Abstract:Countercurrent chromatography is a powerful separation technique, which is used as an alternative or complementary technique to other chromatographic methods due to its high efficiency and preparative capacity. This technique does not require a solid stationary phase and relies simply on the partition of a sample between the two phases of an immiscible solvent system. Thus, it eliminates the complications resulting from the solid support matrix, such as irreversible adsorptive sample loss and deactivation, tailing of solute peaks, and contamination. With these advantages, CCC is gaining popularity as an important separation method. This review summarizes the basic principles and gives the most recent CCC applications in various areas, including the isolation and purification of natural products, development of different elution modes and multidimensional methods, progress in pH‐zone‐refining CCC techniques, bioseparation with aqueous polymer two‐phase systems, enantioseparation with various efficient chiral selectors, and on‐line monitoring of the eluate. Abstract:Counter-current chromatography (CCC) is a unique support-free liquid-liquid partition chromatography winning wide applications in the separation of various components from natural or synthetic mixtures. It has been one of the prime methods for isolating compounds from Traditional Chinese Medicines (TCM) and other comprehensive natural products. Although early CCC models produced a long-standing false image that CCC is a time-consuming technique, rapid and high-performance CCC devices and methods for high throughput analysis of natural mixtures have been advanced. For instances, multi-channel CCC, dual CCC, elution-extrusion CCC, and solvent simplification protocols can provide high throughput CCC analysis and produce high purity of compounds or large natural product libraries for drug discovery. This review summarizes the recent advancements of CCC in the high throughput analysis of natural product with an emphasis on the developments of instruments and methods.

Abstract:Due to many merits of the HSCCC,in the past few decades,HSCCC have dramatic development both in the theory and application.The most critical steps for the separation of HSCCC are the screening of the solvent system and the application of the elution mode.In this paper,we review the requirements of HSCCC,the screening method and the principle,the scope of application of gradient elution,dual-mode elution,recycling elution,extrusion elution in detail.Providing valued information for HSCCC researchers is the aim of this review.
A Novel 9×9 Map-Based Solvent Selection Strategy for Targeted Counter-Current Chromatography Isolation of Natural Products

Abstract:Counter-current chromatography (CCC) is an efficient liquid-liquid chromatography technique for separation and purification of complex mixtures like natural products extracts and synthetic chemicals. However, CCC is still a challenging process requiring some special technical knowledge especially in the selection of appropriated solvent systems. In this work, we introduced a new 9 × 9 map-based solvent selection strategy for CCC isolation of targets, which permit more than 60 hexane-ethyl acetate-methanol-water (HEMWat) solvent systems as the start candidates for the selection of solvent systems. Among these solvent systems, there are clear linear correlations between partition coefficient (K) and the system numbers. Thus, an appropriate CCC solvent system (i.e., sweet spot for K = 1) may be hit by measurement of k values of the target only in two random solvent systems. Besides this, surprisingly, we found that through two sweet spots, we could get a line ("Sweet line") where there are infinite sweet solvent systems being suitable for CCC separation. In these sweet solvent systems, the target has the same partition coefficient (K) but different solubilities. Thus, the better sweet solvent system with higher sample solubility can be obtained for high capacity CCC preparation. Furthermore, we found that there is a zone ("Sweet zone") where all solvent systems have their own sweet partition coefficients values for the target in range of 0.4 < K< 2.5 or extended range of 0.25 < K < 16. All results were validated by using 14 pure GUESSmix mimic natural products as standards and further confirmed by isolation of several targets including honokiol and magnolol from the extracts of Magnolia officinalis Rehd. Et Wils and tanshinone IIA from Salvia miltiorrhiza Bunge. In practice, it is much easier to get a suitable solvent system only by making a simple screening two to four HEMWat two-phase solvent systems to obtain the sweet line or sweet zone without special knowledge or comprehensive standards as references. This is an important advancement for solvent system selection and also will be very useful for isolation of current natural products including Traditional Chinese Medicines.

Salting-in Counter-Current Chromatography Separation of Tanshinones Based on Room Temperature Ionic Liquids

Abstract:Counter-current chromatography (CCC) is a solid support-free liquid-liquid partition chromatography and has wide applications. However, CCC separation is still a challenging process and the selection of appropriate solvent system for separation of target compound(s) is still relatively time-consuming. In this work, we introduced a room temperature ionic liquids-based salting-in strategy for the rapid selection of suitable solvent systems for CCC separation. In the randomly selected solvent systems, such as ethyl acetate-water, n-butanol-water, n-pentanol-water, n-hexanol-water, and n-octanol-water, several ionic liquids such as [AMIM]Cl, [MAMIM]Cl, and [BMIM]Cl can increase the solubility of the solutes in the lower phase, which made a dose-dependent decreasing of partition coefficient of solute in the two-phase solvent system. Thus, it is possible to get a suitable solvent system with sweet K spot such as K=1 only by adding some ionic liquids into the systems. As an example, arctiin, a bioactive lignin component of the fruit of Arctium lappa. L. (Niubangzi in Chinese), was selected and successfully separated by CCC with room temperature ionic liquids-based n-butanol-water systems. It seems a very efficient alternative strategy for the optimization of solvent systems for CCC separation of natural products.
A Computational Fluid Dynamics Study of Binary Adsorption Separation in Chromatography

Abstract:FEMLAB software was applied to incorporate computational fluid dynamics (CFD) into the simulation of the adsorption-separation process in chromatography. Simulations of both linear and nonlinear processes demonstrated that significant dispersion retarded the separation processes, and that a small inlet induced curved solute concentration profiles, resulting in peak tailing of the elution chromatogram. The linear simulation visually confirmed that a small Stanton number might be another factor for peak tailing. For cases with a reasonably small Stanton number, peak tailing became more serious with decreasing Stanton number. Comparisons between the results of the CFD simulation of the nonlinear case, the corresponding conventional 1-D simulation and the experimental result lead to the conclusion that the CFD simulation is useful for a better understanding of the dynamic adsorption and separation process in a chromatographic column.

Abstract:Countercurrent chromatography (or counter-current chromatography, CCC) is a unique support-free liquid-liquid partition chromatography. Since it was invented by Y. Ito in 1960s, CCC has been widely accepted and applied as popular separation and purification technique for natural and synthetic complex. However, up to date there is little study to address on hydrophobic and hydrophilic interactions in CCC process, although hydrophobic interaction chromatography (HIC) and hydrophilic interaction chromatography (or hydrophilic interaction liquid chromatography, HILIC) as solid-support chromatographic techniques are widely applied at different stages of downstream processing. In fact, hydrophobic and hydrophilic interactions might be more popular in CCC separation than that in the liquid chromatography. For example, adding small solvents or additives in two-phase solvent systems may change significantly hydrophobic or hydrophilic interactions between solvents and solutes. Normally, CCC separation employs a light and hydrophobic organic phase as the stationary phase, and a heavy and hydrophilic aqueous phase as the mobile phase. Hydrophobic interactions between the solvent system and solutes (targets) will increase the partition coefficients (K values) of solutes and lengthen retention time, while hydrophilic interactions will reduce the K values and separation time. In this work, we aim to provide a general insight on the hydrophobic and hydrophilic interactions in CCC separation. We also highlight the current advances in utilizing the hydrophobic and hydrophilic interactions for K-targeted CCC separation and purification.

Abstract:In counter-current chromatography, the separation efficiency greatly depends on the partitioning ability of the separated substance between the stationary phase and the mobile phase. Partitioning ability is mainly represented by the parameter partition coefficient which is one of the important parameters to evaluate the separation effect of counter-current chromatography. The scope of the partition coefficient value mainly depends on the solvent system. A suitable solvent system election is, therefore, a critical role in the separation of counter-current chromatography. The existing solvent systems that are widely used are mainly two-phase solvent systems. It is difficult to decide on an appropriate solvent system for separation of compounds with wide polarity range, which promotes the development of the three-phase solvent system in counter-current chromatography. This review mainly described the origin, development history of three-phase solvent system, summarized the volume ratios and volume fractions of the upper, middle and lower phases of nearly 50 three-phase solvent systems, its advantages and disadvantages in counter-current chromatography. In addition, the challenges and future perspectives on three-phase solvent systems in counter-current chromatography also are discussed in this review.This article is protected by copyright. The development of high-speed counter-current chromatography, dual counter-current chromatography, cross-axis coil planet centrifuge, pH-zone-refining counter-current chromatography and spiral disk assembly counter-current chromatography were reviewed, and the latest progresses on high-speed counter-current chromatography coupled with mass spectrometry, evaporative light scattering detection, high-performance liquid chromatography and the two-dimensional counter-current chromatography were introduced.

Abstract:An on-line column-switching counter-current chromatography (CCC) with solid-phase trapping interphase is presented in this paper. The large volume injection is avoided using solid-phase trapping interphase. Thereby, totally different chemical composition biphasic solvent system can be utilized to enhance system orthogonality. In the present work, a 140mL-capacity CCC instrument was used in the first dimension, and a parallel three-coil CCC centrifuge (40mL each coil) was used in the second dimensional separation allowing three injections at the same time. With biphasic solvent system composed of n-hexane: ethyl acetate: methanol: water (1:1:1:1, v/v), five well-separated fractions were obtained in the first dimension. Two fractions were online concentrated and further separated in the second dimension with solvent system composed of methyl tert-butyl ether: acetonitrile: water (2:2:3, v/v), where trifluoroacetic acid (10mM) was added to the upper organic phase as a retainer and triethylamine (10mM) to the aqueous mobile phase as an eluter. Four hydroxyanthraquinones were successfully separated and purified from Chinese medicinal plant Rheum officinale in only one step.

Abstract:Objective: To introduce a new way, which facilitates the systematic search and evaluation of the solvent systems suitable for high-speed countercurrent chromatography (HSCCC). Methods: The software integrated with the thermodynamics solution functions and theory is directive to calculate the composition, physical parameters, such as volume, viscosity, dielectric constants, and polarity parameters of the upper and lower phases. Results: Separation of Guizhi-decoction Fr. A by HSCCC, one of traditional Chinese medcines, is conducted to prove the applicability of this software in selection and optimization of solvent systems. Conclusion: This software can be used to select and optimize the solvent systems of HSCCC. It is supposed to extend this method.

Abstract:Counter-current chromatography (CCC) is a unique, liquid-liquid partition chromatography process. Both the mobile and stationary phases are liquids, so no solid support matrix is used. CCC has gained wide acceptance as a preparative technique in a variety of fields. Because the mobile and stationary phases are both liquids, gradient elution is difficult to perform with CCC. Phase equilibrium must be maintained, so any change in the composition of one phase may induce a compositional change in the other. In this work, a new linear gradient elution method was developed for CCC. Biphasic solvent systems containing heptane, ethyl acetate, methanol, and water (HepEMWat) in various ratios were prepared and used to optimize both isocratic and linear gradient CCC separation with methanol. We first separated a test mixture of four standard compounds with partition coefficients ranging from 0.8 to 7.8. The separation resembled a reversed-phase process, and elution was performed while progressively decreasing the polarity of the mobile phase. Target molecules with small partition coefficients eluted first in the lower phase of the optimized HepEMWat solvent system. Elution of constituents with large partition coefficients was quite slow under isocratic conditions. Separation time was significantly reduced when elution was performed with a linear gradient using methanol and the optimal HepEMWat system. Elution with a 3:7:4:6 (v/v/v/v) HepEMWat system took approximately 200 min. This included an 80-min isocratic step, followed by gradient elution with methanol from 0% to 30%. The optimized methanol linear gradient CCC method was then used to separate a complex mixture of natural products isolated from Sinopodophyllum hexandrum (Royle) Ying roots. Twelve compounds with a wide range of polarities were well-resolved in a single separation. We have developed a convenient and cost-effective strategy for the separation of complex mixtures. No tedious mobile phase preparation step is required. The volume of unused mobile phase is minimal, so little solvent is wasted. The method is an important advance for the separation of mixtures that contain many compounds with a large range of polarities and partition coefficients, which are common features of natural products. Abstract:Solvent system selection is the first step toward a successful counter-current chromatography (CCC) separation. This paper introduces a systematic and practical solvent system selection strategy based on the nonrandom two-liquid segment activity coefficient (NRTL-SAC) model, which is efficient in predicting the solute partition coefficient. Firstly, the application of the NRTL-SAC method was extended to the ethyl acetate/n-butanol/water and chloroform/methanol/water solvent system families. Moreover, the versatility and predictive capability of the NRTL-SAC method were investigated. The results indicate that the solute molecular parameters identified from hexane/ethyl acetate/methanol/water solvent system family are capable of predicting a large number of partition coefficients in several other different solvent system families. The NRTL-SAC strategy was further validated by successfully separating five components from Salvia plebeian R.Br. We therefore propose that NRTL-SAC is a promising high throughput method for rapid solvent system selection and highly adaptable to screen suitable solvent system for real-life CCC separation.

Abstract:Counter-current chromatography (CCC), a unique continuous liquid-liquid partition chromatographic technique, has been widely used in the separation and purification of natural products. This technique has made great progress in recent years. In this paper, the different possible separation methods developed in CCC are described. It includes classical CCC, pH-zone-refining, pH-gradient, ion-pairing, multi-channel and two-dimensional CCC, dual-flow, liquid-liquid-liquid three-phase systems, elution-extrusion, gradient elution, powder direct injection, with some words for the various techniques used in pre- and post-CCC procedures, and detection modes. The basic mechanisms of each method are presented and applications are summarized. The future of CCC developments, including its application in protein purification and enrichment, and new platforms, are also discussed. This review paper aims to aid scientists who are interested in the field of CCC research for natural product isolation.
Multi-channel counter-current chromatography for high-throughput fractionation of natural products for drug discovery.

Abstract:A multi-channel counter-current chromatography (CCC) method has been designed and fabricated for the high-throughput fractionation of natural products without complications sometimes encountered with other conventional chromatographic systems, such as irreversible adsorptive constituent losses and deactivation, tailing of solute peaks and contamination. It has multiple independent CCC channels and each channel connects independent separation column(s) by parallel flow tubes, and thus the multi-channel CCC apparatus can achieve simultaneously two or more independent chromatographic processes. Furthermore, a high-throughput CCC fractionation method for natural products has been developed by a combination of a new three-channel CCC apparatus and conventional parallel chromatographic devices including pumps, sample injectors, effluent detectors and collectors, and its performance has been displayed on the fractionation of ethyl acetate extracts of three natural materials Solidago canadensis, Suillus placidus, and Trichosanthes kirilowii, which are found to be potent cytotoxic to tumor cell lines in the course of screening the antitumor candidates. By combination of biological screening programs and preparative high-performance liquid chromatography (HPLC) purification, 22.8mg 6β-angeloyloxykolavenic acid and 29.4mg 6β-tigloyloxykolavenic acid for S. canadensis, 25.3mg suillin for S. placidus, and 6.8mg 23,24-dihydrocucurbitacin B for T. Kirilowii as their major cytotoxic principles were isolated from each 1000mg crude ethyl acetate exctract. Their chemical structures were characterized by electrospray ionization mass spectrometry, one- and two-dimensional nuclear magnetic resonance. The overall results indicate the multi-channel CCC is very useful for high-throughput fractionation of natural products for drug discovery in spite of the solvent balancing requirement and the lower resolution of the shorter CCC columns.

Abstract:Countercurrent chromatography (CCC) is a support-free liquidliquid chromatography using centrifugal fields to hold the liquid stationary phase. CCC has been widely applied in the separation of various natural and synthetic components using a variety of biphasic liquid systems. The related hexane or heptane/ethyl acetate/methanol or ethanol/water biphasic liquid systems demonstrated their significance in CCC. Gradient is difficult in CCC since any composition change in one phase induces a composition change of the other phase to maintain phase equilibrium. This work provides a new insight into linear gradient elution in CCC that is feasible with some biphasic liquid systems such as selected compositions of the hexane/ethyl acetate/ethanol/water systems. The equations modeling solute motion inside the CCC column are proposed. Particular compositions of the liquid system, namely the hexane/ethyl acetate/ethanol/water 8:2:E:W compositions with E + W = 10, were studied from W = 1 to 9. They showed moderate changes in the upper organic phase compositions. The model is tested with the separation of tanshinones from the rhizome of Salvia miltiorrhiza Bunge. Different linear solvent gradient profiles were experimentally performed between 8:2:5:5 and 8:2:3:7 compositions and the results were evaluated using the proposed model. Five tanshinones including dihydrotanshinone I, cryptotanshinone, tanshinone I, 1,2-dihydrotanshinquinone, and tanshinone IIA have been successfully separated (>95% purities) using a gradient profile optimized by the developed model. The gradient model can be used only with biphasic liquid systems in which one phase shows minimum composition changes when the other phase composition changes notably. This case is not the general case for biphasic liquid systems but can be applied with specific compositions of the quaternary hexane or heptane/ethyl acetate/methanol or ethanol/water most useful CCC liquid systems.

Abstract:A new series of organic-high ionic strength aqueous two-phase solvents systems was designed for separation of highly polar compounds by spiral high-speed counter-current chromatography. A total of 21 solvent systems composed of 1-butanol-ethanol-saturated ammonium sulfate-water at various volume ratios are arranged according to an increasing order of polarity. Selection of the two-phase solvent system for a single compound or a multiple sample mixture can be achieved by two steps of partition coefficient measurements using a graphic method. The capability of the method is demonstrated by optimization of partition coefficient for seven highly polar samples including tartrazine (K=0.77), tryptophan (K=1.00), methyl green (K= 0.93), tyrosine (0.81), metanephrine (K=0.89), tyramine (K=0.98), and normetanephrine (K=0.96). Three sulfonic acid components in D&C Green No. 8 were successfully separated by HSCCC using the graphic selection of the two-phase solvent system.
Temperature-Driven Separation on Countercurrent Chromatography

Abstract:The selection of a biphasic solvent system is the first and most important step for a successful countercurrent chromatography (CCC) separation. The partitioning coefficient of targets and their related impurities on one side, and stationary phase retention (Sf.%) on another, are among the parameters used for solvent system selection. At constant temperature, the Sf.% of a selected solvent system in any CCC instrument is dominated by the rule of Du’s plot, which describes the correlation between Sf.% and flow rate. In a biphasic liquid system, physico-chemical properties such as density, viscosity, and interfacial tension between two phases are temperature dependent. In this article, the effect of temperature on the retention of several typical solvent systems were investigated using an high speed CCC instrument. The results showed that temperature changes could induce density, viscosity, and interfacial tension variation of the biphasic system with consequent changes in the system’s hydrodynamic behavior. As the temperature increased, the density and viscosity of the selected solvent system decreased. This led to a variation in the interfacial tension between two phases depending on the solvent system composition. The Sf.% increased when the temperature increased for the tested solvent systems. Based on the obtained results, an improved Du’s plot was suggested, accounting for the contribution of operating temperature. The prospect of this article provides hands-on strategies for the development of high-performance separation method for HSCCC.
