Abstract:Multi-modal imaging probes through synergistically combining two or more imaging modalities into single nanoparticle, offer possibilities to address multiple issues such as resolution, sensitivity, and tissue penetration. In this study, we report a facile strategy to synthesize a dual-modal fluorescence/magnetic resonance (MR) imaging nanoprobe with bright fluorescence and significantly improved relaxivity, where Gd3+ ions are simply chelated onto the outer surface of the cyclic DTPA dianhydride (cDTPAA) functionalized carbon quantum dots (CQDs). Transmission electron microscopy, Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, and zeta potential measurements validated the successful preparation of CQD–DTPA–Gd. The surface conjugation of CQDs with cDTPAA and further Gd3+ chelation does not perturb core optical properties of CQDs, and the thus prepared CQD–DTPA–Gd contain minimum Gd3+ content (8.06%, w/w), while exhibiting significantly improved longitudinal relaxivity (56.72mM−1s−1). Excellent water solubility, good cell-membrane permeability and negligible cytotoxicity make CQD–DTPA–Gd an ideal dual-modal fluorescence/MR imaging probe, suggesting its potential and significance in practical bioimaging applications and medical translation in the future.
Attaching Double Chain Cationic Gd(Iii)-Containing Surfactants On Nanosized Colloids For Highly Efficient Mri Contrast Agents

Abstract:A new Gd(III)-based metallosurfactant with double quaternary-ammonium-containing long alkyl chains was developed and subjected to the miniemulsion polymerization of vinyl monomers, giving nanosized colloids (50-110 nm) with narrow size distribution. The size of the colloid particles can be easily tuned by adjusting the feeding ratio of metallosurfactant to monomer. SEM and XPS analysis showed that the Gd(III) complexes were attached on the surface of the colloids. ICP-AES analysis further quantified the contents of Gd(III) in the miniemulsions. T1 magnetic resonance imaging (MRI) showed that these Gd(III)-loading colloids exhibited relaxivity of up to 22.77 mM(-1) s(-1), much higher than that of Ominiscan (R) (4.64 mM(-1) s(-1)).
Ln(III) Chelates-Functionalized Carbon Quantum Dots: Synthesis, Optical Studies and Multimodal Bioimaging Applications.

Abstract:The carbon quantum dots termined with amine groups (CQDs-NH2) were synthesized in one-pot hydrothermal method with citric acid as carbon source and branched polyethylenimine (BPEI) as passive agent, which was then covalently linked to 1, 4, 7, 10-tetraazacyclononane (DOTA) framework to yield CQDs-DOTA. The CQDs-DOTA provide excellent ligand scaffolds for the chelation of lanthanide ions, yielding the related complexes CQDs-DOTA-Ln (Ln = Eu, Tb, Yb and Gd). The successful preparation of CQDs-DOTA-Ln was validated by TEM, XRD, XPS, and FT-IR spectroscopy. The CQDs-DOTA-Ln (Ln = Eu, Tb and Yb) exhibited the characteristic emissions of related lanthanide ions, indicating the CQDs could sensitize the luminescence of lanthanide ions. Besides, the CQDs-DOTA-Gd could serve as an excellent T1-weighted MR imaging probe due to the chelation of paramagnetic Gd (III) ions and good hydrophilicity. The cytotoxicity of CQDs-DOTA-Ln was evaluated through MTT assay upon HeLa cells. The images from fluorescence microscopy further verified their applications in bioimaging in vitro. Due to the good biocompatibility, low toxicity, and high contrast efficiency, the CQDs-DOTA-Ln with Vis/NIR fluorescence and MR multi-modal imaging performance could be used as potential contrast agents for clinic applications.
Co‐Assembly of Gd(III)‐Based Metallosurfactant and Conjugated Polymer Nanoparticles in Organosilica Cross‐Linked Block Copolymer Micelles for Highly Efficient MRI and Fluorescent Bimodal Imaging

Abstract:Conformity of two biological imaging entities, magnetic resonance imaging (MRI) and fluorescence imaging, is achieved through co‐assembly of a Gd(III)‐based metallosurfactant, conjugated polymeric nanoparticles, and amphiphilic block copolymer F127 (PEO106PPO70PEO106) followed by crosslinking with organosilica. The cross‐linked micelles with a size around 100 nm exhibit outstanding dispersion stability in aqueous and phosphate buffered saline solutions, bright fluorescence emission, and high relaxivities, providing a new approach to synthesize highly efficient bimodal contrast agents. The relaxivities of the co‐assembled micelles are synergistically enhanced by incorporation of Gd(III) complexes with high hydration number (q = 3) and elongation of rotation correlation time to achieve r1 values up to 105.37 mm−1 s−1 (at 1.5 T), which is over 20 times that of clinically used MRI contrast agents and among the highest values of all the nanoparticular MRI contrast agents. The external PEO layer endows these micelles with very low cytotoxicity for both in vitro and in vivo imaging. Meanwhile, thanks to the enhanced permeability and retention effect originating from their nanoscale sizes, the bimodal contrast agents show a prolonged blood circulation time in vivo and targeted accumulation at tumor regions to display outstanding MRI imaging performance.

A paramagnetic polyoxometalate cluster, K13 [Gd(β2 -SiW11 O39 )2 ], is first encapsulated with a cationic 3-generation dendron (D-3) bearing triethylene glycol monomethyl ether terminal groups through ionic substitution. The formed electrostatic complex, (D-3)13 [Gd(β2 -SiW11 O39 )2 ] (D-GdSiW), which displays an ambivalent micellar structure, is then demonstrated to act as a suitable carrier for the uptake of cationic fluorescent dyes, forming a loaded complex. The dye-loaded complex D-GdSiW-D is thus proven to display high relaxivity as a contrast agent, maintaining its structural stability under physiological conditions with low cell cytotoxicity. Confocal laser scanning microscopic observation also demonstrates the fluorescence imaging property of the prepared complex upon consumption by MCF-7 cells. Both in vivo fluorescence imaging and MRI further confirm that the complex micelle with dual micellar structure may be a suitable liver-specific contrast agent for clinical diagnosis.

Abstract:Suffering from high inherent toxicity generated from heavy metals in semiconductor quantum dots and a complex preparation process, the medical application of conventional gadolinium(III)/semiconductor quantum dot-based magnetic resonance/fluorescence multimodal imaging (MRI/FI) contrast agent has been limited. In this paper, a new kind of biocompatible carbon quantum dot functionalized by gadolinium(III) chelates (named as Gd(III)/CQDs) was obtained through one-step pyrolysis of gadopentetate monomeglumine, which provides simultaneously a carbon source and a gadolinium( III) source. The Gd(III)/CQDs show good magnetic resonance response and photoluminescence properties with longitudinal relaxation rate and quantum yield of 5.5-6.4 mM L-1 S-1 [Gd(III)%, 0.5 mM] and 2.6-8.9%, respectively. The Gd(III)/CQDs with diameters in the range of 2-3 nm can be well dispersed in deionized water, phosphate-buffered saline buffer solution or culture medium to form a stable dispersion, which with low cytotoxicity can penetrate into cells and show green fluorescence (excitation at 488 nm), presenting a great potential to be used as an MRI/FI contrast agent.

Abstract:It is highly desired to develop the dual-modality fluorescence and magnetic resonance imaging (MRI) probes in medical imaging because it can provide high-resolution macroscopical anatomical information and high-sensitivity microscopical optical signal simultaneously. In this study, harmless gadolinium-doped carbon dots (Gd-CDs) were prepared via a convenient one-pot hydrothermal approach for fluorescence/MR bimodal imaging. The derived Gd-CDs exhibit enhanced blue photoluminescence with a quantum yield as high as 69.86% and significantly improved longitudinal relaxivity (r(1) = 14.33 mM(-1)s(-1), 0.5 T) in comparison with commercial Magnevist (Gd-DTPA, r(1) = 4.5 mM(-1)s(-1), 0.5 T). Here Gd3+ ions are simply chelated onto CDs by carboxyl groups. Moreover, unlike the previous reports, Gd3+ chelation does not perturb core optical properties of CDs. Excellent water solubility, good cell-membrane permeability and negligible cytotoxicity make Gd-CDs an ideal dual-modal fluorescence/MR imaging nanoprobe, suggesting its potential and significance in practical biological and clinic applications in the future. One-pot Synthesis of Gadolinium-Doped Carbon Quantum Dots for High-Performance Multimodal Bioimaging

Abstract:In this study, an efficient and mild approach is reported for the facile synthesis of a carbon quantum dot (CQD)-based dual-modal fluorescence (FL)/magnetic resonance (MR) imaging probe by doping Gd(iii) into CQDs through a one-pot pyrolysis process at low temperature. The as-prepared Gd-encapsulated CQDs (GCDs), which have an average diameter of ∼15 nm, are highly water-soluble. The GCDs are observed to have a high MR response with a longitudinal relaxation of 57.42 mM-1 s-1 and a strong fluorescence brightness with an absolute quantum yield of 40% while containing only 1.0% (w/w) of Gd3+ content. Stemming from the minimum Gd-doping and the inert carbon coating, GCDs exhibit excellent biocompatibility and blood compatibility. Dual-modality bioimaging applications of GCDs are successfully demonstrated by the use of HeLa cells and mice as models, revealing their great potential in fundamental biomedical research studies and even clinical fields such as fluorescence-guided surgery and dual-modal FL/MR imaging of blood vessels.

Abstract:A bimodal contrast nanoagent was developed by chelating gadolinium ions to 2-[bis[2-[carboxymethyl-[2-oxo-2-(2-s ulfanylethyl-amino)ethyl] amino] ethyl] amino] acetic acid (DTDTPA)-modified CuInS2/ZnS quantum dots (QDs). The longitudinal relaxivity (r(1)) of the resulted QDs@DTDTPA-Gd nanoparticles (NPs) was calculated to be 9.91 mM(-1) s(-1), which was 2.5 times as high as that of clinically approved Gd-DTPA (3.9 mM(-1) s(-1)). In addition, the in vivo imaging experiments showed that QDs@DTDTPA-Gd NPs could enhance both near-infrared fluorescence and T-1-weighted magnetic resonance (MR) imaging of tumor tissue through passive targeting accumulation. Moreover, the high colloidal and fluorescence stabilities and good biocompatibility indicate that QDs@DTDTPA-Gd NPs have a great potential for use as an efficient nanoagent to integrate the extremely high sensitivity of fluorescence imaging to the high resolution of MR imaging. Integration of bimodal detectability in the same agent of QDs@DTDTPA-Gd NPs can avoid extra stress on the blood clearance mechanisms as the administration of multiple dose of agents.
Gd(III) Functionalized Gold Nanorods for Multimodal Imaging Applications.

Abstract:We demonstrate a novel noncovalent method for producing Gd(III)-functionalized gold nanorods as multimodal contrast agents for MRI and CT imaging. The ligand is connected to the surface of the gold nanorods by a noncovalent bond making the Gd(III) ions directly accessible to water molecules, and resulting in a longitudinal relaxivity as high as 21.3 mM(-1) s(-1). In addition, compared with spherical gold nanoparticles, gold nanorods have more binding sites for Gd(III) ions due to their large surface-to-volume ratio. Benefiting from the advantages of the new type of carry material and the novel fabrication approach, the multimodal imaging probes exhibit a high longitudinal relaxivity r(1) on the order of 1.1 × 10(7) mM(-1) s(-1) on a per-particle basis, which is 24 times higher than that of Gd(III)-ion-functionalized spherical gold nanoparticles. Furthermore, CT imaging shows that such nanoprobes could induce an efficient contrast enhancement when the gold concentration is at least equal to 1.31 mg ml(-1). These results demonstrate that the as-prepared Gd functionalized gold nanorods could provide a new and versatile platform for the development of multimodal imaging probes.
Preparation of Highly Efficient Mri Contrast Agents Through Complexation of Cationic Gd-Iii-Containing Metallosurfactant with Biocompatible Polyelectrolytes

Abstract:Novel contrast agents were developed through assembling of Gd-III-containing metallosurfactant (MS) with biocompatible polyelectrolytes sodium hyaluronate (HA), heparinsodium (HS) and dextran sulfate sodium (DSS). The formed polyelectrolyte-surfactant complexes showed different structural patterns as the charge ratio increased, including spherical aggregates, rod-like aggregates and network patterns in monovalent HA system, while spherical structures emerged in multivalent HS and DSS systems. Energy dispersive spectroscopy analysis and scanning electron microscopy mapping showed the presence of Gd-III in these complexes. Inductively coupled plasma atomic emission spectrometry was further used to quantify the contents of Gd-III in the assemblies. T1 magnetic resonance imaging showed that these Gd-III-loaded complexes exhibited relaxivity of up to 63.81 mm(-1) s(-1), much higher than that of Ominiscan (4.64 mm(-1) s(-1)). The cytotoxicity test in vitro demonstrated the excellent biocompatibility of these complexes, which is essential for clinical application.
Gd3+-Functionalized Gold Nanoclusters for Fluorescence-Magnetic Resonance Bimodal Imaging

Abstract:Multimodal imaging that aims to advance imaging by strategically combining existing technologies with uniquely designed probes has attracted great interest in recent years. In this work, Gd3+-functionalized gold nanoclusters were prepared with a straightforward and scalable approach by using proteins as templates. The impact of the synthesis approach on the conjugation efficiency of Gd chelators to proteins, as well as the fluorescence quantum yield of gold nanoclusters was carefully studied. The effect of protein size on the relaxivities of the probes was also investigated. By using the optimal conditions, dual function probes with intense red fluorescence emission and longitudinal relaxivity as high as 9.7 mM(-1) s(-1) could be obtained. The probes were applied for cell staining and in vivo contrast-enhanced magnetic resonance imaging (MRI). The MR images showed that the dual function probes circulated freely in the blood pool without undesirable accumulation in the liver and spleen. Meanwhile, the material could be removed from the body through renal clearance, making it attractive for practical multimodal imaging.

Abstract:Nowadays, it is highly desired to develop dual-modal fluorescence and magnetic resonance imaging (FI/MRI) probes in medical imaging because it unites the respective advantages of each imaging modality: high sensitivity of FI and superior spatial resolution of MRI. In this study, a facile strategy to fabricate a new bimodal imaging nanoprobe (Gd-CQDs@N-Fe3O4) was reported by integrating the fluorescence ability of carbon quantum dots (CQDs) and T-1 and T-2 contrast-enhancing functionality of Gd(III) ions and Fe3O4 nanoparticles into a single hybrid nanostructure. The hybrid composites were investigated by FT-IR, XRD, TEM, XPS, VSM, and so on, which confirmed that Gd-CQDs@N-Fe3O4 nanoparticles were successfully obtained and exhibited super-paramagnetic property at room temperature. The derived nanoprobes presented an excitation wavelength-independent emission behavior. In addition, r(1) and r(2) relaxivities of the synthesized imaging nanoprobes were measured to be 5.16 and 115.6 rnM(-1) s(-1), which nominated Gd-CQDs@N-Fe3O4 nanocomposites as a suitable T-1-T-2 contrast agent. The Gd-CQDs@N-Fe3O4 nanoparticles combining two synergetic imaging modalities showed great potential in FI/MRI dual-modal imaging for a more complementary and accurate detection.
Facile Preparation of Double Rare Earth-Doped Carbon Dots for MRI/CT/FI Multimodal Imaging

Abstract:The integration of different imaging functions into an ultrasmall nanoplatform with excellent biocompatibility and metabolic performance to fabricate multimodal imaging probes has been a tremendous challenge. Herein, a novel kind of gadolinium- and ytterbium-doped carbon dots (Gd/Yb@ CDs) with diameter of 5.26 0.93 nm was obtained by the convenient one-step hydrothermal process for magnetic resonance imaging (MRI), X-ray computed tomography (CT), and fluorescent imaging (FI). The obtained Gd/Yb@ CDs exhibited not only the excitation-dependent emission and superhigh photostability but also higher longitudinal relaxivity (r(1) = 6.65 s(-1)) and excellent X-ray absorption performance (45.43 HU L CI). The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay, tissue section assessment, and body weight investigation shed light on the superior biocompatibility of these Gd/Yb@CDs. MRI/CT imaging demonstrated these multimodal nanoprobes could give rise to obviously enhanced contrast effect on the xenografted tumor model. Furthermore, biodistribution analysis demonstrated that these nanoprobes could be easily removed from the mice through the kidney elimination pathway. Therefore, the present Gd/Yb@CDs could be served as promising MRI/CT/FI multimodal nanoprobes to provide more accurate and comprehensive diagnosis information.

Abstract:Dual-modal imaging techniques have gained intense attention for their potential role in the dawning era of tumor early accurate diagnosis. Chelate-free robust dual-modal imaging nanoprobes with high efficiency and low toxicity are of essential importance for tumor targeted dual-modal in vivo imaging. It is still a crucial issue to endow Cd-free dual-modal nanoprobes with bright fluorescence as well as high relaxivity. Herein, a facile synthetic strategy was developed to prepare Gd-doped CuInS/ZnS bimodal quantum dots (GCIS/ZnS, BQDs) with optimized properties. The fluorescent properties of the GCIS/ZnS BQDs can be thoroughly optimized by varying reaction temperature, aging time, and ZnS coating. The amount of Gd precursor can be well-controlled to realize the optimized balance between the MR relaxivity and optical properties. The obtained hydrophobic GCIS/ZnS BQDs were surface engineered into aqueous phase with PEGylated dextran-stearyl acid polymeric lipid vesicles (PEG-DS PLVs). Upon the phase transfer, the hydrophilic GCIS/ZnS@PLVs exhibited pronounced near-infrared fluorescence as well as high longitudinal relaxivity (r1 = 9.45 mM(-1) S(-1)) in water with good colloidal stability. In vivo tumor-bearing animal experiments further verified GCIS/ZnS@PLVs could achieve tumor-targeted MR/fluorescence dual-modal imaging. No toxicity was observed in the in vivo and ex vivo experiments. The GCIS/ZnS@PLVs present great potential as bimodal imaging contrast agents for tumor diagnosis.

Abstract:A Zn2+-responsive bimodal magnetic resonance imaging (MRI) and luminescence imaging probe GdL was synthesized. The relaxivity and luminescence properties were examined. In the presence of 0.5 equiv of Zn2+, the longitudinal relaxivity is increased from 3.8 mM(-1) s(-1) to 5.9 mM(-1) s(-1) at 23 MHz and 25 degrees C with 55% enhancement, whereas the fluorescence exhibits a 7-fold increase. The Zn2+ responsive imaging probe shows favorable selectivity and tolerance over a variety of biologically relevant anions and metal ions in physiological pH range for both relaxivity and luminescence. In vitro phantom images and confocal fluorescence images in living cells show that the bimodal Zn2+ probe can effectively enhance T-1-weighted imaging contrast and luminescence imaging effect through Zn2+ coordination with excellent cellmembrane permeability and biocompatibility. Spectral and electrospray ionization mass spectrometry (ESI-MS) studies indicate that two different Zn2+-bound species, (GdL)(2)Zn and GdLZn, are formed when 0.5 and 1 equiv of Zn2+ are bound to GdL complex, respectively. Crystal structural determination and dysprosium-induced O-17 NMR shift (DIS) experiment demonstrate that the increased molecular weight and the improved molecular rigidity upon complexation of Zn2+ with GdL is the primary factor for relaxivity enhancement. Significant enhancement of the luminescence is due to a heavy atom effect and much increased molecular rigidity upon Zn2+ binding to 8-sulfonamidoquinoline chromophore.

Abstract:Fluorescence (FL)/magnetic resonance (MR) dual-modal imaging nanoprobes are significant not only for cutting edge research in molecular imaging, but also for clinical diagnosis with high precision and accuracy. However, synthesis of FL/MR dual-modal imaging nanoprobes that simultaneously exhibit strong fluorescent brightness and high MR response, long-term colloidal stability with uniform sizes, good biocompatibility and a versatile surface functionality has proven challenging. In this study, the well-defined core-shell structured Gd3+ chelate-conjugated fluorescent polymer nanoparticles (Gd-FPNPs) that consist of rhodamine B (RB)-encapsulated poly(methyl methacrylate) (PMMA) cores and Gd3+ chelate-conjugated branched polyethylenimine (PEI) shells, are facilely synthesized via a one-step graft copolymerization of RB-encapsulated MMA from PEI-DTPA-Gd induced by tert-butyl hydroperoxide (TBHP) at 80 °C for 2 h. The mild synthesis route not only preserves the chemical environment for Gd3+ coordination, but also improves optical properties and chemo-/photostability of RB. A high local concentration of outer surface-chelated Gd3+ and their direct interactions with hydrogen protons endow Gd-FPNPs high longitudinal relaxivity (26.86 mM-1 s-1). The uniform spherical structure of Gd-FPNPs facilitates their biotransfer, and their surface carboxyl and amine groups afford them both long-term colloidal stability and cell-membrane permeability. The excellent biocompatibility and FL/MR dual-modal imaging capability of Gd-FPNPs are demonstrated using HeLa cells and mice as models. All the results confirm that Gd-FPNPs fulfill the design criteria for a high-performance imaging nanoprobe. In addition, this study enables such probes to be prepared also by those not skilled in nanomaterial synthesis, and thus promoting the development of novel functional imaging nanoprobes.

Abstract:The design and development of contrast agents for magnetic resonance imaging (MRI) with improved chemical stability and higher contrasting capability for clinical translation compared to conventional contrast agents are still of great interest. In this study, a facile and universal approach was explored for controllable functionalization of red-emissive carbon nanodots (RCNDs) with diethylenetriaminepenta-acetic anhydride (DTPA) for chelation of gadolinium. A series of accurate characterizations were used to control each step of the synthesis. The functionalization did not alter the band gap of the carbon nanodots, preserving their inherent far-red fluorescence. The as-prepared RCND-DTPA-Gd displayed a high colloidal stability with negligible Gd leakage. The nanodots also showed a better magnetic resonance relaxivity than commercial MRI agents. RCND-DTPA-Gd had good biocompatibility in vivo even at high doses. The systemically injected RCND-DTPA-Gd were found to be efficiently excreted through the renal route, a feature that further minimizes the potential toxicity risks. All these properties suggest that carbon nanodots can be well designed as efficient carriers of Gd, resulting in potential clinical tools as dual MRI/fluorescence functional probes for imaging applications. The approach described here could pave the pathway to a flexible strategy for the controllable functionalization of small-sized nanoparticles including carbon dots, rendering them more versatile. This work is expected to promote the future translation of carbon nanodots into clinical trials.
