Abstract:Two-dimensional (2D) materials are of great significance to the materials community as a result of their high surface area and controllable surface properties. However, controlled preparation of biodegradable 2D structures with biological activity is difficult. In this work we demonstrate that by careful selection of building block structures and assembly conditions it is possible to use crystallization-driven self-assembly (CDSA) to assemble well-defined 2D nanostructures from poly(L-lactide) (PLLA)-based diblock glycopolymers. 1D glyco-cylinders and 2D diamond-shaped glyco-platelets are produced, where the underlying formation mechanism is revealed by dissipative particle dynamics simulations. Furthermore, we demonstrate that assembly of the polymers under mild degradation provides a straightforward route to hollow-cored platelets, a morphology that has previously proven laborious to access. The well-defined sizes and shapes of the glyco-platelets allow us to investigate macrophage activation efficiency and demonstrate clear size and shape effects, pointing toward potential applications in immunology.

Abstract:To date, nanotechnology has increasingly been identified as a promising and efficient means to address a number of challenges associated with public health. In the past decade, two-dimensional (2D) biomaterials, as a unique nanoplatform with planar topology, have attracted explosive interest in various fields such as biomedicine due to their unique morphology, physicochemical properties and biological effect. Motivated by the progress of graphene in biomedicine, dozens of types of ultrathin 2D biomaterials have found versatile bio-applications, including biosensing, biomedical imaging, delivery of therapeutic agents, cancer theranostics, tissue engineering, as well as others. The effective utilization of 2D biomaterials stems from the in-depth knowledge of structure-property-bioactivity-biosafety-application-performance relationships. A comprehensive summary of 2D biomaterials for biomedicine is still lacking. In this comprehensive review, we aim to concentrate on the state-of-the-art 2D biomaterials with a particular focus on their versatile biomedical applications. In particular, we discuss the design, fabrication and functionalization of 2D biomaterials used for diverse biomedical applications based on the up-to-date progress. Furthermore, the interactions between 2D biomaterials and biological systems on the spatial-temporal scale are highlighted, which will deepen the understanding of the underlying action mechanism of 2D biomaterials aiding their design with improved functionalities. Finally, taking the bench-to-bedside as a focus, we conclude this review by proposing the current crucial issues/challenges and presenting the future development directions to advance the clinical translation of these emerging 2D biomaterials.

Abstract:A large mumber of experimental results have demonstrated that self-assembly materials exhibit a variety of outstanding electronic, conductive, electroluminescent and photovoltaic properties. Based on such a fact that the self-assembly process, whatever it I s simple or complex, can easily occur, one has designed and synthesized a lot of functional materials including functionalized dye films, organic/ inorganic hybrid films, sensors, solar cells and optical communication elements, and fabricated the corresponding devices. It' s very important to form such a very stable, covalently bonded and structure-controlled material by molecular assembly in the bio-system. This has gradually become a hot-research field of non-biology. It's reasonable to believe that molecular assembly will become an important techndogy and will be put to use in the fields of materials and life sciences. In this review, the recent progress on the structures and applications of organic self-assembly materials, and self-assembly methodologies as well have been briefly introduced.

Abstract:In the past few decades, 2D layer materials have gradually become a central focus in materials science owing to their uniquely layered structural qualities and good optoelectronic properties. However, in the development of 2D materials, several disadvantages, such as limited types of materials and the inability to synthesize large-scale materials, severely confine their application. Therefore, further exploration of new materials and preparation methods is necessary to meet technological developmental needs. Organic molecular materials have the advantage of being customizable. Therefore, if organic molecular and 2D materials are combined, the resulting 2D organic materials would have excellent optical and electrical properties. In addition, through this combination, the free design and large-scale synthesis of 2D materials can be realized in principle. Furthermore, 2D organic materials exhibit excellent properties and unique functionalities along with great potential for developing sensors, biomedicine, and electronics. In this review, 2D organic materials are divided into five categories. The preparation methods and material properties of each class of materials are also described in detail. Notably, to comprehensively understand each material's advantages, the latest research applications for each material are presented in detail and summarized. Finally, the future development and application prospects of 2D organic materials are briefly discussed.

Abstract:Two-dimensional (2D) organic assemblies possess many intriguing properties such as planar structure, extended surface, flexibility and tailorability. The fabrication of 2D organic structures in a controlled manner is crucial to realize their potential functions and applications. In recent years, we have developed three methods to fabricate 2D organic assemblies through the rational design of building blocks. Firstly, hydrotropic anions will induce cationic bolaamphiphiles to form 2D assemblies, because such anions can insert their hydrophobic parts into the assemblies and weaken the electrostatic repulsion between adjacent headgroups. Secondly, the topology of supra-amphiphiles can be easily tuned because of the dynamic nature of non-covalent bonds, thus providing a facile approach for the fabrication of 2D assemblies. Thirdly, through molecular modification or introduction of new non-covalent bonds, we can lower the dimensionality of three-dimensional (3D) networks to form 2D assemblies. It is highly anticipated that these methods can be further applied for the preparation of functional 2D nano/micro materials for optoelectronics.

Abstract:Self-assembling biomolecular soft materials are a novel type of soft matter formed through the self-assembly process by using biomolecules or biomolecular building blocks. The characteristics of bio-sourced origin and assembly driven by weak interactions endow these materials with advantages such as high biocompatibility, reversible assembly, dynamic responsiveness, and controllable microstructures. These properties offer immense potential for development in fields such as biomedicine, tissue engineering, and flexible sensing. This paper concisely reviews the fundamental construction principles of self-assembling biomolecular soft materials and discusses three categories, i.e. nanomaterials, gel materials, and composite materials, by using amino acids and peptides as examples of assembly units. The specific self-assembly molecular mechanisms, material construction strategies, and functional application scenarios of these materials are elucidated. We anticipate that the research on self-assembling soft matter biomolecular materials will evolve from exploring structural units and measuring properties to customizing multifunctional properties and integrating advanced applications. This will lead to the development of novel composite intelligent biomolecular soft matter materials, and further promoting their applications in biomedicine, organic semiconductors, and soft robotics.

Abstract:The use of self-assembly for the construction of functional biomaterials is a highly promising and exciting area of research, with great potential for the treatment of injury or disease. By using multiple noncovalent interactions, coded into the molecular design of the constituent components, self-assembly allows for the construction of complex, adaptable, and highly tunable materials with potent biological effects. This review describes some of the seminal advances in the use of self-assembly to make novel systems for regenerative medicine and biology. Materials based on peptides, proteins, DNA, or hybrids thereof have found application in the treatment of a wide range of injuries and diseases, and this review outlines the design principles and practical applications of these systems. Most of the examples covered focus on the synthesis of hydrogels for the scaffolding or transplantation of cells, with an emphasis on the biological, mechanical, and structural properties of the resulting materials. In addition, we will discuss the distinct advantages conferred by self-assembly (compared with traditional covalent materials), and present some of the challenges and opportunities for the next generation of self-assembled biomaterials.

Abstract:Polymer self-assembly can prepare various shapes and sizes of pores, making it widely used. The complexity and diversity of biomolecules make them a unique class of building blocks for precise assembly. They are particularly suitable for the new generation of biomaterials integrated with life systems as they possess inherent characteristics such as accurate identification, self-organization, and adaptability. Therefore, many excellent methods developed have led to various practical results. At the same time, the development of advanced science and technology has also expanded the application scope of self-assembly of synthetic polymers. By utilizing this technology, materials with unique shapes and properties can be prepared and applied in the field of tissue engineering. Nanomaterials with transparent and conductive properties can be prepared and applied in fields such as electronic displays and smart glass. Multi-dimensional, controllable, and multi-level self-assembly between nanostructures has been achieved through quantitative control of polymer dosage and combination, chemical modification, and composite methods. Here, we list the classic applications of natural- and artificially synthesized polymer self-assembly in the fields of biomedicine and materials, introduce the cutting-edge technologies involved in these applications, and discuss in-depth the advantages, disadvantages, and future development directions of each type of polymer self-assembly.

Abstract:Noncovalent interactions provide a flexible method of engineering various chemical entities with tailored properties. Specific noncovalent interactions between functionalized small molecules, macromolecules or both of them bearing complementary binding sites can be used to engineer supramolecular complexes that display unique structure and properties of polymers, which can be defined as supramolecularly engineered polymers. Due to their dynamic tunable structures and interesting physical/chemical properties, supramolecularly engineered polymers have recently received more and more attention from both academia and industry. In this feature article, we summarize the recent progress in the self-assembly of supramolecularly engineered polymers as well as their biomedical applications. In view of different molecular building units, the supramolecularly engineered polymers can be classified into the following three major types: supramolecularly engineered polymers built by small molecules, supramolecularly engineered polymers built by small molecules and macromolecules, and supramolecularly engineered polymers built by macromolecules, which possess distinct morphologies, definite architectures and specific functions. Owing to the reversible nature of the noncovalent interactions, the supramolecularly engineered polymers have exhibited unique features or advantages in molecular self-assembly, for example, facile preparation and functionalization, controllable morphologies and structures, dynamic self-assembly processes, adjustable performance, and so on. Furthermore, the self-assembled supramolecular structures hold great potential as promising candidates in various biomedical fields, including bioimaging, drug delivery, gene transfection, protein delivery, regenerative medicine and tissue engineering. Such developments in the self-assembly of supramolecularly engineered polymers and their biomedical applications greatly promote the interdiscipline research among supramolecular chemistry, polymer materials, biomedicine, nano-science and technology.

Abstract:Inspired by biological systems, peptides can self‐assemble into various hierarchical nanostructures through non‐covalent interactions and form functional materials exhibiting excellent chemical and physical propertie, which have broad applications in bio‐/nano‐technologic fields. The self‐assembly mechanism, self‐assembly morphology of peptide supramolecula architecture and their various applications have been widely explored with the merit of biocompatible, easy preparation, and controllable functionality. Herein, we introduce the latest research progress of self‐assembling peptide‐based nanomaterials and review their applications in biomedicine and optoelectronics, including tissue engineering, anticancer therapy, biomimetic catalysis, energy harvesting, etc. We believe that this review will inspire the rational design and development of novel peptide‐based functional bio‐inspired materials in the future.

Abstract:Protein self-assembly is a fundamental biological process where proteins spontaneously organize into complex and functional structures without external direction. This process is crucial for the formation of various biological functionalities. However, when protein self-assembly fails, it can trigger the development of multiple disorders, thus making understanding this phenomenon extremely important. Up until recently, protein self-assembly has been solely linked either to biological function or malfunction; however, in the past decade or two it has also been found to hold promising potential as an alternative route for fabricating materials for biomedical applications. It is therefore necessary and timely to summarize the key aspects of protein self-assembly: how the protein structure and self-assembly conditions (chemical environments, kinetics, and the physicochemical characteristics of protein complexes) can be utilized to design biomaterials. This minireview focuses on the basic concepts of forming supramolecular structures, and the existing routes for modifications. We then compare the applicability of different approaches, including compartmentalization and self-assembly monitoring. Finally, based on the cutting-edge progress made during the last years, we summarize the current knowledge about tailoring a final function by introducing changes in self-assembly and link it to biomaterials' performance.

Abstract:Inspired by molecular self-assembly, which is ubiquitous in natural environments and biological systems, self-assembled peptides have become a research hotspot in the biomedical field due to their inherent biocompatibility and biodegradability, properties that are afforded by the amide linkages forming the peptide backbone. This review summarizes the biological advantages, principles, and design strategies of self-assembled polypeptide systems. We then focus on the latest advances in in situ self-assembly of polypeptides in medical applications, such as oncotherapy, materials science, regenerative medicine, and drug delivery, and then briefly discuss their potential challenges in clinical treatment.

Abstract:The rapid expansion of the study of chiral inorganic structures has led to the extension of the functional boundaries of inorganic materials. Nature-inspired self-assembled chiral inorganic structures exhibit diverse morphologies due to their high assembly efficiency and controlled assembly process, and they exhibit superior inherent properties such as mechanical properties, chiral optical activity, and chiral fluorescence. Although chiral self-assembled inorganic structures are becoming more mature in chiral catalysis and chiral optical regulation, biomedical research is still in its infancy. In this paper, various forms of chiral self-assembled inorganic structures are summarized, which provides a structural starting point for various applications of chiral self-assembly inorganic structures in biomedical fields. Based on the few existing research statuses and mechanism discussions on the chiral self-assembled materials-mediated regulation of cell behavior, molecular probes, and tumor therapy, this paper provides guidance for future chiral self-assembled structures to solve the same or similar medical problems. In the field of chiral photonics, chiral self-assembled structures exhibit a chirality-induced selection effect, while selectivity is exhibited by chiral isomers in the medical field. It is worth considering whether there is some correspondence or juxtaposition between these phenomena. Future chiral self-assembled structures in medicine will focus on the precise treatment of tumors, induction of soft and hard tissue regeneration, explanation of the biochemical mechanisms and processes of its medical effects, and improvement of related theories.

Abstract:Ultrathin, 2D organic layers of sub-ten nanometer thicknesses and high aspect ratios have received a great deal of attention for their graphene-like topological features and emerging properties. Rational synthetic strategies have led to the realization of periodic 2D layers with unprecedented structural precision. Herein, recent progress on the synthesis of 2D organic layers, including methods based on both non-covalent and covalent interactions, is summarized, and potential applications are highlighted. Such 2D organic nanostructures have a brilliant future as prospective multifunctional materials, showing great promise as platforms for engineering novel optoelectronic, interfacial, and bioactive properties.

Abstract:Since the discovery of graphene in 2004, two-dimensional (2D) nanostructures have been attracting tremendous interest for a variety of applications, including bio-inspired applications, due to their fascinating electronic, mechanical and optical properties. Especially, graphene and other 2D or quasi-2D nanostructures show excellent conductivity and flexibility. Herein, we highlight the recent impressive progress concerning the use of two representative types of 2D and quasi-2D nanostructures, graphenebased nanosheets and ultrathin polymeric nanosheets, as conductive or/and flexible elements to engineer three dimensional (3D) tissues. The results feature the unique potential of these 2D and quasi-2D nanostructures in the biomaterial tissue engineering research field.

Abstract:Biomass, a renewable hydrocarbon, is one of the favorable sources of advanced carbon materials owing to its abundant resources and diverse molecular structures. Biomass-based two-dimensional carbon nanomaterials (2D-BC) have attracted extensive attention due to their tunable structures and properties, and have been widely used in the design and fabrication of electrochemical sensing platforms. This review embarks on the thermal conversion process of biomass from different sources and the synthesis strategy of 2D-BC materials. The affinity between 2D-BC structure and properties is emphasized. The recent progress in 2D-BC-based electrochemical sensors for health and environmental monitoring is also presented. Finally, the challenges and future development directions related to such materials are proposed in order to promote their further application in the field of electrochemical sensing.

Abstract:Peptide materials, such as self-assembled peptide materials, are very important biomaterials. Driven by multiple interaction forces, peptide molecules can self-assemble into a variety of different macroscopic forms with different properties and functions. In recent years, the research on self-assembled peptides has made great progress from laboratory design to clinical application. This review focuses on the different morphologies, including nanoparticles, nanovesicles, nanotubes, nanofibers, and others, formed by self-assembled peptide. The mechanisms and applications of the morphology transformation are also discussed in this paper, and the future direction of self-assembled nanomaterials is envisioned.
