Abstract:In contrast to traditional breeding, which relies on the identification of mutants, metabolic engineering provides a new platform to modify the oil composition in oil crops for improved nutrition. By altering endogenous genes involved in the biosynthesis pathways, it is possible to modify edible plant oils to increase the content of desired components or reduce the content of undesirable components. However, introduction of novel nutritional components such as omega-3 long-chain polyunsaturated fatty acids needs transgenic expression of novel genes in crops. Despite formidable challenges, significant progress in engineering nutritionally improved edible plant oils has recently been achieved, with some commercial products now on the market.

Abstract:Triglycerides are the main storage form of oil in plant seeds. Both fatty acids and triglycerides possess important functions in the process of plant growth and development. To improve the seed oil content and improve its fatty acid composition, this paper analyzed the research progress on the oil regulation and synthesis metabolism process of plant seeds and summarized the strategies for the improvement of plant seed oil: (a) To regulate carbon distribution by inhibiting the expression of genes encoding key enzymes, allocating carbon sources into the protein synthesis pathway, and enhancing the expression of key genes encoding key enzymes, leading carbon sources into the synthesis pathway of fatty acids; (b) To intervene in lipid synthesis by promoting the biosynthesis of fatty acids and improving the expression level of key genes encoding enzymes in the triacylglycerol (TAG) assembly process; (c) To improve seed oil quality by altering the plant fatty acid composition and regulating the gene expression of fatty acid desaturase, as well as introducing an exogenous synthesis pathway of long chain polyunsaturated fatty acids; (d) To regulate the expression of transcription factors for lipid synthesis metabolism to increase the seed oil content. In addition, this article reviews the key enzymes involved in the biosynthesis of plant fatty acids, the synthesis of triacylglycerol, and the regulation process. It also summarizes the regulatory roles of transcription factors such as WRI, LEC, and Dof on the key enzymes during the synthesis process. This review holds significant implications for research on the genetic engineering applications in plant seed lipid metabolism.

Abstract:Genetic engineering refers to the specific molecular biological modification of DNA sequences. With the rapid development of genetic engineering methods, especially the breakthroughs in guiding endonuclease technology, gene remodeling of crops has become simpler, more precise, and efficient. Genetic engineering techniques can be used to develop crops with superior traits such as high trace elements and high plant nutrients, providing an important tool to meet the needs of nearly 7.6 billion people in the world for crop yield and quality and to achieve sustainable development. This review first introduces transgenic technology and gene editing technology and analyzes the achievements in improving the efficiency of genetic transformation and regeneration in recent years. Then, it focuses on reviewing the applications of related genetic engineering technologies in improving the yield and quality of rice, maize, and wheat. In addition, the problems of genetic engineering technology in crop applications are discussed. The aim is to provide a reference for research on the development of genetic engineering technologies and the improvement of crop yield and quality.

Abstract:Peanut is an important cash crop used in oil, food and feed in our country. The rapid development of sequencing technology has promoted the research on the related aspects of peanut genetic breeding. This paper reviews the research progress of peanut origin and evolution, genetic breeding, molecular markers and their applications, genomics, QTL mapping and genome selection techniques. The main problems of molecular genetic breeding in peanut research worldwide include: the narrow genetic resources of cultivated species, unstable genetic transformation and unclear molecular mechanism of important agronomic traits. Considering the severe challenges regarding the supply of edible oil, and the main problems in peanut production, the urgent research directions of peanut are put forward: The de novo domestication and the exploitation of excellent genes from wild resources to improve modern cultivars; Integration of multi-omics data to enhance the importance of big data in peanut genetics and breeding; Cloning the important genes related to peanut agronomic traits and analyzing their fine regulation mechanisms; Precision molecular design breeding and using gene editing technology to accurately improve the key traits of peanut.

Abstract:Maize oil, which is a blend of fatty acid esters generated from triacylglycerol (TAG), is an important component of maize-derived food, feed, and biofuel. The kernel oil content in commercial high-oil maize hybrids averages ∼8%, which is far lower than that in developed high-oil maize lines (as high as 20%). Advances in high-oil maize genomics and genetics and the development of systems metabolic engineering technologies provide new opportunities for high-oil maize breeding. In this review, we discuss the possibility of using kernels and vegetative tissues as factories to produce TAG, eicosapentaenoic acid, and docosahexaenoic acid. We further propose specific implementation strategies based on the metabolic engineering of other species to develop transgenic and gene-editing products, as well as traditional breeding strategies, for application in high-oil maize breeding programs.

Abstract:It was reviewed the rapeseed explant types,selection markers,transformation methods,the influence factors of regeneration systems and its applications in rapeseed breeding were discussed.It was also mentioned the problems and development in the future of rapeseed transformation.It was found that there were still limitations in the transformation of rapeseeds through the published papers and establishment of transformation system of rapeseeds.Firstly,the process of receptor regeneration was a long period and the frequency of gene transformation was low.Secondly,the introduced gene might be silent and the expression of transferred gene would unstable in the progeny.Thirdly,the scientific evaluation should be made about the bad influence of transgenic rapeseed to human heath and the natural environment.Moreover,more complete law and rule should be made to supervise the environment release of transgenic plants.At the same time,it should meet the needs of people by modifying the trait of rapeseeds.Furthermore,there would be a wide foreground to produce the substitute of energy source by introducing special gene into rapeseed plants.

Abstract:Carotenoids have a range of diverse biological functions and actions, especially playing an important role in human health with provitamin A activity, anti-cancer activity, enhancing immune ability and so on. Human body can't synthesis carotenoids by itself and must absorb them from outside. However, carotenoid contents in many plant are very low, and many kinds of carotenoid are difficult to produce by chemical ways. With the elucidation of carotenoid biosynthetic pathway and cloning genes of relative enzymes from microorganisms and higher plants, it is possible to regulate carotenoid biosynthesis via genetic engineering. This article reviews gene cloning of carotenoid biosynthetic enzymes in microorganisms and higher plants, and advances in the studies of carotenoid production in heterologous microorganisms and crop plants using gene-manipulated carotenoid biosynthesis.

Abstract:Oil palm, a cross-pollinated crop with long generation time, poses a lot of challenges in achieving sustainable oil palm with high yield and quality. The African oil palm (Elaeis guineensis Jacq.) is the most productive and versatile oil-yielding crop in the world, producing more than any other oil-yielding crop. Despite recent challenges, such as stress tolerance, superior oil quality, disease tolerance, and the need for new market niches, there is a growing need to explore and develop new varieties with high yield potential and the genetic diversity required to maintain oil palm yield stability. Breeding is an indispensable part of producing high-quality planting materials to increase oil palm yield. Biotechnological technologies have transformed conventional plant breeding approaches by introducing novel genotypes for breeding. Innovative pre-breeding and breeding approaches, such as identifying candidate genes in wild or land races using genomics tools, can pave the way for genetic improvement in oil palm. In this review, we highlighted the modern breeding tools, including genomics, marker-assisted breeding, genetic engineering, and genome editing techniques in oil palm crops, and we explored certain concerns connected to the techniques and their applications in practical breeding.

Abstract:Since the success of the first genetically modified tobacco in 1983, great advances have been achieved in plant genetic transformation technology. Genetic engineering has become an important tool for plant functional gene research and crop germplasm innovation, which brings a revolution for crop production. Gene-gun-mediated transfer and use of Agrobacterium tumefaciens as a vector are the two common approaches for delivering the expression cassette of gene of interest into plant cells. Both DNA delivery methods insert for-eign DNA into plant genome at random position, and thus have some limitations. The recent advent of ge-nome-editing technologies has enabled to generate mutation, insertion and deletion of small fragments and oth-er modifications at very precise locations within the genome. The new technology, named as "gene editing" or"genome editing", has broad prospects and application value for gene function research and genetic improve-ment. On the basis of introducing plant transgenic technology and its application, this paper emphasizes the technical detail, status and prospect of the new technology of plant gene editing, the CRISPR/Cas9 system.

Abstract:Vegetable crops provide a rich source of essential nutrients for humanity and represent critical economic values to global rural societies. However, genetic studies of vegetable crops have lagged behind major food crops, such as rice, wheat and maize, thereby limiting the application of molecular breeding. In the past decades, genome sequencing technologies have been increasingly applied in genetic studies and breeding of vegetables. In this review, we recapitulate recent progress on reference genome construction, population genomics and the exploitation of multi-omics datasets in vegetable crops. These advances have enabled an in-depth understanding of their domestication and evolution, and facilitated the genetic dissection of numerous agronomic traits, which jointly expedites the exploitation of state-of-the-art biotechnologies in vegetable breeding. We further provide perspectives of further directions for vegetable genomics and indicate how the ever-increasing omics data could accelerate genetic, biological studies and breeding in vegetable crops.
Progress in genetic engineering of peanut (Arachis hypogaea L.)--a review

Abstract:Peanut (Arachis hypogaea L.) is a major species of the family, Leguminosae, and economically important not only for vegetable oil but as a source of proteins, minerals and vitamins. It is widely grown in the semi-arid tropics and plays a role in the world agricultural economy. Peanut production and productivity is constrained by several biotic (insect pests and diseases) and abiotic (drought, salinity, water logging and temperature aberrations) stresses, as a result of which crop experiences serious economic losses. Genetic engineering techniques such as Agrobacterium tumefaciens and DNA-bombardment-mediated transformation are used as powerful tools to complement conventional breeding and expedite peanut improvement by the introduction of agronomically useful traits in high-yield background. Resistance to several fungal, virus and insect pest have been achieved through variety of approaches ranging from gene coding for cell wall component, pathogenesis-related proteins, oxalate oxidase, bacterial chloroperoxidase, coat proteins, RNA interference, crystal proteins etc. To develop transgenic plants withstanding major abiotic stresses, genes coding transcription factors for drought and salinity, cytokinin biosynthesis, nucleic acid processing, ion antiporter and human antiapoptotic have been used. Moreover, peanut has also been used in vaccine production for the control of several animal diseases. In addition to above, this study also presents a comprehensive account on the influence of some important factors on peanut genetic engineering. Future research thrusts not only suggest the use of different approaches for higher expression of transgene(s) but also provide a way forward for the improvement of crops.

Abstract:Improving oil yield and quality is a major goal for crop breeding, and CRISPR/Cas-mediated genome editing has opened a new era for designing oil crops with enhanced yield and quality. CRISPR/Cas technology can not only increase oil production but also enhance oil quality, including enhancing pharmaceutical and health components, improving oil nutrients, and removing allergic and toxic components. As new molecular targets for oil biosynthesis are discovered and the CRISPR/Cas system is further improved, CRISPR/Cas will become a better molecular tool for designing new oil crops with higher oil production, enhanced nutrients, and improved health components. 'CRISPRized' oil crops will have broad applications both in industry (e.g., as biofuels) and in daily human life.

Abstract:Oils extracted from plants have been used predominantly as edible oil. Soybean, peanut, rapeseed mustard, sunflower, safflower, Sesamum, linseed, castor and cotton seed are predominant oil crops. Global status of nine major and minor oil crops has been discussed which includes their classification, contribution, major growing countries and objectives. Major objectives in oil crop improvement are enhancement of seed and oil yield, quality of oil according to its use, i.e. edible or industrial uses, breeding of varieties which fit in different cropping systems and breeding biotic and abiotic stress resistant/tolerant varieties. Achievements in varietal development programme of nine oil crops in India have also been discussed and future research needs to meet the increasing demand have also been highlighted. This review describes developments in use of biotechnological tools in seven edible oil crops, namely, Brassica, soybean, sunflower, groundnut, Sesamum, linseed and safflower and also highlights the prospects of using markers in genetic improvement of these crops. Molecular markers reported for genetic diversity assessment, mapping and tagging genes/QTLs for different qualitative and quantitative traits and their use in marker-assisted selection have been presented.

Abstract:The world's population is projected to increase by two billion by 2050, resulting in food and energy insecurity. Oilseed crops have been identified as key to address these challenges: they produce and store lipids in the seeds as triacylglycerols that can serve as a source of food/feed, renewable fuels, and other industrially-relevant chemicals. Therefore, improving seed oil content and composition has generated immense interest. Research efforts aiming to unravel the regulatory pathways involved in fatty acid synthesis and to identify targets for metabolic engineering have made tremendous progress. This review provides a summary of the current knowledge of oil metabolism and discusses how photochemical activity and unconventional pathways can contribute to high carbon conversion efficiency in seeds. It also highlights the importance of 13C-metabolic flux analysis as a tool to gain insights on the pathways that regulate oil biosynthesis in seeds. Finally, a list of key genes and regulators that have been recently targeted to enhance seed oil production are reviewed and additional possible targets in the metabolic pathways are proposed to achieve desirable oil content and quality.

Abstract:Gene manipulation techniques can be used to increase, decrease, or add specific proteins to the edible parts of transgenic crop plants. With some basic understanding of plant biosynthetic pathways, then, the targeting of genes encoding specific enzymes allows the direct modification of the biochemical composition of foods. At Calgene, we have engineered the chemical composition of canola vegetable oils. Transgenic canola which are otherwise exactly like regular canola plants produce seed with oils a) that are modified in average fatty acid carbon chain length, b) that are modified in content of saturated fatty acids (both lower and higher), or c) that contain structured lipids. In principle, although the gene target may not be obvious, the relative amount of lipids compared to other nutrients can be decreased or increased in foods like peanut or soybean. The oil content of some food products might be modified to enhance levels of medium chain triglycerides or to contain "fish oil" fatty acids--without the cost or olfactory disadvantages. On a broader scope, the amino acid composition of proteins in basic grains is being pursued by several groups. Specific vitamin contents such as Vitamin A or E might be enhanced in basic foodstuffs; type and content of fiber may eventually be manipulated. Specific components such as caffeine or phytic acid conceivably can be eliminated in the source plant, negating the need for processing steps that add cost and that lessen flavor and nutrition. Clearly, the biochemical compositions of foods is complex and varied. Moreover, tailoring foods to better meet our needs is an expensive and lengthy process. The best possible understanding of how nutrition promotes good health is necessary to direct and prioritize our efforts to improve plants as food sources.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract:SUMMARY Vegetable oils are rich sources of polyunsaturated fatty acids and energy as well as valuable sources of human food, animal feed, and bioenergy. Triacylglycerols, which are comprised of three fatty acids attached to a glycerol backbone, are the main component of vegetable oils. Here, we review the development and application of multiple‐level omics in major oilseeds and emphasize the progress in the analysis of the biological roles of key genes underlying seed oil content and quality in major oilseeds. Finally, we discuss future research directions in functional genomics research based on current omics and oil metabolic engineering strategies that aim to enhance seed oil content and quality, and specific fatty acids components according to either human health needs or industrial requirements.

Abstract:Certain plants and microorganisms can produce high amounts of unusual fatty acids (UFAs) such as hydroxy, conjugated, cyclic, and very long-chain polyunsaturated fatty acids, which have distinct physicochemical properties and significant applications in the food, feed, and oleochemical industries. Since many natural sources of UFAs are not ideal for large-scale agricultural production or fermentation, it is attractive to produce them through synthetic biology. Although several UFAs have been commercially or pre-commercially produced in transgenic plants and microorganisms, their contents in transgenic hosts are generally much lower than in natural sources. Moreover, reproducing this success for a wider spectrum of UFAs has remained challenging. This review discusses recent advancements in our understanding of the biosynthesis, accumulation, and heterologous production of UFAs, and addresses the challenges and potential strategies for achieving high UFA content in engineered plants and microorganisms.

Abstract:The quality of vegetables is facing new demands in terms of diversity and nutritional health. Given the improvements in living standards and the quality of consumed products, consumers are looking for vegetable products that maintain their nutrition, taste, and visual qualities. These requirements are directing scientists to focus on vegetable quality in breeding research. Thus, in recent years, research on vegetable quality has been widely carried out, and many applications have been developed via gene manipulation. In general, vegetable quality traits can be divided into three parts. First, commodity quality, which is most related to the commerciality of plants, refers to the appearance of the product. The second is flavor quality, which usually represents the texture and flavor of vegetables. Third, nutritional quality mainly refers to the contents of nutrients and health ingredients such as soluble solids (sugar), vitamin C, and minerals needed by humans. With biotechnological development, researchers can use gene manipulation technologies, such as molecular markers, transgenes and gene editing to improve the quality of vegetables. This review attempts to summarize recent studies on major vegetable crops species, with Brassicaceae, Solanaceae , and Cucurbitaceae as examples, to analyze the present situation of vegetable quality with the development of modern agriculture.
