Two 5^Il6-Tnfalpha Mutant Fusion Proteins with SAPGTP and PG Linkers

Abstract:Using computer,ten linkers with higher flexibility of their influences on the spatial configurations of IL 6 and TNF Δ IN 5′IL6 TNF Δ fusion protein were compared.The linker with amino acid sequence SAPGTP was chosed.5′IL6 SAPGTP TNF Δ fusion protein has been constructed and it has analogous spatial configurations with 5′IL6 PG TNF Δ by the conjecture on computer.By sequentially analysis,the two fusion proteins above proved consistent with what had designed.They were expressed in E.Coli and were purified.Their biological activities and cytotoxities were tested and compared on high expression IL 6R cells.The results showed that 5′IL6 SAPGTP TNF Δ had 2 7 fold biological activity than that of 5′IL6 PG TNF Δ on L929 cells,and had 1 3 fold inhibition ratio than that of 5′IL6 PG TNF Δ on U 937 cells.Meanwhile,5′IL6 SAPGTP TNF Δ and 5′IL6 PG TNF Δ showed 3 7 fold and 2 9 fold cytotoxities respectively than that of TNFα mutant(rhTNFα D11a),which was mutant at the same sites,on high expression IL 6R cell line U 937 .So,the fusion protein 5′IL6 TNF Δ with linker SAPGTP is better than the 5′IL6 TNF Δ With linker PG.
