Abstract:The application of immobilized lipase in the enzymatic production of biodiesel has shown numerous advantages. In this study, surface of Polyacrylonitrile (PAN) hollow membrane was first modified using nitrile-click chemistry in order to fit for interaction with enzyme proteins. Then sodium alginate (SA) was introduced and the membrane was post-treated by CaCl2. When the prepared PAN-PEI-SA-CaCl2 was used for lipase immobilization, the protein loading was 36.90 mg/g, and the enzyme activity reached up to 54.47 U/g, which was 2.5 times as much as that of Novozym® 435. As a result, the constructed immobilized lipase obtained a maximum biodiesel yield of 78.5%, which was 2.4 times that of the Novozym® 435 in transesterification reactions. Moreover, the biodiesel yield decreased by only 11% after the immobilized enzyme was continuously used for 20 times. This study exhibits that this technic has broad application prospects in the field of conversion of biomass resources.
Effect of Immobilization by Modified Ultrastable-Y Molecular Sieve on the Resolution of (R,s)-2-octanol in Nonaqueous Media by Lipase

Abstract:The lipase from Penicillium expansum PED-03 (PEL) was immobilized into a modified ultrastable-Y molecular sieve and the resolution of (R,S)-2-octanol was carried out in a bioreactor in nonaqueous media by the immobilized lipase to investigate the effect of immobilization on the catalytic property of PEL duing the enzymatic reaction. It was found that the conversion (c), enantiomeric excess (ee), and enantioselectivity (E) of the resolution catalyzed by the PEL immobilized on the modified ultrastable-Y molecular sieve were much higher than those of the reaction catalyzed by free PEL and PEL immobilized on other supports. Being immobilized on the modified Ultrastable-Y molecular sieve, the lipase exhibited obvious activity in a wider pH range and at a much higher temperature than the free PEL, and showed a marked enhancement of stability against thermal inactivation, by which the suitable pH value of the buffer used for immobilization could be "memorized". The conversion of the reaction catalyzed by PEL immobilized on the modified ultrastable-Y molecular sieve reached 97.68% of the theoretical value with excellent enantioselectivity (Average E of 8 batches >460) in nonaqueous media at "memorial" pH 9.5, 50 degrees C for 24 h, which showed a good application potential to the production of optically pure (R,S)-2-octanol.

Abstract:The lipase was immobilized on the acetylcellulose/polypropylene composite membrane by Sodium periodate oxidation and the response surface analysis was used to further optimize immobilized conditions,and then discussed the properties of immobilized enzyme membrane.The results indicated that the optimal conditions of the immobilized lipase are:with immobilized enzyme activities for index,the sodium periodate concentration was 0.15 mol/L,activated 60 min,which the lipase concentration was 0.013 g/mL with cross-linking agent concentration was 0.24% under the phosphate buffered solution of pH was 8.0,then after the time of adsorption-crosslinking for 3.1 h of temperature at 4 ℃,separately.The lipase activities were 0.52 U/cm2,and the optimal pH of the immobilized lipase was 9.0,the optimal operating temperature of the immobilized lipase was 35 ℃.
Effect of Hydrophobic or Hydrophilic Performance of Membranes on Lipase Immobilization

Abstract:The difference of hydrophobic and hydrophilic performance of membrane has great effect on the loading, the activity and the stability of lipase immobilized with adsorption. The immobilization of lipase from Candida rugosa (CRL) and from porcine pancreas (PPL) on polypropylene (PP) hollow fiber and on polytetrafluoroethylene (PTFE) , polysulfone (PSF), polyamine (PA), polyacrylonitrile (PAN), polyethersulfone (PES), polyvinylidene fluoride (PVDF) and cellulose acetate (CA) plate membranes were conducted, respectively. The hydrolysis degree of olive oil catalyzed by immobilized enzyme on membrane was used to evaluate the enzyme activity. It is found that mentioned above two kinds of enzymes immobilized on strongly hydrophobic PTFE or PP membrane have a large loading, a high activity and a long half-life time, and the half-life time of CRL immobilized on PTFE membrane is 6 times longer than that of free enzyme. The experimental results also show that the loading of PPL immobilized on strongly hydrophilic CA membrane is larger than that of on PTFE membrane, but the specific activity and the recovery efficiency of activity are lower than that immobilized on PTFE membrane. The loading of lipase immobilized on the PSF or PAN membrane, whose contact angle is in the range of 40°to 50°, is the lowest. The PTFE and PP membrane are suitable to be used for immobilization of above mentioned two kinds of lipases with adsorption, and the appropriate preparation conditions are as follows: the pH of solution containing enzyme is 7.5, the adsorption temperature is 25℃ and the adsorption time is 10 h.
Immobilization and Properties of Lipase from Candida Rugosa on Electrospun Nanofibrous Membranes with Biomimetic Phospholipid Moities

Abstract:Reported here is a protocol to fabricate a biocatalyst with high enzyme loading and activity retention, from the conjugation of electrospun nanofibrous membrane having biomimetic phospholipid moiety and lipase. To improve the catalytic efficiency and activity of the immobilized enzyme, poly(acrylonitrile-co-2-methacryloyloxyethyl phosphorylcholine)s(PANCMPCs) were, respectively, electrospun into nanofibrous membranes with a mean of 90 nm, as a support for enzyme immobilization. Lipase from Candida rugosa was immobilized on these nanofibrous membranes by adsorption. Properties of immobilized lipase on PANCMPC nanofibrous membranes were compared with those of the lipase immobilized on the polyacrylonitrile(PAN) nanofibrous and sheet membranes, respectively. Effective enzyme loading on the nanofibrous membranes was achieved up to 22.0 mg/g, which was over 10 times that on the sheet membrane. The activity retention of immobilized lipase increased from 56.4% to 76.8% with an increase in phospholipid moiety from 0 to 9.6%(molar fraction) in the nanofibrous membrane. Kinetic parameter K., was also determined for free and immobilized lipase. The Km value of the immobilized lipase on the nanofibrous membrane was obviously lower than that on the sheet membrane. The optimum pH was 7.7 for free lipase, but shifted to 8.3-8.5 for immobilized lipases. The optimum temperature was determined to be 35 degrees C for the free enzyme, but 42-44 degrees C for the immobilized ones, respectively. In addition, the thermal stability, reusability, and storage stability of the immobilized lipase were obviously improved compared to the free one.

Abstract:In this work, poly(acrylonitrile-co-acrylic acid) (PANCAA) was electrospun into nanofibers with a mean diameter of 180nm. To create a biofriendly microenvironment for enzyme immobilization, collagen or protein hydrolysate from egg skin (ES) was respectively tethered on the prepared nanofibrous membranes in the presence of 1-ethyl-3-(dimethyl-aminopropyl) carbodiamine (EDC)/N-hydroxyl succinimide (NHS). Confocal laser scanning microscopy (CLSM) was used to verify the surface modification and protein density on the nanofibrous membranes. Lipase from Candida rugosa was then immobilized on the protein-modified nanofibrous membranes by covalent binding using glutaraldehyde (GA) as coupling agent, and on the nascent PANCAA nanofibrous membrane using EDC/NHS as coupling agent, respectively. The properties of the immobilized enzyme were assayed. It was found that different pre-tethered biomacromolecules had distinct effects on the immobilized enzyme. The activity retention of the immobilized lipase on ES hydrolysate-modified nanofibrous membrane increased from 15.0% to 20.4% compared with that on the nascent one, while it was enhanced up to more than quadrupled (activity retention of 61.7%) on the collagen-modified nanofibrous membrane. The kinetic parameter, Km and Vmax, were also determined for the free and immobilized lipases. Furthermore, the stabilities of the immobilized lipases were obviously improved compared with the free one.

Abstract:Nanofibrous membranes with an average diameter of 100 and 180 nm were fabricated from poly(acrylonitrile-co-maleic acid) (PANCMA) by the electrospinning process. These nanofibrous membranes contain reactive groups which can be used to covalently immobilize biomacromolecules. Two natural macromolecules, chitosan and gelatin, were tethered on these nanofibrous membranes to fabricate dual-layer biomimetic supports for enzyme immobilization in the presence of 1-ethyl-3-(dimethyl-aminopropyl) carbodiimide hydrochloride (EDC)/N-hydroxyl succinimide (NHS). Lipase from Candida rugosa was then immobilized on these dual-layer biomimetic supports using glutaraldehyde (GA), and on the nascent PANCMA fibrous membrane using EDC/NHS as coupling agent, respectively. The properties of the immobilized lipases were assayed. It was found that there is an increase of the activity retention of the immobilized lipase on the chitosan-modified nanofibrous membrane (45.6+/-1.8%) and on the gelatin-modified one (49.7+/-1.8%), compared to that on the nascent one (37.6+/-1.8%). The kinetic parameters of the free and immobilized lipases, K(m) and V(max), were also assayed. In comparison with the immobilized lipase on the nascent nanofibrous membrane, there is an increase of the V(max) value for the immobilized lipases on the chitosan- and gelatin-modified nanofibrous membranes. Results also indicate that the pH and thermal stabilities of lipases increase upon immobilization. The residual activities of the immobilized lipases are 55% on the chitosan-modified nanofibrous membrane and 60% on the gelatin-modified one, after 10 uses.

Abstract:One natural biomacromolecule, gelatin, was tethered on the surface of poly(acrylonitrile-co-maleic acid) (PANCMA) ultrafiltration hollow fiber membrane to fabricate dual-layer biomimetic support for enzyme immobilization in the presence of 1-ethyl-3-(dimethyl-aminopropyl) carbodiimide hydrochloride (EDC)/N-hydroxyl succinimide (NHS). Attenuated total reflectance Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy were employed to confirm the chemical changes of the modified PANCMA membrane surfaces. Lipase from Candida rugosa was immobilized on this dual-layer biomimetic support using glutaraldehyde (GA), and on the nascent PANCMA membrane using EDC/NHS as coupling agent, respectively. The properties of the immobilized enzymes were assayed and compared with the free one. It was found that there was an increase of the activity retention of the immobilized lipase on the gelatin-modified membrane (49.2%), compared to that on the nascent membrane (33.9%). The kinetic parameters of the free and immobilized lipases, Km and Vmax, were also assayed. In comparison with the immobilized lipase on the nascent membrane, there was a decrease of the Km value and an increase of the Vmax value for the immobilized lipase on the gelatin-modified membrane. Results also indicated that the pH and thermal stabilities of lipases increased upon immobilization. The residual activities of the immobilized lipases were 55% on the gelatin-modified membrane and 62% on the nascent PANCMA membrane, after 10 uses.

Abstract:Lipase from Candida rugosa was covalently immobilized on the surface of an ultrafiltration hollow fiber membrane fabricated from poly(acrylonitrile-co-maleic acid) (PANCMA) in which the carboxyl groups were activated with 1-ethyl-3-( dimethylaminopropyl) carbodiimide hydrochloride (EDC) and dicyclohexyl carbodiimide (DCC N-hydroxyl succinimide (NHS), respectively. The properties of the immobilized lipase were assayed and compared with those of the free enzyme. The maximum activities were observed in a relatively broader pH value range at high temperatures for the immobilized lipase compared to the free one. It was also found that the thermal and pH stabilities of lipase were improved upon immobilization and at 50 degrees C the thermal inactivation rate constant values are 2.1 x 10(-2) for the free lipase, 3.2 x 10(-3) for the immobilized lipase on the EDC-activated PANCMA membrane and 3.5 x 10(-3) for the immobilized lipase on the DCC/NHS-activated PANCMA membrane, respectively.
Immobilization of Lipase Onto Functional Cyclomatrix Polyphosphazene Microspheres

Abstract:Functional polyphosphazene cyclomatrix microspheres (PMS) containing glucose and dodecyl moieties were utilized for physical adsorption of lipase. In this regard, Candia Rugosa lipase was immobilized onto the above microspheres through physical adsorption, which exhibited catalytic hydrolysis of glycerol triacetate as the model reaction. As observed under transmission electron microscope (TEM) and scanning electron microscope (SEM), the size of all the microspheres ranged from 400 nm to 1 mu m with a mean diameter of 680 nm. Zeta potential analysis demonstrated that the microspheres were negatively charged. The influences of buffer pH, substrate concentration and temperature were also studied along with reusability. The obtained results demonstrated that the immobilized lipase on deprotected glycosylated PMS (D-GPMS) showed the best performance and possessed the highest enzymatic loading activity around 1.0884 mmol L-1 min(-1) g(-1), and retained nearly 50% of its initial activity even after 10 cycles of application. Moreover, the lipase-microsphere complex displayed high pH adaptability from 7 to 9 and temperature stability above 40 degrees C. From the viewpoint of biocompatible PMS support, the GPMS-lipase conjugate have potential applications in field such as food, medicine and environment. (C) 2016 Elsevier B.V. All rights reserved.

Abstract:Porcine pancreatic lipase (PPL) was immobilized onto functionalized ionic liquid-modified silica carrier using gelatinization and physical adsorption. The immobilized lipase was characterized with N2 adsorption–desorption, X-ray diffraction (XRD), scanning electron microscopy (SEM), and Fourier transform infrared spectroscopy (FT-IR) before and after modification and immobilization. The results showed that the modification of the ionic liquid and the introduction of lipase had been successfully approved. The rate of enzymatic reaction and its influencing factors was primarily studied by enzymatic reaction kinetics. Km values of PPL-SiO2@CA and PPL-IM/BF4-SiO2@CA were 4.9 and 3.7 mg/ml, respectively. It indicated that the modification of the functionalized ionic liquid enhanced the affinity between the immobilized enzyme and the substrate. The immobilization efficiency, specific activity, optimum temperature, optimum pH, thermal stability, reusability, and storage stability of the immobilized enzyme were investigated. We found that the stability of the immobilized enzyme was significantly higher than that of the unmodified immobilized enzyme. Specially, PPL-IM/BF4-SiO2@CA maintained good thermal stability and retained more than 92% of its activity at 65 °C after preheating 3 h.Graphical AbstractAbove, the immobilized lipase maintained more than 92% of its initial activity after incubating at 65 °C for 3 h.

Abstract:In this study, an enhanced activity and stability method for immobilizing porcine pancreatic lipase (PPL) was developed based on ZIF-8 encapsulated supramolecular-modified gold nanoparticle complexes (pSC(4)-AuNPs@ZIF-8). Supramolecular calix[4]arene (pSC(4)) can recognize the amino group of PPL through non-covalent force, and this flexible binding method protected the structure of PPL during the immobilization process. Due to the hydrophilic of pSC(4)-AuNPs and hydrophobic of ZIF-8, PPL can maintain a "lid open" conformation, which can enhance the stability of PPL structure and reduce PPL activity loss. ZIF-8 was used to immobilize PPL to avoid the difficult recovery of free PPL. Compared with the native form of PPL, it exhibited 70.6% maintained activity with terrific pH and temperature stability, and had good performance in thermal stability, time stability, and reusability. In addition, three immobilized PPL methods were designed to further clarify the influence of synthetic methods and additives on the activity and stability of PPL. Importantly, the loading rate of pSC(4)-AuNPs@ZIF-8@PPL was up to 51.2% among these immobilized PPL systems. Therefore, pSC(4)-AuNPs@ZIF-8 may serve as a versatile and promising immobilization system for enzymes.
Immobilization of lipase on canvas fiber and its characterization

Abstract:The methods of lipase immobilization with canvas fiber were discussed in detail,and the function of immobilization lipase was determined.The NaIO4 oxidation has been employed in the process of changing the adjacent hydroxyl to aldehyde group.It was showed that through repeat crosslink,the lipase density on the carrier surface was enhanced about 30%.The activity of the immobilized lipase was investigated in catalyzing hydrolysis Oliver oil and soybean oil.The reaction stability was compared with those of the dissolved lipase in catalyzing hydrolysis of soybean oil.Through immobilization,its reaction tolerance to temperature and pH had been raised in some degree.With the lipase protection measure during the cross-link process,the tolerance of lipase to pH,temperature was improved further.The hydrolysis rate was more than 85% in the first run,and after 7 runs of continual reactions under mild conditions,the activity kept 64% of the original.The half-life of the immobilized lipase was more than 140 h.

Abstract:A core/sheath nanofiber membrane with poly[bis(p-methylphenoxy)]phospha-zene (PMPPh) as the sheath and easily spinnable polyacrylonitrile (PAN) as the core was prepared via a coaxial electrospinning process. Field-emission scanning electron microscopy and transmission electron microscopy were used to characterize the morphology of the nanofiber membrane. It was found that the concentration of the PAN spinning solution and the ratio of the core/sheath solution flow rates played a decisive role in the coaxial electrospinning process. In addition, the stabilized core/sheath PMPPh nanofiber membrane was investigated as a support for enzyme immobilization because of its excellent biocompatibility, high surface/volume ratio, and large porosity. Lipase from Candida rugosa was immobilized on the nanofiber membrane by adsorption. The properties of the immobilized lipase on the polyphosphazene nanofiber membrane were studied and compared with those of a PAN nanofiber membrane. The results showed that the adsorption capacity (20.4 ± 2.7 mg/g) and activity retention (63.7%) of the immobilized lipase on the polyphosphazene nanofiber membrane were higher than those on the PAN membrane.
Study on Immobilization of Phospholipase A_1 by Sodium Periodate Oxidation

Abstract:The phospholipase A_1 was immobilized on the cellulose membrane carrier by sodium periodate oxidation and the different influential factors of immobilization condition on the catalytic activity of phospholipase A_1 were studied.The optimal conditions of the immobilized lipase：the sodium periodate concentration was 0.15mol/L,activated 80min,which the lipase concentration was 0.05g/mL under the phosphate buffered solution with pH5.8 under 4℃,crosslinking for 4h.The phospholipase A_1 activities was 4.8U/cm~2.The immobilized lipase characterization：the optimal temperature for the immobilized lipase was 35℃;the optimal pH was 9.0,which drifted 1.0 to the alkaline side than that of free lipase;the immobilized lipase membrane was used for 7 times still with high activity above 65%.The results of SEM images showed that cellulose membrane could better immobilize lipase.

Abstract:In this study, a biphasic enzymatic membrane reactor was made by immobilizing Candida Rugosa lipase onto the dense surface of polysulfone ultrafiltration membrane by filtration and then cross-linking with glutaraldehyde solution. The reactor was further applied for the hydrolysis of olive oil, the performance of which was evaluated in respect of apparent reaction rate based on the amount of fatty acids extracted into the aqueous phase per minute and per membrane surface. It was found that the ultrafiltration and cross-linking process greatly improved the reaction rate per unit membrane area and the enzyme lifetime. The highest reaction rate reached 0.089μmol FFA/mincm2 when the enzyme loading density was 0.098mg/cm2. The results also indicated that the performance of lipase immobilized on the membrane surface was superior to that immobilized in the pores, and the apparent reaction rate and stability of immobilized lipases were improved greatly after cross-linking. It suggested that immobilization of enzymes by filtration and then cross-linking the enzymes onto the membrane surface is a simple and convenient way to prepare a high-activity immobilized enzyme membrane.

Lipase from Candida rugosa was covalently immobilized onto this membrane surface on which the carboxyl groups were activated with 1-ethyl-3-(dimethyl-aminopropyl) carbodiimide hydrochloride/N-hydroxyl succinimide as coupling agent. Using the hydrolysis reaction of p-nitrophenyl palmitate in aqueous and organic media, the properties of the immobilized lipase were assayed and compared with those of the free ones. Compared to the free enzyme (at 37°C), it was found that the maximum activity was observed at 45°C for the immobilized enzyme in the aqueous medium. On the other hand, the amount of added water in the organic medium of heptane showed greater effect on the reaction rate for the free lipase than the immobilized ones. The kinetic constants of the free and immobilized lipases, Km and Vmax, were assayed in the two media. Results indicated that the enzyme activity in heptane increased due to the immobilization and the specific activity was 0.233U/mg for the immobilized lipase, 0.204U/mg for the free ones, respectively. The residual activities of the immobilized enzyme were 62% in aqueous media and 67% in organic media, after 10 reuses.
Protection of Opening Lids: Very High Catalytic Activity of Lipase Immobilized on Core-Shell Nanoparticles.

Abstract:Various hydrophobic supports have been used for lipase immobilization since the active site of lipase can be opened in a hydrophobic environment. Nevertheless, the increase of lipase activity is still limited. This study demonstrates a hyperactivation-protection strategy of lipase after immobilization on poly(n-butyl acrylate)-polyaldehyde dextran (PBA-PAD) core-shell nanoparticles. The inner hydrophobic PBA domain helps to rearrange lipase conformation to a more active form after immobilization into the PAD shell. More importantly, the outer PAD shell with dense polysaccharide chains prevents the immobilized lipase from contact with outside aqueous medium and revert its conformation back to an inactive form. As a result, under optimal conditions the activity of lipase immobilized in PBA-PAD nanoparticles was enhanced 40 times over the free one, much higher than in any previous report. Furthermore, the immobilized lipase retained more than 80 % of its activity after 10 reaction cycles.

Abstract:A specially designed microstructure in the composite membrane with a porous hydrophobic layer and a relatively dense hydrophilic layer is proposed for lipase immobilization. Firstly, the composite membrane was prepared by coating the cellulose acetate layer with the average pore size of 1.40nm on the hydrophobic PTFE layer with the average pore size of 76.3nm. Then, enzymes were absorbed in pores of PTFE layer and deposited on the interface between the two layers by the filtration process. The relatively dense CA layer was used to reject the enzymes controlling the enzyme loading which prevented enzymes from being dissolved into the aqueous phase. The porous PTFE layer supplied a hydrophobic environment and a large specific surface area for the immobilization of lipases which were propitious to the activation of lipase. The activity of immobilized lipase membrane based on hydrolysis of olive oil was assayed in the biphasic membrane reactor and the maximum specific activity (1.20±0.04μmol-FFAmin−1cm−2) was found to be higher than the value reported in some literature. The kinetic parameters of the immobilized lipases, Km and kmax were fitted with the Michaelis–Menten equation. The Thiele modulus (φ) was used to evaluate the effect of the mass transfer through the membrane on the performance of reaction systems. The optimum enzyme loading (0.020±0.002mg-proteincm−2) was obtained with the highest activity and without the diffusion-limited. Furthermore, the immobilized lipases retained 80% residual activity after ten hydrolysis cycles. The composite membrane was easily regenerated and lipases immobilized in the regenerated membrane remained a high activity.
Immobilization of Lipase from Penicillium Expansum and Its Application to Transesterification of Corn Oil

Abstract:Lipase from Penicillium expansum was immobilized by physical adsorption. An inexpensive resin D4020 was selected as the support after screening 20 kinds of supports from different sources for the better immobilization performance. The effects of immobilization conditions on the immobilization efficiency and transesterification activity of the immobilized lipase were systematically studied. The results showed that the optimal enzyme amount, pH, and adsorption time were 0.7 g/g, 9.4, and 4 h, respectively. The transesterification activity of the immobilized lipase could be improved by using 0.5% galactose as a lyoprotectant. Under optimal conditions, the transesterification activity of the immobilized enzyme was 404.4 U/g, whereas the free enzyme did not exhibit transesterification activity. Subsequently, transesterification of corn oil to biodiesel catalyzed by the immobilized lipase was explored. Tert-amyl alcohol was found to be the most suitable reaction medium with a favorable amount of 0.5 ml/g. The optimum enzyme amount, water added, and reaction temperature were 60.6 U/g, 1.2%, and 35 degrees C, respectively. A three-step methanolysis protocol was used and one molar equivalent of methanol was added at a reaction time of 0, 2, and 6 h, respectively, by which a methyl ester yield of 85.0% was obtained after reaction for 24 h. Good operational stability was achieved after the enzyme was immobilized, and 62.8% of its original activity was retained after repeated use for 10 batches. The transesterification activity and operational stability of the Penicillium expansum lipase were enhanced remarkably by immobilization.
