Abstract:Skeletal editing of N-heterocycles has recently received considerable attention, and the introduction of boron atom into heterocycles often results in positive property changes. However, direct enlargement of N-heterocycles through boron atom insertion is rarely reported in the literature. Here, we report a N-heterocyclic editing reaction through the combination boron atom insertion and C-H borylation, accessing the fused BN-heterocycles. The synthetic potential of this chemistry was demonstrated by substrate scope and late-stage diversification of products.

Abstract:The closo ‐dodecaborate dianion is a fundamental icosahedral boron cage with 12 identical B−H vertices. The chemistry and applications of boron clusters have inspired researchers ever since their discovery several decades ago, and the selective modification of the cage positions has remained a major synthetic challenge. A rhodium(III)‐catalyzed B−H functionalization–cyclization cascade of closo ‐dodecaborate amides is reported. The transformations occur chemoselectively at B−H positions in the presence of C−H bonds prone to competitive cyclometalation. Previously inaccessible cage derivatives with B−C(sp 2 ) and B−C(sp 3 ) bonds as well as a fused diboraoxazole ring are obtained in a one‐pot process. The reactions proceed under mild conditions and exhibit complete cage regioselectivity with broad functional group tolerance. These cluster derivatives enable a largely extended investigation of the application of anionic boron clusters in research areas such as photoluminescent materials and medicinal chemistry.

Abstract:The replacement of a CC unit with an isoelectronic BN unit in aromatic systems can give rise to molecules and materials with fascinating properties. We report here the synthesis, characterization, and reactivity of a 1,4,2,3-diazadiborole species, 2, featuring an unprecedented 6π-aromatic BN-heterocyclic moiety that is isoelectronic to cyclopentadienide (Cp−). Bearing an unsymmetrical B=B entity, 2 exhibits reactivity toward oxidants, protic reagents, electrophiles, and unsaturated substrates. This reactivity facilitates the synthesis of a variety of novel mono- and bicyclic organoboron derivatives through mechanisms including ring retention, cleavage/recombination, annulation, and expansion. These findings reveal innovative synthetic routes to BN-embedded aromatic compounds via desymmetrization, affording unique building blocks for synthetic chemistry.

Abstract:The paper presents a new method for the formation of B-N bond by the catalytic insertion of nitrenes into organic boranes. The reaction proceeds most selectively for cyclic boranes with 2-phenylpyridine framework and nitrenes generated in situ by oxidation sulfonamides and sulfamates. The most effective catalysts for the process are rhodium and ruthenium carboxylates of [M2(OOCR)4]X type. Complexes with NTTL ligands derived from S-tert-leucine provide chiral products with stereogenic boron atoms. The developed approach can be useful for the introduction of boron heterocycles into functional materials and biologically active molecules.

Abstract:The diastereoselective synthesis of boron and carbon vicinal stereogenic centers is a great challenge in current synthetic chemistry due to the paucity of efficient synthetic strategies. Herein, a Cu-catalyzed diastereo- and enantioselective B–H bond insertion reaction of ene-yne-ketones as furyl-carbene precursors has been developed using chiral bis(oxazolines) (BOX) as the ligand, thus furnishing a series of densely functionalized products (42 examples) bearing both boron and carbon vicinal stereogenic centers in high enantioselectivity and high diastereoselectivity (up to 99% ee and > 20:1 dr) with good yields (up to 98%).

Abstract:Chiral propargylic boron compounds have a diverse array of potential applications. Although numbers of highly enantioselective methodologies for the synthesis of other organoboron compounds are well established, highly enantioselective synthesis of chiral propargylic boron compounds remains unknown. Herein, we report a catalytic method for the construction of chiral propargylic boron compounds by means of highly enantioselective dirhodium-catalyzed B–H bond insertion reactions using aryl propargylic sulfonylhydrazones as carbene precursors. This method enables the straightforward synthesis of a wide range of stable chiral propargylic boron compounds in yields up to 99% with enantioselectivities up to 97% ee. The operationally simple method features a broad substrate scope, good functional group tolerance, and mild reaction conditions. The boron compounds prepared in this study are inaccessible by other known methods and exhibit high potential utility because they could be efficiently transformed into allenyl borates, triazole borane compounds, and homopropargylic and propargylic alcohols with good enantiomeric fidelity. Our work expands the library of catalytic B–H bond insertion reactions, increases the variety of chiral organoboron compounds that can be synthesized, and provides preliminary information about possible transformations of chiral propargylic boron compounds, thus laying the foundation for wide application of these compounds.

Abstract:Slipping boron into alkyl ethers The carbon–oxygen bonds that hold alkyl ethers together are relatively inert. Lyu et al. report that zinc and nickel can team up to insert boron between the carbon and oxygen using an unusual mechanism. First, a dibromoborane pries open the bond with help from a zinc ion. Next, the nickel stitches together the carbon and boron, and the oxidation of metallic zinc starts the cycle over again. Abstract:A strategy for the photosensitized [2+2]-cycloaddition between styrenyl dihaloboranes and unactivated allylamines to access cyclobutylboronates with control of stereochemistry and regiochemistry is presented. The success of the reaction relies on the temporary coordination between in situ generated dihaloboranes and amines under mild reaction conditions. In addition, cyclobutanes with varying substitution patterns have been prepared using N -heterocycles as directing group. Manipulation of the C−B bond allows for the synthesis of a diverse class of cyclobutanes from simple precursors. Moreover, these reactions lead to the synthesis of complex amines and heteroaromatic compounds, which have significant utility in medicinal chemistry. Finally, a dearomative [4+2]-cycloaddition of naphthalenes using a boron-enabled temporary tethering strategy has also been uncovered to synthesize complex 3-dimensional borylated building blocks.

Abstract:Abstract Compared with the well-developed carbon-stereogenic chemistry, the construction of boron-stereogenic compounds remains undeveloped and challenging. Herein, the previously elusive catalytic enantioselective construction of boron-stereogenic compounds has been achieved through enantioselective desymmetric B–H bond insertion reaction. The B–H bond insertion reaction of 2-arylpyridine-boranes with versatile diazo compounds under chiral copper catalyst can afford boron-stereogenic compounds with good to excellent enantioselectivity. Moreover, the synthetic utility of this reaction is demonstrated by the scalability and downstream transformations. DFT calculations provide insights into the reaction mechanism and the origin of stereoselectivity.

Abstract:Boron-mediated homologation can potentially access almost any kind of chiral centers from readily available boronates via asymmetric carbenoid insertion, followed by versatile transformations of the carbon−boron bonds. However, the current asymmetric boron homologation strategies exhibit limitations, and enantioselective insertion of diversely substituted carbenoids remains challenging. Here we report an enantioconvergent approach for direct insertion of carbon-, oxygen-, nitrogen-, sulfur-, and silicon-substituted carbenoids into carbon−boron bonds. The excellent enantioselectivity is enabled by a new class of chiral oxazaborolidines derived from inexpensive α-amino esters. Computational studies revealed that the non-C2-symmetric oxazaborolidine features a puckered geometry and the cooperative effects of multiple substituents create an asymmetric environment for effective enantioinduction. This method is scalable, and each chiral center can be independently controlled by the chiral oxazaborolidine without being influenced by nearby stereocenters. Besides forming singular chiral centers, iterative operations of this asymmetric homologation simplify synthesis of complex molecules with multiple stereocenters.

Abstract:A first example of boronic acid catalyzed intermolecular aza-Piancatelli rearrangement to access a wide range of trans-4,5-disubstituted cyclopentenones is described. The catalytic system can also be extended to intramolecular aza-Piancatelli rearrangement to afford a wide range of azaspirocycles. Good to excellent yields were obtained in an excellent diastereoselective manner.

Abstract:Transition metal (TM)-catalysed directed hydroboration of aliphatic internal olefins which facilitates the construction of complex alkylboronates is an essential synthetic methodology. Here, an efficient method for the borylation of cyclopropanes involving TM-catalysed directed C-C activation has been developed. Upon exposure to neutral Rh(i)-catalyst systems, N-Piv-substituted cyclopropylamines (CPAs) undergo proximal-selective hydroboration with HBpin to provide valuable gamma-amino boronates in one step which are otherwise difficult to synthesize by known methods. The enantioenriched substrates can deliver chiral products without erosion of the enantioselectivities. Versatile synthetic utility of the obtained gamma-amino boronates is also demonstrated. Experimental and computational mechanistic studies showed the preferred pathway and the origin of this selectivity. This study will enable the further use of CPAs as valuable building blocks for the tunable generation of C-heteroatom or C-C bonds through selective C-C bond activation.

Abstract:A visible‐light‐induced protocol is developed for the construction of C(sp2)−P bonds with heterogeneous boron carbonitrides (BCN). The system features with broad scope and mild reaction conditions, providing various aryl phosphates in moderate to good yields from arenes and arylhalides. Mechanism studies verify a radical process, and C−H bond cleavage is not involved in the rate‐determined step. Furthermore, the strategy allows the late‐stage phosphonation of biologically active compounds and gram‐scale synthesis, which offers a metal‐free arene for valuable phosphorus‐containing compounds.

Abstract:Aromatic boron-containing organic compound C 2 B 2 H 2 with an unusual C=C bond was experimentally synthesized in 2017. Here we investigate the structure and bonding nature of C 2 B 2 H 2 and its derivatives C 2 B 2 R 2 using DFT and VB theory. While the C=C bond in C 2 B 2 R 2 consists of a π bond and a charge-shift (CS) bond, C 2 B 2 F 2 has the lowest LUMO energy and its LUMO is similar with that of ethylene, suggesting that C 2 B 2 F 2 can be an ideal dienophile for the Diels-Alder reaction. Subsequently, the mechanism and stereoselectivity of the Diels-Alder reaction of C 2 B 2 F 2 with 5-substituted cyclopentadienes are studied. Computations demonstrate that these Diels-Alder reactions are feasible thermodynamically and kinetically. The stereoselectivity and distortion angles of C 2 B 2 R 2 exhibit linear correlations with the electronegativity difference between the two substituents bonded to the C(sp 3 ) of cyclopentadiene, suggesting that the stereoselectivity of related Diels-Alder reaction products can be modulated by the substitution of cyclopentadiene. Considering the current interests in boron neutron capture therapy (BNCT), we design six BNCT drugs through the Diels-Alder reaction of C 2 B 2 F 2 with dienes containing peptide fragments. Thus, we demonstrate a kind of new method of designing three-in-one BNCT drugs with the facile Diels-Alder reaction.

Abstract:The synthesis of polysubstituted (hetero)aromatic compounds is essential in various fields, including pharmaceuticals, where such compounds are fundamental to many approved drugs. In this study, we present a novel electrochemical method for sin-gle-carbon insertion targeting various (hetero)aromatic compounds, with a particular focus on pyridines. In this process, the electrochemical oxidation of pyrrole derivatives produces a radical cation intermediate, which then undergoes nucleophilic attack by diazo compounds to yield polysubstituted pyridine derivatives. Notably, the insertion position is influenced by the electronic properties of N-protecting groups, allowing for unprecedented para-selective insertion through the introduction of electron-withdrawing groups. This approach is applicable to various substrates such as indole, imidazole, indene, and cyclo-pentadiene, resulting in the desired carbon-inserted products. Insights from in-situ spectroscopy and theoretical calculations suggest the involvement of distonic radical cation intermediates, facilitating carbon-atom migration on the aromatic ring and enabling insertion at different positions. This study expands the chemical toolkit for synthesizing polysubstituted (het-ero)aromatic compounds and introduces a new concept for single-carbon insertion chemistry.

Abstract:Herein, we disclose a RhIII-catalyzed heteroarylation of C(sp3)–H and C(sp2)–H bonds in heterocycles with organoboron reagents. This protocol displays high efficiency and excellent functional group tolerance. A range of heterocyclic boronates with strong coordinating atoms, including pyridine, pyrimidine, pyrazole, thiophene, and furan derivatives, can be extensively served as the coupling reagents.
