Abstract:Fluorescence immunoassay technology is widely recognized as the most sensitive detection technology, now widely used in clinical testing. The research of quantitative analyzer for fluorescence immune chromatography strips is of great significance. In this thesis, a quantitative analyzer for fluorescence immune chromatography strips has realized, based on photoelectric detection principle and immunochromatographic. The system uses LPC1788 and MSP430 as processors. Using some algorithms such as low-pass filtering, least square method and area method to analyze the detected waveform and eliminate noise with high accuracy. In our experiment, the minimum detection concentration is at several mIU/ml levels, the relative standard deviation is less than 2% and it has good linear response characteristics in a concentration range from 0.5125 to 952mIU/ml while the correlation coefficient R2 is 0.981. The instrument has a good prospect.

Abstract:Hook effect is now widely present in sandwich lateral flow assay reactions, which refers to the phenomenon of false-negative results due to inappropriate antigen-antibody ratios, and can greatly limit the true color development of test strip strips under high sample concentration conditions, thus limiting quantitative detection. In this study, we developed a novel deep learning-based discrimination algorithm to accurately distinguish whether the strips are affected by the hook effect, which not only saves cost and manpower, but also clears the way for subsequent immunochromatographic quantification.

Chemometrics, which uses mathematical tools to deal with the overlapping analytical signals, provides alternative approaches for the resolution of overlapping chromatographic signals. Abstract:Enhancing the sensitivity of immunoassays is an important requirement in the field of immunology, especially in light of rapid developments in genetic testing, making the detection of low-abundance protein biomarkers crucial. Therefore, innovations in highly sensitive immunoassays are imperative. This demand has led to the emergence of single-molecule immunoassays (SMIs), driving advancements in early diagnostic techniques, and ushering in a new era of immunoassays. This review begins by tracing the development of immunoassays and offers a detailed discussion of SMI technology across two distinct pathways: non-chip (SMI without microfluidic chips) and on-chip (SMI with microfluidic chips). Furthermore, we evaluated and compared these methods using two pathways. In addition, this review discusses the significance of SMI techniques in the diagnosis of various diseases and their current applications in laboratory and clinical settings. The progress of SMI in commercial applications and suggestions for innovative directions are also summarized. Despite the considerable potential of SMI, these technologies face challenges in practical application, particularly in developing countries and economically disadvantaged regions. The final section of this review addresses the challenges and prospects of these technologies.

Abstract:Cells are the fundamental unit of life. Owing to cellular heterogeneity, traditional biochemical assays which analyze cells in bulk often overlook the rich information available when single cells are studied. Accurate and reliable determination of the chemical composition in individual cells would greatly improve the probability of discriminating infected cells from healthy ones and provide a solid foundation on study and development in various fields including biochemistry, medicine, and pathology clinic. Microfluidic chip system for single-cell analysis is now attracting broad interests. The ability of integrating the whole process on one microfluidic chip, including single cell injection, lysis, separation and detection of cellular constituents by chip-based CE, offers a new technical platform for single cell analysis. This article provides a review on recent advances in determining intracellular contents in individual cells using microfluidic devices. Focus areas include on-chip single cell manipulation by means of electroosmotic flow (EOF), pressure-driven flow combined with EOF and optical tweezer (OT) to perform sequentially single cell injection, lysis, separation and detection of cellular constituents by chip-based CE with highly sensitive detection. The approaches on on-chip derivatization are also addressed.

Abstract:In recent years, point-of-care testing has played an important role in immunoassay, biochemical analysis, and molecular diagnosis, especially in low-resource settings. Among various point-of-care-testing platforms, microfluidic chips have many outstanding advantages. Microfluidic chip applies the technology of miniaturizing conventional laboratory which enables the whole biochemical process including reagent loading, reaction, separation, and detection on the microchip. As a result, microfluidic platform has become a hotspot of research in the fields of food safety, health care, and environmental monitoring in the past few decades. Here, the state-of-the-art application of microfluidics in immunoassay in the past decade will be reviewed. According to different driving forces of fluid, microfluidic platform is divided into two parts: passive manipulation and active manipulation. In passive manipulation, we focus on the capillary-driven microfluidics, while in active manipulation, we introduce pressure microfluidics, centrifugal microfluidics, electric microfluidics, optofluidics, magnetic microfluidics, and digital microfluidics. Additionally, within the introduction of each platform, innovation of the methods used and their corresponding performance improvement will be discussed. Ultimately, the shortcomings of different platforms and approaches for improvement will be proposed.

Abstract:Technologies based on digital microfluidics (DMF) have made significant advancements in the automated manipulation of microscale liquids and complex multistep processes. Due to their numerous benefits, such as automation, speed, cost-effectiveness, and minimal sample volume requirements, these systems are particularly well suited for immunoassays. In this review, an overview is provided of diverse DMF manipulation platforms and their applications in immunological analysis. Initially, droplet-driven DMF platforms based on electrowetting on dielectric (EWOD), magnetic manipulation, surface acoustic wave (SAW), and other related technologies are briefly introduced. The preparation of DMF is then described, including material selection, fabrication techniques and droplet generation. Subsequently, a comprehensive account of advancements in the integration of DMF with various immunoassay techniques is offered, encompassing colorimetric, direct chemiluminescence, enzymatic chemiluminescence, electrosensory, and other immunoassays. Ultimately, the potential challenges and future perspectives in this burgeoning field are delved into.

Abstract:The global outbreak of coronavirus disease 2019 (COVID-19) has been catastrophic to both human health and social development. Therefore, developing highly reliable and sensitive point-of-care testing (POCT) for detecting severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has become a priority. Among all available POCTs, the lateral flow immunoassay (LFIA, also known as immunochromatography) has proved to be effective due to its accuracy, portability, convenience, and speed. In areas with a scarcity of laboratory resources and medical personnel, the LFIA provides an affordable option for the diagnosis of COVID-19. This review offers a comprehensive overview of methods for improving the sensitivity of SARS-CoV-2 detection using immunochromatography based on nanotechnology, sorted according to the different detection targets (antigens, antibodies, and nucleic acids). It also looks into the performance and properties of the various sensitivity enhancement strategies, before delving into the remaining challenges in COVID-19 diagnosis through LFIA. Ultimately, it seeks to provide helpful guidance in selecting an appropriate strategy for SARS-CoV-2 immunochromatographic detection based on nanotechnology.

Abstract:Point-of-care testing (POCT) is essential for providing a rapid diagnostic result in a prompt on-site diagnosis and treatment. A quick analysis time and a high sensitivity, with a sample-to-answer format, are the most important features for current POCT diagnostic systems. This review covers recent advances in POCT technologies with an emphasis on demonstrated and commercially available POCT diagnostic systems with laboratory quality using lateral flow immunoassay (LFIA). The system includes the integration of nanoparticles (NPs) in lateral flow test strips (LFTSs) and the mechanism through which particles improve the analytical performance of the fabricated strips. Several examples of NP-based LFTSs were selected to illustrate novel concepts or devices with promising applications as screening tools and superior alternatives to existing conventional strategies in clinical analysis, food safety, and environmental monitoring. In each analyte category, detection methods, configuration of LFIA modules, and advantages of POCT systems are reviewed and discussed along with future prospects. This review also discusses novel signal-enhancement strategies, optimal reader systems, and multiplex design prototypes, which have been employed for highly sensitive multiplex assay of LFTSs.
Artificial intelligence reinforced upconversion nanoparticle-based lateral flow assay via transfer learning

Abstract:The combination of upconverting nanoparticles (UCNPs) and immunochromatography has become a widely used and promising new detection technique for point-of-care testing (POCT). However, their low luminescence efficiency, non-specific adsorption, and image noise have always limited their progress toward practical applications. Recently, artificial intelligence (AI) has demonstrated powerful representational learning and generalization capabilities in computer vision. We report for the first time a combination of AI and upconversion nanoparticle-based lateral flow assays (UCNP-LFAs) for the quantitative detection of commercial internet of things (IoT) devices. This universal UCNPs quantitative detection strategy combines high accuracy, sensitivity, and applicability in the field detection environment. By using transfer learning to train AI models in a small self-built database, we not only significantly improved the accuracy and robustness of quantitative detection, but also efficiently solved the actual problems of data scarcity and low computing power of POCT equipment. Then, the trained AI model was deployed in IoT devices, whereby the detection process does not require detailed data preprocessing to achieve real-time inference of quantitative results. We validated the quantitative detection of two detectors using eight transfer learning models on a small dataset. The AI quickly provided ultra-high accuracy prediction results (some models could reach 100% accuracy) even when strong noise was added. Simultaneously, the high flexibility of this strategy promises to be a general quantitative detection method for optical biosensors. We believe that this strategy and device have a scientific significance in revolutionizing the existing POCT technology landscape and providing excellent commercial value in the in vitro diagnostics (IVD) industry.

In this paper, we present a comprehensive review of the recent advances and emerging trends of paper-based POC immunoassays, including the fundamental components and work principles, various detection mechanisms and applications, and existing commercialized devices/products. At last, we envision three promising development directions for paper-based POC immunoassays.

Abstract:Rapid and sensitive detection of hazards in clinical, food, and environmental samples is of great importance in the field of public health. Immunochromatographic assay (ICA), as a rapid diagnostics platform, is currently the most attractive point-of-care sensor due to its rapidness, user-friendly, and low-cost. Although, a variety of "head-to-toe" improvement strategies have sprung up to provide ICA with superior analytical performance in recent years, their respective strengths and weaknesses are difficult to interconnect and integrate, hampering their widespread deployment in ensuring public health. In this review, the momentous strategies to address the inherent limitations of ICA covering increasing sensitivity, enabling quantitative detection of analytes, and multiplexing are presented. Particularly, significant attention is focused on synergistically breaking the bottleneck of practical feasibility of ICA. Furthermore, the unique superiority and latest applications in clinical diagnosis, food safety, and environmental monitoring, as well as perspectives in the development process of the new generation of ICA are also elaborated. Eventually, the main challenges and future-oriented innovations in this field have also received attention with the profound development of the new generation of ICA in public health.

Abstract:The usability of many high-throughput lab-on-a-chip devices in point-of-care applications is currently limited by the manual data acquisition and analysis process, which are labor intensive and time consuming. Based on our original design in the biochemical reactions, we proposed here a universal approach to perform automatic, fast, and robust analysis for high-throughput array-based microfluidic immunoassays. Inspired by two-dimensional (2D) barcodes, we incorporated asymmetric function patterns into a microfluidic array. These function patterns provide quantitative information on the characteristic dimensions of the microfluidic array, as well as mark its orientation and origin of coordinates. We used a computer program to perform automatic analysis for a high-throughput antigen/antibody interaction experiment in 10 s, which was more than 500 times faster than conventional manual processing. Our method is broadly applicable to many other microchannel-based immunoassays. (C) 2013 AIP Publishing LLC.
Recent Application of Artificial Intelligence in Non-Gynecological Cancer Cytopathology: A Systematic Review

Abstract:State-of-the-art artificial intelligence (AI) has recently gained considerable interest in the healthcare sector and has provided solutions to problems through automated diagnosis. Cytological examination is a crucial step in the initial diagnosis of cancer, although it shows limited diagnostic efficacy. Recently, AI applications in the processing of cytopathological images have shown promising results despite the elementary level of the technology. Here, we performed a systematic review with a quantitative analysis of recent AI applications in non-gynecological (non-GYN) cancer cytology to understand the current technical status. We searched the major online databases, including MEDLINE, Cochrane Library, and EMBASE, for relevant English articles published from January 2010 to January 2021. The searched query terms were: "artificial intelligence", "image processing", "deep learning", "cytopathology", and "fine-needle aspiration cytology." Out of 17,000 studies, only 26 studies (26 models) were included in the full-text review, whereas 13 studies were included for quantitative analysis. There were eight classes of AI models treated of according to target organs: thyroid (n = 11, 39%), urinary bladder (n = 6, 21%), lung (n = 4, 14%), breast (n = 2, 7%), pleural effusion (n = 2, 7%), ovary (n = 1, 4%), pancreas (n = 1, 4%), and prostate (n = 1, 4). Most of the studies focused on classification and segmentation tasks. Although most of the studies showed impressive results, the sizes of the training and validation datasets were limited. Overall, AI is also promising for non-GYN cancer cytopathology analysis, such as pathology or gynecological cytology. However, the lack of well-annotated, large-scale datasets with Z-stacking and external cross-validation was the major limitation found across all studies. Future studies with larger datasets with high-quality annotations and external validation are required.

Abstract:Artificial intelligence (AI) is a reality of our times, and it has been successfully implemented in all fields, including medicine. As a relatively new domain, all efforts are directed towards creating algorithms applicable in most medical specialties. Pathology, as one of the most important areas of interest for precision medicine, has received significant attention in the development and implementation of AI algorithms. This focus is especially important for achieving accurate diagnoses. Moreover, immunohistochemistry (IHC) serves as a complementary diagnostic tool in pathology. It can be further augmented through the application of deep learning (DL) and machine learning (ML) algorithms for assessing and analyzing immunohistochemical markers. Such advancements can aid in delineating targeted therapeutic approaches and prognostic stratification. This article explores the applications and integration of various AI software programs and platforms used in immunohistochemical analysis. It concludes by highlighting the application of these technologies to pathologies such as breast, prostate, lung, melanocytic proliferations, and hematologic conditions. Additionally, it underscores the necessity for further innovative diagnostic algorithms to assist physicians in the diagnostic process.

The combination of immunoassays and microfluidics affords a promising platform for multiple, sensitive, and automatic point-of-care (POC) diagnostics. In this review, we focus on the description of recent achievements in microfluidic chips for immunoassays categorized by their detection method. Following a brief introduction to the basic principles of each detection method, we examine current microfluidic immunosensor detection systems in detail. We also highlight interesting strategies for sensitive immunosensing configurations, multiplexed analysis, and POC diagnostics in microfluidic immunosensors.
