Abstract:Optically active aryl alcohols are a kind of important chiral building blocks for the synthesis of chiral drugs. In recent years, the preparations of enantiopure aryl alcohols have become one of the focus points in organic chemistry. Compared with conventional chemical processes, biocatalysis is more attractive due to its high enantioselectivity, mild and safe reaction conditions and less environmental hazards. Asymmetric bioreduction of aryl ketones is the most effective method for the synthesis of enantiopure aryl alcohols. This paper mainly reviews the recent progress in the preparation of chiral aryl alcohols by asymmetric bioreduction catalyzed by microbial whole-cells, enzymes, genetically engineered bacteria or yeasts, as well as immobilized cells. The effects of substrate co-solvents such as organic solvents, surfactants and ionic liquids on asymmetric bioreduction are further summarized. The prospect of the preparation of enantiopure aryl alcohols by asymmetric bioreduction is also discussed.

Abstract:Abstract Chiral iodoarenes have been steadily increasing in importance in recent years, especially in enantioselective synthesis and catalysis. Since the development of the concept of chiral iodine(I/III) catalysis, the use of various chiral aryliodine frameworks has been explored in this area. Abstract:As a structural analog of oxazoline, imidazoline (4,5-dihydroimidazole) has received much attention in the rational design of chiral ligands. The additional N-substituent provides broader space for fine-tuning of electronic and steric effects, and it offers a good handle for immobilizing onto solid supports. In the past decades, imidazoline ring has emerged as a powerful candidate for the design of chiral nitrogen-containing ligands, as well as a significant alternative for oxazoline ring. Various chiral imidazoline ligands have been designed and utilized in asymmetric organic reactions. These new catalysts can not only be applied in classical reactions, but also be employed to develop new organic reactions with high enantioselectivities. This review provides an overview of chiral imidazoline ligands. Their applications in asymmetric synthesis are also summarized.

Abstract:A variety of chiral bisamide ligands have been prepared and widely used in asymmetric catalytic syntheses with highly regio-and enantioselective products, especially in the transition-metal-catalyzed asymmetric allylic alkylation reactions. The achievements in the asymmetric allylic alkylation reactions with various nucleophiles have been reviewed. The applications of some typical chiral bisamides in decarboxylative asymmetric allylic alkylation of enol carbonates, [3+2] cycloaddition of vinyl cyclopropanes and alkylidene azlactones, and aldol reactions are also introduced.

Abstract:Chiral benzothiazepines constitute the core structures of many foremost pharmaceuticals with diverse biological activities endowed by their unique scaffolds, which poses a great challenge to organic chemists and pharmaceutical researchers. This review provides a concise overview for the asymmetric synthesis of chiral benzothiazepine derivatives, focusing on advances in asymmetric catalysis, including metal catalysis, small-molecule organocatalysis and enzymatic catalysis. The catalytic asymmetric reactions, involving asymmetric epoxidation, reduction, dihydroxylation, hydrogenation, aldol reaction and other sulfa-Michael addition, have emerged as powerful strategies for the rapid construction of chiral benzothiazepine through single or multistep reactions. The booming asymmetric synthetic methodology affords us instructive clues for the highly efficient preparation of chiral benzothiazepines, facilitating their large-scale preparation and diversity-oriented synthesis.

Among the plethora of reported synthetic methods, asymmetric catalysis has emerged as one of the most efficient and direct ways to obtain optically pure chiral compounds. This article mainly reviews the recent progress of asymmetric catalysis made by Chinese chemists from the following aspects: the development of new chiral ligands, transition metal-catalyzed asymmetric reactions, asymmetric organocatalytic reactions, and new strategies for development of novel chiral ligands/catalysts, new chiral drugs and agricultural chemicals.

In this review, we summarize the recent development of chiral phosphoric acid-catalyzed synthesis of a wide range of axially chiral biaryls, heterobiaryls, vinylarenes, N-arylamines, spiranes, and allenes with high efficiency and excellent stereoselectivity.

Abstract:The stereoselective synthesis of β-amino acids has attracted major attention among the synthetic community in recent years. This review provides an overview of the important advances in chiral β-amino acid synthesis over the past decade. Abstract:In the last few years, the research area of chiral β-aminophosphines capable of promoting a wide range of diverse organic transformations has attracted more attention. Their derivatives constitute attractive reagents towards this end, due to their stability, low toxicity and ease of handling. These novel β-aminophosphine derivatives are potentially useful as organocatalysts and ligands in metal-catalysed reactions. In particular, chiral β-aminophosphines have recently emerged as powerful phosphine catalysts in asymmetric synthesis and catalysis. The asymmetric transformations to which metal complexes of these ligands have been applied include palladium-catalysed allylic substitutions and copper-catalysed 1,4-additions to enones among others. This review summarizes the most significant developments in the area of synthesis and application of chiral β-aminophosphine derivatives, providing a detailed overview of the current state of the art and including future aspects.

Abstract:The axially chiral allene framework has garnered significant attention from synthetic community, due to its wide applications in natural products, organic synthesis, pharmaceuticals, and materials science. Among the structurally diverse chiral allene compounds, heteroatom-substituted axially chiral allenes represent a large and versatile subclass. While the utility involving heteroatom-substituted chiral allenes has been extensively explored, the asymmetric synthesis of these valuable chiral frameworks has posed a long-standing challenging. Benefited from the development of asymmetric catalysis, many efficient synthetic methods in enantioselective catalysis as well as a variety of competent reaction partners recently have emerged. This review primarily summarizes the contributions made to date regarding the heteroatom-substitued axially chiral allenes in their catalytic asymmetric syntheses and their synthetic applications.

Abstract:Chiral pincer complexes, characterized by their rigid tridentate coordination framework, have emerged as powerful catalysts in asymmetric synthesis. This review provides a comprehensive overview of recent advancements in the development of chiral pincer-type ligands and their corresponding transition metal complexes. We highlight the latest progress in their application across a range of catalytic asymmetric reactions, including the (transfer) hydrogenation of polar and non-polar bonds, hydrophosphination, alkynylation, Friedel-Crafts reactions, enantioselective reductive cyclization of alkynyl-tethered cyclohexadienones, enantioselective hydrosilylation, as well as Aza–Morita–Baylis–Hillman reactions. The structural rigidity and tunability of chiral pincer complexes enable precise control over stereoselectivity, resulting in high enantioselectivity and efficiency in complex molecular transformations. As the field advances, innovations in ligand design and the exploration of new metal centers are expected to expand the scope and utility of these catalysts, bearing significant implications for the synthesis of enantioenriched compounds in pharmaceuticals, materials science, and beyond.

Abstract:Notwithstanding the common use of boronic acids in catalysis, the design and synthesis of efficient chiral boronic acids remains a formidable challenge. Herein we disclose a rational design and a concise synthesis of a series of novel C2-symmetric disubstituted boronic acids, or borinic acids. Chiral borinic acids have been found to be excellent agents that can achieve highly enantioselective desymmetrization of 2,2-disubstituted-1,3-propanediols, providing chiral diols with a quaternary stereocenter, which has been regarded in recent years as challenging. A wide range of substrates are tolerated in this reaction, providing a series of chiral diols with high enantioselectivity. The presence of the tetracoordinate borinate in the catalytic cycle has been proven by 11B NMR studies, and a stereocontrol model has been proposed.

Abstract:Chiral alcohols are important intermediates for various fine chemicals and pharmaceuticals. Integrating chemical catalysis and efficient enzyme catalysis in sequential systems for the synthesis of chiral alcohols is considered an ecofriendly and promising approach. Herein, employing a highly selective carbonic anhydrase II and different chemical catalysts, we constructed three general chemoenzymatic sequential systems for chiral alcohol compound synthesis from alkanes, alkenes, and alkynes, respectively. Compared to classical approaches, the combination of chemical catalysis and promiscuous carbonic anhydrase catalysis is simple and efficient since it requires only mild reaction conditions and avoids expensive chiral ligands and cumbersome operation steps. In this integrated approach, a wide variety of readily available aryl alkanes, alkenes, and alkynes are transformed into valuable chiral alcohols with excellent enantioselectivity of up to 99% (nearly all above 90%). This unified strategy of combining enzymatic and chemical catalyses advances the general chemoenzymatic process for powerful and important chemical transformations.

Abstract:The study presents a new synthetic approach to chiral α‐tertiary alkyl aminonitriles via catalytic asymmetric isomerization of cyanoketimines. It features a complementary method to the catalytic asymmetric Strecker reaction and umpolung addition of cyanoketimine. The utility of this method was illustrated by the development of a concise enantioselective synthesis of vildagliptin. Comprehensive Summary Chiral α‐tertiary aminonitriles are valuable synthetic intermediates. They are also found in various structures of biologically active molecules. Therefore, numerous reports of catalytic asymmetric synthesis of chiral α‐aminonitriles continuously emerged during the past few decades. Great strides have been made for the synthesis of chiral α‐aryl and α‐branched alkyl aminonitriles. However, efficient methods for catalytic asymmetric synthesis of chiral α‐linear alkyl aminonitriles remain limited. We herein report a new synthetic approach to chiral α‐tertiary alkyl aminonitriles via catalytic asymmetric isomerization of cyanoketimines. The synthetic value of this method was illustrated by application to a concise catalytic asymmetric synthesis of vildagliptin.
