The Neuroprotective Effect of BDNF on Retinal Ganglion Cells Damaged by Experimental Acute Hyper-intraocular Pressure

Abstract:Objective To study the neuroprotective effect of Brain-Derived Neurotrophic Factor(BDNF) on retinal ganglion cells(RGCs) damaged by experimental acute hyper-intraocular pressure(HIOP).Methods Sixteen rabbits were divided randomly into BDNF or PBS groups.Experimental acute HIOP model was performed in the right eyes by perfusing balanced salt solution into anterior chamber.The all left eyes served as the control group.3.75μg of BDNF or 5μl of 0.1M phosphate buffered solution(PBS)was injected respectively into vitreous body cavity of right eyes of BDNF or PBS group before perfusion.All rabbits were euthanatized on 14th day.All rabbits were labelled retrogradely with 30% horseradish peroxidase in 2% Dimethyl Sulfoxide 48 hours before sacrifice and then counted the density of RGCs of inferior half retina located 2,4,6mm from the optic disc was counted.Results At the distance of 2,4,6mm from optic disc,the density of RGCs of normal control group was(1 619±377),(771±232),(358±122) per mm~2 respectively,and that in BDNF or PBS group was(1 304±246),(669±283),(283±93) per mm~2 respectively or(1 002±410),(627 ±211),(264±107) per mm2respectively.At the distance of 2 mm,the density of RGCs of BDNFgroup was significantly higher than that in the PBS group.Conclusions BDNF could protect RGCsfromdamage induced by experi mental acute hyper-intraocular pressure and enhance their survival.
