Abstract:Leaf senescence happen in the late stage of leaf development.During the process of leaf senescence,chlorophyll and other macromolecules are degraded,photosynthetic capacity declines and nutrients of the degraded products tissues are transferred from senescing to young tissues and reproductive organs.Leaf senescence is influenced by the leaf developmental stage and environmental stresses,like dark,drought,salinity,low temperature,ozone,nutrient deficiency and pathogen infection,etc.Reactive oxygen species(ROS) accumulates during leaf senescence with reduced antioxidase activities(SOD,CAT and APX).Unfavorable conditions might accelerate and lead to a premature senescence,inducing expression of senescence associated genes(SAGs),speeding up the plant maturation and decreasing the crop yield.The measures utilized in production practice,i.e.nitrogen management,plant intensity,and chemical regulator are involved in the process.In this review,we summarized the advancement in this field.
Shade Effects on Peanut Yield Associate with Physiological and Expressional Regulation on Photosynthesis and Sucrose Metabolism

Abstract:Intercropping improves land utilization with more crops grown together; however, shorter crops in intercropping experience stress, being shaded by the taller crops. Systematic changes in phenotype, physiology, yield, and gene regulation under shade stress in peanut are largely unknown, although shade responses have been well analyzed in model plants. We exposed peanut plants to simulated 40% and 80% shade for 15 and 30 days at the seedling stage, flowering stage, and both stages. Shade caused the increased elongation growth of the main stem, internode, and leaf, and elongation was positively associated with auxin levels. Shade stress reduced peanut yield. Further comparative RNA-seq analyses revealed expressional changes in many metabolism pathways and common core sets of expressional regulations in all shade treatments. Expressional downregulation of most genes for light-harvesting and photosynthesis agreed with the observed decreased parameters of photosynthesis processes. Other major regulations included expressional downregulation of most core genes in the sucrose and starch metabolism, and growth-promoting genes in plant hormone signal pathways. Together, the results advance our understanding of physiological and molecular regulation in shade avoidance in peanut, which could guide the breeding designing in the intercropping system.

Optimized nitrogen application ameliorates the photosynthetic performance and yield potential in peanuts as revealed by OJIP chlorophyll fluorescence kinetics

Abstract:Background: Nitrogen (N) is a crucial element for increasing photosynthesis and crop yields. The study aims to evaluate the photosynthetic regulation and yield formation mechanisms of different nodulating peanut varieties with N fertilizer application. Method: The present work explored the effect of N fertilizer application rates (N0, N45, N105, and N165) on the photosynthetic characteristics, chlorophyll fluorescence characteristics, dry matter, N accumulation, and yield of four peanut varieties. Results: The results showed that N application increased the photosynthetic capacity, dry matter, N accumulation, and yield of peanuts. The measurement of chlorophyll a fluorescence revealed that the K-phase, J-phase, and I-phase from the OJIP curve decreased under N105 treatment compared with N0, and WOI, ET0/CSM, RE0/CSM, ET0/RC, RE0/RC, φPo, φEo, φRo, and Ψ0 increased, whereas VJ, VI, WK, ABS/RC, TR0/RC, DI0/RC, and φDo decreased. Meanwhile, the photosystem activity and electron transfer efficiency of nodulating peanut varieties decreased with an increase in N (N165). However, the photosynthetic capacity and yield of the non-nodulating peanut variety, which highly depended on N fertilizer, increased with an increase in N. Conclusion: Optimized N application (N105) increased the activity of the photosystem II (PSII) reaction center, improved the electron and energy transfer performance in the photosynthetic electron transport chain, and reduced the energy dissipation of leaves in nodulating peanut varieties, which is conducive to improving the yield. Nevertheless, high N (N165) had a positive effect on the photosystem and yield of non-nodulating peanut. The results provide highly valuable guidance for optimizing peanut N management and cultivation measures.

Abstract:Leaf senescence, the final stage of leaf development, is one of the adaptive mechanisms formed by plants over a long period of evolution. Leaf senescence is accompanied by various changes in cell structure, physiological metabolism, and gene expressions. This process is controlled by a variety of internal and external factors. Meanwhile, the genes and plant hormones involved in leaf aging affect the quality, yield and stress resistance in horticultural plants. Leaf senescence mediated by plant hormones affected plant quality at both pre-harvest and post-harvest stages. Exogenous plant growth regulators or plant hormone inhibitors has been applied to delay leaf senescence. Modification of related gene expression by over-expression or antisense inhibition could delay or accelerate leaf senescence, and thus influence quality. Environmental factors such as light, temperature and water status also trigger or delay leaf senescence. Delaying leaf senescence could increase chloroplast lifespan and photosynthesis and thus improve source strength, leading to enhanced yield. Accelerating leaf senescence promotes nutrient redistribution from old leaves into young leaves, and may raise yield under certain circumstances. Many genes and transcriptional factors involved in leaf senescence are associated with responses to abiotic and biotic stresses. WRKY transcriptional factors play a vital role in this process and they could interact with JA signalling. This review summarized how genes, plant hormones and environmental factors affect the quality, yield. Besides, the regulation of leaf senescence holds great promise to improving the resistance to plant biotic and abiotic stresses.

Abstract:A recombinant inbred line (RIL) population with 215 lines derived from a cross between a highly yield line Zheng8903 (female parent) and a variety Yuhua No. 4 (male parent) was used in this research. The RILs population was grown in three environments to make a genetics analysis of yield from F-7 to F-9. Jointly segregating analysis was used to analyze the genetics of yield in the combination of Zheng8903xYuhua No. 4 using the method of major gene plus polygene mixed inheritance model. The results indicated that the optimum model of yield trait was two major genes plus polygenes mixed inheritance models and its expression could be greatly influenced by different environments. The heritability of major gene for the yield trait in the RILs was 70.73, 40.29 and 37.79% at Zhengzhou, Sanya and Yuanyang, respectively. The yield had significant major gene effect and the results implied that not only should the two major genes' effects be considered but also the polygene's effect should be considered in breeding to increase peanut yield.

Abstract:Field experiments with a peanut cultivar(Arachis hypogaea L.cv Qinghua5) under plastic film mulching were systematically conducted to investigate the effects of different sowing date photosynthetic characteristics and Yield in Peanut.The results indicated that: there were significant differences in the photosynthetic characteristics and yield of peanut under different sowing dates.In the sowing date of 10 May,Schlorophyll SPAD,LAI,Photosynthesis rate of single leaf and yield were biggest in all treatments.when the sowing time was later or earlier,their contents would be reduced.This study considered that 10th May is the suitable time for sowing peanut to get abundant harvest.

Abstract:Introduction: In maize and peanut intercropping, shading emerges as a critical factor for restricting peanut growth, yield, and quality. Methods: This study investigated the impact of 30% shade on shade-tolerant [Huayu 22 (HY22) and Fuhua 12 (FH12)] and shade-sensitive [Nonghua 11 (NH11) and Nonghua 5(NH5)] peanut varieties, with non-shaded condition as the control (CK). The effects of shade stress on plant morphology, photosynthetic characteristics, dry-matter accumulation, chloroplast ultra-microstructure, yield, and quality of different shade-tolerant peanut varieties were examined. Results: Compared to that in the control, shade stress led to an elongation of the main stem, shortening of the lateral branches, and reduction in the leaf area. However, these changes were less significant in the shade-tolerant than in the shade-sensitive peanut varieties, with minimal effect on the elongation of the main stem height and shortening of the lateral branches. Differences in leaf area became significant during the later stages of shade stress, particularly pronounced in the shade-sensitive peanut varieties. To enhance light capture by leaves, the shade-tolerant peanut varieties exhibited increased chlorophyll content and chloroplast grain-layer numbers. The decrease in the chlorophyll a/b ratio was more pronounced in the shade-tolerant than in the shade-sensitive peanut varieties, with significant differences. However, reduced activities of ribulose 1,5-biphosphate (RuBP) carboxylase/oxygenase and fructose 1,6-biphosphate aldolase (FBA) resulted in decreased net photosynthetic rates, particularly evident in the shade-sensitive peanut varieties during the late shade period. Shade stress led to decreased dry-matter accumulation, reduced weight of 100 fruits and kernels, and a significant decline in yield in the shade-sensitive cultivars. Shading also affected peanut-kernel quality. Compared with that in the control, the protein content increased and amino-acid (except cysteine) content decreased in the shade-tolerant cultivars. Discussion: Under shade stress, shade tolerant peanut varieties have increased the yield by improving the photosynthetic efficiency, which provided a reference for rational selection of shade tolerant peanut varieties in maize and peanut intercropping system.

Abstract:Peanut is an important oil and economic crop in China. Currently, the domestic production of peanut remains far below the needs of consumers. Thus, further improving the yield per unit area is a crucial approach to meet the rising market demand. Seed size related traits are important agronomic traits in peanut, fundamentally contributing to improving yield per unit area. This review summarized the research progress on the regulatory pathways of seed size in plants, molecular markers, genetic linkage map construction, and QTL mapping of seed size related traits in peanut. We discussed the frontline challenges and opportunities for the coming researches of peanut seed related traits and the perspectives of yield improvement in peanut.

Abstract:Peanut, as an oilseed, have high nutritional value for humans and livestock and poultry. The seed yield (SY) of peanut decreases under drought stress conditions, and the zinc (Zn) nano-fertilizer can reduce the negative impacts of water deficiency on peanut plants and improves its SY. This experiment carried out as split plot based on randomized complete block design with three replications in Astara, Iran, during the 2018 and 2019 cropping seasons. Three irrigation regimes at 50 mm (IR 50 ), 75 mm (IR 75 ) and 100 mm (IR 100 ) evaporation from class A evaporation pan in main plot and three levels of Zn nano-chelate, including 0.5 g. l −1 ; (Zn 0.5 ), 1 g. l −1 ; (Zn 1 ) and 1.5 g. l −1 ; (Zn 1.5 ) in sub plot, comprised experimental treatments. The results indicated that the highest pod yield (PY), SY, oil yield (OY) and protein yield (PRY) of peanut (3633, 2900, 1582 and 826 kg. ha −1 , respectively) were obtained in response to Zn nano-chelate with a dose of 1.0 g. l −1 in IR 50 treatment. Nonetheless, under foliar spraying of Zn 1 and severe drought stress conditions (IR 100 ) were observed the highest proline (14.84 μmol g −1 FW), superoxide dismutase (53.42 Unit mg −1 protein min −1 ), catalase (72.87 nmol mg −1 protein min −1 ), peroxidase (64.17 nmol mg −1 protein min −1 ), soluble sugar content (172.04 mg g −1 FW), soluble protein content (114.16 mg g −1 DW) and anthocyanin pigments (11.20 μmol g −1 FW). Generally, application of Zn 1 treatment, through increasing antioxidant enzymes activity and non-enzymatic antioxidants, improved peanut SY. Hence, foliar application of Zn nano-chelate could be recommended to enhance peanut yield.

Abstract:Peanut production plays a crucial role in global food security, particularly in developing countries, where it provides essential nutrition and income. This paper examines the optimization of peanut production through genetic advancements, climate adaptation strategies, and sustainable practices. The primary objective is to increase yields by addressing challenges related to climate change, pests, and resource constraints. Globally, peanut production is hindered by rising temperatures, irregular rainfall, and declining soil quality, impacting both yield and quality. Developing countries, especially in Africa and Asia, face additional challenges, such as limited access to advanced agricultural technologies, inadequate infrastructure, and insufficient support for smallholder farmers. The vital issues include genetic vulnerabilities to pests, climate stress, and inefficient water use. Recent genetic research has provided insights into breeding more resilient, drought-resistant varieties, offering hope for improving yields, despite environmental challenges. The adoption of climate adaptation strategies, precision farming, and integrated pest management is essential for boosting productivity. These, along with optimized irrigation and nutrient management, have significantly impacted peanut production in resource-limited settings. Additionally, drought-resistant varieties have proven crucial, enabling farmers to increase resilience and yields in areas facing climate stress. In conclusion, optimizing peanut production requires continued investment in genetic advancements, infrastructure, and sustainable practices. Future efforts should focus on improving climate adaptation and sustainable farming techniques for long-term success.

Abstract:Peanut is an important cash crop used in oil, food and feed in our country. The rapid development of sequencing technology has promoted the research on the related aspects of peanut genetic breeding. This paper reviews the research progress of peanut origin and evolution, genetic breeding, molecular markers and their applications, genomics, QTL mapping and genome selection techniques. The main problems of molecular genetic breeding in peanut research worldwide include: the narrow genetic resources of cultivated species, unstable genetic transformation and unclear molecular mechanism of important agronomic traits. Considering the severe challenges regarding the supply of edible oil, and the main problems in peanut production, the urgent research directions of peanut are put forward: The de novo domestication and the exploitation of excellent genes from wild resources to improve modern cultivars; Integration of multi-omics data to enhance the importance of big data in peanut genetics and breeding; Cloning the important genes related to peanut agronomic traits and analyzing their fine regulation mechanisms; Precision molecular design breeding and using gene editing technology to accurately improve the key traits of peanut.

Abstract:Leaf senescence can impact crop production by either changing photosynthesis duration,or by modifying the nutrient remobilization efficiency and harvest index.The doubling of the grain yield in major cereals in the last 50 years was primarily achieved through the extension of photosynthesis duration and the increase in crop biomass partitioning,two things that are intrinsically coupled with leaf senescence.In this review,we consider the functionality of a leaf as a function of leaf age,and divide a leaf’s life into three phases:the functionality increasing phase at the early growth stage,the full functionality phase,and the senescence and functionality decreasing phase.A genetic framework is proposed to describe gene actions at various checkpoints to regulate leaf development and senescence.Four categories of genes contribute to crop production:those which regulate （I） the speed and transition of early leaf growth,（II） photosynthesis rate,（III） the onset and （IV） the progression of leaf senescence.Current advances in isolating and characterizing senescence regulatory genes are discussed in the leaf aging and crop production context.We argue that the breeding of crops with leaf senescence ideotypes should be an essential part of further crop genetic improvement.

Abstract:Peanuts ( Arachis hypogaea ) are an essential oilseed crop known for their unique developmental process, characterized by aerial flowering followed by subterranean fruit development. This crop is polyploid, consisting of A and B subgenomes, which complicates its genetic analysis. The advent and progression of omics technologies—encompassing genomics, transcriptomics, proteomics, epigenomics, and metabolomics—have significantly advanced our understanding of peanut biology, particularly in the context of seed development and the regulation of seed-associated traits. Following the completion of the peanut reference genome, research has utilized omics data to elucidate the quantitative trait loci (QTL) associated with seed weight, oil content, protein content, fatty acid composition, sucrose content, and seed coat color as well as the regulatory mechanisms governing seed development. This review aims to summarize the advancements in peanut seed development regulation and trait analysis based on reference genome-guided omics studies. It provides an overview of the significant progress made in understanding the molecular basis of peanut seed development, offering insights into the complex genetic and epigenetic mechanisms that influence key agronomic traits. These studies highlight the significance of omics data in profoundly elucidating the regulatory mechanisms of peanut seed development. Furthermore, they lay a foundational basis for future research on trait-related functional genes, highlighting the pivotal role of comprehensive genomic analysis in advancing our understanding of plant biology.

Nitrogen application in pod zone improves yield and quality of two peanut cultivars by modulating nitrogen accumulation and metabolism

Abstract:Cultivated peanut (Arachis hypogaea L.) represents one of the most important oil and cash crops world-widely. Unlike many other legumes, peanuts absorb nitrogen through their underground pods. Despite this unique feature, the relationship between yield and nitrogen uptake within the pod zone remains poorly understood. In our pot experiment, we divided the underground peanut part into two zones-pod and root-and investigated the physiological and agronomic traits of two peanut cultivars, SH11 (large seeds, LS) and HY23 (small seeds, SS), at 10 (S1), 20 (S2), and 30 (S3) days after gynophores penetrated the soil, with nitrogen application in the pod zone. Results indicated that nitrogen application increased pod yield, kernel protein content, and nitrogen accumulation in plants. For both LS and SS peanut cultivars, optimal nitrogen content was 60 kg·hm- 2, leading to maximum yield. LS cultivar exhibited higher yield and nitrogen accumulation increases than SS cultivar. Nitrogen application up-regulated the expression of nitrogen metabolism-related genes in the pod, including nitrate reductase (NR), nitrite reductase (NIR), glutamine synthetase (GS), glutamate synthase (NADH-GOGAT), ATP binding cassette (ABC), and nitrate transporter (NRT2). Additionally, nitrogen application increased enzyme activity in the pod, including NR, GS, and GOGAT, consistent with gene expression levels. These nitrogen metabolism traits exhibited higher up-regulations in the large-seeded cultivar than in the small-seeded one and showed a significant correlation with yield in the large-seeded cultivar at S2 and S3. Our findings offer a scientific basis for the judicious application and efficient utilization of nitrogen fertilization in peanuts, laying the groundwork for further elucidating the molecular mechanisms of peanut nitrogen utilization.
Dynamic changes of seed development, oil accumulation and fatty acid composition in peanut under soil water deficit

Abstract:Soil water deficit is a key environmental factor limiting peanut yield and quality, which can occur at any growth stage of peanut. But the exact mechanism of soil water deficit affecting the formation of peanut yield and quality remains unclear. In this study, the seed development, yield components, oil accumulation and fatty acid composition of common (HY25; FH18) and high oleic acid varieties (KN71; HY52) under soil water deficit throughout the growth period were investigated. It was found that the decrease of pod number and 100-pod weight per plant was the main factor leading to the reduction in peanut yield under soil water deficit. The number of oil bodies, maximum oil accumulation rate and oil content were significantly reduced, especially in drought-sensitive peanut varieties. The down-regulation of enzyme activities on the Kennedy pathway was the main factor hindering oil synthesis. Peanut varieties with lower levels of FAD2 transcripts might more sensitive to drought stress in terms of fatty acid metabolism. Under soil water deficiency, high oleic acid peanut oleate synthase activity was reduced, oleic acid metabolizing enzyme activity was elevated, which lead to decreased oleic acid content and the ratio of oleic acid to linoleic acid (O/L), and impaired lipid quality. Among them, the lipid quality of HY52 was most severely compromised. In contrast, the common varieties exhibited opposite enzyme activity patterns, with increases in oleic acid content and O/L, and improved lipid quality. This study elucidated the response mechanism of peanut grain development and oil metabolism to soil water deficit, which can provide theoretical basis and technical support for realizing high quality and stable yield of peanut under adversity.

Abstract:Background In China, double-seed (DS) sowing (i.e., sowing two seeds per hole) has been conventionally performed towards the erect-plant-type peanuts to increase the low germination rate due to poor seed preservation conditions. However, the corresponding within-hole plant competition usually limits the subsequent plant growth and the final yield. We developed a high-yield cultivation system of single-seed (SS) precision sowing to solve this paradox, saving 20% of seeds and increasing yields by more than 10% relative to the conventional DS sowing. Methods To explore the mechanisms of these two different cropping patterns in peanut yields, we conducted transcriptomic and physiological comparisons in the seeding plant leaf and root tissues between SS precision sowing and standard DS sowing treatments. Results After assembly, each library contained an average of 43 million reads and generated a total of 523,800, 338 clean reads. After GO and Kyoto Encyclopedia of Genes and Genomes pathway analysis, we found the key genes for biotic and abiotic stress showed higher expression in roots of plants grown under the SS precision sowing treatment, including genes encoding disease resistance, oxidation-reduction, hormone related, and stress response transcription factors and signaling regulation proteins. In particular, the resveratrol synthesis genes related to stress and disease resistance appeared induced in roots under the SS sowing treatment. Conclusion These data indicated that Aspergillus flavus resistance and stress tolerance in roots under SS precision sowing were enhanced compared with roots under the DS sowing treatment. This work benefits the development of underground pods and thus increasing peanut yields.

Abstract:Better management of N fertilizer is essential for improving crop productivity. Wheat (Triticum aestivum L.)-peanut (Arachis hypogaea L.) relay intercropping rotation systems are a mainstay of the measures to improve the economic and food security situation in China. Therefore, a 2-year field study (2015-2017) was conducted to evaluate the effect of different N fertilizer management regimes on the photosynthetic characteristics and uptake and translocation of N in peanut in the wheat-peanut rotation system. We used common compound fertilizer (CCF) and controlled-release compound fertilizer (CRF) at the same N-P2O5-K2O proportion (The contents of N, P2O5, and K2O in the two kinds of fertilizer were 20, 15, and 10%, respectively.). The fertilizer was applied on the day before sowing, at the jointing stage or the flag leaf stage of winter wheat, and at the initial flowering stage of peanut in various proportions, with 0 kg N ha-1 as the control. Results showed that split applications of N significantly increased leaf area index (LAI) and chlorophyll content and improved photosynthetic rate, thus increasing the pod yield of peanut. Topdressing N at the jointing stage (S1) or at the flag leaf stage of wheat (S2) and supplying part of the N at the initial flowering stage of peanut increased pod yield. Withholding N until the flag leaf stage (S2) did not negatively affect wheat grain yield; however, it increased N accumulation in each organ and N allocation proportions in the peanut pod, ultimately improving pod yield. With the same N-P2O5-K2O proportion and equivalent amounts of nutrient, CRF can decreased malondialdehyde (MDA) and maintain a relatively high LAI and chlorophyll content at the late growth stage of peanut, prolong the functional period of peanut leaves and delay leaf senescence, resulting in an increase of pod yield over that with CCF. At S1, CRF resulted in a better pod yield than CCF by 9.4%, and at S2 it was 12.6% higher. In summary, applying N fertilizer in three splits and delaying the topdressing fertilization until the flag leaf stage of winter wheat increases total grain yields of wheat and peanut. This method could therefore be an appropriate N management strategy for wheat-peanut relay intercropping rotation systems in China.

Abstract:Drought stress and nutrient deficiency are two main factors that restrict the growth and yield of peanut (Arachis hypogaea L.) in arid and semi-arid regions. However, the effects of N fertilizer on root growth and pod yield of peanut under different water conditions are unclear. The growth, root morphology, and pod yield of peanut were studied under different water and N combinations. Two soil water treatments were tested: Well-watered (WW, 80%-85% field capacity) and drought stress conditions (DS, 55%-60% field capacity). Three N treatments were tested: no N application (NN), moderate N (MN, 90 kg ha-1), and high N (HN, 180 kg ha-1). Results showed that pod yield of peanut was limited by drought and N deficiency. Compared with NN, peanut shoot DM and pod yield were improved by MN under DS, with an average increase range of pod yield of 16.2% in the 2 yr. The annual average root length density (RLD) in the deeper soil layer (40-100 cm) was 5.5% higher under DS-MN than DS-NN treatment. The N use efficiency (NUE) was 12.3% higher under WW than DS treatment, and the NUE of MN was significantly higher than that of HN under different water treatments. Root lengths in 40-60 and 60-80 cm soil layers were also positively correlated with pod yield (r = 0.66** and 0.47**, respectively) and NUE (r = 0.60* and 0.85**, respectively). In this study, root growth in deeper soil was improved by MN application under DS, thus increasing yield and NUE of DS conditions.
