Abstract:Embryonic stem (ES) cells and ES cell-derived differentiated cells can be used in tissue regeneration approaches. However, inflammation may pose a major hurdle. To define the inflammatory response of ES and ES cell-derived vascular cells, we exposed these cells to LPS. With the exception of MIF no significant cytokine mRNA levels were observed either at baseline or after stimulation. Further experiments revealed that these cells do not express TLR4. Analysis of the DNA methylation status of the TLR4 upstream region showed increased methylation. Moreover, in vitro methylation suppressed TLR4 promoter activity in reporter gene assays. ChIP assays showed that in this region histones H3 and H4 are hypoacetylated in ES cells. Interestingly, 5-aza-dC or TSA partially relieves this gene repression. Finally, the increased levels of TLR4 observed in ES cells after treatment with 5-aza-dC or TSA confer responsiveness to LPS, as induction of IL-6 and TNFα mRNA was detected in endotoxin stimulated ES cells.

Abstract:Introduction: Recent researches identified the existence of perivascular adipose-derived stem cells (ADSCs) which could differentiate into vascular lineages and participate in vascular remodeling. Single-cell mRNA analysis revealed cellular heterogeneity of subcutaneous ADSCs in respect to cell clustering and cell differentiation. However, such analysis of perivascular ADSCs has not been investigated at a single-cell level. Hypothesis: There is a significant difference among perivascular ADSCs subpopulations in respect to vascular-lineage differentiation. Methods: We performed droplet-based single-cell profiling of subcutaneous and perivascular adipose stromal cells and compared ADSCs regarding their heterogeneity, gene ontology, and cell fate trajectory by applying single-cell analysis as well as in vitro and in vivo assays. Results: Single-cell analysis uncovered 4 perivascular ADSCs subpopulations including Dpp4+ , Col4a2+ / Icam1+ , Clec11a+ / Cpe+ and Sult1e1+ cells. Notably, the Clec11a + subpopulation comprised the bulk of perivascular ADSCs, while was hardly presented in subcutaneous ADSCs. Further gene-set enrichment analysis suggested Clec11a + ADSCs were potentially involved with TGF-β signaling pathways and pseudotemporal analysis predicted that Clec11a + subpopulation lay at the end of the differential trajectory towards smooth muscle cells (SMCs). In vitro assays displayed that perivascular ADSCs could differentiate into SMCs via CLEC11A regulation when treated by TGF-β1. To further elucidate the role of the Clec11a + subpopulation in SMCs differentiation, we labeled CLEC11A+ and CLEC11A- perivascular ADSCs by lentivirus transfection and isolated them by FACS assay. CLEC11A+ cells showed the greater capability of SMCs differentiation in response to TGF-β1 in vitro and enhanced neointima formation when transplanted to the adventitial side of guidewire injured arteries. Conclusions: The present study depicted the unique heterogeneity of perivascular ADSCs and the novel role of the Clec11a + subpopulation, providing a supplement for the relationship between perivascular ADSCs and vascular SMCs.

Abstract:OBJECTIVE:Human embryonic stem cells (hESCs) have recently been reported as an unlimited source of mesenchymal stem cells (MSCs). The present study not only provides an identical and clinically compliant MSC source derived from hESCs (hESC-MSCs), but also describes the immunomodulative effects of hESC-MSCs in vitro and in vivo for a carbon tetrachloride (CCl(4))-induced liver inflammation model.METHODS:Undifferentiated hESCs were treated with Rho-associated kinase (ROCK) inhibitor and induced to fibroblast-looking cells. These cells were tested for their surface markers and multilineage differentiation capability. Further more, we analyzed their immune characteristics by mixed lymphocyte reactions (MLRs) and animal experiments.RESULTS:hESC-MSCs show a homogenous fibroblastic morphology that resembles bone marrow-derived MSCs (BM-MSCs). The cell markers and differentiation potential of hESC-MSCs are also similar to those of BM-MSCs. Unlike their original cells, hESC-MSCs possess poor immunogenicity and can survive and be engrafted into a xenogenic immunocompetent environment.CONCLUSIONS:The hESC-MSCs demonstrate strong inhibitory effects on lymphocyte proliferation in vitro and anti-inflammatory infiltration properties in vivo. This study offers information essential to the applications of hESC-MSC-based therapies and evidence for the therapeutic mechanisms of action.

Abstract:Highly differentiated mature vascular smooth muscle cells (VSMCs) are the predominant type of cells constituting arterial walls, which are essential for maintaining the structural and functional integrity of blood vessels. VSMCs demonstrate a notable degree of adaptability following vascular damage, a characteristic that plays a crucial role in the progression of vascular remodeling. Advances in single-cell RNA sequencing in both healthy and pathological vascular tissues have offered profound insights into the complexity of VSMCs, revealing a more intricate diversity than previously recognized. In response to injury, VSMCs undergo dedifferentiation and exhibit pluripotent markers. This review summarizes the researches that have employed lineage tracing alongside single-cell sequencing analysis to explore the dynamics of vascular damage. The primary focus of this study was on the process of dedifferentiation in VSMCs, with particular attention to its underlying mechanisms. The discussion included the impact of microenvironmental cues, the control of transcription factors, and the various molecular pathways involved in VSMCs dedifferentiation. Herein, we provide a comprehensive analysis of cells dedifferentiated from adult VSMCs upon vascular injury.

Abstract:In the face of extraordinary advances in the prevention, diagnosis, and treatment of human diseases, devastating cardiopulmonary diseases continue to deprive people of health. Research in developmental biology has led to the discovery of stem cells, which provide a platform to understand the basic mechanism of those diseases and give a hope for potential clinical applications. Three main types of stem cells have been characterized, including the embryonic stem (ES) cells, embryonic germ stein cells, and tissue-resident, or "adult," stem cells, on the basis of their origin and pluripotent ability. The source of the ES cells is the inner cell mass of a blastocyst, which is a very early embryo that contains 200-250 cells and is shaped like a hollow sphere. Embryonic germ line cells come from a human fetus that is 5-9 weeks old and are derived from tissue that would have developed into the ovaries or testes. "Adult" stem cells are derived from the umbilical cord and placenta or from blood, bone marrow, skin, and other tissues. A major distinction between the ES cells and the germ line cells is the capacity of the former to develop into every type of tissue found in an adult, whereas the latter are less versatile. Similarly, progenitor cells are characterized as pluripotent cells that can replicate and differentiate into a restricted number of cell types. This chapter will focus on the methods, mechanisms, and applications of ES cells derived from both human and mouse origins that differentiate into vascular cells.

Abstract:Inflammation and TNF-alpha signaling play a central role in most of the pathological conditions where cell transplantation could be applied. As shown by initial experiments, embryonic stem (ES) cells and ES-cell derived vascular cells express very low levels of TNF-alpha receptor I (TNFRp55) and thus do not induce cytokine expression in response to TNF-alpha stimulation. Transient transfection analysis of wild-type or deletion variants of the TNFRp55 gene promoter showed a strong activity for a 250-bp fragment in the upstream region of the gene. This activity was abolished by mutations targeting the Sp1/Sp3 or AP1 binding sites. Moreover, treatment with trichostatin A (TSA) led to a pronounced increase in TNFRp55 mRNA and promoter activity. Overexpression of Sp1 or c-fos further enhanced the TSA-induced luciferase activity, and this response was attenuated by Sp3 or c-jun coexpression. Additional experiments revealed that TSA did not affect the Sp1/ Sp3 ratio but caused transcriptional activation of the c-fos gene. Thus, we provide the first evidence that ES and ES-cell-derived vascular cells lack cytokine expression in response to TNF-alpha stimulation due to low levels of c-fos and transcriptional activation of Sp1 that can be regulated by inhibition of histone deacetylase activity.
Innate immunity in human embryonic stem cells: comparison with adult human endothelial cells

Abstract:Treatment of human disease with human embryonic stem cell (hESC)-derived cells is now close to reality, but little is known of their responses to physiological and pathological insult. The ability of cells to respond via activation of Toll like receptors (TLR) is critical in innate immune sensing in most tissues, but also extends to more general danger sensing, e.g. of oxidative stress, in cardiomyocytes. We used biomarker release and gene-array analysis to compare responses in hESC before and after differentiation, and to those in primary human endothelial cells. The presence of cardiomyocytes and endothelial cells was confirmed in differentiated cultures by immunostaining, FACS-sorting and, for cardiomyocytes, beating activity. Undifferentiated hESC did not respond with CXCL8 release to Gram positive or Gram negative bacteria, or a range of PAMPs (pathogen associated molecular patterns) for TLRs 1-9 (apart from flagellin, an activator of TLR5). Surprisingly, lack of TLR-dependent responses was maintained over 4 months of differentiation of hESC, in cultures which included cardiomyocytes and endothelial cells. In contrast, primary cultures of human aortic endothelial cells (HAEC) demonstrated responses to a broad range of PAMPs. Expression of downstream TLR signalling pathways was demonstrated in hESC, and IL-1beta, TNFalpha and INFgamma, which bypass the TLRs, stimulated CXCL8 release. NFkappaB pathway expression was also present in hESC and NFkappaB was able to translocate to the nucleus. Low expression levels of TLRs were detected in hESC, especially TLRs 1 and 4, explaining the lack of response of hESC to the main TLR signals. TLR5 levels were similar between differentiated hESC and HAEC, and siRNA knockdown of TLR5 abolished the response to flagellin. These findings have potential implications for survival and function of grafted hESC-derived cells.

Abstract:Human induced pluripotent stem cells (iPSCs) can be differentiated into vascular endothelial and smooth muscle cells (iEC and iVSMC). The defined genetic makeup and the ease of genetic engineering in multiple cell types on an isogenic background make these cells potentially powerful tools for studying molecular mechanisms underlying vascular function and disease. While several studies have characterized iEC and iVSMC and compared them with other cultured cells, no studies have analyzed these cells in conjunction with native human cells. In this study, we characterized iEC, iVSMC, and fractions of native human arterioles by examining large panels of marker genes. iEC and iVSMC were differentiated (n=4) from a custom-made iPSC line. Differentiated cells were sorted using VE-cadherin ( CDH5 ) to obtain iEC (positive selection) and iVSMC (negative selection). Arterioles were isolated from adipose tissues obtained from 4 humans, and the endothelial and vessel remainder fractions were separated by scraping. RNA-seq was performed on all samples. We focused the analysis on 10 and 15 marker genes for EC and VSMC, respectively, that we curated from the literature. iEC exhibited robust expression of 9 of the 10 EC markers such as CD34 (FPKM, 137±58), PECAM1 (153±32), and VWF (44±7), while iVSMC had negligible expression of EC marker genes. Conversely, iVSMC exhibited robust expression of 11 of the 15 VSMC marker genes, while the expression of these genes in iEC was depleted by several fold. Eight of the 10 EC marker genes were enriched in the endothelial fraction of native arterioles by several fold compared with intact arterioles, while 12 of the 15 VSMC marker genes were substantially depleted in the endothelial fraction. EC marker genes were significantly depleted and VSMC marker genes enriched in the vessel remainder fraction. In summary, iEC and iVSMC, as well as endothelial and vessel remainder factions from native human arterioles, exhibit robust expression of large panels of EC and VSMC marker genes based on agnostic RNA-seq analysis. These novel findings indicate that iEC, iVSMC, and endothelial and vessel remainder factions of native human arterioles are relevant tools for studying molecular regulation in specific vascular cell types.
Derivation of vascular smooth muscle and endothelial cells from mouse embryonic stem cells

Abstract:Objective: To investigate the feasibility of obtaining smooth muscle and endothelial cells from differentiated mouse embryonic stem cells (ESCs). Methods: Flk-1 was used as a marker for vascular precursors. Flk-1 positive cells were then sorted from embryoid bodies by flow cytometery, and induced to vascular smooth muscle cells and endothelial cells with platelet derived growth factor and vascular endothelial growth factor, respectively. After induction, the markers for smooth muscle and endothelial cells were analyzed by immunofluorescent staining, and the differentiation efficiency and purity were analyzed by flow cytometery. Results: It was revealed by flow cytometry that about 50% of cells in day 4 embryoid bodies expressed Flk-1,and (95.9 ± 3.5) % of α-SMA positive smooth muscle cells and (59.1 ± 4.8)% of CD31 positive endothelial cells were obtained from differentiation of sorted Flk-1 positive cells. Conclusion: Vascular smooth muscle and endothelial cells can be obtained from differentiated mouse ESCs, providing a new cell source for tissue engineered blood vessel construction.

Abstract:Background: Peripheral vascular disease remains a leading cause of vascular morbidity and mortality worldwide despite advances in medical and surgical therapy. Besides traditional approaches, which can only restore blood flow to native arteries, an alternative approach is to enhance the growth of new vessels, thereby facilitating the physiological response to ischemia. Methods: Actin CreER /R26 VT2/GK3 “Rainbow” reporter mouse was used for unbiased in vivo survey of injury-responsive vasculogenic clonal formation. Prospective isolation and transplantation were used to determine vessel-forming capacity of different populations. Single-cell RNA sequencing was used to characterize distinct vessel-forming populations and their interactions. Results: Two populations of distinct vascular stem/progenitor cells (VSPCs) were identified from adipose-derived mesenchymal stromal cells (MSCs): VSPC1 is CD45-Ter119-Tie2+PDGFRa-CD31+CD105 high Sca1 low , which gives rise to stunted vessels (incomplete tubular structures) in a transplant setting, and VSPC2 which is CD45-Ter119-Tie2+PDGFRa+CD31-CD105 low Sca1 high and forms stunted vessels and fat. Interestingly, co-transplantation of VSPC1 and VSPC2 is required to form functional vessels that improve perfusion in the mouse hindlimb ischemia model. Similarly, VSPC1 and VSPC2 populations isolated from human adipose tissue could rescue the ischemic condition in mice. Conclusions: These findings suggest that autologous co-transplantation of synergistic VSPCs from non-essential adipose tissue can promote neovascularization and represents a promising treatment for ischemic disease.

Abstract:Introduction: Given the limited success to date in using non-specific membrane makers, in identifying cardiac stem cells, the significance of finding a much more potential cardiac mesenchymal stem cell (C-MSC) maker from transcription factors in nuclear is very important. Hypothesis: We want to test the feasibility of using GATA4, an early cardiac-specific transcription factor, as biomarker to identify a subpopulation of C-MSC that could be highly efficacious for cardiovascular repair. Methods: A lentiviral vector in which the expression of eGFP reporter is driven by a 1kb fragment of the mouse GATA4 promoter was constructed. Murine C-MSC with high GFP intensity were sorted by FACS. Results: Western blot shows that the C-MSC population with high GFP intensity exhibits higher GATA4 protein expression as compared with unsorted C-MSC. Interestingly, we observed that sorted GATA4high C-MSC also exhibit higher HIF-1α protein expression as compared with unsorted C-MSC. We compared hypoxia- and serum deprivation induced-apoptosis between sorted GATA4high and unsorted C-MSC by TUNEL staining. We found that the GATA4high exhibited lower rate of TUNEL positivity as compared with unsorted C-MSC, suggesting GATA4high C-MSC are more resistant to hypoxia- and serum deprivation-induced apoptosis in comparison with unsorted C-MSC. In mouse model of myocardial infarction by permanent left coronary ligation, GATA4high C-MSC or unsorted C-MSC was injected into the border zone area. Left ventricular ejection fraction (EF) were serially (days 1 and 14) assessed using echocardiography. At day 1, no differences in EF were observed between GATA4high C-MSC and unsorted C-MSC group (n=9, ns). At day 14, left ventricular ejection fraction in GATA4high C-MSC cell-treated group was improved about 11.2% compared with the unsorted C-MSC group (n=9, P Conclusions: Our results suggest that GATA4high, as compared with unsorted C-MSC, exhibit enhanced survival capacity which will consequently lead to improvement of heart contractile function in the infarcted heart, therefore, GATA4high C-MSC can be considered as a much more potential progenitor for heart repair.

Abstract:Objective To master the technique of mouse embryonic stem (ES) cells differentiate into endothelial cells,which would be a new therapeutic approach for cardiovascular disease.Methods Expression of selfrenewal marker genes in E 14 cells was assessed.Expression of vascular endothelial growth factor receptor 2 (Flk 1) in monolayer differentiation on day 4 and vascular endothelial cadherin (VE-cadherin) on day 8 were detected.On day 8,differentiation cells were also observed under phase contrast microscopy (PCM) and transmission electron microscope (TEM).ES cells and endothelial-specific molecular markers were assessed by RT-PCR at different time-points.Results As self-renewal marker genes were expressed in E14 cells,E14 cells was identified to maintain their selfrenewal pluripotency.The marker gene of letarl,Flk1 was expressed on differentiation day 4.On differentiation day 8 the marker gene VE-cadherin was expressed and as observed under PCM endothelial cells with spindle shape and TEM with Weibel-Palade body,thus were the major populations generated after VEGF induction,and E14 cells were confirmed differentiated into mature endothelial cells.The expressions of genes octamer binding transcription factor 4 (Oct4),Flk1 and VE-cadherin were detected on differentiation day 2,4,6,8 and 10.Conclusions As VE-cadherin gene was expressed in monolayer on differentiation day 8,E14 cells were confirmed differentiated into endothelial cells,which would be a new therapeutic approach for cardiovascular disease.

Abstract:Objective To explore the conditions and characteristics of endothelial cell (EC) differentiation as well as new blood vessel formation from in vitro differentiation of mouse embryonic stem (ES) cells. Methods Two-dimensional and three-dimensional differentiation systems of mouse ES cells were established respectively,and then platelet-endothelial cell adhesion molecule1 (PECAM-1) immunofluorescent staining and DiI-acetylated low-density lipoprotein (DiI-Ac-LDL) labeling of differentiated cells were performed to observe the characteristics of EC differentiation and blood vessel formation. Results In the two-dimensional differentiation system, ES cells differentiated into ECs in the absence of exogenous growth factors. The ECs were localized in areas with high cell density. Some differentiated ECs grew as a colony instead of endothelial networks, others connected to form EC networks confirmed by PECAM-1 immunofluorescent staining. In the suspended embryoid bodies(EBs)culture system of three-dimensional differentiation, three structures were found without addition of exogenous growth factors: cord-like structure, vessel-like structure and disordered cell mass. The three-dimensional reconstruction of these EBs' cryosections showed massive vascular networks. In the typeⅠcollagen culture system of three-dimensional differentiation, EBs derived from ES cells displayed sprouting angiogenesis, which was dependent on cocktails of exogenous growth factors. Conclusion In these two-and three-dimensional differentiation systems in vitro, EC differentiation and blood vessel formation are similar to the in vivo process, which includes vasculogenesis and angiogenesis. Thus these two systems may serve as ideal models for further research on mechanisms of vascular development.
Sca-1(+) Progenitors Derived from Embryonic Stem Cells Differentiate into Endothelial Cells Capable of Vascular Repair after Arterial Injury

Abstract:Background— Embryonic stem cells possess the ability to differentiate into endothelium. The ability to produce large volumes of endothelium from embryonic stem cells could provide a potential therapeutic modality for vascular injury. We describe an approach that selects endothelial cells using magnetic beads that may be used therapeutically to treat arterial injury. Methods and Results— Large numbers of endothelial cells (ECs) with high purity were produced using Sca-1 + cells isolated with magnetic beads from predifferentiated embryonic stem cells (ESCs) cultured in α-MEM containing 10 ng/mL VEGF 165 for a minimum of 21 days (esEC). The transcription regulator histone deacetylase (HDAC3) was essential for VEGF-induced EC differentiation. Immunofluorescence or fluorescence-activated cell sorter (FACS) analysis revealed that esECs expressed a full range of EC lineage-specific markers including CD31, CD106, CD144, Flk-1, Flt-1, and von Willebrand factor (vWF). FACS analysis confirmed that 99% of esECs were CD31-positive and 75% vWF-positive. Furthermore, almost all cells were positive for DiI-acLDL uptake. When matrigel containing esECs was subcutaneously implanted into mice, various vessel-like structures were observed indicating their endothelial cell like phenotype. In keeping with this, when esECs infected with adenovirus-LacZ were injected into denuded femoral arteries of mice, they were found to form a neo-endothelium that covered the injured areas (86%±13.6%), which resulted in a 73% decrease in neointimal area 2 weeks after injury. Conclusions— We conclude that Sca-1 + cells can differentiate into functional ECs via activation of HDAC3, accelerating re-endothelialization of injured arteries and reducing neointima formation.

Abstract:Embryonic stem (ES) cells are highlighted as promising cell sources for regenerative medicine. Here, we focused on providing the platform that forced ES cells to reproduce the vascular organization process, leading to efficiency and safety evaluation as preclinical testing of biological agents. Murine ES cell-derived embryoid bodies on matrigel, but not collagen or gelatin, could be differentiated into sprouting blood vessels without the addition of growth factors. The expression of endothelial cell marker CD31 and smooth muscle marker alpha-smooth muscle actin was partially colocalized and started to increase 7 days after culture on matrigel, accompanied by the induction of a number of growth factors, such as vascular endothelial growth factor, fibroblast growth factor-2, hepatocyte growth factor, transforming growth factor-beta, and angiopoietin-1. Moreover, notch-related genes, such as Del1 or Del4 (delta-like 1/4) and hey1 or hey2 (hairy/enhancer of split related TRPW motif 1/2), were upregulated in a similar time course. The treatment of neutralizing antibodies against these growth factors failed to inhibit the differentiation into the sprouting blood vessels, whereas arginine-glycine-aspartic peptide, a selective inhibitor for the alphavbeta3-integrins, did inhibit differentiation. An anticancer drug to inhibit angiogenesis, TNP-470, also blocked the vascular formation in this model. ES cells could reproduce the vascular organization process on the biosynthetic scaffolds, such as matrigel, without the addition of growth factors. In the future, a human ES-based tissue model would be an optional tool for the screening of pharmaceutical drugs for vascular disease.

Abstract:Significance: Since the discovery of vascular stem cells, there has been considerable advancement in comprehending the nature and functions of these cells. Due to their differentiation potential to repair endothelial cells and to participate in lesion formation during vascular remodeling, it is crucial to elucidate vascular stem cell behaviors and the mechanisms underlying this process, which could provide new chances for the design of clinical therapeutic application of stem cells. Recent Advances: Over the past decades, major progress has been made on progenitor/vascular stem cells in the field of cardiovascular research. Vascular stem cells are mostly latent in their niches and can be bioactivated in response to damage and get involved in endothelial repair and smooth muscle cell aggregation to generate neointima. Accumulating evidence has been shown recently, using genetic lineage tracing mouse models, to particularly provide solutions to the nature of vascular stem cells and to monitor both cell migration and the process of differentiation during physiological angiogenesis and in vascular diseases. Critical Issues: This article reviews and summarizes the current research progress of vascular stem cells in this field and highlights future prospects for stem cell research in regenerative medicine. Future Directions: Despite recent advances and achievements of stem cells in cardiovascular research, the nature and cell fate of vascular stem cells remain elusive. Further comprehensive studies using new techniques including genetic cell lineage tracing and single-cell RNA sequencing are essential to fully illuminate the role of stem cells in vascular development and diseases. Abstract:Vascular, resident stem cells are present in all 3 layers of the vessel wall; they play a role in vascular formation under physiological conditions and in remodeling in pathological situations. Throughout development and adult early life, resident stem cells participate in vessel formation through vasculogenesis and angiogenesis. In adults, the vascular stem cells are mostly quiescent in their niches but can be activated in response to injury and participate in endothelial repair and smooth muscle cell accumulation to form neointima. However, delineation of the characteristics and of the migration and differentiation behaviors of these stem cells is an area of ongoing investigation. A set of genetic mouse models for cell lineage tracing has been developed to specifically address the nature of these cells and both migration and differentiation processes during physiological angiogenesis and in vascular diseases. This review summarizes the current knowledge on resident stem cells, which has become more defined and refined in vascular biology research, thus contributing to the development of new potential therapeutic strategies to promote endothelial regeneration and ameliorate vascular disease development.

Abstract:Cardiovascular disease is a major cause of death worldwide. Many cardiovascular events, such as stroke and coronary heart disease, strongly correlate with loss of blood supply, leading to loss of cell function, organ failure, and eventually death.1 Vascular regeneration, which includes restoration of normal vascular structure and function, the reversal of vascular senescence, and the growth of new blood vessels may be a promising approach for the treatment of these diseases.2 Stem/progenitor cells are able to differentiate into vascular cell lineages, which may contribute to the regenerative process and could be useful for the disease treatment.3 Recently, several publications in Arteriosclerosis, Thrombosis, and Vascular Biology and other journals have demonstrated great progress in research into stem/progenitor cells for vascular regeneration.4–6 In the present article, we will highlight these updated publications, providing insights into the role of stem/progenitor cells in restoration of vascular function under pathophysiological conditions and the contribution of these cells to lesion formation in the vessel wall through the mechanisms of cell migration and differentiation.Since the discovery of hemangiogenic cells in the bone marrow, researchers have been searching for other such pools of circulating progenitors, such as those that may contribute to vascular repair after ischemic injury. The study of circulating endothelial progenitors, termed endothelial progenitor cells (EPCs), became an extremely attractive field of research after they were first isolated by Asahara et al7 in 1997, using magnetic bead selection based on expression of specific cell surface antigens.
