Abstract:Znic (Zn) alloys with good cytocompatibility and suitable degradation rate have been a kind of biodegradable metal with great potential for clinical applications. This paper summarizes the biological role of degradable Zn alloy as bone implant materials, discusses the mechanical properties of different Zn alloys and their advantages and disadvantages as bone implant materials, and analyzes the influence of different processing strategies (such as alloying and additive manufacturing) on the mechanical properties of Zn alloys. This paper provides systematic design approaches for biodegradable Zn alloys as bone implant materials in terms of the material selection, product processing, structural topology optimization, and assesses their application prospects with a view to better serve the clinic.

Abstract:Zinc (Zn)-based biodegradable materials show moderate degradation rates in comparison with other biodegradable materials (Fe and Mg). Biocompatibility and non-toxicity also make them a viable option for implant applications. Furthermore, Pure Zn has poor mechanical behavior, with a tensile strength of around 100-150 MPa and an elongation of 0.3-2%, which is far from reaching the strength required as an orthopedic implant material (tensile strength is more than 300 MPa, elongation more than 15%). Alloy and composite fabrication have proven to be excellent ways to improve the mechanical performance of Zn. Therefore, their alloys and composites have emerged as an innovative category of biodegradable materials. This paper summarizes the most important recent research results on the mechanical and biological characteristics of biodegradable Zn-based implants for orthopedic applications and the most commonly added components in Zn alloys and composites.

Abstract:Zinc (Zn) is a new generation of biodegradable metal as temporary biomedical implants with a promising degradation rate. However, its clinical applications have been limited because of the insufficient mechanical properties. Considering the degradation property and biocompatibility, we proposed Zn–Sr alloys after extrusion treatments to simultaneously improve the mechanical strength and ductility. The in vitro and in vivo degradation and biocompatibility were also evaluated using electrochemical and immersion corrosion tests, various cell and bacterial models, together with subcutaneous and femoral implantations in rats. Results showed that the extruded Zn-0.7Sr alloys exhibited two times higher mechanical strengths (∼120 MPa) and better ductility (∼10%) than the pure Zn counterparts. The Zn–Sr alloys provided enhanced in vitro and in vivo biocompatibility along with promising antibacterial properties.

Abstract:In the last decade, iron and magnesium, both pure and alloyed, have been extensively studied as potential biodegradable metals for medical applications. However, broad experience with these material systems has uncovered critical limitations in terms of their suitability for clinical applications. Recently, zinc and zinc-based alloys have been proposed as new additions to the list of degradable metals and as promising alternatives to magnesium and iron. The main byproduct of zinc metal corrosion, Zn2+, is highly regulated within physiological systems and plays a critical role in numerous fundamental cellular processes. Zn2+ released from an implant may suppress harmful smooth muscle cells and restenosis in arteries, while stimulating beneficial osteogenesis in bone. An important limitation of pure zinc as a potential biodegradable structural support, however, lies in its low strength (σUTS ~ 30 MPa) and plasticity (ε < 0.25%) that are insufficient for most medical device applications. Developing high strength and ductility zinc with sufficient hardness, while retaining its biocompatibility, is one of the main goals of metallurgical engineering. This paper will review and compare the biocompatibility, corrosion behavior and mechanical properties of pure zinc, as well as currently researched zinc alloys.

Abstract:Zn and its alloys have recently been used as a new class of biodegradable biomedical metals besides magnesium and iron alloys, owing to their moderate corrosion rate and good mechanical properties. In recent years, researchers have rigorously studied the design, processing, and degradation mechanism of Zn alloys, but their biocompatibility has not been well explored. Past research on the biocompatibility of Zn alloys focused on in vitro cytotoxicity, hemolysis, and coagulation, and only a few materials were implanted into animals for characterizing the histocompatibility. Biocompatibility involves complex local and systemic reactions, such as cells, tissues, blood, and immunity. In addition to the physical and chemical properties of the material, the biocompatibility is also affected by interactions between the material and body. In this paper, the chemical and phase compositions of degradable zinc alloys were analyzed, and the biological evaluation methods were clarified. In view of the recent studies on zinc alloy biocompatibility, future research directions were proposed.

Abstract:Traditional inert materials used in internal fixation have caused many complications and generally require removal with secondary surgeries. Biodegradable materials, such as magnesium (Mg)-, iron (Fe)- and zinc (Zn)-based alloys, open up a new pathway to address those issues. During the last decades, Mg-based alloys have attracted much attention by researchers. However, the issues with an over-fast degradation rate and release of hydrogen still need to be overcome. Zn alloys have comparable mechanical properties with traditional metal materials, e.g., titanium (Ti), and have a moderate degradation rate, potentially serving as a good candidate for internal fixation materials, especially at load-bearing sites of the skeleton. Emerging Zn-based alloys and composites have been developed in recent years and in vitro and in vivo studies have been performed to explore their biodegradability, mechanical property, and biocompatibility in order to move towards the ultimate goal of clinical application in fracture fixation. This article seeks to offer a review of related research progress on Zn-based biodegradable materials, which may provide a useful reference for future studies on Zn-based biodegradable materials targeting applications in orthopedic internal fixation.

Abstract:Zn-based biodegradable alloys or composites have the potential to be developed to next-generation orthopedic implants as alternatives to conventional implants to avoid revision surgeries and to reduce biocompatibility issues. This review summarizes the current research status on Zn-based biodegradable materials. The biological function of Zn, design criteria for orthopedic implants, and corrosion behavior of biodegradable materials are briefly discussed. The performance of many novel zinc-based biodegradable materials is evaluated in terms of biodegradation, biocompatibility, and mechanical properties. Zn-based materials perform a significant role in bone metabolism and the growth of new cells and show medium degradation without the release of excessive hydrogen. The addition of alloying elements such as Mg, Zr, Mn, Ca, and Li into pure Zn enhances the mechanical properties of Zn alloys. Grain refinement by the application of post-processing techniques is effective for the development of many suitable Zn-based biodegradable materials.

Abstract:Biodegradable metals and their alloys have gained significant attention as promising candidates for biomedical applications. Notably, Magnesium, Zinc, and Iron stand out as the most prominent biodegradable materials, prompting extensive research into novel alloy development. These metals exhibit inherent biodegradability, acceptable biocompatibility, and commendable mechanical integrity, rendering them suitable for implant use. In this comprehensive review, we delve into recent advancements in biodegradable implants within the biomedical domain, with a primary focus on orthopedic and cardiovascular applications. The paper extensively explores the in vivo and in vitro degradation behavior, mechanical properties, and corrosion mechanism of these materials. Additionally, we discuss progress in surface modification and fabrication techniques, vital in optimizing implant performance. However, certain limitations associated with alloying materials, especially Zinc and Iron, have hindered their widespread implementation. We underscore the importance of overcoming these challenges to unlock the full potential of these materials. Furthermore, we present the promise of new metals, such as Molybdenum, which exhibit remarkable properties, opening doors to implants with superior mechanical characteristics. Overall, this review emphasizes the growing significance of biodegradable metallic implants, showcasing their potential to address diverse clinical needs. Abstract:Biodegradable zinc (Zn) alloys are promising implant materials to restore bone defects in orthopedics. Adding silver (Ag) to Zn not only enhances its mechanical properties but also improves its antibacterial properties. However, the regular pattern of Ag composition on the properties of Zn-xAg alloys remains unclear. In this paper, Zn-Ag alloys with different Ag contents ranging from 0.5 to 6.0% were prepared by casting process and were systematically investigated both in vitro and in vivo. Mechanical tests suggest that the increase of Ag content enhanced the yield strength (YS), ultimate tensile strength (UTS), Vickers hardness, and elongation. Compared with pure Zn, the maximum YS, UTS, Vickers hardness, and elongation were achieved at Zn-6Ag, which were increased by 267%, 295%, 115%, and 172%, respectively. The degradation rate increased and the antibacterial properties were enhanced accordingly. All 25% and most 50% extracts of Zn-Ag alloys significantly promoted the proliferation of MC3T3-E1 cells, except for 50% Zn-6Ag extracts showing cytotoxicity. In the rat femoral condyle model, Zn-2Ag and Zn-4Ag alloys exhibited enhanced osteogenic performance at the early stage of implantation compared with pure Zn. Zn-Ag alloys (especially Zn-2Ag and Zn-4Ag) are identified as promising bone repair materials.

Abstract:In recent years, zinc alloys have shown great potential as orthopedic plant materials because of their excellent biocompatibility, osteoinductive and biodegradation activities in vivo. The physiological function, degradation rate and mechanical properties of pure zinc were introduced in this review. Taking the gap between the above indicators and the clinical needs of orthopedics as the clue and the alloy elements as the classification basis, the existing achievements of researchers to improve the properties of zinc-based materials by changing the microstructure of materials and triggering the corresponding strengthening mechanism, and using the physiological synergistic function of added elements were expounded in this paper. The standard formulation, alloy design improvement and the introduction of new technologies such as additive manufacturing in the field of medical zinc based materials were discussed in order to meet the diversified needs of clinical diagnosis and treatment.

Abstract:Zn-based biomaterials have emerged as promising new types of bioresorbable metallics applicable to orthopedic devices, cardiovascular stents, and other medical applications recently. Compared to other degradable metallic biomaterials (i.e. Mg- or Fe-based), Zn biomaterials have a more appropriate corrosion rate without hydrogen gas evolution. Here, we evaluated the potential of Zn-based metallics as medical implants, both in vitro and in vivo, alongside a standard benchmark Mg alloy, AZ31. The mechanical properties of the pure Zn were not strong enough, but were significantly enhanced (microhardness >70 kg/mm2, strength >220 MPa, elongation >15%) after alloying with Sr or Mg (1.5 at. %), surpassing the minimal design criteria for load-bearing device applications. The corrosion rate of Zn-based biomaterials was about 0.4 mm/year, significantly slower than that of AZ31. The measured cell viability and proliferation of three different human primary cells fared better for Zn-based biomaterials than AZ31 using both direct and indirect culture methods. Platelet adhesion and activation on Zn-based materials were minimal, significantly less than on AZ31. Hemolysis ratio of red cells (<0.5%) after incubation with Zn-based materials was also well below the ISO standard of 5%. Moreover, Zn-based biomaterials promoted stem cell differentiation to induce the extracellular matrix (ECM) mineralization process. In addition, in vivo animal testing using subcutaneous, bone, and vascular implantations revealed that the acute toxicity and immune response of Zn-based biomaterials were minimal/moderate, comparable to that of AZ31. No extensive cell death and foreign body reactions were observed. Taken together, Zn-based biomaterials may have a great potential as promising candidates for medical implants.

Abstract:Zinc (Zn) is a promising bioresorbable implant material with more moderate degradation rate compared to magnesium (Mg) and iron (Fe). However, the low mechanical strength and localized degradation behavior of pure Zn limit its clinical applications. Alloying is one of the most effective ways to overcome these limitations. After screening the alloying element candidates regarding their potentials for improvement on the degradation and biocompatibility, we proposed Fe as the alloying element for Zn, and investigated the in vitro and in vivo performances of these alloys in both subcutaneous and femoral tissues. Results showed that the uniformly distributed secondary phase in Zn-Fe alloys significantly improved the mechanical property and facilitated uniform degradation, which thus enhanced their biocompatibility, especially the Zn-0.4Fe alloy. Moreover, these Zn-Fe alloys showed outstanding antibacterial property. Taken together, Zn-Fe alloys could be promising candidates as bioresorbable medical implants for various cardiovascular, wound closure, and orthopedic applications.
In Vitro Evaluation of the Feasibility of Commercial Zn Alloys As Biodegradable Metals

Abstract:In this work, three widely used commercial Zn alloys（ZA4-1, ZA4-3, ZA6-1） were purchased and prepared by hot extrusion at 200 °C. The microstructure, mechanical properties, corrosion behaviors,biocompatibility and hemocompatibility of Zn alloys were studied with pure Zn as control. Commercial Zn alloys demonstrated increased strength and superb elongation compared with pure Zn. Accelerated corrosion rates and uniform corrosion morphologies were observed in terms of commercial Zn alloys due to galvanic effects between Zn matrix and α-Al phases. 100% extracts of ZA4-1 and ZA6-1 alloys showed mild cytotoxicity while 50% extracts of all samples displayed good biocompatibility. Retardant cell cycle and inhibited stress fibers expression were observed induced by high concentration of Zn 2+ releasing during corrosion. The hemolysis ratios of Zn alloys were lower than 1% while the adhered platelets showed slightly activated morphologies. In general, commercial Zn alloys possess promising mechanical properties, appropriate corrosion rates, significantly improved biocompatibility and good hemocompatibility in comparison to pure Zn. It is feasible to develop biodegradable metals based on commercial Zn alloys.

Abstract:Biodegradable metals (BMs) gradually degrade <i>in vivo</i> by releasing corrosion products once exposed to the physiological environment in the body. Complete dissolution of biodegradable implants assists tissue healing, with no implant residues in the surrounding tissues. In recent years, three classes of BMs have been extensively investigated, including magnesium (Mg)-based, iron (Fe)-based, and zinc (Zn)-based BMs. Among these three BMs, Mg-based materials have undergone the most clinical trials. However, Mg-based BMs generally exhibit faster degradation rates, which may not match the healing periods for bone tissue, whereas Fe-based BMs exhibit slower and less complete <i>in vivo</i> degradation. Zn-based BMs are now considered a new class of BMs due to their intermediate degradation rates, which fall between those of Mg-based BMs and Fe-based BMs, thus requiring extensive research to validate their suitability for biomedical applications. In the present study, recent research and development on Zn-based BMs are reviewed in conjunction with discussion of their advantages and limitations in relation to existing BMs. The underlying roles of alloy composition, microstructure, and processing technique on the mechanical and corrosion properties of Zn-based BMs are also discussed.

Abstract:Zn-3Cu-xMg (x=0, 0.1, 0.5 and 1.0wt.%) alloys were developed as potential biodegradable metallic materials in this study. The mechanical properties, corrosion behavior and in vitro cytocompatibility of Zn-3Cu-xMg alloys were studied systematically to evaluate the feasibility as biodegradable implant materials. The secondary phase in as-cast and as-extruded Zn-3Cu alloy was CuZn5 phase. Mg2Zn11 phase newly formed and precipitated by Mg addition. The volume fraction of Mg2Zn11 phase increased gradually with increasing Mg concentration. As a result, yield strength was improved from 213.7 to 426.7MPa and increased by 99.7% while elongation decreased from 47.1% to 0.9%. Besides, biocompatibility was improved apparently and in vitro corrosion rates increased from 11.4 to 43.2μmyear−1, which is more suitable for clinic application. The present research indicated that the newly developed alloys could be promising candidates for biomedical use due to the proper mechanical properties, degradation rate and acceptable biocompatibility.

Abstract:Zinc-based materials have shown great promise as metallic implants for cardiovascular stents, orthopedic devices, and various other medical applications. As an excellent biodegradable metal, zinc holds significant potential in the development of biodegradable implants due to its superior biocompatibility and enhanced corrosion resistance compared to magnesium. This study investigates the impact of alloying elements such as aluminum (Al) and copper (Cu) on the properties of zinc alloys, focusing on their mechanical performance and in vitro corrosion behavior. The zinc-aluminum-copper alloy was synthesized using a sintering process to enhance its mechanical strength and corrosion resistance in physiological environments. Metal powders of zinc, aluminum, and copper were compressed into pellets and subsequently heated in a high-temperature furnace at 400 °C to form the alloy. To mitigate oxidation, the metals were compacted into cylindrical shapes using a pelletizer, ensuring that only the outermost layer was oxidized while the interior remained oxide free. The resulting Zn-2Al-8Cu alloy exhibited the highest hardness value of 121.91 HV. The compressive yield strength, measured using a universal testing machine (UTM), was found to be 245.17 MPa for the Zn-2Al-8Cu alloy. Corrosion resistance was evaluated through electrochemical tests in NaCl solution, with the Zn-4Al alloy demonstrating the lowest corrosion rate at 0.551 mm/year. Microstructural and surface morphological analyses were conducted using x-ray diffractometer (XRD) and field-emission scanning electron microscopy (FE-SEM). These findings indicate that the addition of aluminum and copper to zinc significantly improves both mechanical properties and corrosion resistance, making these alloys suitable for medical implant applications. The microstructural examination revealed fine, uniformly distributed phases, which contribute to the enhanced mechanical strength and corrosion resistance. Abstract:Biodegradable zinc (Zn) alloys stand out as promising contenders for biomedical applications due to their favorable mechanical properties and appropriate degradation rates, offering the potential to mitigate the risks and expenses associated with secondary surgeries. While current research predominantly centers on the in vitro examination of Zn alloys, notable disparities often emerge between in vivo and in vitro findings. Consequently, conducting in vivo investigations on Zn alloys holds paramount significance in advancing their clinical application. Different element compositions and processing methods decide the mechanical properties and biological performance of Zn alloys, thus affecting their suitability for specific medical applications. This paper presents a comprehensive overview of recent strides in the development of biodegradable Zn alloys, with a focus on key aspects such as mechanical properties, toxicity, animal experiments, biological properties, and molecular mechanisms. By summarizing these advancements, the paper aims to broaden the scope of research directions and enhance the understanding of the clinical applications of biodegradable Zn alloys.
