Abstract:[Objective] To clone and express an esterase gene e22 from marine bacterium Altererythrobacter epoxidivorans CGMCC 1.7731T and characterize the enzyme.[Methods] Screening of the genome of strain CGMCC 1.7731T obtained an esterase-encoding gene,e22.The gene was amplified by PCR and cloned into the expression vector pET-28a.Then,the recombinant plasmid was transformed into Escherichia coli BL21(DE3) for heterogenetic expression.After purified by affinity chromatography,the enzyme was characterized.[Results] The amino acids sequence analysis indicated E22 belongs to Family Ⅱ of lipolytic enzyme.The enzymatic property assay revealed that E22 had maximum activity using p-nitrophenyl butyrate as substrate.It was an alkaline esterase that had highest activity at pH 10.5.The optimal reaction temperature was 55 C.After incubated at 60 C for 2 h,E22 still remained over 50％ activity,which showed its moderate thermal stability.The activity of E22 was not significantly affected by the presence of 1％ methanol,1％ Triton X-100 or 0.1％ SDS.On the contrary,the addition of 10 mmol/L Ba2+ and Ca2+ would apparently inhibit the catalytic capacity of E22.[Conclusion] E22 is a novel marine esterase with excellent properties such as thermostability,and alkaline,organic solvent and detergent tolerance.These characteristics suggest its application prospects in the industrial production.

A Novel Halotolerant Thermoalkaliphilic Esterase from Marine Bacterium Erythrobacter Seohaensis SW-135

Abstract:A novel esterase gene, e69, was cloned from Erythrobacter seohaensis SW-135, which was isolated from a tidal flat sediment of the Yellow Sea in Korea. This gene is 825 bp in length and codes for a 29.54 kDa protein containing 274 amino acids. Phylogenetic analysis showed that E69 is a new member of the bacterial lipolytic enzyme family IV. This enzyme exhibited the highest level of activity toward p-nitrophenyl (NP) butyrate but little or no activity toward the other p-NP esters tested. The optimum temperature and pH of the catalytic activity of E69 were 60°C and pH 10.5, respectively. The enzyme exhibited stable activity over a wide range of alkaline pH values (7.5-9.5). In addition, E69 was found to be a halotolerant esterase as it exhibited the highest hydrolytic activity in the presence of 0.5 M NaCl and was still active in the presence of 3 M NaCl. Moreover, it possessed some degree of tolerance to Triton X-100 and several organic solvents. Through homology modeling and comparison with other esterases, it was suggested that the absence of the cap domain and its narrow substrate-binding pocket might be responsible for its narrow substrate specificity. Sequence and structural analysis results suggested that its high ratio of negatively to positively charged residues, large hydrophobic surface area, and negative electrostatic potential on the surface may be responsible for its alkaline adaptation. The results of this study provide insight into marine alkaliphilic esterases, and the unique properties of E69 make it a promising candidate as a biocatalyst for industrial applications.
The Study of Transesterification Catalyzed by Rhodobacter Sphaeroides Esterase

Abstract:In this study, a carboxyl esterase from Rhodobacter sphaeroides was introduced as a catalyst in organic solvent, and the transesterification of 1-phenethyl alcohol and vinyl acetate was served as a model reaction. It was found that changes in used solvent and temperature influence the activity of the esterase and it was observed that the highest activity was obtained when acetonitrile was served as the solvent with the catalytic temperature of 45℃. However, after four days’ exposure in organic solvent, the relative activity of the esterase decreases to 58.3%, which indicates that the esterase is partially denatured due to the toxicity of organic solvent. Six days’ reaction carried out in optimal condition shows that the molar conversation reaches to 47.9%and the E value is 64. It demonstrated that this carboxyl esterase displays high enantioselectivity toward R-1-phenetyl alcohol and can be applied to industrial manufacturing as a novel biocatalyst.

Abstract:A novel esterase gene (e25) was identified from Altererythrobacter epoxidivorans CGMCC 1.7731(T) by genome sequence screening. The e25 gene is 948 nucleotides in length and encodes a 315amino acid protein (E25) with a predicted molecular mass of 33,683 Da. A phylogenetic tree revealed that E25 belongs to the hormone-sensitive lipase (HSL) family of lipolytic enzymes. An activity assay of E25 showed that it exhibited the highest catalytic efficiency when using p-nitrophenyl caproate (C6) as a substrate. The optimum pH and temperature were determined to be approximately pH 9 and 45 degrees C, and the K-m and V-max values were 0.12mM and 1,772 mu mol/min/mg, respectively. After an incubation at 40 degrees C for 80min, E25 retained 75% of its basal activity. The enzyme exhibited good tolerance to metal cations, such as Ba2+, Ca2+, and Cu2+ (10mM), but its activity was strongly inhibited by Co2+, Ni2+, Mn2+, and Zn2+. The E25 enzyme was stimulated by glycerol and retained over 60% of its basal activity in the presence of 1% Tween-80 and Triton X-100. Overall, the activity of E25 under alkaline conditions and its organic solvent and detergent tolerance indicate that E25 could be useful as a novel industrial catalyst in biotechnological applications.
Structure Guided Protein Engineering Increases Enzymatic Activities of the SGNH Family Esterases

Abstract:Esterases and lipases hydrolyze short-chain esters and long-chain triglycerides, respectively, and therefore play essential roles in the synthesis and decomposition of ester bonds in the pharmaceutical and food industries. Many SGNH family esterases share high similarity in sequences. However, they have distinct enzymatic activities toward the same substrates. Due to a lack of structural information, the detailed catalytic mechanisms of these esterases remain barely investigated. In this study, we identified two SGNH family esterases, CrmE10 and AlinE4, from marine bacteria with significantly different preferences for pH, temperature, metal ion, and organic solvent tolerance despite high sequence similarity. The crystal structures of these two esterases, including wild type and mutants, were determined to high resolutions ranging from 1.18 Å to 2.24 Å. Both CrmE10 and AlinE4 were composed of five β-strands and nine α-helices, which formed one compact N-terminal α/β globular domain and one extended C-terminal domain. The aspartic residues (D178 in CrmE10/D162 in AlinE4) destabilized the conformations of the catalytic triad (Ser-Asp-His) in both esterases, and the metal ion Cd2+ might reduce enzymatic activity by blocking proton transfer or substrate binding. CrmE10 and AlinE4 showed distinctly different electrostatic surface potentials, despite the similar atomic architectures and a similar swap catalytic mechanism. When five negatively charged residues (Asp or Glu) were mutated to residue Lys, CrmE10 obtained elevated alkaline adaptability and significantly increased the enzymatic activity from 0 to 20% at pH 10.5. Also, CrmE10 mutants exhibited dramatic change for enzymatic properties when compared with the wide-type enzyme. These findings offer a perspective for understanding the catalytic mechanism of different esterases and might facilitate the industrial biocatalytic applications.

Abstract:High-efficiency ammonifiers are imperative for the treatment of wastewater with high organic nitrogen content. Ammonification characteristics and the corresponding protease production and activity of a Bacillus amyloliquefaciens was studied using simulated wastewater. High ammonification was observed with more than 500 mg/ L NH4+ released from tryptone medium after 18-h cultivation and almost all the organic nitrogen was transformed to NH4+-N and biomass. Single factor experiments were performed to optimize the conditions. For ammonification, pH 5, 25-37 degrees C, and a C:N ratio < 15 were optimal; for protease production, pH 7, 30 degrees C and addition of 2 mM Mn2+ were favorable, whereas pH 8, 50-60 degrees C and the presence of 1 mM Mn2+ showed the highest protease activity. Moreover, protease inhibition experiment indicated that Bacillus amyloliquefaciens DT simultaneously produces metalloprotease and serine protease. Correlation analysis indicated that protease production and anunonification were tightly coupled with cell growth during the logarithmic growth phase. However, temperature had different influence on the correlations between protease secretion and cell growth. These findings are expected to help in designing a high-efficiency reactor for the treatment of wastewater with high organic nitrogen and enhance our understanding of the production and activity of proteases during the ammonification process.

Abstract:Optically active form of α-cyano-3-phenoxybenzyl alcohol (CPBA), building block of pyrethroid insecticides, was synthesized as its acetate by the combination of anion-exchange resin-catalyzed transcyanation between m-phenoxybenzaldehyde and acetone cyanohydrin, and lipase-catalyzed enantioselective transesterification of the resulting cyanohydrin with vinyl acetate. Through a screening of enzymes, Alcaligenes sp. lipase showed the highest activity and the conversion exceeded 50%. Effects of solvents and temperatures on this reaction were studied. Among four kinds of solvent, diisopropyl ether was a best choice. The optimal temperature was 50°C. Although the external diffusion limitation could be excluded by raising the rotational speed, internal diffusion could not be ignored, since the enzyme was an immobilized one on the particles with a considerably large diameter. The e.e. values of CPBA ester were measured by polarimeter and NMR.

Abstract:The transesterification of α-d-glucose with divinylsuccinate, divinyladipate and divinylsebacate in pyridine at 55 °C for 3 days was catalyzed by an alkaline protease from Bacillus subtilis to give corresponding 6- O -vinyl glucose esters at 30%, 53% and 35% yield, respectively. The stereo-selectivity of the alkaline protease toward the α-anomer was affected by the acyl donor chain length. 6- O -Vinylsuccinyl-d-glucose was mixture of α- and β-anomers (α/β=44/56), the other two products were the pure α-d-glucose derivatives.

Abstract:This paper describes highly selective transesterification reactions, catalyzed by an alkaline protease from Bacillus subtilis in pyridine, of several mono- and di-saccharides with divinyl dicarboxylates ranging from 4 to 10 carbon atoms. A series of polymerizable vinyl fatty acid sugar esters were obtained with good selectivity and high yields. Most products had high proportions of the alpha anomer. The influences of the enzymes, solvents, temperature, and acyl donor chain length on the reaction were studied. Abstract:Catalyzed by Rhodococcus erythropolis AJ270, an amidase-containing microbial whole cell catalyst in neutral phosphate buffer at 30 degrees C, a number of prochiral alpha-substituted alpha-aminomalonamides underwent highly efficient and enantioselective hydrolytic desymmetrization to afford functionalized alpha-tetrasubstituted alpha-amino acids in 74-98% chemical yields and 94.0 to >99.5% ee. The presence of a free alpha-amino (NH2) substituent in the substrates was deemed important to ensure high biocatalytic efficiency and enantioselectivity. The synthetic application of biocatalytic desymmetrization was demonstrated by practical chemical transformations of (R)-2-amino-2-carbamoylpent-4-enoic acid to alpha-substituted serine analogues and a bioactive diamino alcohol derivative.

Abstract:Through regioselective acylation of the primary hydroxyl groups in troxerutin, a series of vinyl troxerutin esters were synthesized in nonaqueous medium. The influence parameters such as enzyme source, the amount of the enzyme, solvents, chain length of acyl donors, water content of solvents and reaction time were investigated. The results indicated that alkaline protease from Bacillus subtilis performed highest activity in pyridine with water content lower than 1%. As decreasing of the chain length of the acyl donor, the transesterification reached equilibrium more rapidly and yields were improved.

Abstract:Alkaline proteases are the most important group of industrial enzymes that hydrolyse the peptide bond of proteins into small peptides. The industrial demand of alkaline protease predominantly from the microbial origin has been recently increased and enhances the research for alkaline protease with high stability at extreme industrial conditions. Thus, this study is aimed to characterize the alkaline protease from bacterial isolate, Bacillus subtilis VBC7 screened from dairy waste dumped soil. Extracellular alkaline protease production was carried out in alkaline broth by submerged fermentation. The production medium with 10% ground nut extract at pH 10 and 40 °C enhanced the alkaline protease activity (712 U/mL) than other wastes such as coconut pulp extract and sesame seed extract. This optimized media increased the bacterial growth rate and activity of alkaline protease compared to the unsupplemented basal medium. Further, alkaline protease was partially purified and assessed their molecular weight (~30 k Da) in 12 % SDS -PAGE. The enzyme activity was observed at pH 10 and 40 °C and stable over a wide range of pH (5-12), and temperatures (10-70 °C). In addition, enzyme activity was stimulated in the presence of Mg2+, Ca2+ and Mn2+ and was unpretentious after treating with surfactants (SDS, Triton X-100, Tween 80), organic solvents (ethanol, methanol, chloroform, acetone and hexane) and protease inhibitors (EDTA and β-mercaptoethanol). These compatible features could lead the enzyme as a potential candidate among the industrial sectors.

Abstract:An extracellular lipase was purified to homogeneity with a purification factor of 5.5-fold from a bacterial strain Serratia marcescens ECU1010. The purified lipase is a dimer with two homologous subunits, of which the molecular mass is 65kDa, and the pI is 4.2. The pH and temperature optima were shown to be pH 8.0 and 45°C, respectively. Among p-nitrophenyl esters of fatty acids with varied chain length, the lipase showed the maximum activity on p-nitrophenyl myristate (C14). The lipase was activated by some surfactants such as Gum Arabic, polyvinyl alcohol (PVA) and Pg350me, but not by Ca2+. The enzyme displayed pretty high stability in many water miscible and immiscible solvents. This is a unique property of the enzyme which makes it extremely suitable for chemo-enzymatic applications in non-aqueous phase organic synthesis including enantiomeric resolution. Several typical chiral compounds were tested for kinetic resolution with this lipase, consequently giving excellent enantioselectivities (E=83 ∼>100) for glycidyl butyrate (GB), 4-hydroxy-3-methyl-2-(2-propenyl)-2-cyclopenten-1-one acetate (HMPCA), naproxen methyl ester (NME) and trans-3-(4′-methoxyphenyl) glycidic acid methyl ester (MPGM).

Abstract:The peptide bond formation of N-protected non-coded amino acids having different structures as acyl donor substrates that is catalyzed by thermoase in organic media was investigated. In these reactions, N-protected l-α-non-coded amino acids, including l-Orn, l-Cit, α-aminobutyric acid (l-α-Abu) and phenylalanine homologues, were used as the acyl donors and phenylalanine derivatives were used as the acyl acceptors. This kind of enzymatic reactions cannot be carried out in an aqueous buffer due to the rigid specificity of proteases to coded amino acids in water. The results demonstrated that the substrate specificity of proteases could be broadened in organic solvents. In addition, the factors that influenced these protease-catalyzed reactions, including structures of the substrates, water content and the bases used, were systematically studied. Our work provided important evidence for broadening the application of protease in organic synthesis.

Abstract:Using enrichment procedures, a lipolytic strain was isolated from a stinky tofu brine and was identified as Bacillus amyloliquefaciens (named B. amyloliquefaciens Nsic-8) by morphological, physiological, biochemical tests and 16S rDNA sequence analysis. Meanwhile, the key enzyme gene (named lip BA) involved in ester metabolism was obtained from Nsic-8 with the assistance of homology analysis. The novel gene has an open reading frame of 645 bp, and encodes a 214-amino-acid lipase (LipBA). The deduced amino acid sequence shows the highest identity with the lipase from B. amyloliquefaciens IT-45 (NCBI database) and belongs to the family of triacylglycerol lipase (EC 3.1.1.3). The lipase gene was expressed in Escherichia coli BL21(DE3) using plasmid pET-28a. The enzyme activity and specific activity were 250 ± 16 U/ml and 1750 ± 153 U/mg, respectively. The optimum pH and temperature of the recombinant enzyme were 9.0 and 40 °C respectively. LipBA showed much higher stability under alkaline conditions and was stable at pH 7.0–11.0. The Km and Vmax values of purified LipBA using 4-nitrophenyl palmitate as the substrate were 1.04 ± 0.06 mM and 119.05 ± 7.16 μmol/(ml min), respectively. After purification, recombinant lipase was immobilized with the optimal conditions (immobilization time 3 h at 30 °C, with 92 % enzyme recovery) and the immobilized enzyme was applied in biodiesel production. This is the first report of the lipase activity and lipase gene obtained from B. amyloliquefaciens (including wild strain and recombinant strain) and the recombinant LipBA with the detailed enzymatic properties. Also the preliminary study of the transesterification shows the potential value in biodiesel production applications.
Detergent-compatible, organic solvent-tolerant alkaline protease from Bacillus circulans MTCC 7942: Purification and characterization

Abstract:Proteases are now recognized as the most indispensable industrial biocatalyst owing to their diverse microbial sources and innovative applications. In the present investigation, a thermostable, organic solvent-tolerant, alkaline serine protease from Bacillus circulans MTCC 7942, was purified and characterized. The protease was purified to 37-fold by a three-step purification scheme with 39% recovery. The optimum pH and temperature for protease was 10 and 60 °C, respectively. The apparent molecular mass of the purified enzyme was 43 kD as revealed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The Km and Vmax values using casein-substrate were 3.1 mg/mL and 1.8 µmol/min, respectively. The protease remained stable in the presence of organic solvents with higher (>3.2) log P value (cyclohexane, n-octane, n-hexadecane, n-decane, and n-dodecane), as compared to organic solvents with lower (<3.2) log P value (acetone, butanol, benzene, chloroform, toluene). Remarkably, the protease showed profound stability even in the presence of organic solvents with less log P values (glycerol, dimethyl sulfate [DMSO], p-xylene), indicating the possibility of nonaqueous enzymatic applications. Also, protease activity was improved in the presence of metal ions (Ca(2+), Mg(2+), Mn(2+)); enhanced by biosurfactants; hardly affected by bleaching agents, oxidizing agents, and chemical surfactants; and stable in commercial detergents. In addition, a protease-detergent formulation effectively washed out egg and blood stains as compared to detergent alone. The protease was suitable for various commercial applications like processing of gelatinous film and as a compatible additive to detergent formulation with its operative utility in hard water.
