Ursolic Acid Induces Differentiation of B16 Melanoma Cells

Abstract:[Objective] To evaluate the differentiation effect of Ursolic Acid (UA) on B16 melanoma cells. [Methods] MTT method was used to determine the inhibitory rate of cell proliferation. The B16 cells′s morphological and electron microscope changes, melanin synthesis, activity of tyrosinase and tumorigencity in mice were assessed after UA-exposed B16 cells for different time. [Results] UA at concentration 4~64 μM showed different inhibition to B16 melanoma cells(P 0.05), B16 cells treated with UA for 12, 24 and 48 h, the IC50 value was 58.05 μM, 35.13 μM, 12.17 μM, respectively. After exposed to UA, the morphological of B16 cells changed greatly and rich mitochondria, rough endoplasmic reticulum and elastic filament appeared in the cells, however, in control group, the above-mentioned structures couldn′t be observed except numerous freely scattered polyribosomes. Moreover, UA could increase melanin biosynthesis (P 0.01 vs control) and up-regulate tyrosinase activity (P 0.01 or 0.05 vs control) of B16 cells. Furthermore, UA decreased vivo tumorigencity of B16 (P 0.01 vs control). [Conclusion] UA can strongly induce differentiation of B16 cells, but the mechanisms should be needed for further investigated.
