Abstract:A versatile method for the synthesis of enantioenriched N-H sulfoximines is reported. The approach stems from the organomagnesium-mediated ring opening of novel cyclic sulfonimidate templates. The reactions proceed in high yield and with excellent stereofidelity with alkyl, aryl, and heteroaryl Grignard reagents. The chiral auxiliary is readily removed from the resultant sulfoximines via an unusual oxidative debenzylation protocol that utilizes molecular oxygen as the terminal oxidant. This provides a general strategy for the synthesis of highly enantioenriched N-H sulfoximines.

Abstract:As the aza-analogues of sulfoxides, sulfilimines bearing S(IV) stereogenic centers hold tremendous potential in synthetic chemistry and medicinal chemistry. However, the catalytic asymmetric synthesis of chiral sulfilimines are much less explored compared with chiral sulfoxides. In addition to the well-established asymmetric imidation of sulfides, catalytic enantioselective sulfur functionalization of sulfenamides has received increasing attention and developed rapidly over the past two years, which provides an efficient and alternative approach to chiral sulfilimine synthesis. This Highlight article summarizes and discusses the most recent advances in the catalytic asymmetric S-C bond-forming reactions for the preparation of chiral sulfilimines from sulfenamides.

Abstract:Sulfinamidines are promising aza-SIV chiral building blocks in asymmetric synthesis and drug discovery. However, no report has documented their enantioselective synthesis. Here we present an enantioselective synthesis of sulfinamidines via electrophilic amination of sulfenamides using an enantiopure N-H oxaziridine. The resulting enantiomerically enriched primary sulfinamidines are configurationally stable at 90 oC in solution. We also demonstrate a one-pot, three-component, enantioselective synthesis of sulfinamides using N-H oxaziridine reagents.

Abstract:A process has been designed and demonstrated for the asymmetric synthesis of sulfinamides using quinine as auxiliary. A variety of chiral sulfinamides including N-alkyl sulfinamides with diverse structure were prepared in good yields and excellent enantioselectivity starting from easily available and inexpensive reagents. The auxiliary quinine could be recovered and recycled.

Abstract:A new chiral sulfinyl, transfer auxiliary derived from readily available phenylglycine was developed. This auxiliary is utilized to synthesize a diverse array of alkyl- and arylsulfinamides and sulfinylferrocenes in high yields and excellent ee's. Abstract:Process development for a scalable and green synthesis of chiral tert-butanesulfinamide (TBSA) on a multikilogram scale is reported. The process is based on the identification of a chiral sulfinyl transfer agent, benzo[1,3]oxathiozin-2-one, that contains active and differentiated S-N and S-O bonds, allowing the synthesis to proceed under mild reaction conditions. This method is practical and overcomes the disadvantages of earlier methods deploying harsh reaction conditions and hazardous and toxic reagents.

A series of sulfinamides were obtained with good outcomes (up to 99% yield, >19:1 dr, and 98:2 er). Abstract:Sulfilimines are versatile synthetic intermediates and important moieties in bioactive molecules. However, their applications in drug discovery are underexplored, and efficient asymmetric synthetic methods are highly desirable. Here, we report a transition metal–free pentanidium-catalyzed sulfur alkylation of sulfenamides with exclusive chemoselectivity over nitrogen and high enantioselectivity. The reaction conditions were mild, and a wide range of enantioenriched aryl and alkyl sulfilimines were obtained. The synthetic utility and practicability of this robust protocol were further demonstrated through gram-scale reactions and late-stage functionalization of drugs.

Abstract:Achieving the preparation of enantiomerically enriched S-stereogenic compounds is a long-standing objective in stereoselective synthesis, owing to the fundamental importance and broad applications of these chiral scaffolds in various fields. Despite recent significant advancements, catalyst-controlled stereoselective synthesis of S-stereogenic compounds remains to be a considerable challenge, particularly by means of small-molecule catalysts. Herein, we disclosed an organocatalytic strategy for highly practical and enantioselective sulfinylation of alcohols and amines through the activation of sulfinates by forming mixed sulfinic anhydrides. Tuning the structure of the reactive species with sterically congested moieties, a simple, naturally occurring quinine catalyst effectively controls the chemo- and enantioselectivity over the nucleophilic S–O and S–N bond constructions, affording a wide range of chiral sulfinyl derivatives with excellent optical purities. Notably, the protocol readily facilitates the coupling with various natural products and commercial drugs that could offer an attractive strategy for the late-stage diversification of important biologically intriguing molecules.

Abstract:Sulfoximines are experiencing steadily increasing use in the development of pharmaceuticals and agrochemicals. Although recently a number of synthetic methods to access this versatile motif have been disclosed, only N H-sulfoximines have been considered as the ultimate targets. Here, we report an approach toward enantiopure N -substituted sulfoximines via direct stereoretentive S -alkylation of parent sulfinamides with zinc carbenoids. Mechanistically, a carbon–sulfur bond is formed in the course of 1,2-metallate rearrangement featuring an unusual migration of the S-atom in the transient zincate complex. The approach accommodates a large variety of differently substituted sulfinamides and features excellent functional group compatibility.

Abstract:Ru(II)-catalyzed enantioselective C–H functionalization involving an enantiodetermining C–H cleavage step remains undeveloped. Here we describe a Ru(II)-catalyzed enantioselective C–H activation/annulation of sulfoximines with α-carbonyl sulfoxonium ylides using a novel class of chiral binaphthyl monocarboxylic acids as chiral ligands, which can be easily and modularly prepared from 1,1′-binaphthyl-2,2′-dicarboxylic acid. A broad range of sulfur-stereogenic sulfoximines were prepared in high yields with excellent enantioselectivities (up to 99% yield and 99% ee) via desymmetrization, kinetic resolution, and parallel kinetic resolution.
