Relation and Regulation of Development of Alveolar Type II Cell and Expression of SPmRNAs in Fetal Rabbit Lung

Abstract:Objective To explore the relation of alveolar typeⅡ cell development and SP mRNAs expression in fetal rabbit lung, and to investigate the regulatory effects of epidermal growth factor (EGF), thyrotropin releasing hormone (TRH), and dexamethasone (Dex) on them. Methods Pregnant rabbits were divided into EGF treated, TRH treated, Dex treated, and control groups with 16 in each. EGF, TRH, Dex, and saline were given intravenously respectively, at 22-24 d or 24-26 d after gestation. Ultrastructure of alveolar type Ⅱ cells in fetal lungs was observed using electron microscopy, and the levels of SP A, SP B, and SP C mRNA were detected by RT PCR and Southern blotting. Results Alveolar type Ⅱ cells of fetal lungs in EGF, TRH, and Dex treated groups contained more lamellar bodies and less glycogen than those in control group at 27 d after gestation. SP B mRNA levels from EGF, TRH, and Dex treated fetal lungs were also significantly higher than those from control group, but no significant difference of SP A and SP C mRNA levels were found among the four groups. In 25 d lung, only few type Ⅱ cell in Dex treated group contained a few small lamellar bodies, and no lamellar body was found in other treated groups and control group. However, glycogen was very abundant in every group. Also, only Dex appeared to increase the expression of SP B mRNA at a lower level, and SP B mRNA was undetectable in other groups in 25 d lung. But the expression of SP A and SP C mRNA was at a high level in all groups. Conclusion The time of expression for SP A and SP C mRNA is earlier than that of SP B mRNA in fetal rabbit lung. SP B mRNA expression is lower than SP A or SP C mRNA expression. The accumulation of detectable SP B mRNA and the increase of SP B mRNA level correspond to the appearance and the increment of lamellar body numbers in type Ⅱ cell. It suggests that the increase of SP B mRNA level may be a marker of differentiation and mature of type Ⅱ cell in fetal lung. Prenatal maternal treatment with Dex, EGF, and TRH can promote the development of type Ⅱ cell and increase the expression of SP B mRNA in rabbit fatal lung, but the up regulation of Dex may be more significant at the early stage of gestation.
