Abstract:In this study, a method to enrich forsythiaside A (FA) from the ball-milled powder of Forsythia suspensa leaves was established. Forsythiaside A was extracted by heating and stirring, and the extraction process was optimized by response surface methodology using the independent variables of extraction temperature, extraction time, liquid-to-material ratio, ethanol concentration, and pH value of the solution, respectively. Then five types of macroporous resins with different polarities were studied to explain the adsorption mechanism by static adsorption and desorption experiments. Different models were used to study the static adsorption thermodynamics and kinetic process of AB-8 resin on FA extracts, and optimal conditions of the process were optimized. Finally, the antioxidant abilities of purified FA were evaluated. The results showed that the optimum extraction conditions were 2.2 h, 53.4 °C, 45.7 mL/g, 60% ethanol, and pH 6, and the extraction efficiency of forsythiaside A could achieve 120.96 mg/g. The AB-8 macroporous resin was the most suitable resin with high adsorption and desorption rates. It was found that the Temkin model was able to calculate the adsorption parameters accurately. The adsorption kinetic process of the resin was in accordance with the pseudo-second-order kinetic model. The optimal process conditions were as follows: the optimum loading concentration of 5.83 mg/mL, the loading rate of 1 BV/h, and the loading volume of 7.51 BV; in the desorption process, deionized water was used to remove impurities first, and then, 30% ethanol was used with an elution rate of 1 BV/h. The purity of FA increased from 20.52 to 81.58%. The purified FA exhibited superior scavenging ability for DPPH radicals and ABTS radicals. Cytotoxicity experiments proved that it was safe and nontoxic and was capable of promoting cell growth. In conclusion, the present study provides a theoretical basis for industrially extracting and purifying FA on a reliable basis.

Abstract:The enzyme-assisted extraction of rosemary acid from Perilla frutescens was optimized applying one-factor-at-a-time method (OFAT) and response surface methodology (RSM) and the antioxidant activity of the rosemary acid was investigated. Preliminary investigations were conducted into the effect of enzyme dosage, hydrolysis temperature, time and pH on the extraction yield of rosemary acid. Furthermore, the optimization of process parameters for extracting rosemary acid by cellulase hydrolysis of Perilla frutescens was done by RSM using Box Behnken design. Meanwhile, the antioxidant activity was assessed by superoxide anion and 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assays. The optimal conditions for rosemary acid extraction were determined as follows:enzyme dosage, 3%;hydrolysis temperature, 45℃;time, 12 min;and pH, 4. The maximum extraction yield of rosemary acid of 0.617%was obtained in experiments conducted under these optimal conditions, being not significantly different from the predicted value (0.621%). The rosemary acid showed strong scavenging capacity on superoxide anion and DPPH radicals, thereby having potent antioxidant activity.
Antioxidant Capacities of Loquat Flower Extracts Using Different Solvents

Abstract:DPPH,ABTS,FRAP and F-C methods were used to analyze the antioxidant capacity of loquat flowers extracts.The results showed that water extracts exhibited the highest antioxidant capacity,followed by methanol extracts and ethanol extracts in the three methods(DPPH,ABTS,F-C).In the FRAP method the methanol extracts showed higher antioxidant capacity than the water extracts.That may be due to different extract solvents and different reaction mechanisms.In general,the ethanol extracts exhibited the lowest antioxidant capacity.

Abstract:The extracts containing oxidation resisting constituents were obtained from the leaves of Aesulus Chinensis Bunge by different extracting methods. The activities of oxidation resistance of the extracts were measured by using rape oil as a substrate. It was found that the extracts obtained by 70% ethanol with an added amount of 0. 08% showed the highest activity and better than 0. 2% BHT. The extracts contain phenolic compounds, tannins,organic acids, flavonoids, phytosteriols, etc.

Abstract:The objectives were to investigate the antioxidative activity of Forsythia suspensa and to identify the active fractions using the bioassay-linked fractionation method. BuOH and CH_2Cl_2 fractions from F.suspensa appeared potent antioxidant effects against the DPPH radical,showing the IC_(50) values of 79.69 and 27.75 μg/mL,respectively.Moreover,they recovered cell viability declined by SIN-1 treatment dose-dependently(P 0.01),suggesting the protective roles from cellular oxidative damage.

Abstract:In this research, the conditions for extraction of phenolics from leaves of Ficus virens were optimized using response surface methodology (RSM). The extraction abilities of phenolics (EAP) and flavonoids (EAF), the 2,2-diphenyl-1-pierylhydrazyl (DPPH) free-radical scavenging potential, and the ferric reducing/antioxidant power (FRAP) were used as quality indicators. The results of single-factor experiments showed that temperature, ethanol concentration, extraction time, and the number of extraction cycles were the main influencing variables, and these provided key information for the central composite design. The results of RSM fitted well to a second degree polynomial model and more than 98% of the variability was explained. The ideal extraction conditions for EAP, EAF, DPPH free-radical scavenging potential, and FRAP were obtained. Considering the four quality indicators overall, the ideal extraction conditions were 58% ethanol at 57 °C for 37 min with three extraction cycles. At the ideal extraction conditions, the values of EAP, EAF, DPPH free-radical scavenging potential, and FRAP were 5.72%, 3.09%, 58.88 mg ascorbic acid equivalent (AAE)/g dry weight (DW), and 15.86 mg AAE/g DW, respectively. In addition, linear correlations were observed between EAP, EAF, and antioxidant potential.

Abstract:In this paper orthogonal experiment method was used to study the extraction process in(alcohol)-water system of folium Artemisiae argyi.Its contents of total flavonoids and total polysaccharides were measured by colorimetry.And it was found that different factors impacting on extraction were as followed: the concentration of alcohol,extraction temperature,extraction time and the ratio of materials for extraction liquid.The anti-oxidant activities of the extraction liquid were also studied by the chemical(luminescence) method.Results showed that folium Artemisiae argyi contains abundant of flavonoid and polysaccharide,and its extract possesses strong anti-oxidant activity.Therefore the researh and development of folium Artemisiae argyi are of great significance.

Abstract:It was found that all the extracts of persimmon leaves from ether, petroleum ether, ethyl acetate, butanol showed strong antioxidation effects, among them, petroleum ether extract was the best one, and when the additive quantity of the extract reached to 0.14%, its antioxidation perfermance was stronger than that of 0.02% BHT. The components of antioxidation resistance mainly were phenols, tannins and flavonoids.
Effect of Extracting Solvent on the Quality of Lentinula edodes Oil Based on Chemical Composition, in vitro Antioxidant Activity and Correlation Analysis

Abstract:In this study, effects of 4 solvents (petroleum-ether, n-hexane, ethyl-acetate, and chloroform) on the chemical characterizations and in vitro antioxidant capacities of oil were assessed to determine the optimal extraction solvent for L. edodes oil. Three data analysis techniques including principal component analysis, hierarchical cluster analysis, and multiple linear regression, were applied to determine the relationship between the nutrient and antioxidant capacity. The results showed that chloroform extracted L. edodes oil exhibited the largest amount of α-tocopherol, flavones, and unsaponifiable matter, chloroform was thus confirmed desirable for extracting L. edodes oil rich in nutrition. While based on the best DPPH and ABTS, the ethyl-acetate extracted oil show the strongest antioxidant property. More than that, the results also showed that different extraction solvents could induce large variations in minor components and free radical scavenging activity among the test oils, and the total phenol content was found positively correlated to the antioxidant capacity of L. edodes oil, which could be well predicted by all MLR models. These findings revealed the influence of solvent on the chemical characterization and in vitro antioxidant capacity of L. edodes oil, providing a theoretical foundation for future applications of L. edodes oil.

Abstract:Antioxidation activity of extracts from kumquat with flavone or limonin as solvent was studied by the iodine method.Results indicate the extracts from kumquat have antioxidation effect.The extract by flavone exhibits obvious antioxidation for oil when the extract dosage is 0.02%,and it has synergistic action with VC,VE.The extract by limonin exhibits weak antioxidation for oil when the dosage is 0.08%,and it has not synergistic action with VC,VE,and the stability is no good.

Abstract:Two new monoepoxylignans, forsythialan A (1) and B (2), were isolated from the methanol extract of the fruits of Forsythia suspensa Vahl, along with a known compound, phillyrin (3). Their structures were established by extensive spectroscopic analyses. The compounds 1 and 2 showed potent antioxidant effects against peroxynitrite-induced oxidative stress in renal epithelial cell.

Abstract:Besides active substances, Forsythia suspensa is rich in dietary fiber (DF), but it is often wasted or discarded and not put to good use. In order to improve the function of Forsythia DF, it was modified using alkaline hydrogen peroxide (AHP) and cellulase (EM). Compared to the control DF (ODF), the DF modified using AHP (AHDF) and EM (EMDF) had a looser microstructure, lower crystallinity, and higher oil holding capacity (OHC) and cation exchange capacity (CEC). The AHP treatment significantly increased the water holding capacity (WHC) and water swelling ability (WSA) of the DF, while the EM treatment achieved just the opposite. Moreover, the functional properties of AHDF and EMDF, including their cholesterol adsorption capacity (CAC), nitrite ion adsorption capacity (NAC), glucose adsorption capacity (GAC), glucose dialysis retardation index (GDRI), α-amylase inhibitory activity, and DPPH radical scavenging activity, were far better than those of ODF. Together, the results revealed that AHP and EM modifications could effectively improve or enhance the physicochemical and functional properties of Forsythia suspensa DF.

Abstract:Fructus forsythia essential oil (FEO) with excellent antibacterial activity was rarely reported. The objective of the present study was to investigate the antibacterial activity and the antibacterial mechanism of FEO against two food-borne pathogenic bacteria, Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) in vitro. When treated FEO, the zones of inhibition (ZOI) of E. coli (20.5 ± 0.25 mm) and S. aureus (24.3 ± 0.21 mm) were much larger than control (p < 0.05). The minimum inhibitory concentrations (MICs) of FEO were 3.13 mg/mL and 1.56 mg/mL for E. coli and S. aureus, respectively. The antibacterial mechanism of FEO against E. coil was due to the changes in permeability and integrity of cell membrane leading to the leakage of nucleic acids and proteins. With the superior antibacterial activity of FEO, the nano-encapsulation method has been applied in FEO. When compared to FEO and blank chitosan nanoparticles, FEO-loaded nanoparticles (chitosan to FEO of 1:1) can effectively inhibit the growth of E. coil above 90% at room temperature. It is necessary to consider that FEO and FEO-loaded nanoparticles will become promising antibacterial additives for food preservative, cosmetic, and pharmaceutical applications.

Abstract:The optimal conditions of flavonoid extraction from leaves of prunus virginiana by solvothermal method and its antioxygenation were studied.The results through primary factor experiments and orthogonal tests showed that the best technical conditions for flavonoids extraction could be obtained with 70% alcohol,ratio of raw materials and solution as 1g:40mL,time of extraction 2.5h,temperature of extraction at 80 ℃,and times of extraction was two.The antioxygenation of the flavonoids from leaves of prunus virginiana was studied in the soybean oil.The graph of POV showed that the flavonoid from leaves of prunus virginiana was a good antioxidant.

Abstract:Antioxidants are organic compounds that help to prevent lipid oxidation and improve the shelf-life of edible oils and fats. Currently, synthetic antioxidants were used as oil stabilizing agent. However, synthetic antioxidants have been causing various health risks. As a result, natural antioxidants such as most parts of olive plant, green tea, sesame, medicinal plants were plays an important role to retard lipid oxidation. The palm oil was continuously frying at 180 °C for 6 days using Lepidium sativum (0.2%w/v) and Aframomum corrorima (0.3%w/v) seeds extracts as antioxidant. The physicochemical properties of oil in the herbal extract additive group significantly maintained the oil quality during frying compared to the normal control and the food sample containing group. The L. sativum extract had a greater oil stability compared to A. corrorima extract. However, the frying oil without herbal extract significantly increase the physicochemical properties of oil such as iodine value, acid value, free fatty acid, total polar compounds, density, moisture content, pH etc. during repetitive frying. The antioxidant activity of the plant extract was outstanding, with an IC50 value in the range of 75-149.9 μg/mL when compared to the standard butyl hydroxy anisole, which had an IC50 value in the range of 74.9 ± 0.06-96.7 ± 0.75 μg/mL. The total phenolic and flavonoid content of the extract for L. sativum was 128.6 ± 0.00 mg GAE/g, 127.0 ± 0.00 mg QE/g, and 130.16 ± 0.001 mg GAE/g, 105.76 ± 0.02 mg QE/g, respectively. The significant effect of the plant extract on the degradation of oil and the formation of free fatty acids was confirmed by Fourier transform infrared spectroscopy. The result of these study revealed that the ethanolic crude extract of L. sativum and A. corrorima had a potential natural antioxidant to prevent the degradation of palm oil.

Abstract:Objective: To study the extraction and anti-oxidation of total flavones from hazel's flower.Methods: The optimum extraction conditions of total flavones from hazel's flower were investigated with orthogonal experimental design.Fenton reaction was used to produce and detect hydroxyl radicals(·OH),and pyrogallol system was used to produce and detect the superoxide radical anion(O2-·).Results: The optimal extraction condition was: the alcohol concentration 50%,temperature 70℃,extraction time 5h and dosage liquor ratio 1:20.The order of the factors which affected extraction was: temperature alcohol concentration extraction time dosage liquor ratio.The capacity of scavenging hydroxyl free radical of flavones was far more than that of vitamin C,and the supreme scavenging rate of superoxide anion was 80.13%.Conclusion: The optimum extraction condition of total flavones from hazel's flower was investigated,and the total flavones from hazel's flower had a strong capacity of scavenging superoxide anion and hydroxyl free radical.
Analysis of Forsythia suspensa fruit and leaf extracts using UHPLC-Q-Exactive-Orbitrap/MS: In vivo antioxidant activity on D-galactose-induced aging mice

Abstract:Making health-enhancing tea from Forsythia suspensa leaves has been a tradition of Chinese folk culture for centuries. However, these leaves were not officially recognized as a new food source until 2017 by the Chinese government. In this study, ethyl acetate fractions from Forsythia suspensa fruit and leaves exhibited excellent antioxidant activity in vitro antioxidant assays and in vivo D-galactose-induced aging mice model. The antioxidant activity of the leaves was higher than that of fruit both in vitro and in vivo. The chemical constituents present in these ethyl acetate fractions were comprehensively analyzed using UHPLC-Q-Exactive-Orbitrap/MS. A total of 20 compounds were identified, among which forsythoside E, (+)-epipinoresinol, dihydromyricetin, chlorogenic acid, and ursolic acid were exclusively detected in the ethyl acetate fraction of Forsythia suspensa leaves, but absent in the ethyl acetate fraction derived from its fruit. This study provides theoretical support for the utilization of Forsythia suspensa fruit and leaves.

Abstract:Antioxidation activity of the total flavonoids of Glycyrrhiza on edible oils was studied using peroxide value (POV) by Schaal oven-storage test. The results showed that the total flavonoids are of good antioxidant effects on four edible oils. Furthermore, the activity on lard is the strongest and dose-dependent. The total flavonoids of Glycyrrhiza added with vitamin C, citric acid or tartaric acid exhibit remarkable synergistic antioxidation activity in lard. The antioxidation activity of the total flavonoids synergized with synthetic antioxidants such as BHA, BHT or PG is proved to be better than single antioxidant used.

Abstract:In this study, effect of some solvents (ethanol, acetonitrile and water) for the extraction of phenolic compounds was investigated. Solvent volume, temperature and duration of the extraction process have a positive correlation with the amounts of phenolic compounds and antioxidant activity. The amounts of phenolic compounds, especially oleuropein in the leaf extract is higher than the fruit extract, and the concentration of oleuropein content in leaves and fruits extract was 4900 and 1800 mg/L, respectively. The oleuropein extracted from olive leaves was purified with liquid-liquid extraction to above 98%, and its concentration in leaves was 9.8 w/w/% in dried leaves.
