Abstract:A mild, efficient, and environmentally benign one-pot synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones by a three-component domino reaction of 4-aminocoumarins, arylglyoxal monohydrates, and 1,3-dicarbonyl compounds in refluxing ethanol without the use of catalyst is reported. Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. The attractive features of this protocol are operational simplicity, short reaction time, easy purification of products and good to excellent yields.

Abstract:An efficient tandem process was developed to synthesize diversified benzo[a]xanthones from 2-methyl-3-(1-alkynyl)chromones with electron-deficient chromone-fused dienes. This unusual reaction, involving multiple steps and not requiring the use of transition metal catalysts or an inert atmosphere, results in the formation of three new C-C bonds and one C-O bond.

Abstract:Utilizing simple precursors for the exquisite construction of molecules with diversity and complexity is urgent. Herein, a novel A3-X type reaction for functionalized 3-haloquinolines has been developed by PhI (OAc)2-mediated four-component tandem addition/cyclization/oxidation/aromatization process with simple aromatic amine, aldehyde, alkyne and halide salt. Broad functional group tolerance, very mild condition, easy for further transformation illustrate the concision, high efficiency and practicability of this A3-X type reaction.

Abstract:A one-pot synthesis of substituted (dihydro)thieno[2,3-c]chromene derivatives was developed through a Lewis base catalyzed domino Michael addition/intramolecular cyclization reaction between 3-nitro-2H -chromenes and thioamides. The desired (dihydro)thieno[2,3-c]chromene derivatives were prepared with a large substrate scope (22 examples) and excellent diastereoselectivity in moderate to excellent yields.

Abstract:An In(III)‐catalyzed [2+3+1] annulation strategy is developed for the diastereoselective construction of diversely functionalized tetrahydroxanthones bearing chromone rings. This protocol is based on a three‐component reaction involving various 3‐formylchromones and α‐methylstyrenes via a carbonyl‐ene and formal [4+2] cycloaddition reaction. Notable features of this reaction include its wide substrate scope, high tolerance towards functional groups, atom economy, and excellent diastereoselectivity. In addition, the synthesized tetrahydroxanthones are readily transformed into 2‐hydroxybenzophenone products.

Abstract:Accomplished herein is a rearrangement strategy for the highly efficient assembly of synthetically cumbersome while medicinally significant 2-amino-3-formyl chromones via palladium-catalyzed ring-opening, rearrangement, and cyclization process. Such a sequence enables the formation of one C(sp2)–O bond and one C(sp2)–C(sp2) bond, and reconstruction of the benzo-γ-pyrone moiety in a single operation, thus producing the difunctionalized chromone-incorporated derivatives. The reaction proceeds in a shorter reaction time (30 min for 3-iodochromones in most cases) in a highly atom- and step-economical manner. The synthetic application of the current protocol is further demonstrated by late-stage modification of pharmaceuticals and their intermediates, gram-scale reactions, transformations of functional groups, as well as the synthesis of bioactive molecules and drugs. Mechanistic studies indicate that a nucleophilic ring-opening process of the benzo-γ-pyrone moiety, phenoxy anion intermediate, and an intramolecular palladium-catalyzed cyclization process might be involved in the present reaction system.

Under this strategy, a wide scope of spiro[indoline‐3,4′‐pyrano[3,2‐b]chromene] derivatives, which combined chromone, pyran, indolone in one molecule, could be obtained in moderate to excellent yields (up to 94 %) with moderate enantioselectivities (up to 95 % ee). In addition, scaled‐up experiment also confirmed the synthetic practicality of this synthetic strategy.
