Effects of Pretreatment with Taxol And/Or Cytochalasin B on the Vitrification of Buffalo Oocytes

Abstract:Oocytes vitrification is vital for the establishment of "oocytes bank", which can be potentially used for in vitro fertilization and production of nuclear transfer embryos. However, no methods used for oocytes vitrification is efficient, especially with respect to developmental potential. One of the important reasons is that oocyte cytoskeleton is damaged during vitrification, subsequently correct spindle/chromosome configuration may not happen after thawing. We hypothesized that the cytoskeleton stabilizing agent Taxol and/or cytochalasin B (CB) may maintain the stability of metaphase II buffalo oocyte microtubules and protect spindle / chromosome morphology integrity, which can improve oocyte developmental competence after in vitro fertilization (IVF). Cumulus-oocytes complexes (COCs) were recovered by aspiration from buffalo follicles (diameter, 2-6mm) using a 10mL disposable syringe with an 18-gauge needle. Buffalos oocytes were cultured for 22 hours for in vitro maturation. After removing cumulus granulosa cells, oocytes with the first polar body were divided into four groups randomly. The control group was not pretreated, where as other 3 groups were pretreated with 0.5 µM taxol, 1.0 µM taxol, and 1.0 µM taxol+10mM CB for 20 minutes. Vitrification solution composed of 20% dimethyl sulfoxide plus 20% ethylene glycol supplemented with 0.5 M trehalose. Three-step addition was performed, and oocytes were vitrified using glass microscopic grids(GMP). After thawing, the oocytes were cultured in in vitro maturation solution for additional 2 hours. Then the ratios of survival, cleavage, blastocysts and spindle/chromosome morphology integrity of the thawed oocytes were measured by immunofluorescence and IVF experiments. The results showed that there was no significantly difference among all groups in oocyte survival rate. Cleavage rate in 1.0 µM taxol group(42.18%) and 1.0 µM taxol+CB group(44.30%) were significantly higher than that in the control(21.91%) and 0.50 µM taxol group(28.05%), but there was no major variation between the first 2 groups and the last 2 groups(P>0.05). 1.0 µM taxol+CB group had the highest blastocyte rate among the control, 0.50 µM taxol and 1.00 µM taxol group (13.92% vs 4.11%, 4.88%, 9.38%, P<0.05). The ratio of spindle/chromosome morphology integrity in 1.0 µM taxol group and 1.0 µM taxol+CB group were higher than that in the control (50.00% vs. 41.17%, 33.78%; P<0.05). But no significant difference was seen between the two treated groups (P>0.05). From the above results, we knew that the 0.50 µM taxol group yielded negligible results, so we did not check this group's ratio of spindle/chromlsome morphology integrity. In conclusion, 1.0 µM taxol plus 10mM cytochalasin B pretreated buffalo oocytes can improve meiotic spindle/chromosome morphology integrity of vitrified-thawed oocytes, and their developmental ability after IVF.
