Abstract:Objective: To investigate the influence of lead exposure on the immune function of lymphocytes and erythrocytes in preschool children. Materials and methods: A group of 217 children three to six years of age from a rural area were given a thorough physical examination and the concentration of lead in blood samples taken from each subject was determined. The indices of lymphocyte immunity (CD 3 + CD 4 + , CD 3 + CD 8 + CD 4 + CD 8 + , CD 3 − CD 19 + ) and erythrocyte immunity (RBC-C 3 b, RBC-IC, RFER, RFIR, CD 35 and its average fluorescence intensity) of 40 children with blood lead levels above 0.483 μmol/L were measured and compared with a control group. Results: The blood lead levels of the 217 children ranged from 0.11 μmol/L to 2.11 μmol/L. The CD 3 + CD 4 + and CD 4 + CD 8 + cells were lower ( P <0.01) and the CD 3 + CD 8 + cells were higher in the lead-poisoned subjects than those in the control group ( P <0.05). CD 3 + and CD 3 − CD 19 + did not show significant differences. Although the RBC-C 3 b rosette forming rate was lower and the RBC-IC rosette forming rate was higher in the lead-poisoned group, this difference could not be shown to be statistically significant ( P >0.05). RFIR was found to be lower in the lead-poisoned group ( P <0.01). Compared with the control group, the positive rate of CD 35 was not found to be significantly different in a group of 25 lead-poisoned children ( P >0.05), while the average fluorescence intensity was lower in the lead-poisoned group ( P <0.05). Conclusion: Lead exposure can result in impaired immune function of T lymphocytes and erythrocytes in preschool children.

Abstract:OBJECTIVE:To observe the influence of lead exposure on the activity of nitric oxide synthase (NOS) in different brain regions of rat. METHODS:By establishing a series of rat models exposed to different low levels of lead (drinking water containing 0.025%, 0.050%, 0.075% of lead acetate) during developing period, NOS activities in hippocampus, cerebellum, cerebral cortex and brain stem were studied. RESULTS:On the 21st day after birth, NOS activities in hippocampus of three levels of lead exposed groups [(1.53 +/- 0.20), (1.66 +/- 0.23), (1.88 +/- 0.32) U/mg pro respectively], and in cerebellum [(0.87 +/- 0.24), (0.85 +/- 0.09), (0.91 +/- 0.18) U/mg pro respectively] were significantly lower than those of control group [(2.36 +/- 0.18), (1.41 +/- 0.18) U/mg pro, respectively, P < 0.01]. NOS activities in cerebral cortex of 0.075% group [at 7, 14, 21 d of age [(1.29 +/- 0.14), (1.03 +/- 0.15), (0.69 +/- 0.10) U/mg pro] were significantly lower than those in control group [(2.54 +/- 0.31), (1.64 +/- 0.22), (1.24 +/- 0.14) U/mg pro respectively], and 0.025% group [(2.42 +/- 0.19), (1.59 +/- 0.17), (1.27 +/- 0.12) U/mg pro respectively], and 0.050% group [(2.56 +/- 0.53), (1.77 +/- 0.19), (1.24 +/- 0.10) U/mg pro respectively, P < 0.05]. There were no significant differences among control, 0.025%, and 0.050% groups (P > 0.05). Lead exposure had no influence on NOS activity in brain stem at the same age (P > 0.05). CONCLUSION:NOS activities in hippocampus, cerebellum and cerebral cortex were inhibited by low level lead exposure and the degree of the effect was related to Pb exposure time and/or level of Pb exposed.
Study on the Neurotoxic Effects of Low-Level Lead Exposure in Rats

Abstract:Objective: To investigate effects of developmental lead exposure on nitric oxide synthase (NOS) activity in different brain regions and on N-methyl-D-aspartate (NMDA) receptor mRNA expression in the hippocampus of rats. On the basis of these observations, we explored possible mechanisms by which lead, exposure leads to impaired learning and memorizing abilities in children. Methods: A series of rat animal models exposed to low levels of lead during the developing period was established (drinking water containing 0.025%, 0.05% and 0.075% lead acetate). NOS activities in the hippocampus, the cerebral cortex, the cerebellum and the brain stem were determined with fluorescence measurement and levels of mRNA expression of the NMDA receptor 2A (NR2A) subunit and NMDA receptor 2B (NR2B) subunit in the rat hippocampus were measured with Retro-translation (RT-PCR). Results: There were no differences in the body weight of rat pups between any of the groups at any given time ( P >0.05). The blood lead level of Pb-exposed rat pups showed a systematic pattern of change: at 14 d of age, it was lower than that at 7 d of age, then rising to the peak level at 21 d and finally falling to lower levels at 28 d. The hippocampal NOS activities of lead-exposed groups were all lower than that of the control group on the 21 st and 28th day ( P <0.01). NOS activities in the cerebellum of lead-exposed groups were all lower than that of the control group on the 21st, and 28th day ( P <0.001) and the NOS activity of the 0.025% group was significantly lower than that of the 0.05% groups on the 28th day ( P <0.05). NOS activity in the cerebral cortex of the 0.075% group was significantly lower than that of the control, 0.025% and 0.05% groups on the four day spans ( P <0.001). There was no significant difference of NOS activity in the brain stem between any lead-exposed group and the control group on the four day spans. In the 0.05% and the 0.075% groups the level of NR2A mRNA expression was higher than that in the control group at 7 d and 14 d of age ( P <0.05). In the 0.025% group, the level of NR2A was found to be higher than that in the control group at 7 d of age only ( P <0.05). No significant differences were found for the levels of NR2B mRNA expression between any of the groups at any given time. Conclusions: NOS activity in the hippocampus, the cerebral cortex and the cerebellum are inhibited by lead exposure. The degree of the inhibitory effect depends on the time span of exposure and the lead concentration. Developmental low-level lead exposure was found to raise the level of NR2A mRNA expression in the hippocampus of rats. Developmental low-level lead exposure does not affect the level of NR2B mRNA expression in the hippocampus.

Abstract:The effects of environmental lead on the immune system of young children were assessed by determining the peripheral blood lymphocytes CD3+, CD4+, CD8+, B(CD19+) counts, and natural killer (CD16+CD56+) cells in 35 preschool children whose mean blood lead level was 140.6 microg/L. The results were compared to an age- and sex-matched control group with a mean blood lead level of 64.3 microg/L. Compared to the controls, a significant reduction in the percentage of CD4+ cells and a significant increase of CD8+ cells were seen in the high-lead group. The negative correlation between the percentage of CD4+ cells and blood lead levels was found to be significant (p<0.01). These results suggest that exposure to environmental lead might result in alterations in the immune function of young children.

Abstract:Children were divided into two groups based on their blood lead levels(< 10 or > or = 10 mg/dl). The mental development indexes(PDI, VIQ, PIQ and FIO) were significantly decreased in the high lead group. Simple correlation analysis indicated that there was a negative association between blood lead levels and PDI, VIQ, PIQ and FIO(r = -0.1458, -0.3330, -0.2316 and -0.3133 respectively). But multiple regression analysis showed that the indexes of mental development were not associated with blood lead levels. It is concluded that lead is probably one of the risk factors hindering mental development. But the effect is weak while the blood lead level is not high enough.

Abstract:OBJECTIVE:To know the blood lead levels (BLL) of preschool children in Zhejiang Province between 1997 to 2003.METHODS:The BLL of preschool children was investigated in this province with cluster sampling in 2003, and compared with the results with that of 1997. The BLL was determined with graphite oven atom absorption spectrography method.RESULT:The average BLL of 2013 preschool children in 2003 was (0.34 +/- 0.13) micromol/L, similar with that of 1997 [(0.35 +/- 0.26)micromol/L (P >0.05)]. There were 274 children with BLL >or= 0.483 micromol/L, and the rate of lead poisoning was 13.61 %, lower than that of 1997 (when there were 448 children with BLL >or= 0.483 micromol/L, and the rate of lead poisoning was 23.84%). In 2003, the rates of lead poisoning among preschool children of Hangzhou, Shangyu, Zhoushan and Changshan were 7.69%, 15.37%, 10.87% and 20.15 %, respectively. There were significant differences among the four areas (P < 0.01). In comparison with the results of 1997, the lead poisoning rates of Hangzhou and Shangyu children decreased significantly, while that of Changshan children increased significantly (P <0.01), and that of Zhoushan children didn't change significantly. The highest rate of lead poisoning was found in children of 4-6 years group as 6 years ago.CONCLUSION:The rate of lead poisoning among preschool children has decreased significantly now in Zhejiang Province. But the condition of BLL in preschool children is still far from optimistic. The environmental lead pollution in remote mountain areas and island areas should be highly concerned by the government.

Abstract:【Objective】 To investigate the features of laboratory indexes and intelligence quotient(IQ) in children whose parents exposed to lead.【Methods】 The students aged 7 to17 years old were divided into three groups(61 for each group):Group one(whose parents were exposed to lead),group two(whose parents were not exposed to lead).The two groups studied in a school nearby a battery factory.Control group was selected(whose age and gender were matched to the above group).【Results】 Blood lead level(PbB),urine lead level(PbU),free erythrocyte porphyry(FEP) were higher and Hb,MCHC(Hb/PCV) were lower in group one and two compared with control group,which demonstrated that prolonged exposure to lead concentration may disturb the metabolic of porphyrin and hereby inhibit synthesis of haemoglobin.The frequency of low IQ was 14.75%,4.92%,4.92% in group one,group two and control group respectively.The average IQ in controls was 106.16,and 105.20 in control group and group two.However,in group one average IQ was 98.59 in children whose mother exposed to lead(n=16),and 101.91 in children whose father exposed to lead(n=45).【Conclusions】 Long term lead exposure may result in children's hemoglobin decrease and influence mental development in children.It is necessary for government and society to take effective measures to protect children's physical and mental health from lead pollution.

[Effects of Lead Exposure on Their Brain-Stem Auditory Evoked Potential in Children].

Abstract:OBJECTIVE:To determine if low lead exposure can influence auditory development in children. METHODS:Children were divided into two groups, i.e., low (< 0.483 micromol/L) and high (>or= 0.483 micromol/L) blood lead levels with atomic spectrophotometry (AAS). Latencies and latent intervals between peak waves of their brain-stem auditory evoked potentials (BAEP) in the two groups were measured and compared. RESULTS:There was positive correlation between blood lead levels and BAEP in wave III at right ear, wave V at both right and left ears and latent intervals between peak waves I to V, with correlation coefficients of 0.2991, 0.2748, 0.3248, 0.2064 and 0.2412, respectively. There was significant difference in latencies of wave III at right ear, wave V at both right and left ears and latent intervals between peak waves I to V at both right and left ears and those between waves of III to V at right ear in the low blood lead group. Latencies and latent intervals between peak waves were longer in the high blood level group (with a mean of 0.78 micromol/L and a range of 0.58 to 1.42 micromol/L) than those in the low one (with a mean of 0.30 micromol/L and a range of 0.17 to 0.45 micromol/L). CONCLUSION:Elevated blood lead level could cause prolongation of latencies and latent intervals between peak waves of BAEP and slowdown of the nerve conduction velocity, and so auditory development was influenced.
Influence of exposure to environmental lead on serum immunoglobulin in preschool children.

Abstract:Serum immunoglobulin (IgG, IgM, and IgE) concentrations of 38 preschool children with blood lead levels ⩾0.48μmol/L (10μg/dL) were examined and compared to 35 preschool children with blood lead levels ⩽0.48μmol/L. No differences in serum concentrations of IgG, IgM, and IgE in the populations were observed, but IgG, IgM, and IgE of male and female children from the high blood lead level group were compared to those of controls and the results showed that IgG and IgM were significantly lower in the high blood lead level group of females than in the controls, while IgE was significantly higher in the high blood lead level group of females than in the controls (P<0.05). No correlation between blood lead concentration and serum immunogloblins IgG and IgM was demonstrated, but a statistically significant relationship between IgE and blood lead level was found in this population. These data indicate that the effect of lead on IgG, IgM, and IgE was stronger in females than in males and lead could play a role in this process by stimulating IgE production.
The Effect of Lead on Brainstem Auditory Evoked Potentials in Children

Abstract:OBJECTIVE To determine whether lead affects brainstem auditory evoked potentials (BAEPs) in low-to-moderate lead exposed children. METHODS BAEPs were recorded from 114 asymptomatic children aged 1 - 6 years. Average values were calculated for peak latency (PL) and amplitude (Amp). Whole blood lead (PbB) levels were assessed by graphite furnace atomic absorption spectroscopy. Based on their PbB levels, subjects were divided into low lead (PbB < 100 micro g/L) and high lead subgroups (PbB > or = 100 micro g/L). RESULTS The PbB levels of the 114 subjects ranged from 32.0 to 380.0 micro g/L in a positively skewed distribution. The median of PbB levels was 90.0 micro g/L while the arithmetic average was 88.0 micro g/L. Of the subjects, 43.0% (49/114) had levels equal to or greater than 100 micro g/L. Bilateral PLs I, V, and III of the left ear in the high lead subgroup were significantly longer than those in the low lead subgroup (P < 0.05). A positive correlation was found between PbB levels and bilateral PLs I, V and III of the left ear (P < 0.05), after controlling for age and gender as confounding factors. A significant and positive correlation between PbB levels and PL I of the left ear, even when PbB levels were lower than 100 micro g/L, in the low subgroup (r = 0.295, P = 0.019) was also found. CONCLUSIONS Lead poisoning in children younger than 6 years old is a very serious problem to which close attention should be paid. The indications that lead prolongs partial PLs may imply that lead, even at PbB levels lower than 100 micro g/L, impairs both the peripheral and the central portions of the auditory system. BAEPs may be a sensitive detector of subclinical lead exposure effects on the nervous system in children.

Abstract:Children are more susceptible to environmental pollutants than adults because of their special physiological structure and behavior.Moreover,children are particularly vulnerable to neurotoxic effects of heavy metals due to their developing nervous system.Studies reported that lead has negative impacts on neurodevelopment,and there are potential interactions between exposures to lead and other metals.This paper reviewed relevant studies in recent years about prenatal and postnatal lead exposure and co-exposure with other heavy metals and provided future research directions.
Adverse Health Effects of Lead Exposure on Physical Growth, Erythrocyte Parameters and School Performances for School-Aged Children in Eastern China.

Abstract:We conducted a cross-sectional study with 395 completely matched student samples enrolled from a public primary school in Nanjing of eastern China, including questionnaires, blood samples, growth indexes and school performances, all of which were used for the analysis of variance (ANOVA) and general linear model (GLM). The results showed that factors, such as gender, age, parents' education, residential passive smoking and picky eaters, had significant impacts on the blood lead levels (BPbs). As for the linear and non-linear dose-response relationship between BPbs and erythrocyte parameters, we found a positive association between BPbs and red blood cell count (RBC count) and mean corpuscular hemoglobin concentration (MCHC) but a negative association between BPbs and hemoglobin (HGB), hematocrit (HCT), mean corpuscular volume (MCV) and mean corpuscular hemoglobin (MCH). When BPbs increased by 10 mu g/L, the RBC count increased by 0.18 x 10(12)/L, while HGB and HCT decreased by 1.19 g/L and 0.41% for boys, respectively. As for girls, corresponding increases in RBC count was 0.05 x 10(12)/L, while HGB and HCT decreased by 0.82 g/L and 0.23%. Meanwhile, for both boys and girls, MCHC increased by 2.55 g/L, while MCV and MCH levels decreased by 0.41 fL and 0.12 pg each. Furthermore, a remarkable adverse effect (p < 0.05) was observed on children's school performances as a result of increased BPbs. As BPbs increased by 10 mu g/L, children's scores for Chinese, Math and English decreased by 0.42 points, 0.39 points and 0.87 points, respectively. In summary, our study indicated that lead exposure can have adverse health effects on children's erythrocyte parameters, BMI, and school performances.

Abstract:Objective To explore the effects of environmental lead exposure on immune system in preschool children. Methods The blood lead levels of 217 preschool children were determined by graphite furnance atomic absorption spectrophotometry. The distribution of T_lymphocyte subsets: CD 3+,CD 3+CD 4+,CD 3+CD 8+, B cells (CD 3- CD 19+), NK cells (CD 3-CD 16+CD 56-) were analyzed by flow cytometer, the levels of serum immunoglobulin G and immunoglobulin M were determined by scattering turbidimetry, the levels of serum immunoglobulin E were examined with ELISA. Results The mean level of blood lead of 217 preschool children was (0.46±0.27)μmol/L(range:0.11～2.71 μmol/L). The blood lead levels of 63 preschool children were ≥0.48 μmol/L. 38 preschool children among 63 preschool children with blood lead level of ≥0.48 μmol/L were selected as the high_blood_lead group, 35 preschool children with blood lead levels of 0.48 μmol/L were served as low_blood_lead group. The comparative study on the distribution of T_lymphocyte subsets was carried out in these two groups. Lower percentage of CD 3+CD 4+cells, lower ratio of CD 3+CD 4+ cells to CD 3+CD 8+ cells, higher percentage of CD 3+CD 8+ cells were found in preschool children with blood lead level of ≥ 0.48 μmol/L compared with those of control group (P0.01 or P0.05). No significant differences were observed in the percentages of CD 3-CD 19+ cells, CD 3-CD 16+CD 56+ cells, and the levels of IgG, IgM, IgE between high_blood_lead group and control group (P0.05). Condusion The blood lead levels of ≥0.48 μmol/L presented adverse effects on the T_lymphocyte subsets.
Low-Level Environmental Lead Exposure and Children’s Intellectual Function: An International Pooled Analysis

Abstract:Lead is a confirmed neurotoxin, but questions remain about lead-associated intellectual deficits at blood lead levels < 10 microg/dL and whether lower exposures are, for a given change in exposure, associated with greater deficits. The objective of this study was to examine the association of intelligence test scores and blood lead concentration, especially for children who had maximal measured blood lead levels < 10 microg/dL. We examined data collected from 1,333 children who participated in seven international population-based longitudinal cohort studies, followed from birth or infancy until 5-10 years of age. The full-scale IQ score was the primary outcome measure. The geometric mean blood lead concentration of the children peaked at 17.8 microg/dL and declined to 9.4 microg/dL by 5-7 years of age; 244 (18%) children had a maximal blood lead concentration < 10 microg/dL, and 103 (8%) had a maximal blood lead concentration < 7.5 microg/dL. After adjustment for covariates, we found an inverse relationship between blood lead concentration and IQ score. Using a log-linear model, we found a 6.9 IQ point decrement [95% confidence interval (CI), 4.2-9.4] associated with an increase in concurrent blood lead levels from 2.4 to 30 microg/dL. The estimated IQ point decrements associated with an increase in blood lead from 2.4 to 10 microg/dL, 10 to 20 microg/dL, and 20 to 30 microg/dL were 3.9 (95% CI, 2.4-5.3), 1.9 (95% CI, 1.2-2.6), and 1.1 (95% CI, 0.7-1.5), respectively. For a given increase in blood lead, the lead-associated intellectual decrement for children with a maximal blood lead level < 7.5 microg/dL was significantly greater than that observed for those with a maximal blood lead level > or = 7.5 microg/dL (p = 0.015). We conclude that environmental lead exposure in children who have maximal blood lead levels < 7.5 microg/dL is associated with intellectual deficits.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Background</h3><p>Lead exposure is one of the most concerning public health problems worldwide, particularly among children. Yet the impact of chronic lead exposure on the thyroid status and related intelligence quotient performance among school-age children remained elusive.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Objective</h3><p>The aim of this study was to evaluate the influence of lead exposure on the thyroid hormones, amino acid neurotransmitters balances, and intelligence quotient (IQ) among school-age children living nearby a lead-zinc mining site. Other factors such as rice lead levels, mothers' smoking behavior, and diet intake were also investigated.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Methods</h3><p>A total of 255 children aged 7-12 years old were recruited in this study. Blood lead level (BLL), thyroid hormones including free triiodothyronine (FT3), free thyroxine (FT4) and thyroid stimulating hormone (TSH), and amino acid neurotransmitters such as glutamate (Glu), glutamine (Gln), and γ-aminobutyric acid (GABA) were measured using graphite furnace atomic absorption spectroscopy (GFAAS), chemiluminescence immunoassay, high performance liquid chromatography (HPLC). Raven's standard progressive matrices (SPM) and the questionnaire were used to determine IQ and collect related influence factors.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>The average BLL of children was 84.8 μg/L. The occurrence of lead intoxication (defined as the BLL ≥ 100 μg/L) was 31.8%. Serum TSH levels and IQ of lead-intoxicated children were significantly lower than those without lead toxicity. The GABA level of girls with the lead intoxication was higher than those with no lead-exposed group. Correlation analyses revealed that BLL were inversely associated with the serum TSH levels (R= -0.186, p < 0.05), but positively related with IQ grades (R = 0.147, p < 0.05). Moreover, BLL and Glu were inversely correlated with IQ. In addition, this study revealed four factors that may contribute to the incidence of lead intoxication among children, including the frequency of mother smoking (OR = 3.587, p < 0.05) and drinking un-boiled stagnant tap water (OR = 3.716, p < 0.05); eating fresh fruits and vegetables (OR = 0.323, p < 0.05) and soy products regularly (OR = 0.181, p < 0.05) may protect against lead intoxication.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusion</h3><p>Lead exposure affects the serum TSH, GABA levels and IQ of school-aged children. Developing good living habits, improving environment, increasing the intake of high-quality protein and fresh vegetable and fruit may improve the condition of lead intoxication.</p>

Investigation on Health Risk by Environmental Lead Pollution in Children

Abstract:Objective To assess the health risk of children living in lead polluted city of China.Methods The blood lead levels of children in certain industrialized city were calculated using IEUBKwin software developed by Environmental Protection Agency,US.Then the incidence of mild mental retardation was obtained with the method introduced in Lead-Assessing the Environmental Burden of Disease at National and Local Levels published by World Health Organization.Results The probability ratio that the blood lead level over 10 μg/dl(toxicity threshold),was up to 65.04%,which was higher than the average level in China.Otherwise,the incidence of mild mental retardation was also higher than that of other cities in China,which was 3.2 times higher than the estimated incidence for East Asia by WHO.Conclusions It was suggested that the lead pollution in this city was quite serious,and the health risk caused by lead pollution was quite big in children.Consequently,the control measures must be taken for blocking the development of lead pollution in this area.

Abstract:Tinfoil manufacturing and electronic waste (e-waste) recycling remain rudimentary processes in Zhejing Province, China, which could account for elevated blood lead levels (BLLs) and health impacts on children. We assessed the potential health risks of lead in tinfoil manufacturing and e-waste recycling areas. 329 children in total aged 11-12 who lived in a tinfoil manufacturing area (Lanxi), an e-waste recycling area (Luqiao) and a reference area (Chun'an) were studied. Lead levels in children's blood were determined by inductively coupled plasma mass spectrometer (ICP-MS). Blood urea nitrogen (BUN), serum creatinine, serum calcium, delta-Aminolaevulinic acid (delta-ALA) and intelligence quotient (IQ) were also measured. Geometric mean of BLLs in Lanxi, Luqiao and Chun'an were 8.11 mu g/dL, 6.97 mu g/dL, and 2.78 mu g/dL respectively, with 35.1%, 38.9% and 0% of children who had BLLs above 10 mu g/dL. The BLLs in exposed areas were much higher than those in the control area. Lanxi children had higher creatinine and calcium than Chun'an children, and Luqiao children had higher delta-ALA and lower calcium than Chun'an children. No significant differences of IQ were observed between Lanxi, Luqiao and Chun'an, however a negative relationship between BLLs and IQ was shown for the study children. The results indicated that lead pollution from e-waste recycling and tinfoil processing appears to be a potential serious threat to children's health. (C) 2012 Elsevier B.V. All rights reserved.

Abstract:To assess the strength of the association between blood lead and children's IQ, a meta-analysis of the studies examining the relationship in school age children was performed. Emphasis was given to the size of the effect, since that allows comparisons that are informative about potential confounding and effect modifiers. Sensitivity analyses were also performed. A highly significant association was found between lead exposure and children's IQ (P < 0.001). An increase in blood lead from 10 to 20 micrograms/dl was associated with a decrease of 2.6 IQ points in the meta-analysis. This result was robust to inclusion or exclusion of the strongest individual studies and to relaxing the age requirements (school age children) of the meta-analysis. Adding eight studies with effect estimates of 0 would still leave a significant association with blood lead (P < 0.01). There was no evidence that the effect was limited to disadvantaged children and there was a suggestion of the opposite. The studies with mean blood lead levels of 15 micrograms/dl or lower in their sample had higher estimated blood lead slopes, suggesting that a threshold at 10 micrograms/dl is implausible. The study with the lowest mean blood lead level was examined using nonparametric smoothing. It showed no evidence of a threshold down to blood lead concentrations of 1 microgram/dl. Lead interferes with GABAergic and dopaminergic neurotransmission. It has been shown to bind to the NMDA receptor and inhibit long-term potentiation in the hippocampal region of the brain. Moreover, experimental studies have demonstrated that blood levels of 10 micrograms/dl interfere with a broad range of cognitive function in primates. Given this support, these associations in humans should be considered causal.
