1024 ANGIOTENSION II TYPE 2 RECEPTOR-INTERACTING PROTEIN, ATIP, IN HEMATOPOIETIC CELLS IMPROVED GLUCOSE INTOLERANCE IN TYPE 2 DIABETIC MICE

Abstract:Objectives: We cloned AT2 receptor-interacting protein (ATIP) as a protein interacting specifically with the C-terminal tail of the AT2 receptor. The roles of ATIP in regulating glucose metabolism have never been examined. Here, we examined the possible roles of ATIP in hematopoietic cells in the pathogenesis diabetes mellitus, focusing on adipose tissues. Design and Methods: We employed wild-type (WT, C57BL/6J) mice, ATIP-transgenic (ATIP-Tg, based on C57BL/6J strain) mice, chimeric type 2 diabetic mice, KKAy, repopulated with bone marrow (BM) prepared from ATIP-Tg, WT or KKAy mice. Oral glucose tolerance test and F4/80 staining were performed, and morphological changes and cytokine levels of epididymal and retroperitoneal white adipose tissues were examined. Glucose uptake was evaluated by uptake of 2-[3H] deoxy-D-glucose (DG). Results: ATIP-Tg mice showed a lower body weight compared with WT mice. Moreover, adipose tissue weight was less and the size of adipocyte of ATIP-Tg was smaller than those of WT mice. Significant increase in 2-[3H]DG uptake was observed in white adipose tissue of ATIP-Tg mice. Chimeric KKAy with BM-ATIP-Tg demonstrated improved glucose intolerance compared with BM-WT and BM-KKAy. The size of adipocyte was smaller in BM-ATIP-Tg chimeric KKAy. 2-[3H]DG uptake was significantly increased in BM-ATIP-Tg KKAy. Number of macrophage and expression of inflammatory cytokine were less in the adipose tissue of BM-ATIP-Tg KKAy. Conclusions: ATIP in hematopoietic cells may have a crucial role in glucose metabolism at least in part via reducing invasion of inflammatory cells such as macrophages in adipose tissue.
