Abstract:Three hexakisimidazolium-incorporated molecular cages o -, m - and p - HiCages have been prepared through the condensation of 1,3,5-tri(imidazolyl)benzene and o -, m - or p -xylylene dibromide. The new hexacationic molecular cages are highly water-soluble. It is demonstrated that among various purines m - and p - HiCages have the ability to bind uric acid selectively in water. The binding constants their complexes for uric acid are determined by isothermal titration calorimetric experiments to be (5.86 ± 0.30) × 10 4 M -1 and (2.61 ± 0.06) × 10 4 M -1 , and the structure of both complexes is confirmed by X-ray diffraction analysis. These imidazolium-derived molecular cages exhibit low cytotoxicity and hemolysis rate, providing the possibility for subsequent biological applications.

Abstract:A triangular-prism shaped cage was self-assembled by the formation of a dynamic covalent bond, namely a hydrazone, in acidic aqueous solution. The hexacationic host bears a number of relatively acidic protons pointing inside the cage cavity, which is able to accommodate an iodide anion selectively in water over other halide anions such as F- , Cl- , and Br- , which are more hydrated. As a comparison, a macrocycle analogue bearing fewer positive charges and fewer hydrogen bond donors shows no anion binding ability. As inferred from the single-crystal structure and theoretical calculations, the ability of the cage to selectively recognize iodide anions results from the combination of electrostatic forces, C-H⋅⋅⋅I- hydrogen bonds, and the hydrophobic effect.
Robust organic cages prepared using hydrazone condensation display sulfate/hydrogenphosphate selectivity in water

Abstract:Two robust hexacationic cages incorporating either urea or isophthalamide motifs were synthesized via a short and high-yielding synthetic pathway using hydrazone condensation reactions in water for the cage forming step. Stability testing revealed that the cages are stable to a range of stimuli in water and in organic solvents. The urea containing cage can bind anions in pure water, and displays strong and selective binding of SO42– over HPO42–. The isophthalamide containing cage binds SO42– only weakly in 1:1 D2O:d6-DMSO but displays strong and cooperative binding of two HPO42– anions. Combined quantum mechanical/annealed molecular dynamics simulations suggest that the remarkable differences in anion selectivity are largely a result of the differing flexibilities of the two cages.

Abstract:A water-soluble cage containing three naphthalenediimide (NDI) units was synthesized in a one-pot manner without chromatographic purification, during which six irreversible C-N bonds formed simultaneously via an SN2 reaction. The cage was observed to be capable of accommodating a variety π-electron rich guests in a peripheral manner in water. However, for linear guests including I3- and I2, the cage is able to form an inclusion complex. Besides, in the solid state, the cage can absorb vapor of I2.

Abstract:Abstract Developing efficient adsorbents to capture radioactive iodine produced from nuclear wastes is highly desired. Here we report the facial synthesis of a hexacationic imidazolium organic cage and its iodine adsorption properties. Crucial role of counteranions has been disclosed for iodine capture with this cage, where distinct iodine capture behaviors were observed when different counteranions were used. Mechanistic investigations, especially with the X-ray crystallographic analysis of the iodine-loaded sample, allowed the direct visualization of the iodine binding modes at the molecular level. A network of multiple non-covalent interactions including hydrogen bonds, halogen bonds, anion···π interactions, electrostatic interaction between polyiodides and the hexacationic skeleton of the cage are found responsible for the observed high iodine capture performance. Our results may provide an alternative strategy to design efficient iodine adsorbents.

Abstract:Hyperuricemia, characterized by elevated uric acid levels and subsequent crystal deposition, contributing to conditions such as gout, cardiovascular events, and kidney injury, poses a significant health threat, particularly in developed countries. Current drug options for treatment are limited, with safety concerns, leading to suboptimal therapeutic outcomes in symptomatic hyperuricemia patients and a lack of pharmaceutical interventions for asymptomatic cases. Distinguishing from the previous drug design strategies, we directly target uric acid, the pathological molecule of hyperuricemia, resulting in a pyrimidine derivative capable of increasing the solubility and excretion of uric acid by forming a complex with it. Its prodrug showed an anti-hyperuricemia activity comparable to benzbromarone and a favorable safety profile in vivo. Our finding provides a strategy purely based on organic chemistry to address the largely unmet therapeutic needs on novel anti-hyperuricemia drugs.

Abstract:Self-assembly by imine condensation in aqueous media is a formidable task because of the labile nature of imines in the presence of water. Here, by taking advantage of multivalence and ligand preorganization, basket-shaped triscationic cage molecules are self-assembled in high yields in both water and organic solvent, by condensing a hexaformyl and bisamine. These cages, especially the chiral ones, are stable or inert in aqueous solution, that is, no decomposition was observed upon dilution, precipitation, or exposure to competitive amines or aldehydes. Such water-compatibility allows the hosts to take advantage of the hydrophobic effect to accommodate hydrophobic guests. The chiral cage S-2(3+) selectively binds and distinguishes one of two enantiomers, opening up opportunities for applications such as chiral compound separation. Chiral narcissistic self-sorting and sergeants-and-soldiers effects occur during cage formation when two amino precursors are involved in self-assembly.

Abstract:Selective anion recognition remains a key challenge in supramolecular chemistry: only a very small number of systems that can function in water are known, and these nearly always preferentially bind hydrophobic anions. In this work, we report three robust hexa-cationic cages that can be prepared on scales up to 14 g in two simple and high-yielding steps from commercially-available materials. One of these cages displays unusually strong sulfate binding in water (Ka = 12,000 M–1), and demonstrates high selectivity for this anion over H2PO4–/HPO42– in DMSO/buffer mixtures. These results demonstrate that relatively large, three-dimensional supramolecular hosts can be prepared in high yields and on large scales, and can be highly potent receptors.

Abstract:Strongly hydrated anions are challenging targets for molecular receptors. In nature, sulfate binding proteins use multiple neutral NH hydrogen bonds to surround the guest, resulting in sub-millimolar to sub-micromolar sulfate affinities. Synthetic receptors, however, have not yet achieved comparable efficiency and selectivity for sulfate solely by relying on hydrogen bonds or other ion-dipole interactions. Here we report charge-neutral macrotricyclic hexa-urea cages which exhibit strong sulfate affinities (6000 to 9000 M–1) in water with exceptional selectivity over hydrophobic anions. Unlike sulfate binding proteins where a low-polarity microenvironment enhances sulfate binding, the urea cages achieve high sulfate affinities with the urea binding sites exposed to water. The work demonstrates that synthetic receptors operating solely by neutral hydrogen bond donors can emulate the efficiency and selectivity of naturally occurring systems.

Abstract:An ester–imine cage was synthesized from hexapyrrolic tripodal precursor and characterized with NMR spectroscopy and X–ray crystallography. This novel molecular cage displays good stability in wet organic solvents and exhibits moderate to strong affinity (10 3 –10 6 M– 6 ) for anions in chloroform. Abstract:Solute binding proteins can capture strongly hydrated substrates from water with high affinity. Notable examples are the sulfate binding proteins (SBPs), which use neutral NH and OH groups to stabilize highly charged sulfate anions without salt-bridge formation. Synthetic receptors are yet to achieve comparable capabilities solely by hydrogen bonds or other ion-dipole interactions. Here we report charge-neutral urea cages which have strong affinities for CrO42–, SO42– and Cl– up to micromolar and sub-micromolar levels in water, with tunable anion selectivity based on size match. This is despite the fact that these urea cages are much smaller than proteins and have their anion binding sites exposed to water, demonstrating synthetic receptors’ hydrogen bonding potency that emulates nature’s anion binders.

Abstract:Abstract In this paper, we report a unique type of core-shell crystalline material that combines an inorganic zeolitic cage structure with a macrocyclic host arrangement and that can remove trace levels of iodine from water effectively. These unique assemblies are made up of an inorganic Archimedean truncatedhexahedron ( tcu ) polyhedron in the kernel which possesses six calixarene-like shell cavities. The cages have good adaptability to guests and can be assembled into a series of supramolecular structures in the crystalline state with different lattice pore shapes. Due to the unique core-shell porous structures, the compounds are not only stable in organic solvents but also in water. The characteristics of the cages enable rapid iodine capture from low concentration aqueous I 2 /KI solutions (down to 4 ppm concentration). We have studied the detailed process and mechanism of iodine capture and aggregation at the molecular level. The facile synthesis, considerable adsorption capacity, recyclability, and β- and γ-radiation resistance of the cages should make these materials suitable for the extraction of iodine from aqueous effluent streams (most obviously, radioactive iodide produced by atomic power generation).

Abstract:The utilization of a confined cavity in a coordination cage to manipulate the reaction pathways can nicely complement the synthetic shortage of site-selective reactions. However, harmonization of the transition state in the cavity is a significant challenge. To address this issue, we have designed an imidazolium-functionalized cage with a regular octahedral geometry composed of 12 imidazolium linkers and 6 Cu4-calix[4]arene nodes. As a catalyst, the diisopropylphenylimidazolium functionality enables an efficient N-chlorination, and the created cavity favors a six-membered cyclic transition state for chlorine atom transfer, boosting the site-selective C(sp3)–H chlorination of amides/sulfonamides in the water. Our mechanistic investigations have disclosed the rate-determining N–Cl activation mode within the cavity for the chlorine atom transfer pathways.

Abstract:A tricationic cage was synthesized via click chemistry. The cage has a preorganized cavity, which is surrounded by six relatively acidic C-H bonds. In organic solvent, the cage can recognize a variety of anions, among which the Cl- anion has the highest binding affinity due to size selectivity. The cage is also able to selectively recognize the Cl- anion in water, even though the binding affinity is just moderate.

Abstract:Zr-based metal–organic frameworks (Zr-MOFs) have been widely used as ion adsorbents for the removal or extraction of toxic and/or radionuclide species from aqueous solutions. However, the mechanisms by which uranyl cations (UO 2 2+ ) interact with Zr-MOFs have not been established. In this work, the nature of the bonding of uranyl cations with a Zr-MOF was determined using density functional theory for nineteen structurally distinct candidate complexes. The results showed that in all cases the binding energy was of the order of 1.5 eV, but depended on the specific bonding site. The most stable structure involved coordination of the uranyl cation and two structurally distinct oxygens in the Zr-MOF metal node. It was also found that higher degree of deprotonation in Zr-MOF correlated with higher binding energy between the Zr-MOF and uranyl cations. These insights can aid in the design of Zr-MOFs with optimized features for efficient capture of uranyl cations.

Abstract:ZrSiO4-based ceramics have been considered as one of the candidate nuclear waste forms, while the immobilization behavior of mixed-valent uranium in the structure of the ceramics was unclear. Herein, ZrSiO4–U ceramics with a general formula of Zr1-4x(U6+xU5+2x)SiO4 were designed and synthesized. The evolutions of phase and microstructure depending on the content of U were investigated. The ceramics with 0 ≤ x < 0.04 showed a single ZrSiO4 phase, while U3O8 phase was detected when x ≥ 0.04. The results demonstrated that the solubility limit of U in ZrSiO4 ceramic was up to 1.51 at.% or 11.06 wt%. The calculated lattice parameters increased with the increase of U content. The increased parameters provided the evidence of the immobilization of both U5+ and U6+ into ZrSiO4 crystal lattice by replacing Zr4+. Furthermore, the obtained ZrSiO4–U ceramics exhibited good aqueous durability (∼10−5 g m−2 d−1). The results indicated that ZrSiO4-based ceramics can be employed to effectively immobilize mixed-valent U.

Abstract:The field of molecular cages has attracted increasing interest in relation to the development of biological applications, as evidenced by the remarkable examples published in recent years. Two key factors have contributed to this achievement: First, the remarkable and adjustable host–guest chemical properties of molecular cages make them highly suitable for biological applications. This allows encapsulating therapeutic molecules to improve their properties. Second, significant advances have been made in synthetic methods to create water-soluble molecular cages. Achieving the necessary water solubility is a significant challenge, which in most cases requires specific chemical groups to overcome the inherent hydrophobic nature of the molecular cages which feature the organic components of the cage. This can be achieved by either incorporating water-solubilizing groups with negative/positive charges, polyethylene glycol chains, etc.; or by introducing charges directly into the cage structure itself. These synthetic strategies allow preparing water-soluble molecular cages for diverse biological applications, including cages' anticancer activity, anticancer drug delivery, photodynamic therapy, and molecular recognition of biological molecules. In the review we describe selected examples that show the main concepts to achieve water solubility in molecular cages and some selected recent biological applications.

Theoretical Studies on the Complexation of Uranyl with Typical Carboxylate and Amidoximate Ligands

Abstract:Understanding of the bonding nature of uranyl and various ligands is the key for designing robust sequestering agents for uranium extraction from seawater. In this paper thermodynamic properties related to the complexation reaction of uranyl(VI) in aqueous solution (i.e. existing in the form of UO2(H2O)52+) by several typical ligands (L) including acetate (CH3CO2−), bicarbonate (HOCO2−), carbonate (CO32−), CH3(NH2)CNO− (acetamidoximate, AO−) and glutarimidedioximate (denoted as GDO2−) have been investigated by using relativistic density functional theory (DFT). The geometries, vibrational frequencies, natural net charges, and bond orders of the formed uranyl-L complexes in aqueous solution are studied. Based on the DFT analysis we show that the binding interaction between uranyl and amidoximate ligand is the strongest among the selected complexes. The thermodynamics of the complexation reaction are examined, and the calculated results show that complexation of uranyl with amidoximate ligands is most preferred thermodynamically. Besides, reaction paths of the substitution complexation of solvated uranyl by acetate and AO− have been studied, respectively. We have obtained two minima along the reaction path of solvated uranyl with acetate, the monodentate-acetate complex and the bidentate-acetate one, while only one minimum involving monodentate-AO complex has been located for AO− ligand. Comparing the energy barriers of the two reaction paths, we find that complexation of uranyl with AO− is more difficult in kinetics, though it is more preferable in thermodynamics. These results show that theoretical studies can help to select efficient ligands with fine-tuned thermodynamic and kinetic properties for binding uranyl in seawater.
