Abstract:Obesity is a major health concren worldwide and is an important risk factor for heart diseases. The objective of this study was to evaluate the role of endothelin-1 receptor A (ETA) in obesity-associated cardiac dysfunctions. Cardiac specific ETA receptor knock-out (ETAKO) mice and their age- and gender-matched controls (C57BL/6J mice) were randomly assigned to receive either high- fat (45% fat) diet (HFD) or low-fat (10% fat) diet (LFD) starting from their 5-6 weeks of age for 24 weeks. Abstract:Background: Though autophagy including the selective mitophagy is essential for cellular homeostasis, its role in obese heart function remains controversial. Beclin-1 plays a cardinal role in conventional autophagy and has recently been implicated in alternative autophagy and mitophagy. This study was designed to examine the impact of Beclin-1 haploinsufficiency using heterozygous deletion of Beclin-1 (BCN-KO) on high fat diet (HFD)-induced cardiac anomalies. Methods and Results: Adult wild type (WT) and BCN-KO mice were fed low fat diet (LFD) or HFD for 12 weeks. HFD triggered comparable body weight gain in WT and BCN-KO mice. However, BCN-KO improved HFD-induced glucose intolerance and countered against HFD-induced cardiomyocyte contractile dysfunction manifested as decreased peak shortening, +dL/dt, -dL/dt and prolonged TR 90 . Levels of Beclin-1 were downregulated following HFD feeding although such effect was masked by BCN haploinsufficiency. Levels of autophagy markers LC3B-II/I and Atg5 were downregulated in a comparable manner in both HFD groups. Interestingly, Rab9, an essential regulator for alternative autophagy, was upregulated in HFD groups with a more pronounced effect in BCN-KO mice. Level of p62 was increased by HFD in WT but not BCN-KO group. Furthermore, the mitochondrial coupling protein for LC3B Bnip3 (also a key molecule for alternative autophagy-induced mitophagy) was upregulated in HFD groups with a more pronounced effect in BCN-KO mice. Moreover, Pink1 and AMPK phosphorylation, two key regulators in mitophagy were decreased by HFD in WT but not BCN-KO group. In line with these results, PGC-1 a (a marker of mitochondrial biogenesis) was downregulated by HFD, the effect of which was rescued by BCN-KO. Conclusion: Taken together, our data revealed that HFD led to cardiac contractile defect accompanied with dampened conventional autophagy and facilitated alternative autophagy. Our data suggested that adaptive mitophagy may play a pivotal role in BCN haploinsufficiency-induced beneficial effect. Such as beneficial effect was achieved through promoting HFD-induced alternative autophagy, which may contribute to increased mitophagy under HFD intake possibly via AMPK, Rab9 and Bnip3-mediated signaling pathways.

Abstract:Introduction: Obesity increases the chances of developing cardiovascular disease and strokes. The overabundance of adipose tissue that occurs in obese individuals increases oxidative stress, which trigger upregulation of antioxidant proteins, Sesn2 and Nrf2. Hypothesis: Obesity-related oxidative stress triggers cardiac Sesn2 and Nrf2 to function in cardioprotective properties. Methods: Cardiomyocyte specific Sesn2 knock out (cSesn2 -/- ) and overexpression (tTa-tet-Sesn2) C57BL/6J mice, and their wildtype littermates (Sesn2 f/f and tet-Sesn2) were subjected to either a high fat diet (HFD) or chow diet. Seahorse XF Analyzer for mitochondrial function and IonOptix Calcium and Contractility System for cardiomyocytes contractile functions. Results: The inducible cardiomyocyte overexpression of Sesn2 (tTa-tet-Sesn2) versus tet-Sesn2 mice gained less weight with HFD. Echocardiography data showed no significant differences in systolic function between tet-Sesn2 and tTa-tet-Sesn2 with HFD or chow diet. However, HFD induced diastolic dysfunction of tet-Sesn2 but not tTa-tet-Sesn2 hearts. Intriguingly, cardiac Nrf2 levels were upregulated in tTa-tet-Sesn2 but not tet-Sesn2 hearts with HFD. Conversely, cSesn2 -/- versus Sesn2 f/f showed lower Nrf2 levels and greater cardiac dysfunction with HFD. MitoSOX staining demonstrated that tTa-tet-Sesn2 versus tet-Sesn2 maintain redox homeostasis in the heart with HFD, while cSesn2 -/- versus Sesn2 f/f showed more reactive oxygen species (ROS) in the heart with HFD. Mitochondrial respiration was impaired in obese cSesn2 -/- versus Sesn2 f/f cardiomyocytes. Moreover, the contractility and transient calcium flux signaling was impaired in obese cSesn2 -/- versus Sesn2 f/f cardiomyocytes. tTa-tet-Sesn2 versus tet-Sesn2 cardiomyocytes demonstrated resistance to HFD-induced contractile dysfunctions, impaired calcium homeostasis and activation of the inflammation-related signaling NF-κB. Conclusions: Sesn2 and Nrf2 modulate redox homeostasis and inflammatory response during obesity pathogenesis. There is a feedforward loop relationship between Sesn2 and Nrf2 in the heart with HFD-induced obesity. Cardiac Sesn2/Nrf2 signaling ameliorates cardiac dysfunction caused by obesity.

Abstract:Endothelin (ET)-1 is implicated in the pathophysiology of cardiovascular diseases although its role in obesity anomalies has not been fully elucidated. This study was designed to examine the impact of ET-1 receptor A (ETA) ablation on obesity-induced changes in cardiac geometry and contractile function, as well as the mechanisms involved with a focus on autophagy. Cardiomyocyte-specific ETA receptor knockout (ETAKO) and WT mice were fed either low-fat (10% calorie from fat) or high-fat (45% calorie from fat) diet for 24 weeks. Glucose tolerance test was examined to confirm insulin resistance. High-fat diet intake compromised myocardial geometry (enlarged left ventricular diameters in systole and diastole), morphology (cardiac hypertrophy, increased wall thickness and interstitial fibrosis), contractile function (reduced fractional shortening, ejection fraction and cardiomyocyte shortening) and intracellular Ca2+ handling, the effect of which was significantly attenuated by ETAKO. TUNEL staining revealed overt apoptosis in high-fat-fed group, the effect was reverted by ETAKO. Western blot analysis noted that high-fat intake downregulated leptin receptor and PPARγ, insulin signaling (elevated basal/dampened insulin-stimulated phosphorylation of Akt and IRS1), phosphorylation of AMPK, ACC, upregulated GATA-4, ANP, NFATc3, PPARα, m-TOR/p70s6k signaling, which were attenuated by ETAKO with the exception of AMPK/ACC. Furthermore, high-fat intake suppressed cardiac autophagy, which was abrogated by ETAKO. In cultured murine cardiomyocytes, palmitic acid challenged mimicked high-fat diet-induced hypertrophic and autophagic responses, the effect of which were abolished by the ETA receptor antagonist BQ123 or mTOR inhibitor rapamycin. These results suggest that inhibition of ETA rescues high-fat intake-induced cardiac anomalies possibly through autophagy regulation.

Abstract:Obesity, a state of excessive delivery of energy to adipose tissues, imposes a devastating health threat and cardiovascular function including the increased prevalence of hypertension, insulin resistance, diabetes mellitus, cancer, inflammation, sleep dyspnea and other cardiovascular diseases. Approximately one-third of world populations are obese while effective management remains dismal. A better understanding of the obesity and metabolic disease pathophysiology seems to be essential for the proper management of obesity and cardiometabolic diseases. Knowledge gained over the last decades for the mechanisms that underpin obesity and metabolic diseases has provided a framework for energy imbalance, oxidative stress, apoptosis and sympathetic overactivation in the pathophysiology of these metabolic anomalies. Of particular interest is the role of autophagy, a conservative homeostatic process for cellular quality control through disposal and recycling of cellular components in the maintenance of energy homeostasis and organ function. Autophagy, in particular selective autophagy such as mitophagy, manages stress by selectively ridding the cells of potentially toxic proteins, lipids and organelles. Defects in autophagy and mitophagy may contribute to the development of cardiovascular anomalies of metabolic disorders prompting the pathogenesis of obesity, insulin resistance, diabetes, and atherosclerosis. Here we will highlight the changes of autophagy in particular mitophagy receptor protein FUN-14 containing domain 1 (FUNDC1) in response to nutrient stress, and changes of FUNDC1 may alter the course of cardiovascular function in obesity and metabolic diseases. Finally, the potential of pharmacological modulation of autophagy in the management of metabolic diseases and their cardiovascular complications will be addressed.

Abstract:Cardiac aging is manifested as cardiac remodeling and contractile dysfunction although precise mechanisms remain elusive. This study was designed to examine the role of endothelin-1 (ET-1) in aging-associated myocardial morphological and contractile defects. Echocardiographic and cardiomyocyte contractile properties were evaluated in young (5-6 months) and old (26-28 months) C57BL/6 wild-type and cardiomyocyte-specific ET(A) receptor knockout (ETAKO) mice. Cardiac ROS production and histology were examined. Our data revealed that ETAKO mice displayed an improved survival. Aging increased plasma levels of ET-1 and Ang II, compromised cardiac function (fractional shortening, cardiomyocyte peak shortening, maximal velocity of shortening/relengthening and prolonged relengthening) and intracellular Ca(2+) handling (reduced intracellular Ca(2+) release and decay), the effects of which with the exception of ET-1 and Ang II levels was improved by ETAKO. Histological examination displayed cardiomyocyte hypertrophy and interstitial fibrosis associated with cardiac remodeling in aged C57 mice, which were alleviated in ETAKO mice. Aging promoted ROS generation, protein damage, ER stress, upregulated GATA4, ANP, NFATc3 and the autophagosome cargo protein p62, downregulated intracellular Ca(2+) regulatory proteins SERCA2a and phospholamban as well as the autophagic markers Beclin-1, Atg7, Atg5 and LC3BII, which were ablated by ETAKO. ET-1 triggered a decrease in autophagy and increased hypertrophic markers in vitro, the effect of which were reversed by the ET(A) receptor antagonist BQ123 and the autophagy inducer rapamycin. Antagonism of ET(A), but not ET(B) receptor, rescued cardiac aging, which was negated by autophagy inhibition. Taken together, our data suggest that cardiac ET(A) receptor ablation protects against aging-associated myocardial remodeling and contractile dysfunction possibly through autophagy regulation.

Abstract:The cysteine protease cathepsin K has been implicated in pathogenesis of cardiovascular disease. We hypothesized that ablation of cathepsin K protects against obesity-associated cardiac dysfunction. Wild-type mice fed a high-fat diet exhibited elevated heart weight, enlarged cardiomyocytes, increased left ventricular wall thickness, and decreased fractional shortening. All these changes were reconciled in cathepsin K knockout mice. Cathepsin K knockout partly reversed the impaired cardiomyocyte contractility and dysregulated calcium handling associated with high-fat diet. Additionally, cathepsin K knockout alleviated whole-body glucose intolerance and improved insulin-stimulated Akt phosphorylation in high-fat diet-fed mice. High-fat feeding increased the expression of cardiac hypertrophic proteins and apoptotic markers, which were inhibited by cathepsin K knockout. Furthermore, high-fat feeding resulted in cathepsin K release from lysosomes into the cytoplasm. In H9c2 myoblasts, silencing of cathepsin K inhibited palmitic acid-induced release of cytochrome c from mitochondria and expression of proapoptotic signaling molecules. Collectively, our data indicate that cathepsin K contributes to the development of obesity-associated cardiac hypertrophy and may represent a potential target for the treatment to obesity-associated cardiac anomalies.

Abstract:Obesity, defined as accumulation of excessive body fat, has grown dramatically and imposes a devastating, pandemic health threat. If uncorrected, obesity may confer an increased susceptibility to the onset and development of insulin resistance, hypertension, type 2 diabetes, and the cadre of metabolic syndrome. More importantly, evidence from our group and other laboratories has demonstrated that obesity contributes to the development of cardiac anomalies. Although multiple signaling pathways have been identified as responsible for cardiac dysfunction in obese individuals, the precise mechanism still remains elusive.The autophagy-lysosome pathway, which regulates the degradation of large intracellular organelles and protein aggregates, is essential for the maintenance of cardiomyocytes under physiological and pathological conditions. Recent studies have suggested that the autophagy pathway plays a pivotal role in the development of cardiac anomalies induced by obesity. In the present chapter, we focus on how autophagy is involved in the regulation of cardiac function in obesity and metabolic syndrome.

Abstract:Obesity has become an epidemic and is associated with cardiovascular anomalies. Obesity-induced low-grade chronic inflammation and macrophage infiltration into the heart plays a critical role in the development of cardiac dysfunction. Because the adaptor protein caspase recruitment domain-containing protein 9 (CARD9) regulates the innate immunity in macrophages via activation of pro-inflammatory cytokines, we hypothesized that CARD9 mediates chronic inflammation and plays a detrimental role in obesity-associated cardiac dysfunction. Male C57BL/6 wild-type (WT) and CARD9 knockout (CARD9-/-) mice were fed normal chow (ND, 12% fat) or a Western diet (WD, 45% fat) for 5 months starting at 4 weeks of age. At the end of 5-month WD feeding, cardiac geometry and function were evaluated using echocardiography. Immunofluorescence assay was performed to detect macrophage infiltration in the heart. Heart tissue homogenates, plasma, and supernatants from cultured macrophages were collected to measure the activities of pro-inflammatory cytokines by ELISA kits. Western immunoblotting analyses were performed on isolated peritoneal macrophages and heart tissue to dissect the pro-inflammatory signaling pathways. WD feeding induced insulin resistance and glucose intolerance in the WT mice and the effects were attenuated in the CARD9-/- mice. Myocardial fractional shortening and ejection fraction were significantly compromised in the WD-fed WT mice and CARD9 knockout preserved cardiac function. CARD9 knockout also significantly decreased the number of infiltrated macrophages in the heart. In addition, myocardium-, plasma- and macrophage-derived cytokines IL-6, IL-1β and TNFα were attenuated in the CARD9-/- mice compared to the WT mice. WD feeding resulted in increased protein phosphorylation levels of p38 MAPK in the heart and macrophages which were suppressed in the CARD9-/- mice. Taken together, these results suggest that CARD9 knockout ameliorates obesity-induced cardiac dysfunction, potentially through reduction of macrophage infiltration and suppression of p38 MAPK phosphorylation in the heart.
Inhibition of ER Stress Using Tauroursodeoxycholic Acid Rescues Obesity-Evoked Cardiac Remodeling and Contractile Anomalies through Regulation of Ferroptosis

Abstract:Interrupted ER homeostasis contributes to the etiology of obesity cardiomyopathy although it remains elusive how ER stress evokes cardiac anomalies in obesity. Our study evaluated the impact of ER stress inhibition on cardiac anomalies in obesity. Lean and ob/ob obese mice received chemical ER chaperone tauroursodeoxycholic acid (TUDCA, 50 mg/kg/d, p.o.) for 35 days prior to evaluation of glucose sensitivity, echocardiographic, myocardial geometric, cardiomyocyte mechanical and subcellular Ca 2+ property, mitochondrial integrity, oxidative stress, apoptosis, and ferroptosis. Intracellular Ca 2+ governing domains including sarco(endo)plasmic reticulum Ca 2+ -ATPase (SERCA) were monitored by 45 Ca 2+ uptake and immunoblotting. Our results noted that TUDCA alleviated myocardial remodeling (fibrosis, hypertrophy, enlarged LVESD), echocardiographic anomalies (compromised fractional shortening and ejection fraction), cardiomyocyte contractile dysfunction (amplitude and velocity of cell shortening, relengthening time) and intracellular Ca 2+ anomalies (compromised subcellular Ca 2+ release, clearance and SERCA function), mitochondrial damage (collapsed membrane potential, downregulated mitochondrial elements and ultrastructural alteration), ER stress (GRP78, eIF2α and ATF4), oxidative stress, apoptosis and ferroptosis [downregulated SLC7A11, GPx4 and upregulated transferrin receptor (TFRC)] without affecting global glucose sensitivity and serum Fe 2+ in obese mice. Obesity-evoked change in HSP90, phospholamban and Na + -Ca 2+ exchanger was spared by the chemical ER chaperone. Moreover, in vitro results noted that TUDCA, PERK inhibitor GSK2606414, TFRC neutralizing antibody and ferroptosis inhibitor LIP1 mitigated palmitic acid-elicited changes in lipid peroxidation and mechanical function. Our findings favored a role for ferroptosis in obesity cardiomyopathy downstream of ER stress.

Abstract:Type 2 diabetes mellitus (T2D) contributes to sustained inflammation and myopathic changes in the heart although the precise interplay between the two remains largely unknown. This study evaluated the impact of deficiency in CD74, the cognate receptor for the regulatory cytokine macrophage migration inhibitory factor (MIF), in T2D-induced cardiac remodeling and functional responses, and cell death domains involved. WT and CD74<sup>-/-</sup> mice were fed a high fat diet (60% calorie from fat) for 8 weeks prior to injection of streptozotocin (STZ, 35 mg/kg, i.p., 3 consecutive days) and were maintained for another 8 weeks. KEGG analysis for differentially expressed genes (DEGs) reported gene ontology term related to ferroptosis in T2D mouse hearts. T2D patients displayed elevated plasma MIF levels. Murine T2D exerted overt global metabolic derangements, cardiac remodeling, contractile dysfunction, apoptosis, pyroptosis, ferroptosis and mitochondrial dysfunction, ablation of CD74 attenuated T2D-induced cardiac remodeling, contractile dysfunction, various forms of cell death and mitochondrial defects without affecting global metabolic defects. CD74 ablation rescued T2D-evoked NLRP3-Caspase1 activation and oxidative stress but not dampened autophagy. In vitro evidence depicted that high glucose/high fat (HGHF) compromised cardiomyocyte function and promoted lipid peroxidation, the effects were ablated by inhibitors of NLRP3, pyroptosis, and ferroptosis but not by the mitochondrial targeted antioxidant mitoQ. Recombinant MIF mimicked HGHF-induced lipid peroxidation, GSH depletion and ferroptosis, the effects of which were reversed by inhibitors of MIF, NLRP3 and pyroptosis. Taken together, these data suggest that CD74 ablation protects against T2D-induced cardiac remodeling and contractile dysfunction through NLRP3/pyroptosis-mediated regulation of ferroptosis.

CARD9 Knockout Ameliorates Myocardial Dysfunction Associated with High Fat Diet-Induced Obesity.

Abstract:Obesity is associated with chronic inflammation which plays a critical role in the development of cardiovascular dysfunction. Because the adaptor protein caspase recruitment domain-containing protein 9 (CARD9) in macrophages regulates innate immune responses via activation of pro-inflammatory cytokines, we hypothesize that CARD9 mediates the pro-inflammatory signaling associated with obesity en route to myocardial dysfunction. C57BL/6 wild-type (WT) and CARD9(-/-) mice were fed normal diet (ND, 12% fat) or a high fat diet (HFD, 45% fat) for 5months. At the end of 5-month HFD feeding, cardiac function was evaluated using echocardiography. Cardiomyocytes were isolated and contractile properties were measured. Immunofluorescence was performed to detect macrophage infiltration in the heart. Heart tissue homogenates, plasma, and supernatants from isolated macrophages were collected to measure the concentrations of pro-inflammatory cytokines using ELISA kits. Western immunoblotting analyses were performed on heart tissue homogenates and isolated macrophages to explore the underlying signaling mechanism(s). CARD9 knockout alleviated HFD-induced insulin resistance and glucose intolerance, prevented myocardial dysfunction with preserved cardiac fractional shortening and cardiomyocyte contractile properties. CARD9 knockout also significantly decreased the number of infiltrated macrophages in the heart with reduced myocardium-, plasma-, and macrophage-derived cytokines including IL-6, IL-1β and TNFα. Finally, CARD9 knockout abrogated the increase of p38 MAPK phosphorylation, the decrease of LC3BII/LC3BI ratio and the up-regulation of p62 expression in the heart induced by HFD feeding and restored cardiac autophagy signaling. In conclusion, CARD9 knockout ameliorates myocardial dysfunction associated with HFD-induced obesity, potentially through reduction of macrophage infiltration, suppression of p38 MAPK phosphorylation, and preservation of autophagy in the heart.

Abstract:Background: Macrophages are key players in obesity-associated cardiovascular diseases, which are marked by inflammatory and immune alterations. However, the pathophysiological mechanisms underlying macrophage's role in obesity-induced cardiac inflammation are incompletely understood. Our study aimed to identify the key macrophage population involved in obesity-induced cardiac dysfunction and investigate the molecular mechanism that contributes to the inflammatory response. Methods: In this study, we used single-cell RNA-sequencing analysis of Cd45+CD11b+F4/80+ cardiac macrophages to explore the heterogeneity of cardiac macrophages. The CCR2+ (C-C chemokine receptor 2) macrophages were specifically removed by a dual recombinase approach, and the macrophage CCR2 was deleted to investigate their functions. We also performed cleavage under target and tagmentation analysis, chromatin immunoprecipitation-polymerase chain reaction, luciferase assay, and macrophage-specific lentivirus transfection to define the impact of lysozyme C in macrophages on obesity-induced inflammation. Results: We find that the Ccr2 cluster undergoes a functional transition from homeostatic maintenance to proinflammation. Our data highlight specific changes in macrophage behavior during cardiac dysfunction under metabolic challenge. Consistently, inducible ablation of CCR2+CX3CR1+ macrophages or selective deletion of macrophage CCR2 prevents obesity-induced cardiac dysfunction. At the mechanistic level, we demonstrate that the obesity-induced functional shift of CCR2-expressing macrophages is mediated by the CCR2/activating transcription factor 3/lysozyme 1/NF-κB (nuclear factor kappa B) signaling. Finally, we uncover a noncanonical role for lysozyme 1 as a transcription activator, binding to the RelA promoter, driving NF-κB signaling, and strongly promoting inflammation and cardiac dysfunction in obesity. Conclusions: Our findings suggest that lysozyme 1 may represent a potential target for the diagnosis of obesity-induced inflammation and the treatment of obesity-induced heart disease.

Abstract:ER stress is defined as an imbalance between protein synthesis and protein folding capacity, resulting in the accumulation of misfolded or unfolded protein in ER. Temperate ER stress through UPR enhances the folding capacity of ER and restores the ER homeostasis, exerting a cell protection role. While prolonged ER stress lead to inflammation, cell dysfunction and apoptosis, which takes part in the progression of insulin resistance and atherosclerosis. In the condition of obesity or hyperhomocysteinemia, ER stress in adipose tissue, hypothalamus, liver and other tissue or organs causes the dysregulation of adipokines secretion as well as abnormal receptor or post-receptor signal transduction, which plays an significant role in cardiometabolic disease.

Abstract:Background: Toll-like receptor 4 (TLR4) is an innate proinflammatory mediator found in a wide variety of cell types including cardiomyocytes. The autophagy-lysosome pathway, a major pathway governing protein and organelle degradation and recycling, plays a pivotal role in maintaining cardiac homeostasis under physiological and pathological conditions. The aim of this study was to evaluate the impact of TLR4 knockout (TLR4 -/- ) on high fat diet (HFD)-induced cardiomyopathy and the underlying mechanisms involved, with a focus on inflammation and autophagy pathways. Methods: Wild type (WT) and TLR4 -/- mice were fed on low fat diet (LFD) or HFD for 12 weeks. Metabolic rate and glucose tolerance were measured in WT and TLR4 -/- mice at the end of fat diet intake. Echocardiographic, cardiomyocyte mechanical function, morphological feature, aconitase activity, ROS generation and immunoblotting were assessed. Results: TLR4 -/- did not prevent HFD-induced obesity and insulin resistance, as evidenced by body weight gain and glucose intolerance. In addition, there was little difference in metabolic parameters (V O2 , V CO2 , RER, energy expenditure and physical activity) between WT and TLR4 -/- mice fed with HFD. However, TLR4 -/- alleviated HFD-elicited cardiac hypertrophy and contractile dysfunction. HFD-feeding caused extensive mitochondrial injury and ROS generation, which were alleviated by TLR4 -/- . TLR4 -/- dramatically attenuated inflammation signaling (up-regulated p-IKβ, NF-κB and p-JNK) in the face of HFD intake. Cardiac autophagy was significantly suppressed by HFD, as evidenced by up-regulated p-mTOR, down-regulated p-AMPK, Atg5, Atg12, LC3BII, and up-regulated P62 in HFD-induced cardiomyopathy. Furthermore, in vitro study revealed that the TLR4 inhibitor-TAK-242 reconciled palmitic acid-induced cardiomyocyte autophagy and contractile anomalies. Conclusions: Our results suggested that TLR4 -/- does not prevent HFD-induced obesity and insulin resistance. However, TLR4 is a culprit factor for cardiac dysfunction following HFD intake. The cardioprotective effect of TLR4 -/- against HFD-induced cardiomyopathy is associated with attenuation of myocardial inflammation and recovery of cardiac autophagy activity.

Abstract:Autophagy plays an important role in the pathogenesis of cardiovascular diseases. Dysregulation of autophagy may have a huge effect on cardiac hypertrophy induced by overload pressure although reports on autophagy and cardiac hypertrophy have been contradictory. Some studies showed that autophagy activation attenuated cardiac hypertrophy. However, others suggested that inhibition of autophagy would be protective. Different research models or different pathways involved could be responsible for it. Cardiac hypertrophy may be alleviated through regulation of autophagy. This review aims to highlight the pathways and therapeutic targets identified in the prevention and treatment of cardiac hypertrophy by regulating autophagy.
Disruption of calpain reduces lipotoxicity-induced cardiac injury by preventing endoplasmic reticulum stress.

Abstract:Diabetes and obesity are prevalent in westernized countries. In both conditions, excessive fatty acid uptake by cardiomyocytes induces cardiac lipotoxicity, an important mechanism contributing to diabetic cardiomyopathy. This study investigated the effect of calpain disruption on cardiac lipotoxicity. Cardiac-specific capns1 knockout mice and their wild-type littermates (male, age of 4weeks) were fed a high fat diet (HFD) or normal diet for 20weeks. HFD increased body weight, altered blood lipid profiles and impaired glucose tolerance comparably in both capns1 knockout mice and their wild-type littermates. Calpain activity, cardiomyocyte cross-sectional areas, collagen deposition and triglyceride were significantly increased in HFD-fed mouse hearts, and these were accompanied by myocardial dysfunction and up-regulation of hypertrophic and fibrotic collagen genes as well as pro-inflammatory cytokines. These effects of HFD were attenuated by disruption of calpain in capns1 knockout mice. Mechanistically, deletion of capns1 in HFD-fed mouse hearts and disruption of calpain with calpain inhibitor-III, silencing of capn1, or deletion of capns1 in palmitate-stimulated cardiomyocytes prevented endoplasmic reticulum stress, apoptosis, cleavage of caspase-12 and junctophilin-2, and pro-inflammatory cytokine expression. Pharmacological inhibition of endoplasmic reticulum stress diminished palmitate-induced apoptosis and pro-inflammatory cytokine expression in cardiomyocytes. In summary, disruption of calpain prevents lipotoxicity-induced apoptosis in cardiomyocytes and cardiac injury in mice fed a HFD. The role of calpain is mediated, at least partially, through endoplasmic reticulum stress. Thus, calpain/endoplasmic reticulum stress may represent a new mechanism and potential therapeutic targets for cardiac lipotoxicity.
