Abstract:Six novel chiral 1,3,4-oxadiazole thioether derivatives in yields of 55%～67% were synthesized by Michael addition-elimination reactions of 5-substituted-2-mercapto-1,3,4-oxadiazoles(as a nucleophile) and chiral 5-(l-menthyloxy)-3,4-dibromo-2(5H)-furanone.The structures were characterized by 1H NMR,13C NMR,IR and HR-MS.

Abstract:The chiral 1R,5S-6-alkyl-6-aza-2R-menthoxy-3-oxa-4-oxobicyclo[3,1,0]hexane (5a similar to 5d) containing two stereogenic centers were obtained in 41%similar to 51% yields with ex., >= 98% via the tandem asymmetric Michael addition and internal nucleophilic substitution reaction of the chiron 3 with the primary amine 4 as a nucleophile. After the effective reduction of compounds 5 by LiAlH4 in THF, the target molecules, meso-N-alkyl-2,3-bis(hydroxymethyl)aziridines (6a similar to 6d) were obtained in 66%similar to 91% yields. The chemical structures of 5 and 6 were readily confirmed by analytical and spectroscopic data. The proposed structures of optically active compounds were consistent with the stereochemistry and configuration of their molecules further confirmed by the X-ray crystallography of 5a and 6c. These results could provide a new synthetic route to the functionalized optically active aziridine derivatives.

Abstract:Four novel 3-(5-(substitudelythio)-1,3,4-oxadiazol-2-yl)-2H-chromen-2-one were designed and synthesized.Under the optimized Conditions:solvent was DMF,acid binding agent was K2CO3,reaction temperature was 65℃,reaction time was 4 h,the molar ratio of 3-(5-mercapto-1,3,4-oxadiazol-2-yl)-2H-chromen-2-ones to halogenated hydrocarbons was 1.0: 1.2.Their structures were characterized by 1H NMR,13C NMR,IR and elemental analyses.

Abstract:1,2‐Oxazetidines are valuable small strained molecules that could be used to create new and conventionally difficult‐to‐access chemical transformations. Currently, asymmetric methods towards this class of heterocycles are very rare. Herein, we report a general and practical method to access a series of structurally diverse chiral 1,2‐oxazetidines from readily available chiral epoxides and α‐bromo esters in 3–4 steps by using mild Mitsunobu reactions as an efficient ring‐closure approach to form the highly strained four‐membered rings. The new method is operationally simple, and a range of N‐nosyl‐protected 3‐ and 4‐substituted as well as 3,4‐disubstituted chiral 1,2‐oxazetidines could be conveniently prepared in gram‐scale with excellent enantioselectivities (93–99% ee) and good overall yields for the first time.

Abstract:Chiral 1-amino-2-propanols 4-5 were prepared by racemic 1-amino-2-propanol and chiral tartaric acid. Then, chiral N-dichloroacetyl-2-substituted-5-methyl-1,3-oxazolidines 6-11 were synthesized by the reaction of chiral 1-amino-2-propanol with ketone and dichloroacetyl chloride. The structures of the compounds were characterized by IR, H-1 NMR, C-13 NMR, MS and elemental analysis. The configuration of 7 was determined by X-ray crystallography.

Abstract:Chiral 2‐amino‐butanols (4 and 5) were obtained via the isolation of diastereomeric salt. Then, chiral compounds (6–9) were synthesized by a sequential procedure involving condensation of chiral 2‐amino‐butanol with ketone and dichloroacetyl chloride. All the compounds were characterized by IR, 1H NMR, 13C NMR, and element analysis. The absolute configurations of (S)‐8 was determined by X‐ray crystallography.

Abstract:We have discovered a novel three-component reaction yielding 6 chiral oxime ether 5-(S)-menthyloxy-4-oxime ether-3-bromo-2(5H)-furanone(12a～12f) in a one-pot procedure with chiral-5-menthyloxy-3,4-dibrome-2(5H)-furanone, aldehyde, hydroxylamine hydrochloride, and the yield is 50%～80%. Due to the special structure and the presence of nitrogen atom, those compounds can be used for many reaction and provide a kind of feasible approach for the synthesis and study of chiral chelate compound and chiral oxime ether. The target compounds were characterized by IR, 1H NMR, 13C NMR and HRMS.

Abstract:The tandem asymmetric Michael addition/intramolecular nucleophilic substitution of the novel chiral source, 3-bromo-5-(1-menthyloxy)-2 (5H)-furanone(1) with some carbon nucleophiles has been investigated. The asymmetric reaction can yield optically pure 3-bicycle [3.3.0]octene derivatives containing two novel stereogenic chiral centers. The chemical structure and configuration of these 3-bicyclo[3.3.0]octene compounds were identified on the basis of their elemental analytical data and spectroscopic data, such as [alpha](D)(20), UV, IR, H-1 NMR, C-13 NMR, MS and X-ray crystallography. This asymmetric reaction can provide an important strategy in synthesis of some optically pure 3-bicyclo[3.3.0]octene compounds and some biologically active molecules.

Abstract:Reaction of 1,5-benzothiazepines, containing 2-phenyl-1,2,3-triazole 2a–d, with aryl nitrile oxides in CH2 Cl2 at room temperature leads to a series of novel 1,2,4-oxadiazolo[4,5-d]-1,5-benzothiazepine derivatives 3a-l in good yields. The products were characterized by IR, 1H NMR, MS, elemental analyses, X-ray and their spectrum characters were discussed.

Abstract:N-Substituted-5-aryl-1,3,4-oxadiazole-2-amine was synthesized by a three-step reaction in overall yields of 42.0%~68.7%.The products were characterized by IR,~1H NMR,MS and elemental analysis.Furthermore,the single crystal X-ray study of 5-phenyl-1,3,4-oxadiazole-2-amine(4a) indicated that this compound belonged to 1,3,4-oxadiazole derivatives,not to 1,4-dihydro-1,2,4,5-tetrazine derivatives.

Abstract:The synthetic method of the novel chiral synthon, 5- l -menthyloxy-3-chloro-2-(5H)-furanone 5a and its application in asymmetric reactions were investigated. 5a is easily obtained in highly optical purity, and acts as a stable acceptor of Michael addition with oxygen nucleophiles in tandem double Michael addition / internal nucleophilic substitution to offer the spiro-cyclopropane derivative containing four stereogenic centers 8, which it is difficult to obtain by routine methods. The synthetic methods for 5a and 8 are reported in detail and the new compounds are identified on the basis of their analytical data and spectroscopic data, such as UV, IR, 1 H NMR, 13 C NMR, MS and elementary analysis. The absolute configuration of the interesting spiro-cyclopropanes, spiro [1-chloro-4-( l -menthyloxy)-5-oxo-6-oxa-biscyclo[3.1.0]hexane-2,3′-(4′-/-menthyloxy-5′- l -menthyloxybutyrolactone)] 8 was established by X-ray crystallography. This result can provide important synthetic strategy in synthesis of some complex molecules containing spiro-cyclopropane skeleton with multiple chiral centers.

Abstract:A facile method for preparation of 5-methyl-5-[5-(tert-butyldimethylsiloxymethyl)-3-isoxazolyl]-3-aryl-4-(4-methyloxyphenyl)-4,5-dihydro-1,2,4-oxadiazoles (4a similar to 4e) via 1,3-dipolar cycloaddition of Schiff bases derived from 3-acetyl-5-hydroxymethylisoxazoles and nitrile oxides in situ generated from oximes was described. 5-(tert-Butyldimethylsiloxymethyl)-3-(1,5-diaryl-4,5-dihydropyrazol-3-yl)isoxazoles (6a similar to 6i) were also prepared through cycloaddition of aryl hydrazine and alpha,beta-unsaturated ketones (5) derived from 3-acetyl-5-hydroxylmethylisoxazoles. The structures of all new compounds were characterized by H-1 NMR, C-13 NMR, MS, IR and HRMS spectra.

Abstract:The spiro[ 1-bromo-4-l-mentholoxy-5-oxo-6-oxabicyclo[3.1.0] hexane-2,3 '-(4 ' -ethoxy-5 ' -1-menthyloxybutyrolactone)] 4 was synthesized via the tandem double Michael addition/internal nucleophilic substitution of chiral synthon 1, 5-1-menthyloxy- 3-bromo-2 (SH)-furanone, with nucleophilic ethanol. The two synthetic routes to compound 4 and its analytical data of structure are reported. The absolute configuration of 4 was determined by X-ray crystallography. These results provide a valuable strategy for synthesizing some biologically active molecules.

Abstract:An efficient bifunctional cinchona alkaloid derived thiourea-promoted enantioselective conjugate addition of nitroalkanes to indolylidenecyanoacetates has been developed under neat conditions. The process leads to synthetically interesting densely functionalized 3,3'-disubstituted oxindoles with creation of up to three stereogenic centers.

Abstract:The direct asymmetric vinylogous Michael addition of 3,5-dialkyl-substituted 4-nitroisoxazoles with alpha,beta-unsaturated aldehydes catalyzed by a diphenylprolinol TBS ether organocatalyst is described, giving products with moderate to good yields and up to excellent enantioselectivities (96% ee). This approach provides an easy access to highly functionalized chiral isoxazole derivatives.

Abstract:1,3,4-oxadiazolines are found to possess significant biological activities such as antibacterial,antifungal effects.According to these,we designed and synthesized some new 1,3,4-oxadiazoline derivatives.First,Ethyl 4-methylbenzoate was prepared by reaction of 4-methyl benzoic acid with anhydrous ethanol in the presence of sulfuric acid.This propound reacted with hydrazine hydrate in the presence of anhydrous ethanol as solvent to give 4-methylhydrazin(1).In the presence of anhydrous ethanol,(1) reacted with aromatic aldehydes to produce corresponding hydrazones(2a~2j).Then 2a~2j cyclodehydrated with acetic anhydride to afford the title compounds(3a~3j).The mild reactions and good yields are the merits of this method.The title structures of 3a~3j were confirmed by elementary analyses,IR,~(1)H NMR,and MS spectroscopy.
