Abstract:As a structural analog of oxazoline, imidazoline (4,5-dihydroimidazole) has received much attention in the rational design of chiral ligands. The additional N-substituent provides broader space for fine-tuning of electronic and steric effects, and it offers a good handle for immobilizing onto solid supports. In the past decades, imidazoline ring has emerged as a powerful candidate for the design of chiral nitrogen-containing ligands, as well as a significant alternative for oxazoline ring. Various chiral imidazoline ligands have been designed and utilized in asymmetric organic reactions. These new catalysts can not only be applied in classical reactions, but also be employed to develop new organic reactions with high enantioselectivities. This review provides an overview of chiral imidazoline ligands. Their applications in asymmetric synthesis are also summarized.

Abstract:The ПҖ-stacking interaction is one of the most important intramolecular and intermolecular noncovalent interactions in organic chemistry. It plays an important role in stabilizing some structures and transition states in certain reactions via both intramolecular and intermolecular interactions, facilitating different selectivities, such as chemo-, regio-, and stereoselectivities. This minireview focuses on the recent examples of the ПҖ-stacking interaction-controlled asymmetric synthesis, including auxiliary-induced asymmetric synthesis, kinetic resolution, asymmetric synthesis of helicenes and heterohelicenes, and multilayer 3D chiral molecules.

Abstract:The precise control of Z and E configurations of the carbon-carbon double bond in alkene synthesis has long been a fundamental challenge in synthetic chemistry, even more pronounced when simultaneously striving to achieve enantioselectivity [( Z , R ), ( Z , S ), ( E , R ), ( E , S )]. Moreover, enantiopure non-natural α-amino acids are highly sought after in organic and medicinal chemistry. In this study, we report a ligand-controlled stereodivergent synthesis of non-natural α-quaternary amino acids bearing trisubstituted alkene moieties in high yields with excellent enantioselectivity and Z / E selectivities. This success is achieved through a palladium/copper–cocatalyzed three-component assembly of readily available aryl iodides, allenes, and aldimine esters by simply tuning the chiral ligands of the palladium and copper catalysts.

Abstract:Comprehensive computational studies were carried out to explore the mechanisms of enantioselective Cu/Pd and stereodivergent Cu/Ir dual-catalytic syntheses of α,α-disubstituted α-amino acids (α-AAs). A chiral copper azomethine ylide undergoes facile α-allylation with racemic π-allylpalladium species or stereopure π-allyliridium complex to stereoconvergently or stereodivergently furnish single/double stereocenters, respectively. Stereoselectivity at the α-center is controlled by the facial selectivity of α-allylation with respect to the prochiral nucleophile. Despite apparently similar transition-state assemblies, computational models and distortion/interaction analyses disclose versatile modes of stereoinduction wherein the copper azomethine ylide species can face-selectively intercept metal-π-allyl intermediates utilizing attractive dispersion interactions and/or sterically caused distortions. Generation of the β-stereocenter in the Cu/Ir system relies on a stereospecifically generated allyliridium complex and electronically controlled branched-to-linear selectivity, while the dual Cu/Pd system yields a linear monochiral product due to steric factors and π-π stacking interactions. The studies demonstrate on a molecular level how ligand-encoded chiral information is transferred to the α-/β-sites of the resulting α-AAs and how the mode of regio-/stereoselection is altered by differences in transition-metal-stabilized coupling partners. To facilitate studies of stereoselective catalysis, a suite of analytical tools to extract controlling factors for asymmetric induction is demonstrated.

Abstract:A novel chiral C(2)-symmetric 1,4-diamine with multistereogenic centers at the backbone of the ligand has been synthesized from cheap natural product D-mannitol through multistep transformations. Its diimine derivative (3 a) was found to be highly effective for the enantioselective control of the copper-catalyzed asymmetric aziridination of olefin derivatives with PhI==NTs as the nitrene source, affording the corresponding N-sulfonylated azirindine derivatives in good to excellent yields with up to 99 % ee (ee=enantiomeric excess). The catalyst system discovered in the present work was also extended to a one-pot enantioselective aziridination by using sulfonamide/iodobenzene diacetate as the nitrene source. In this case, most reactions proceeded smoothly to give the corresponding products in moderate yields with good to excellent enantiomeric excesses (75-96 % ee).

Abstract:Enantiodivergent synthesis to prepare both enantiomers of a chiral molecule using a single chiral source has aroused wide interest. Great achievements have been achieved by tuning non-chiral parameters such as solvents, additives, metal cations, anions, temperature and even reaction time. However, reversal of enantioselectivity by variation of the achiral subunits of a chiral ligand while keeping the stereogenic components unchanged has met less success. Herein, we report the ligand-ring-size controlled enantiodivergent aza-Friedel–Crafts alkylation reaction of 3-aminophenols with imines. Under the same conditions, Trost′s ligand afforded the ( R )-products, while its azetidine analogue delivered the opposite ( S )-enantiomers. By using this protocol, the two enantiomers of products were readily prepared in good yields with excellent enantioselectivities. Control experiments and DFT calculations were performed to shed light on the origin of enantiodivergence. The space and size of the ligand ring, which have been identified as essential factors impacting the non-covalent interaction between the substrate and the catalyst ligand, are found to determine the configuration of the products..

Abstract:Asymmetric synthesis of enantioenriched azo compounds bearing tetrasubstituted stereocenter was achieved through chiral N,N′-dioxide/metal Lewis acid promoted interrupted Japp-Klingemann reaction of aryldiazonium tetrafluoroborate salts with nucleophiles under mild conditions. This protocol features wide substrate scope and good functional group compatibility. Azaarene-containing chiral azo compounds were stable enough in Japp-Klingemann reaction condition. The key to success of the reaction was the employment of metal salt/N,N′-dioxide ligand and the dual-task roles of the base. Moreover, the X-ray crystal structure of Ni(II)/N,N′-dioxide/substrate complex confirmed that the substrate was activated by bidentate coordination, which shed light on the origin of chiral control of the reaction.

Abstract:Chiral bisoxazolines have been widely applied ligands to asymmetric catalysis and a series of optically active bisoxazolines with rigid backbones and different chelate sizes were synthesized. The effect of their chelate sizes on the enantioselectivity was investigated in the asymmetric aziridination. The results indicate that tuning chelate size can not only change the enantioselectivity, but also change the enantiofacial selectivity completely.

Abstract:This work presents the first enantio-and diastereoselective assembly of multi-layer folding targets through asymmetric catalytic C-C bond formation. Pd[(S)-BINAP]Cl-2 is found to be an efficient catalyst for the Suzuki-Miyaura coupling between phosphinyl bromides and benzothiadiazole boronic esters for this asymmetric assembly. The structure of the resulting chiral multi-layer folding framework is unambiguously determined by X-ray analysis and shows a nearly parallel pattern of three layers and quasi-syn and -anti configurations. Good to excellent diastereo-and enantioselectivities (up to > 20:1 dr and > 99:1 er) are achieved.
