Abstract:Two new 2-arylbenzofurans, 4-(7-methoxy-3,5-dimethyl-benzofuran-2-yl)phenol (1), 4-(6-methoxy-3,5-dimethylbenzofuran-2-yl)phenol (2), together with two known 2-arylbenzofurans (3 and 4) were isolated from the stems of Nicotiana tabacum. Their structures were determined by means of HRESIMS and extensive 1D and 2D NMR spectroscopic studies. Compounds 1-4 were tested for their anti-tobacoo mosaic virus (TMV) activities and cytotoxicity activities. The results showed that compound 2 exhibited high anti-TMV activities with inhibition rates of 36.4% at the concentration of 20 mu M. This rate is higher than that of positive control. Compounds 1-4 showed moderate-to-weak cytotoxicities against some tested human tumor cell lines with IC50 values in the range of 2.2-8.4 mu M.

Abstract:Three new isobenzofurans (1-3), together with four known phenylpropanoids (4-7) were isolated from the roots of Nicotiana tabacum. Their structures were determined by means of HRESIMS and extensive 1D and 2D NMR spectroscopic studies. Compounds 1-6 were tested for their anti-tobacco mosaic virus (anti-TMV) activities and cytotoxicity activities. The results showed that compounds 5 and 6 exhibited high anti-TMV activities with inhibition rates of 35.1 and 33.4%, respectively. The cytotoxicities of compounds 1-7 against five human tumor cell lines (NB4, A549, SHSY5Y, PC3 were also tested. Compounds 1-7 showed weak inhibitory activities against some tested human tumor cell lines with IC50 values in the range of 3.8-9.6 mu M. [GRAPHICS] .

The results revealed that compounds 1 and 2 showed good inhibition with IZD of 15.2 ± 2.5, and 12.8 ± 2.2 mm, respectively.

The results revealed that compounds 1 and 2 showed high anti-TMV activity with inhibition rate of 35.8 and 32.6%. These rates are close to that of positive control. Compounds 1 and 2 also showed moderate to weak inhibitory activities against some tested human tumor cell lines with IC50 values in the range of 1.5–6.0 μM.

Abstract:Two new aromatic sesquiterpenes, 7-isopropyl-2,2,5-trimethyl-2 H -chromene ( 1 ) and 7-isopropyl-5- (methoxymethyl)-2,2-dimethyl-2 H -chromene ( 2 ), were isolated from the leaves of Nicotiana tabacum (a mutant tobacco for vitamin E synthesis gene knockout by CRISPR-Cas9 gene editing). Their structures were determined by spectroscopic methods, including extensive 1D and 2D NMR techniques. Interestingly, compounds 1 and 2 have a weak bean fragrance, and they have the potential value for use as an essence for Heat Not Burning (HNB) cigarettes.

Abstract:Three new isobenzofurans (1-3), together with four known furan derivatives (4-7) were isolated from the stems of oriental tobacco. Their structures were determined by means of HRESIMS and extensive 1D and 2D NMR spectroscopic studies. Compounds 1-7 were tested for their anti-tobacoo mosaic virus (TMV) activities and cytotoxicity activities. The results revealed that compounds 2 and 3 showed high anti-TMV activities with inhibition rates of 35.4, and 34.2%. These rates are higher than that of positive control. Other compounds also showed anti-TMV activities with inhibition rates in the range of 18.6 similar to 22.7%. These rates are close to that of positive control. Compounds 1-7 also showed moderate-to-weak inhibitory activities against some tested human tumor cell lines with IC50 values in the range of 2.8-9.3 mu M.

Abstract:Four types of 2-arylbenzofurans (1-4), including a new compound, tobarylbenzofuran A (1), were isolated from the leaves of oriental tobacco (a variety of Nicotiana tabacum L). Their structures were elucidated by spectroscopic methods, including extensive 1D and 2D NMR techniques. The two compounds were tested for their cytotoxicity against five human tumors (NB4, A549, SHSY5Y, PC3, and MCF7) cell lines. The results showed that compound 1 showed notable inhibitory effect against SHSY5Y and MCF7 cell lines, with IC50 values of 3.5 and 1.8 mu M, respectively.

The results showed that compounds 2, 3, and 5 exhibited high anti-TMV activity with inhibition rates of 33.6, 35.8, and 36.7%. Abstract:Two new sesquiterpene glycosides, 11R, 12-dihydroxy-6(7)-spirovetiven-8-one-9-O-beta-D-glucopyranoside (1) and rishitin M1-13-O-beta-D-glucopyranoside (2), have been isolated from the leaves of Nicotiana tabacum. Their structures were established by spectroscopicmethods including H-1, C-13, and 2D NMR. The absolute configuration of the side chain in compound 1 was determined as R by NOESY and NOE difference spectra.

Abstract:Two new megastigmane glycosides, (6 R ,7 E ,9 R )-3-oxo- α -ionyl-9-O- α -L-rhamnopyranosyl-(1′′→4′)- β -D-glucopyranoside ( 1 ) and (6 R ,7 E ,9 R )-3-oxo- α -ionyl-9-O- β -D-glucopyranosyl-(1′′→6′)- β -D-glucopyranoside ( 2 ), together with six known analogues ( 3 – 8 ) were isolated from the leaves of Nicotiana tabacum . The structures of all metabolites were determined by comprehensive analysis of NMR and MS spectroscopic data as well as by comparison with those of previously reported. The in vitro anti-inflammatory activity of all isolates was evaluated using a lipopolysaccharide (LPS)-induced RAW264.7 cell inflammatory model, and the compounds 1 , 3 , 7 , and 8 exhibited inhibition of LPS-induced NO production in RAW264.7 macrophage cells with IC 50 values of 42.3-61.7 μM (positive control, dexamethasone, IC 50 = 21.3 ± 1.2 μM).

Abstract:Three new polymeric isopropenyl benzofurans, 4-methyl-2,4-bis(5,6-dimethoxy-2-benzofuranyl)-1-pentene, stenocephalin A (1), 4,6-dimethyl-2,4,6-tri(5,6-dimethoxy-2-benzofuranyl)-1-heptene, stenocephalin B (2) and 4,6,8-trimethyl-2,4,6,8-tetra(5,6-dimethoxy-2-benzofuranyl)-1-nonene, stenocephalin C (3), together with seven known compounds (4-10) were isolated from the roots of Ligularia stenocephala. The structures of the new compounds were elucidated on the basis of spectral evidence, especially on 2D NMR. In addition, the cytotoxic activity and the anti-bacterial activity of compounds 2, 3, 5 and 6 were tested.
