Interplay of Repeat Expansion Genes in Spinocerebellar Ataxia Type 3 (P5.079)

Abstract:Objective: To investigate repeat alterations in non-pathogenic allele of ATXN3 gene and other CAG repeat expansion genes in spinocerebellar ataxia type 3 (SCA3). Background: SCA3 is caused by the pathogenic CAG repeat expansion in ATXN3 , and is the most common subtype of SCA in China and worldwide. Pathogenic expansion in SCA3 could interfere with the DNA repair mechanism, leading to repeat instability, and finally causing phenomenon of anticipation and somatic mosaicism. However, whether this pathogenic expansion could lead to repeat alterations in trans in the non-pathogenic allele of ATXN3 or other repeat expansion genes remain unknown. Design/Methods: We recruited 685 molecularly confirmed SCA3 patients and 300 unrelated controls from China, and we studied the length of repeat in the non-pathogenic allele of ATXN3 and in 8 other CAG repeat expansion genes ( ATXN1 , ATXN2 , CACNA1A , ATXN7 , PPP2R2B , TBP , ATN1 , and HTT ) in these subjects. The distribution of CAG repeat and frequency of large normal alleles (LNA) were analyzed using Mann-Whitney U test and Chi-square test, respectively, between SCA3 patients and controls. Results: We found that SCA3 patients have a decreased alteration of repeat in non-pathogenic allele of ATXN3 (repeat distribution: 21.70 ± 7.10 in controls vs. 19.92 ± 6.97 in SCA3 patients, p = 0.000; LNA frequency: 27.52% in controls vs. 20.53% in patients, p = 0.001). Consistently, SCA3 patients also have a shorter CAG repeat length in HTT (repeat distribution: 18.10 ± 2.10 in controls vs. 17.24 ± 1.96 in patients, p = 0.000; LNA frequency: 2.48% in controls vs. 1.02% in SCA3 patients, p = 0.006). We did not find differences in the rest of the repeat expansion genes. Conclusions: The findings provide novel evidence to prove biological connections among CAG repeat expansion related genes and demonstrate the complex interplay of repeat expansion genes.
