipase could be kept above 60% and the stability was good. At the same time, through temperature stability of lipase, found that the lipase was stable at 25℃- 35℃, its activity could reach more than 70%. When activity of enzyme reaches to maximum 107.6±9.57%, the temperature was 30℃.

Abstract:An enzymatic extract from Aspergillus niger 3T5B8 was produced by Solid State Fermentation (SSF) in aerated columns, using wheat bran as substrate. A combination of extracts produced using three different process conditions varying temperature, pH and aeration formed the final extract (Mixture). The Mixture was concentrated by an ultrafiltration process that partially purified and provided an efficient recovery of the enzymatic activities of xylanase (88.89%), polygalacturonase (89.3%), β-glucosidase (93.15%), protease (98.68%) and carboxymethylcellulase (CMCase) (98.93%). SDS-PAGE analysis showed 15 visible protein bands in the crude and concentrated Mixture with molecular weights ranging from 15.1 to 104.6 kDa. Thin layer chromatography confirmed the effective action of β-glucosidase and xylanase hydrolysis activities over cellobiose and xylan, respectively. A central composite design (CCD) with two variables and four replicates at the center points was used to determine the optimal temperature and pH for CMCase and β-glucosidase. The optimal temperature was 78.9 °C and pH 3.8 for CMCase and 52.8 °C and pH 4.8 for β-glucosidase, respectively.

Abstract:By single-and-multiple-factor orthogonal experiments, the optimum fermentation conditions of aminoacylase-producing Aspergillus oryzae were determined as (p/g L -1): glucose 40, sucrose 10, soluble starch 20, peptone 2.5, potato juice 1 000 mL, inoculation volume 4%, fermentation temperature 30°C and rotation speed 100 r min -1. The highest total enzyme activity, 7 338 u, was obtained after fermentation for 42 h and increased by 279% compared with the original value of 2 627 u before optimization. The optimal pH and temperature of aminoacylase were 7.0 and 40°C, respectively. Low-concentration Co 2+ (5 × 10 -4 mol/L)) activated the aminoacylase remarkably.
Studies on the Submerged Culture Medium Optimization of Ferulic Acid Esterases Production by Aspergillus Niger XTY

Abstract:In order to enhance the production of ferulic acid esterases by Aspergillus niger XTY, response surface methodology was applied to optimize the submerged cultivation medium. A fractional factorial design was used to investigate the main factors affecting ferulic acid esterases yield of Aspergillus niger XTY. The results showed that two factors played important roles in the production of ferulic acid esterases, which including bean dregs and peptone. Central composite design and response surface analysis were applied to establish the second-order regression equation model of the yield of ferulic acid esterases. Based on this model, the optimal fermentation conditions were obtained as follows: 2.26% bean dregs, 1.035% peptone, 0.3%KH2PO4, 0.8%Na2HPO4·7H2O, 0.01%NaCl and 0.02%MgSO4·7H2O. Under the optimum conditions, the specific activity of ferulic acid esterases increased nearly 1.56 times to 944.56 U/g and the experimental values were consistent with the predicted values. These results suggest that the predicted model was reliable and suitable for the optimization of ferulic acid esterases.

Abstract:Single factor experiment method and uniform design methodology were applied to optimize the fermentation of Aspergillus niger C112 for β-glucosidase. Many factors affected the fermentation of β-glucosidase, including the inoculation method(the vitality of strains), inoculum concentration, type and concentration of carbon and nitrogen sources, fermentation time and temperature, and initial pH, et al. The results show that the seed inoculation effectively improved the strains vitality and thus raising the enzyme production capacity; One of important factors which affected the enzyme production was the carbon particle size; Higher β-glucosidase activity was obtained when corn-core particle diameter was greater than 0.180 mm; The optimal fermentation conditions were as follows: corncore 42 g/L, malt extract 18 g/L, initial pH 4.87, cultivation for 120 hours under the temperature of 30 ℃. With the optimal fermentation conditions, the ultimate β-glucosidase activity reached to 12.925 U/mL,increasing by 79.0 % than before.

Abstract:The liquid-state fermentation optimal conditions of Asp. niger FH11 producing p-glucanase were obtained as follows: a 250mL triangle flash containing 50mL medium volume was kept shaking for 200r/min, at 32C , the fermentation initial pH 6. 0, the fermentation period was 50h. As a result, the activity of p-glucanase in fermentation broth reached 86 U/mL.

Abstract:Xylanase is an enzyme that acts on xylan and degrades it into xylose. Xylanase may be produced by several different microbes such as bacteria and fungi. Xylanase has significant applications in food industries for improving dough handling and improving the quality of baked products in the extraction of starch, coffee, and plant oils. Xylanase, along with pectinase and cellulose, has enormous application in the clarification of fruit juices. In the present study, xylanase was produced by solid-state fermentation using lemon peel as substrates. Aspergillus Niger was induced to produce xylanase by frequent sub-culturing on a medium containing 2% xylan. OFAT was analyzed by using several fermentation parameters such as moisture content, particle size, incubation period, incubation temperature, peptone concentration, and pH of the extraction buffer. Under optimized conditions, the maximum xylanase activity was found for particle size of 1.7mm, moisture content of 80%, peptone concentration in nutrient solution at 0.3%, and extraction pH of 7.0. Hence, xylanase can be further subjected to down streaming processes and therefore for various applications.

Abstract:Response surface methodology was used to optimize a medium for β-glucosidase production of Aspergillus niger ZJ1. In the first optimization step,the influence of straw powder, barley flour,wheat bran,(NH4)2SO4 and pH on β-glucosidase production was evaluated using a fractional factorial design. Straw powder and(NH4)2SO4 had negative effect on β-glucosidase production,while other components had no significant effect on β-glucosidase production. The path of steepest ascent was used to approach the optimal region of the medium composition. In the third step,the optimal concentrations of straw powder and(NH4)2SO4 were determined by a central composite design and response surface analysis. The optimized composition of fermentation medium was(g/L):straw powder 7.02,wheat bran 16.65,barley flour 16.65,(NH4)2SO4 2.44,KH2PO4 0.5,MgSO4·7H2O 0.5. The maximum enzyme activity predicted was 403.7U/mL under the optimized cultivation medium composition.

Abstract:Proteases, also known as proteinases or proteolytic enzymes, belong to a group of hydrolases. It can be applied in numerous fields and industries. Solid-state fermentation (SSF) is recognized as an effective method to produce protease. The ultimate aim of this study is to optimize the production of protease from Aspergillus niger under solid-state fermentation (SSF) by utilizing shrimp shell powder as a solid substrate. It was found that the produced protease from SSF was slightly alkaline. The correlation between factors operating parameters (incubation temperature, inoculum size, moisture content) for enzyme production is analyzed using statistical software, Minitab 16. A 23 full factorial experimental design was employed, and the enzyme produced was optimized by the method of desirability function. The optimal conditions for protease production of 3.7 U/mg were 35 °C of incubation temperature, 60% of initial moisture content, and 1.0 inoculum size. It is concluded that SSF protease was successfully produced from Aspergillus niger by utilizing shrimp waste as substrate. Through optimization study, moisture content, the interaction between incubation temperature and moisture content, interaction between moisture content and inoculum size significantly impact protease production.

Abstract:Fatty acid ethyl esters are important flavor chemicals in strong-flavor baijiu. Microorganisms are the main contributors to ester synthesis during baijiu manufacture. However, the ester synthesis was unstable between batches. This was owing to a limited knowledge of the mechanisms for ester synthesis by microorganisms. In this work, a fatty acid ethyl ester synthesizing Aspergillus niger strain CGMCC (China General Microbiological Culture Collection) 3.4309 was identified. The conversion ratios of ethyl valerate, ethyl caproate, ethyl caprylate, and ethyl caprate were 7.87, 29.20, 94.80, and 85.20%, respectively, under the optimized conditions. A comparison of transcriptomes under the initial and optimized ester synthetic conditions indicated that 23 genes were upregulated in transcription level and encoded enzymes with potential abilities for ester synthesis. Eleven of the enzymes were expressed, and three of them, numbered An605, An1097, and An3131, showed the ability to catalyze fatty acid ethyl ester synthesis under aqueous phase, with capric acid as the preferred substrate. The possible enzymatic catalytic mechanism was proposed based on homology modeling and molecular docking. This study reported for the first time that A. niger showed the ability to efficiently catalyze the synthesis of short- and medium-chain fatty acid ethyl esters in aqueous phase, identified the key enzymes, and analyzed the basic enzymatic properties. This is helpful to promote the application of related microorganisms and enzyme resources in the baijiu industry.

Abstract:Xylanase is a member of an important family of enzymes that has been used in many biotechnological processes. However, the overall cost of enzyme production has been the main problem in the industrial application of enzymes. To obtain maximum xylanase production, statistical approaches based on the Plackett–Burman design and response surface methodology were employed. The results of the statistical analyses demonstrated that the optimal conditions for increased xylanase production were the following: inoculum size, 3.8%; maize meal, 4.5%; histidine, 0.6%; methanol, 1%; culture volume, 20%; bean pulp, 30 g L−1; and Tween‐80, 0.8%; and pH 5.0. Verification of the optimization demonstrated that 3273 U mL−1 xylanase was observed under the optimal conditions in shake flask experiments. SDS–PAGE results showed that the size of xylanase protein was about 23 kDa. The results showed that the xylanase produced by fermentation came from Aspergillus Niger by MALDI‐TOF‐MS. The optimized medium resulted in 2.1‐ and 1.4‐fold higher the activity of xylanase compared with the unoptimized medium (the main nutrients are maize meal and bean pulp) and laboratory medium (the main nutrients are yeast extract and peptone), respectively. The optimization of fermentation conditions is an effective means to reduce production cost and improve xylanase activity.
