Abstract:A new flavonoid, 8-(3-hydroxymethyl-2-butenyl)-5,7,2',4'-tetrahydroxyflavanone (1), along with seven known compounds, (2R)-3α,7,4'-Trihydroxy -5-methoxy-8-prenylflavanone (2), (2R)-3β,7,4'- Trihydroxy-5-methoxy-8-prenylflavanone (3), 3,7-dihydroxycoumarin (4), Shandougenines B (5), Specionin (6), Kushenol N (7) and Kushenol I (8) were isolated from the roots of Sophora flavescens. Among them, Compound 5 was isolated from Leguminous plants, and compounds 4 and 6 were isolated from Sophora flavescens for the first time. Their structures were elucidated by the aid of NMR, HRMS, HMBC and CD spectroscopic methods.

Abstract:OBJECTIVE To study the chemical constituents of the flavonoids from Sophora tonkinensis. METHOD The compounds were isolated by chromatography on silica gel, Sephadex LH-20 column and identified by spectroscopic analysis. RESULT Eight compounds were isolated and their structures were identified as tonkinochromane I (1), glabrol (3), lupinifolin (2), tonkinensisol (4), 8-C-prenylkaempferol (5), 7,2'-dihydroxy-4'-methoxy-isoflavanol (6), formononetin (7), and genistein (8), respectively. CONCLUSION Compound 1 was a new compound. And compound 6 was firstly isolated from the genus Sophora. Compounds 2, 3 and 5 were isolated from S. tonkinensis for the first time.

Abstract:Two new isoprenylated flavanones, tonkinochromanes J (1) and K (2), and a new isoprenylated dihydrochalcone, tonkinochromane L (3), were isolated from the roots of Sophora tonkinensis along with four known compounds (4-7). Their structures were determined by means of spectroscopic analyses, including HRMS, IR, 1H and 13C NMR and 2D experiments (COSY, HSQC, and HMBC), and comparison with known related compounds.

Abstract:Phytochemical study on the roots of Sophora tonkinensis Gagnep. led to the isolation of fifteen compounds, including eleven flavonoids (1–11) and four phenolic acids (12–15). The structures of these compounds were elucidated based on NMR and HR-ESIMS analysis, further supported by comparison with previous literatures. Among them, compounds 2, 3, 4, 6, 9, 12 and 13 were firstly discovered from S. tonkinensis, compound 8 was firstly discovered from the genus Sophora. Furthermore, the chemotaxonomic significance of the obtained compounds was discussed in detail.

Abstract:Six undescribed lavandulylated flavonoids (1–6) were isolated from the roots of Sophora flavescens . Remarkably, compounds 1 and 2, which were composed of a flavane unit and a phloroglucinol unit, were the first reported dimers. Compounds 3 and 4 were the first reported neoflavonoids with lavandulyl units. Compounds 5 and 6 were chalcone with oxidized lavandulyl units. Their structures were fully characterized by cumulative analyses of UV, IR, HRESIMS, NMR and ECD spectroscopic data, along with computational calculations through density functional theory. Compounds 1 and 2 showed significant protein tyrosine phosphatase-1B inhibitory activities with IC 50 values of 2.669 and 3.596 μM, respectively.

Abstract:A new flavonol, tonkinensisol, was isolated from the roots of Sophora tonkinensis, together with three known compounds named as bayin, vitexin and lupeol. Their structures were elucidated on the basis of spectroscopic evidence. Additionally, tonkinensisol showed moderate cytotoxicity suppressing the proliferation of HL-60 cells in vitro.

Abstract:Two new flavanones, tonkinochromanes A (1) and B (2), and a new chalcone. tonkinochromane C (3), were isolated from the roots and rhizomes of Sophora tonkinensis. Their structures were elucidated by spectroscopic methods including 2D-NMR and MS experiments. Of the three isoprenyl substituents in these compounds on rings A and B, one to two were found to be ring-fused in the form of either dihydro- or regular 2,2-dimethyl-2H-pyran moieties.

Abstract:Three new flavonoid glycosides-soyaflavonosides A (1), B (2), and C (3)-together with 23 known ones were obtained from the 70% EtOH extract of Flos Sophorae (Sophora japonica, Leguminosae). Their structures were elucidated by chemical and spectroscopic methods. Among the known isolates, 14, 18, 20, 22, and 26 were isolated from the Sophora genus for the first time; 12, 19, 24, and 25 were obtained from the species firstly. Moreover, NMR data for compounds 18 and 26 are reported for the first time here. Meanwhile, compounds 4, 8-13, 15, 16, 19, 21, and 22 presented obvious inhibitory effects on TG accumulation in HepG2 cells. Analysis of the structure-activity relationship indicated that all of the quercetin glycosides examined in this study possess significant activity that is not significantly influenced by the amount of glycosyl present, whereas increasing the amount of glycosyl reduced the activities of isorhamnetin glycosides and orobol. In addition, a high dose (30 μmol/l) of kaempferol was found to inhibit HepG2 cell growth, while a low dose (10 μmol/l) was observed to decrease TG accumulation.

Abstract:Sophora flavescens Aiton is a plant in the Leguminosae family. As a traditional Chinese medicine, it is used to treat eczema, bloody stool, skin pruritus, and so on. By studying non-alkaloid components in the roots of S. flavescens, we obtained a total of 49 compounds (1-49), including three undescribed flavonoids (13, 15 and 18), five undescribed isopentenyl flavonoids (32, 34, 38, 39 and 48), two known coumarins (1-2), three phenolic acids (3-5), one known isopentenyl flavonoids (19-31, 33, 35-37, 40-47 and 49). On the basis of chemical evidences and spectral data analysis (UV, ECD, Optical rotation data, 1D/2D-NMR and HR-ESI-MS), the structures of undescribed compounds were elucidated. The inhibitory effect of compounds 1-49 on LPS induced NO production in RAW 264.7 cells was detected. Compounds 11, 19, 21-24, and 28-30 showed significant inhibitory effects, and the IC50 values of compounds 11 and 22 even reached 4.58 ± 0.66 and 4.53 ± 0.66 μM. This study suggests that flavonoids may be the main component that exerts anti-inflammatory effects in the non-alkaloid extraction layer of the extract from the roots of S. flavescens.

Methods Constituents were isolated by means of extraction with different solvents and column chromatography,and identified by spectral analysis. Results From the acetone soluble fraction,10 compounds were isolated and identified as sophoradiol(Ⅰ),maltol(Ⅱ),α-acetylpyrrole(Ⅲ),hexacosanic acid(Ⅳ),ceryl alcohol(Ⅴ),β-sitosterol(Ⅵ),glycerol-α-monohexacosanate(Ⅶ),octacosanol(Ⅷ),genistein(Ⅸ) and kaempferol(Ⅹ).(Conclusion)Compound Ⅱ,Ⅲ,Ⅳ,Ⅴ,Ⅶ,Ⅷ and compound Ⅰ were isolated from the plant and its fruit respectively for the first time.

Abstract:Five novel pterocarpan derivatives (1–5) and three known pterocarpans were isolated from the roots of Sophora flavescens Ait. Their structures were elucidated based on extensive spectroscopic data (UV, IR, 1D and 2D NMR, and HR-ESI-MS). Notably, compounds 1–5 were the first reported natural products, which possessed unique pterocarpan-glucose-long chain aliphatic acid structures. The kinds of aliphatic acid were determined by comparison with authentic aliphatic acids using HPLC-ELSD analysis after acid hydrolysis. The absolute configurations of 1–5 were confirmed by analysis of their ECD spectra. All compounds were evaluated against lipopolysaccharide (LPS)-induced TNF-α production in RAW264.7 cells, and compound 1 showed significant inhibitory activity with inhibition rate of 86.22%, IC50 = 0.79 μmol/L (dexamethasone as positive control, inhibition rate of 71.38%).This article is protected by copyright. Abstract:Four water-soluble polysaccharides, designated as SF1, SF2, SF3 and SF4, were efficiently extracted from the roots of Sophora flavescens by mechanochemistry under the conditions of rotational speed of 400rpm, grinding time of 10min, powder to ball weight ratio of 1:20, and Na2CO3 loading of 7wt%. The results obtained indicated that all of these four acid heteropolysaccharides are composed of rhamnose, arabinose, xylose, mannose, glucose and galactose, with the average molecular weights of 400.9, 98.6, 99.3, 42.7kDa, respectively. In vitro, SF4 showed the most significant scavenging activity on superoxide radical, ABTS, and DPPH radical, while SF3 had the most significant scavenging activity on hydroxyl radical. Immunological tests demonstrated that SF1, SF2, SF3 and SF4 significantly stimulated nitric oxide production without cytotoxicity in macrophages and promoted splenocyte proliferation. These data suggest that the four polysaccharides fractions have the potential as novel natural sources of antioxidative and immunopotentiating agents.

Abstract:A new isoflavonolignan glycoside, butesuperin B-7' '-O-β-glucopyranoside ( 1) along with a known isoflavonolignan butesuperin A ( 2), were isolated from the roots of Sophora tonkinensis. The structure of the new compound was elucidated using spectroscopic met hods, mainly 1D and 2D NMR. Compound 1 showed week cytotoxicity against BEL-7404 and NCI-H460 cel ls in MTT assay.

Abstract:Sophora flavescens AITON (kurara) has long been used to treat various diseases. Although several research findings revealed the biosynthetic pathways of its characteristic chemical components as represented by matrine, insufficient analysis of transcriptome data hampered in-depth analysis of the underlying putative genes responsible for the biosynthesis of pharmaceutical chemical components. In this study, more than 200 million fastq format reads were generated by Illumina's next-generation sequencing approach using nine types of tissue from S. flavescens, followed by CLC de novo assembly, ultimately yielding 83,325 contigs in total. By mapping the reads back to the contigs, reads per kilobase of the transcript per million mapped reads values were calculated to demonstrate gene expression levels, and overrepresented gene ontology terms were evaluated using Fisher's exact test. In search of the putative genes relevant to essential metabolic pathways, all 1350 unique enzyme commission numbers were used to map pathways against the Kyoto Encyclopedia of Genes and Genomes. By analyzing expression patterns, we proposed some candidate genes involved in the biosynthesis of isoflavonoids and quinolizidine alkaloids. Adopting RNA-Seq analysis, we obtained substantially credible contigs for downstream work. The preferential expression of the gene for putative lysine/ornithine decarboxylase committed in the initial step of matrine biosynthesis in leaves and stems was confirmed in semi-quantitative polymerase chain reaction (PCR) analysis. The findings in this report may serve as a stepping-stone for further research into this promising medicinal plant.

Abstract:Sophora flavescens Ait (Ku-Shen in Chinese) is a popular traditional Chinese herbal medicine in China for a long history. It shows significant pharmacological activities in the treatment of dysentery, eczema, fever, jaundice, vulvar swelling, gastrointestinal hemorrhage and inflammatory disorders. Alkaloids and flavonoids have been identified as virtual components, especially isoprenoid flavanonols are a class of characteristic compounds for S. flavescens. However, few studies have focused on isoprenoid flavonoids analyses and no comprehensive review has yet been published. In the current review, we systematically summarized the isoprenoid flavonoids, a total of 55 compounds have been isolated from S. flavescens, particularly an isoprenyl and a lavandulyl group in backbone structures. Further pharmacological activities, qualitative and quantitative chemical analyses research will contribute to the development of natural isoprenoid flavonoid products in S. flavescens.
Extraction and FT-IR Spectral Identification of Flavonoids Extracted from the Buds of Sophora japonica Linn.

Abstract:Flavonoids from the buds of Sophora japonica Linn.were extracted by three different methods,namely alkali extraction followed by acid sedimentation,ethanol refluxing and ultrasound-assisted extraction.The extraction conditions,including pH value of solvent and the kind of stabilizer were researched as well.Furthermore,with rutin as comparison,the flavonoids content of different extracts from buds of Sophora japonica L.and their FT-IR spectra had been determined.The results showed that both the yield and purity of the products by alkali extracting followed by acid sedimentation with the extract adjusted to pH value 9 containing borax as stabilizer were higher than others.Meanwhile,the typical FT-IR spectra of every sample had specific absorption peak pattern,intensity and location,related to different extracting technology.Among them,the FT-IR spectrum of flavonoids obtained under the above optimum extraction technology was similar to that of standard rutin.Therefore,quantitative analysis along with their FT-IR spectra provide a favorable foundation for quality control of flavonoids from the buds of Sophora japonica Linn..
