Abstract:Stripe rust, caused by Puccinia striiformis f. sp. tritici, is a continuous threat to global wheat production. In 2021, the epidemic of wheat stripe rust in China affected approximately 4.5 million hectares, resulting in severe yield losses. When confronted with the epidemic, tracing the sources of the pathogen can offer valuable insights for disease prevention and control. This study was conducted to analyze the genetic structure, aerodynamics, geographical features, and cultivation practices of the pathogen population in various wheat-producing regions, and to further reveal the spread patterns of the stripe rust pathogens in China. The findings indicated an overall trend of the pathogen dissemination from the west to the east. The pathogen was primarily spread from the northwestern region to the Huang-Huai-Hai region through the Guanzhong Plain and the NanXiang Plain. Meanwhile, the pathogen was also spread eastward from the southwestern region to the lower reaches of the Yangtze River, utilizing the Jianghan Plain as a bridge and the Yangtze River Valley in southwestern Anhui as the main pathway. Furthermore, the pathogen spread northward into Shandong under the driving force of the southeast winds. The findings of this study may provide valuable insights for the integrated management of wheat stripe rust in China.

Abstract:Stripe rust is one of the most destructive wheat diseases in China, negatively affecting the production safety and causing yield losses of wheat. Thus, it is important to analyze the environmental suitability and dynamic changes of wheat stripe rust in China. The occurrence of stripe rust is affected by multiple factors. Therefore, this study combined data from various disciplinary fields such as remote sensing, meteorology, biology, and plant protection to evaluate the environmental suitability of stripe rust in China using species distribution models. The study also discusses the importance and effect of various variables. Results revealed that meteorological factors had the greatest impact on the occurrence of stripe rust, especially temperature and precipitation. Wheat growth factors have a greater impact from April to August. Elevation has a greater impact in summer. The ensemble model results were better than the single model, with TSS and AUC greater than 0.851 and 0.971, respectively. Overlapping analysis showed that the winter stripe rust suitable areas were mainly in the Sichuan Basin, Northwestern Hubei, Southern Shaanxi, and Southern Henan wheat areas. In spring, the suitable areas of stripe rust increased in Huang-Huai-Hai and the middle and lower reaches of the Yangtze River and Guanzhong Plain, and the development of northwestern wheat areas such as Xinjiang and Gansu slightly lagged behind. In summer, wheat threatened by stripe rust is mainly in late-ripening spring wheat areas in Gansu, Ningxia, Qinghai, and Xinjiang. This study can provide a scientific basis for optimizing and improving the comprehensive management strategy of stripe rust.

Abstract:Wheat rusts, including leaf, stripe, and stem rust, have been a threat to global food security due to their devastating impact on wheat yields. In recent years, significant strides have been made in understanding wheat rusts, focusing on disease spread mechanisms, the discovery of new host resistance genes, and the molecular basis of rust pathogenesis. This review summarizes the latest approaches and studies in wheat rust research that provide a comprehensive understanding of disease mechanisms and new insights into control strategies. Recent advances in genetic resistance using modern genomics techniques, as well as molecular mechanisms of rust pathogenesis and host resistance, are discussed. In addition, innovative management strategies, including the use of fungicides and biological control agents, are reviewed, highlighting their role in combating wheat rust. This review also emphasizes the impact of climate change on rust epidemiology and underscores the importance of developing resistant wheat varieties along with adaptive management practices. Finally, gaps in knowledge are identified and suggestions for future research are made. This review aims to inform researchers, agronomists, and policy makers, and to contribute to the development of more effective and sustainable wheat rust control strategies.

Abstract:Puccinia striiformis (Pst) is a devastating biotrophic fungal pathogen that causes wheat stripe rust. It usually loves cool and moist places and can cause 100% crop yield losses in a single field when ideal conditions for disease incidence prevails. Billions of dollars are lost due to fungicide application to reduce stripe rust damage worldwide. Pst is a macrocyclic, heteroecious fungus that requires primary (wheat or grasses) as well as secondary host (Berberis or Mahonia spp.) for completion of life cycle. In this review, we have summarized the knowledge about pathogen life cycle, genes responsible for stripe rust resistance, and susceptibility in wheat. In the end, we discussed the importance of conventional and modern breeding tools for the development of Pst-resistant wheat varieties. According to our findings, genetic engineering and genome editing are less explored tools for the development of Pst-resistant wheat varieties; hence, we highlighted the putative use of advanced genome-modifying tools, i.e., base editing and prime editing, for the development of Pst-resistant wheat.

Abstract:Stripe rust caused by Puccinia striiformis f. sp. tritici (Pst) is one of the most devastating foliar diseases in wheat. Host resistance is the most effective strategy for the management of the disease. To screen for accessions with stable resistance and identify effective stripe rust resistance loci, a genome-wide association study (GWAS) was conducted using a panel of 140 Chinese wheat landraces. The panel was evaluated for stripe rust response at the adult-plant stage at six field-year environments with mixed races and at the seedling stage with two separate predominant races of the pathogen, and genotyped with the genome-wide Diversity Arrays Technology markers. The panel displayed abundant phenotypic variation in stripe rust responses, with 9 landraces showing stable resistance to the mixture of Pst races at the adult-plant stage in the field and 10 landraces showing resistance to individual races at the seedling stage in the greenhouse. GWAS identified 12 quantitative trait loci (QTL) significantly (P ≤ 0.001) associated to stripe rust resistance using the field data of at least two environments and 18 QTL using the seedling data with two races. Among these QTL, 10 were presumably novel, including 4 for adult-plant resistance mapped to chromosomes 1B (QYrcl.sicau-1B.3), 4A (QYrcl.sicau-4A.3), 6A (QYrcl.sicau-6A.2) and 7B (QYrcl.sicau-7B.2) and 6 for all-stage resistance mapped to chromosomes 2D (QYrcl.sicau-2D.1), 3B (QYrcl.sicau-3B.3), 3D (QYrcl.sicau-3D), 4B (QYrcl.sicau-4B), 6A (QYrcl.sicau-6A.1) and 6D (QYrcl.sicau-6D). The landraces with stable resistance can be used for developing wheat cultivars with effective resistance to stripe rust.

Abstract:Stripe rust (caused by Puccinia striiformis f. sp. tritici ) is one of the most severe diseases affecting wheat production. The disease is best controlled by developing and growing resistant cultivars. Chinese wheat ( Triticum aestivum ) landraces have excellent resistance to stripe rust. The objectives of this study were to identify wheat landraces with stable resistance and map quantitative trait loci (QTL) for resistance to stripe rust from 271 Chinese wheat landraces using a genome-wide association study (GWAS) approach. The landraces were phenotyped for stripe rust responses at the seedling stage with two predominant Chinese races of P. striiformis f. sp. tritici in a greenhouse and the adult-plant stage in four field environments and genotyped using the 660K wheat single-nucleotide polymorphism (SNP) array. Thirteen landraces with stable resistance were identified, and 17 QTL, including eight associated to all-stage resistance and nine to adult-plant resistance, were mapped on chromosomes 1A, 1B, 2A, 2D, 3A, 3B, 5A, 5B, 6D, and 7A. These QTL explained 6.06–16.46% of the phenotypic variation. Five of the QTL, QYrCL.sicau-3AL , QYrCL.sicau-3B.4 , QYrCL.sicau-3B.5 , QYrCL.sicau-5AL.1 and QYrCL.sicau-7AL , were likely new. Five Kompetitive allele specific PCR (KASP) markers for four of the QTL were converted from the significant SNP markers. The identified wheat landraces with stable resistance to stripe rust, significant QTL, and KASP markers should be useful for breeding wheat cultivars with durable resistance to stripe rust.

Abstract:Stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), is one of the most destructive diseases of wheat worldwide. The disease is most preferably managed by developing and growing cultivars with high-level, durable resistance. To achieve high-level and long-lasting resistance, we developed a wheat line, RIL-Yr64/Yr15, by pyramiding Yr64 and Yr15, both on the chromosome 1BS and providing high resistance to all tested Pst races. To validate RIL-Yr64/Yr15 possessing both genes, we crossed it to Avocet S (AvS). The F4 RILs from this cross were phenotyped with Pst races under controlled greenhouse conditions and also under natural Pst infection in the field. The population was genotyped with SSR markers previously reported to be linked to the resistance gene loci and with additional SSR and SNP-KASP markers along chromosome 1B. Both phenotype and genotype data confirmed the copresence of Yr64 and Yr15 in RIL-Yr64/Yr15, and the high-resolution linkage map dissected the chromosomal regions and traced their origins. New lines possessing these genes were selected from the F5 population of cross AvS × RIL-Yr64/Yr15 by marker-assisted selection. These lines with the two highly effective genes should be more useful than individual gene lines for developing high-level, durable resistant wheat cultivars.

Abstract:Abstract Background Stripe rust, caused by the fungal pathogen Puccinia striiformis f. sp. tritici ( Pst ), is a serious foliar disease of wheat. Identification of novel stripe rust resistance genes and cultivation of resistant cultivars are considered to be the most effective approaches to control this disease. In this study, we evaluated the infection type (IT), disease severity (DS) and area under the disease progress curve (AUDPC) of 143 Chinese wheat landrace accessions for stripe rust resistance. Assessments were undertaken in five environments at the adult-plant stage with Pst mixture races under field conditions. In addition, IT was assessed at the seedling stage with two prevalent Pst races (CYR32 and CYR34) under a controlled greenhouse environment. Results Seventeen accessions showed stable high-level resistance to stripe rust across all environments in the field tests. Four accessions showed resistance to the Pst races CYR32 and CYR34 at the seedling stage. Combining phenotypic data from the field and greenhouse trials with 6404 markers that covered the entire genome, we detected 17 quantitative trait loci (QTL) on 11 chromosomes for IT associated with seedling resistance and 15 QTL on seven chromosomes for IT, final disease severity (FDS) or AUDPC associated with adult-plant resistance. Four stable QTL detected on four chromosomes, which explained 9.99–23.30% of the phenotypic variation, were simultaneously associated with seedling and adult-plant resistance. Integrating a linkage map of stripe rust resistance in wheat, 27 QTL overlapped with previously reported genes or QTL, whereas four and one QTL conferring seedling and adult-plant resistance, respectively, were mapped distantly from previously reported stripe rust resistance genes or QTL and thus may be novel resistance loci. Conclusions Our results provided an integrated overview of stripe rust resistance resources in a wheat landrace diversity panel from the southern autumn-sown spring wheat zone of China. The identified resistant accessions and resistance loci will be useful in the ongoing effort to develop new wheat cultivars with strong resistance to stripe rust.

Abstract:Since the 1st green revolution,pest problems were getting more serious as significant reduction of natural regulation function in rice ecosystem resulted from susceptible high yielding varieties and heavy use of chemical fertilizers and pesticides in large areas.To reduce pest problems,technology and strategies for pest management have been going through four phases,chemical control,primary integrated pest control,pest management and ecological engineering.However,new problems including serious yield losses caused by small insects,simultaneous outbreaks of multiple diseases,succession of weeds and genetic resistant problems although heavy yield losses have been retrieved.In order to accomplish the sustainable intensified agriculture,plant protection workers should work together with scientists in other disciplines to enhance natural regulating functions in rice ecosystems,coordinated innovation to develop new technology and management system for implementing green plant protection strategies from the view of healthy rice ecosystems.

Abstract:Establishing a species-diversified cropping system to control crop diseases, insect pests and weeds is an important approach to sustainable agricultural development. This paper reviewed the researches on paddy field species-diversified cropping systems at home and abroad, and discussed the controlling effects and mechanisms of multiple species coexistence on rice diseases, pests and weeds control. The multiple species coexistence models such as rice-fish, rice-duck, rice-azolla-fish and rice-azolla-duck had effective controlling effects on Thanatephorus cucumeris (Frank) Donk, Nilaparata lugens Stal, Chaphalocrocis medinalis Gueneeand, and weeds. Other models like intercropping rice with Zizania caduciflora L. and with other wetland crops also had effective effects in controlling the occurrence and spread of Pyricularia grisea. Abstract:Rust fungi cause significant damage to wheat production worldwide. In order to mitigate disease impact and improve food security via durable resistance, it is important to understand the molecular basis of host–pathogen interactions. Despite a long history of research and high agricultural importance, still little is known about the interactions between the stripe rust fungus and wheat host on the gene expression level. Here, we present analysis of the molecular interactions between a major wheat pathogen—Puccinia striiformis f. sp. tritici (Pst)—in resistant and susceptible host backgrounds. Using plants with durable nonrace-specific resistance along with fully susceptible ones allowed us to show how gene expression patterns shift in compatible versus incompatible interactions. The pathogen showed significantly greater number and fold changes of overexpressed genes on the resistant host than the susceptible host. Stress-related pathways including MAPK, oxidation–reduction, osmotic stress, and stress granule formation were, almost exclusively, upregulated in the resistant host background, suggesting the requirement of the resistance-countermeasure mechanism facilitated by Pst. In contrast, the susceptible host background allowed for broad overrepresentation of the nutrient uptake pathways. This is the first study focused on the stripe rust pathogen–wheat interactions, on the whole transcriptome level, from the pathogen side. It lays a foundation for the better understanding of the resistant/susceptible hosts versus pathogenic fungus interaction in a broader sense.

Abstract:Identifying sources of phytopathogen inoculum and determining their contributions to disease outbreaks are essential for predicting disease development and establishing control strategies. Puccinia striiformis f. sp. tritici (Pst), the causal agent of wheat stripe rust, is an airborne fungal pathogen with rapid virulence variation that threatens wheat production through its long-distance migration. Because of wide variation in geographic features, climatic conditions, and wheat production systems, Pst sources and related dispersal routes in China are largely unclear. In the present study, we performed genomic analyses of 154 Pst isolates from all major wheat-growing regions in China to determine Pst population structure and diversity. Through trajectory tracking, historical migration studies, genetic introgression analyses, and field surveys, we investigated Pst sources and their contributions to wheat stripe rust epidemics. We identified Longnan, the Himalayan region, and the Guizhou Plateau, which contain the highest population genetic diversities, as the Pst sources in China. Pst from Longnan disseminates mainly to eastern Liupan Mountain, the Sichuan Basin, and eastern Qinghai; that from the Himalayan region spreads mainly to the Sichuan Basin and eastern Qinghai; and that from the Guizhou Plateau migrates mainly to the Sichuan Basin and the Central Plain. These findings improve our current understanding of wheat stripe rust epidemics in China and emphasize the need for managing stripe rust on a national scale.

Abstract:Stripe rust is one of the most destructive diseases for wheat crops in China. Two stripe rust physiological strains, i.e. CYR30 (intern. name: 175 E 191) and CYR31 (intern. name: 293 E 175) have been the dominant and epidemic physiological strains since 1994. One Aegilops tauschii accession (SQ-214) from CIMMYT was found immune from or highly resistant to Chinese new stripe rust races CYR30 and CYR31 at adult stage. SQ-214 was crossed with a highly susceptible Ae. tauschii accession As-80. Analysis of data from F-1-F-2 populations of SQ-214/As-80 revealed that the resistance was controlled by a single dominant gene. To exploit the resistance for wheat breeding, SQ-214 was crossed with Chinese Spring (CS) and backcrossed by two Chinese commercial wheat varieties MY26 and SW3243. The resistance from SQ-214 was suppressed in the F-1 hybrids (CS/SQ-214) and the F-2 population of CS/SQ-214//MY26. However, the resistance of SQ-214 was expressed in several F-2 individuals of CS/SQ-214//SW3243. Eleven advanced lines with high level of resistance to the Chinese stripe rust CYR30 and CYR31 have been developed. This result suggested that SW3243 does not suppress the expression of the Chinese stripe rust and should be used as wheat germplasm for exploiting resistance of Ae. tauschii in wheat breeding. The gliadin electrophoretic pattern of the eleven advanced lines with high stripe rust resistances was compared with their parents SQ-214, CS and SW3243 by acid polyacrylamide gel clectrophoresis. The omega-gliadin bands of Gli-D(t)1 in Ae. tauschii SQ-214 were transferred to some advanced lines and freely expressed in common wheat genetic background. One of advanced lines possesses a null Gli-D1 allele, where the omega-gliadin bands encoding by the Gli-D1 allele were absent. The potential utilization of this advanced line for wheat quality and stripe rust resistance breeding is also discussed in this paper.

Abstract:The Rice black-streaked dwarf viral disease(RBSDV),transmitted by the small planthopper Laodelphax striatellus Fallen,is one of the most destructive diseases in rice.In order to control this disease,a provincial collaborative research was conducted across the Zhejiang province,China,during 2000 to 2005.The epidemiology,causes of resurgence and damage-yield loss relationship were understood and were applied to design the field sampling and forecast methods,and to determine the economic thresholds of the disease through control of the insect vector.A provincial disease alert system including the GPS,GIS and RS approaches,and an estimation model for virus-infestation in rice were established and were used for long-term forecast of the disease.A new management strategy was proposed which integrated the extension of RBSDV disease-and vector-resistant varieties,improvement of seedling nursery,healthy cultivation of diseased hill gap-filling by removing tillers from nearby healthy hills,and control of insect vectors in the seedlings stage to protect rice plants from the viral disease in field stages.An integrated RBSDV disease management technical system was therefore shaped for the Yangtze Delta region,and applied in large scale,effectively reduced the disease occurrence and damage,achieved significant economic and social effects.

To obtain a better understanding about different effects of redox homeostasis and photosystem (PS) to Pst infection in wheat, we investigated the differences in photosynthesis and the antioxidant defense system in wheat cultivar Chuanmai42 (CM42) in response to two Chinese Pst races known as CYR32 and V26. The results showed that V26-infected wheat accumulated a higher reactive oxygen species (ROS), cell death, and energy dissipation than CYR32-infected wheat when compared with the control. Furthermore, we found that the activities of three antioxidant enzymes (APX, GR, and GPX) and four resistance-related enzymes in CYR32-infected wheat were significantly higher than that in V26-infected wheat. In addition, quantitative RT-PCR indicated that the expression levels of two genes associated with resistant stripe rust in CYR32-infected wheat were clearly higher than that in V26-infected wheat. Compared with CYR32-infected wheat, lower photochemical efficiencies were observed in V26-infected wheat at the adult stage. Meanwhile, only a marked decline in D1 protein was observed in V26-infected wheat. We therefore deduced that wheat with stripe rust resistance could maintain high resistance and photosynthetic capacity by regulating the antioxidant system, disease-resistant related enzymes and genes, and the levels of PSII reaction center proteins.

Abstract:Puccinia triticina , which is the causative agent of wheat leaf rust, is widely spread in China and most other wheat-planting countries around the globe. Cultivating resistant wheat cultivars is the most economical, effective, and environmentally friendly method for controlling leaf rust-caused yield damage. Exploring the source of resistance is very important in wheat resistance breeding programs. In order to explore more effective resistance sources for wheat leaf rust, the resistance of 112 wheat accessions introduced from the U.S. National Plant Germplasm System were identified using a mixture of pathogenic isolates of THTT, THTS, PHTT, THJT and THJS which are the most predominant races in China. As a result, all of these accessions showed high resistance at seedling stage, of which, ninety-nine accessions exhibited resistance at adult plant stage. Eleven molecular markers of eight effective leaf rust resistance genes in China were used to screen the 112 accessions. Seven effective leaf rust resistance genes Lr9 , Lr19 , Lr24 , Lr28 , Lr29 , Lr38 and Lr45 were detected, except Lr47 . Twenty-three accessions had only one of those seven effective leaf rust resistance gene. Eleven accessions carried Lr24 + Lr38 , and 7 accessions carried Lr9 + Lr24 + Lr38 , Lr24 + Lr38 + Lr45 , Lr24 + Lr29 + Lr38 and Lr19+Lr38 + Lr45 respectively. The remaining seventy-one accessions had none of those eight effective leaf rust resistance genes. This study will provide theoretical guidance for rational utilization of these introduted wheat accessions directly or for breeding the resistant wheat cultivars.

Abstract:Stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), is an airborne fungal disease which always destructs leaf and leads to stagnation of grain filling, decreasing of kernel weight, thin seed and yield loss. Winter wheat Huixianhong is a special germplasm with special characteristics of tolerance or resistance against stripe rust. In order to understand the effect on Huixianhong from stripe rust, we designed two kinds of treatment, inoculation of stripe rust fungi (IH) and artificial immune by bactericide (CK) to study the dynamic of disease process, the grain filling and the thousand-kernel weight (TKW). Our results showed that the incubation period of Hongxinahui was 13.5 days. The prevalence increased from 32.9% at 15 days after jointing to 80.0% at 9 days after booting, and reached to 97.0% at the heading stage. The infection type (IT) was 7 to 9 at the beginning of anthesis. The severity, leaf withered area ratio and disease index at the 15th day after anthesis were 67.17%, 98.17% and 0.6717, respectively. The IH maximum increasing rate of leaf necrosis and chlorosis area was from heading beginning stage to anthesis beginning stage, which increased from 18.66 mm2·d-1 to 21.04 mm2·d-1. The maximum rate of grain filling was 1.25 mg·d-1 at the 18th day after anthesis, which was earlier than that of CK by 3.3 days. The IH thousand-kernel weight Loss (TKWL) was more than that of CK by 6.19%, the stage of heading and amature were 3.0 days and 4.5 days earlier than CK, respectively. The stripe rust infection seriously destructed the photosynthetic function of leaf at the earlier stage of grain filling, i.e. at the beginning of anthesis, which led to the most important biomass loss and the grain filling rate decrease. As far as stripe rust is concerned, Huixianhong is a high susceptible, easily been infected, seriously showing symptoms and most quickly epidemic type but can successfully complete grain filling in high quality. It is a very useful germplasm for creating and selecting special breeding materials against stripe rust.

Abstract:Peanuts play a pivotal role as an economic crop on a global scale, serving as a primary source of both edible oil and protein. Peanut rust (Puccinia arachidis Speg.) disease constitutes a significant global biotic stress, representing a substantial economic threat to the peanut industry by inducing noteworthy reductions in seed yields and compromising oil quality. This comprehensive review delves into the distinctive characteristics and detrimental symptoms associated with peanut rust, scrutinizing its epidemiology and the control strategies that are currently implemented. Notably, host resistance emerges as the most favored strategy due to its potential to surmount the limitations inherent in other approaches. The review further considers the recent advancements in peanut rust resistance breeding, integrating the use of molecular marker technology and the identification of rust resistance genes. Our findings indicate that the ongoing refinement of control strategies, especially through the development and application of immune or highly resistant peanut varieties, will have a profound impact on the global peanut industry.

Abstract:Abstract Background Stripe rust (yellow rust) is a significant disease for bread wheat ( Triticum aestivum L.) worldwide. A genome-wide association study was conducted on 240 Chinese wheat cultivars and elite lines genotyped with the wheat 90 K single nucleotide polymorphism (SNP) arrays to decipher the genetic architecture of stripe rust resistance in Chinese germplasm. Results Stripe rust resistance was evaluated at the adult plant stage in Pixian and Xindu in Sichuan province in the 2015–2016 cropping season, and in Wuhan in Hubei province in the 2013–2014, 2016–2017 and 2018–2019 cropping seasons. Twelve stable loci for stripe rust resistance were identified by GWAS using TASSEL and GAPIT software. These loci were distributed on chromosomes 1B, 1D, 2A, 2B, 3A, 3B, 4B (3), 4D, 6D, and 7B and explained 3.6 to 10.3% of the phenotypic variation. Six of the loci corresponded with previously reported genes/QTLs, including Sr2/Yr30/Lr27 , while the other six ( QYr.hbaas-1BS , QYr.hbaas-2BL , QYr.hbaas-3AL , QYr.hbaas-4BL.3 , QYr.hbaas-4DL , and QYr.hbaas-6DS ) are probably novel. The results suggest high genetic diversity for stripe rust resistance in this population. The resistance alleles of QYr.hbaas-2AS , QYr.hbaas-3BS , QYr.hbaas-4DL , and QYr.hbaas-7BL were rare in the present panel, indicating their potential use in breeding for stripe rust resistance in China. Eleven penta-primer amplification refractory mutation system (PARMS) markers were developed from SNPs significantly associated with seven mapped QTLs. Twenty-seven genes were predicted for mapped QTLs. Six of them were considered as candidates for their high relative expression levels post-inoculation. Conclusion The resistant germplasm, mapped QTLs, and PARMS markers developed in this study are resources for enhancing stripe rust resistance in wheat breeding.

Abstract:Stripe or yellow rust (YR) caused by Puccinia striiformis tritici (Pst) is an important foliar disease affecting wheat production globally. Resistant varieties are the most economically and environmentally effective way to manage this disease. The common winter wheat (Triticum aestivum L.) cultivar Luomai 163 exhibited resistance to the Pst races CYR32 and CYR33 at the seedling stage and showed a high level of adult plant resistance in the field. To understand the genetic basis of YR resistance in this cultivar, 142 F5 recombinant inbred lines (RILs) derived from cross Apav#1 × LM163 and both parents were genotyped with the 16K SNP array and bulked segregant analysis sequencing. The analysis detected a major gene, YrLM163, at the seedling stage associated with the 1BL.1RS translocation. Additionally, three genes for resistance at the adult plant stage were detected on chromosome arms 1BL (Lr46/Yr29/Pm39/Sr58), 6BS, and 6BL in Luomai 163, whereas Apav#1 contributed resistance at a quantitative trait locus (QTL) on 2BL. These QTL explained YR disease severity variations ranging from 6.9 to 54.8%. The kompetitive allele-specific PCR (KASP) markers KASP-2BL, KASP-6BS, and KASP-6BL for the three novel loci QYr.hzau-2BL, QYr.hzau-6BS, and QYr.hzau-6BL were developed and validated. QYr.hzau-1BL, QYr.hzau-2BL, and QYr.hzau-6BS showed varying degrees of resistance to YR when present individually or in combination based on genotype and phenotype analysis of a panel of 570 wheat accessions. Six RILs combining resistance alleles of all QTL, showing higher resistance to YR in the field than Luomai 163 with disease severities of 10.7 to 16.0%, are important germplasm resources for breeding programs to develop YR-resistant wheat varieties with good agronomic traits.
