Abstract:A catalytic annulation cascade is achieved that affords an efficient access to phthalides from readily available benzoic acids and saturated ketones. The reaction is catalyzed by a bimetallic Ir/Cu system and proceeds via the combination of Cu-catalyzed dehydrogenation of ketones and Ir-catalyzed direct C-H functionalization under the assistance of the weakly coordinating carboxyl followed by the beta-H elimination and intramolecular Michael addition. This method shows a broad scope of substrates and good functional group tolerance. The protocol furnishes an alternative strategy for the construction of diverse 3-substituted phthalides.

Abstract:An unprecedented Rh(III)-catalyzed C–H activation/cycloaddition of benzamides and methylenecyclopropanes for the selective synthesis of spiro dihydroisoquinolinones and furan-fused azepinones is reported. The process features simple starting materials, mild conditions, and high efficiency and is external oxidant free. The products could be easily converted to other biologically interesting heterocycles. Control experiments and kinetic isotope effect studies were conducted and a plausible mechanism is proposed.

Abstract:A facile rhodium-catalyzed CH activation/intramolecular annulation strategy for N-methoxydihydropyrimidin-2-one was well realized in water media. Moreover, this Rh(III)-catalytic system facilitated the construction of N-methoxydihydropyrimidin-2-one scaffolds reminiscent of unnatural amino alcohols, transformation to pyran-2-one fused heterocycles as well as late-stage modification of drug molecules. Also, the protocol was easily scaled up to a gram-scale level. And H2O was proposed to act as an alternative coordination molecule to accelerate the generation of active Rh(III) catalyst species.

Abstract:We report a novel rhodium-catalyzed rearrangement involving N-substituted 2-thiopyridones and diazoesters. Abstract:Transition-metal-catalyzed C-H activation synthesis of heterocyclic spiro[4,4]nonanes has persistently witnessed the use of additional stoichiometric transition-metal oxidant when employing C=C bond as the Spiro ring closure site. Herein, we have addressed the issue by reporting a redox-neutral strategy for spiro[indene-1,4'-oxa-zolidinones] synthesis via Rh(III)-catalyzed coupling of 4-phenyl-1,3-oxazol-2(3H)-ones with alkynes. The synthesis features a broad substrate scope and high regiospecificity.

Abstract:Comprehensive SummaryA rhodium‐catalyzed denitrogenative carbonylation of 1,2,3‐thiadiazole with alkynes to construct (benzo)thiochromenones is developed. Both terminal and internal alkynes were compatible in this [3+2+1] cycloaddition, affording a broad range of poly‐substituted thiochromenones. Unsymmetrical alkyne substrates exhibited good regioselectivity. It is noteworthy that the present strategy could be used to construct both monocyclic and benzo‐thiochromenones, and it is a novel example of denitrogenative transannulation with CO insertion.

Abstract:Herein, we developed a ligand-promoted Rh(III)-catalyzed C(sp3)-H thiolation of 8-methylquinolines. The effect of ligands on improving the activity of the catalytic centers has been studied in detail and proven to be significant. Various substituents are well tolerated under this reaction condition to provide potential precursors for organic synthesis. The mechanistic study suggests that the reaction may proceed through a five-membered rhodacycle intermediate via thiolation twice.

Abstract:A facile access to highly fused tetracyclic indeno-1,2-benzothiazines has been established via a Rh(III)-catalyzed C-H bond activation and intramolecular annulation cascade between sulfoximides and all-carbon diazo indandiones. This strategy is characterized by the fact that the diazo coupling partners do not require preactivation, along with its high efficiency, broad substrate generality, and facile transformation. Particularly, the highly conjugated tetracyclic products demonstrate good optical properties and can easily enter cells to emit bright fluorescence for live cell imaging.

Abstract:We report herein a chiral Cp x Rh III -catalyzed asymmetric sp 2 C–H bond activation/annulation reaction of free NH-sulfoximines with hypervalent iodonium ylides under external ligand-free conditions. Efficient kinetic resolution (KR) was observed for the reactions of racemic sulfoximines bearing an asymmetric S -aryl- S -alkyl moiety, which also delivered the expected fused tricyclic ( S )-aza-heterocycles and the corresponding chiral ( R )-NH-sulfoximines in mostly good yields and with excellent enantioselectivities. Using this catalytic protocol, structurally unprecedented 10-membered S-stereogenic benzothiazecines with good enantioselectivities could be achieved upon one-pot multi-step oxidation reactions. Mechanistically, an enantiodetermined C–H cleavage and the subsequent formation of a chiral five-membered rhodacycle intermediate were supposed through systematic deuterium (D) labelling studies.

Abstract:In this paper, Rh2(OAc)4-catalyzed [2,3]-sigmatropic rearrangement of sulfur ylides derived from N-tosylhydrazones and sulfides is reported. A series of tosylhydrazones derived from aldehydes were successfully used for [2,3]-sigmatropic rearrangement by reaction with either allylic phenyl sulfides or propargyl phenyl sulfides. The reaction conditions were optimized and afforded the products in moderate to good yields. In addition, a novel and convenient approach for the synthesis of cyclobutenones and cyclopropanes has been developed through direct oxidation of the rearrangement products.

Abstract:A rhodium(III)-catalyzed cross-coupling of benzyl thioethers and aryl carboxylic acids through the two directing groups is reported. Useful structures with diverse substituents were efficiently synthesized in one step with the cleavage of four bonds (CH, CS, OH) and the formation of two bonds (CC, CO). The formed structure is the privileged core in natural products and bioactive molecules. This work highlights the power of using two different directing groups to enhance the selectivity of a double CH activation, the first of such examples in cross-oxidative coupling.

Abstract:A copper-mediated CS/NS bondforming reaction via CH activation that uses elemental sulfur has been developed. The addition of TBAI was found to be crucial for the success of this transformation. The method is scalable, shows excellent functional group tolerance, and is compatible with heterocycle substrates, providing efficient and practical access to benzoisothiazolones. The direct diversification of the benzoisothiazolone products into a variety of sulfur-containing compounds is also demonstrated.

Abstract:We report herein a new strategy of the Rh(III)-catalyzed C-H activation/cyclization of indoles and pyrroles, for the divergent synthesis of privileged heterocycles. A simple derivation of indoles and pyrroles to N-carboxamides with oxidative bidentate directing group could enable rhodacycle formation and late-stage redox-neutral cyclization with alkynes, alkenes and diazo compounds, for access to five- and six-membered fused heterocycles, such as pyrimido[1,6-a]indol-1(2H)-one, 3,4-dihydropyrimido[1,6-a]indol-1(2H)-one, and 1H-imidazo[1,5-a]indol-3(2H)-ones. Kinetic isotope effect study was conducted, and a plausible mechanism was proposed. Furthermore, this protocol was applied to concise synthesis of 5-HT3 receptor antagonist in gram-scale.

Abstract:The development of an efficient strategy for facile access to quinoline-based bis-heterocycles holds paramount importance in medicinal chemistry. Here in, we describe a unified approach for accessing 8-(indol-3-yl)methyl-quinolines by integrating Cp*Rh(III)-catalyzed C(sp3)–H bond activation of 8-methylquinolines followed by nucleophilic cyclization with o-ethynylaniline derivatives. Remarkably, methoxybiaryl ynones under similar catalytic conditions delivered quinoline tethered spiro[5.5]enone scaffolds via a dearomative 6-endo-dig C-cyclization. Moreover, leveraging this method for C8(sp2)−H bond activation of quinoline-N-oxide furnished biologically relevant oxindolyl-quinolines. This reaction proceeds via C(sp2)–H bond activation, regioselective alkyne insertion, oxygen-atom-transfer (OAT) and intramolecular nucleophilic cyclization in a cascade manner. One C−C, one C−N and one C=O bonds were created with concomitant formation of a quaternary center.
