Sensory Nerves Sensitive to Capsaicin Involve in the Visceral Hypersensitivity in Adult Rats Exposed to Chronic Unpredictable Stress

Abstract:Objective To find the evidences of involvement of neurogenic inflammation in changes of visceral sensitivity in adult rats exposed to chronic unpredictable stress (CUS).Methods The sensory denervation model of rats treated with capsaicin were exposed to CUS for 21 days.The visceral sensitivity was assessed by abdominal withdrawal reflex (AWR) score,and inflammation of whole colon mucosa was assessed by tissue myeloperoxidase (MPO) activity.The mast cell (MC) was stained with toludine blue-erosin and its ultrastructure change was observed using electron microscopy.oxantrazole,a stabilizer of MC membrane,was used to investigate the possible role of MC.Results ① After one week of CUS,the AWR score was significantly increased at rectal distention of 0.4 ml (2.56±0.50),0.8 ml (3.76±0,50) and 1.2 ml (3.89±0.40) compared with controls (1.25±0.50,1.41±0.30,1.74±0.40,respectively,all P values <0.05).Capsaicin administration,however,inhibited the elevation of AWR score induced by CUS (P<0.05).② The MPO activity of whole colon mucosa was increased by CUS,whereas it was inhibited by capsaicin and/or doxantrazole administration (P<0.05).③ The number of MCs in proximal colon of the rats exposed to CUS was increased,but it was inhibited by doxantrazole.④Electron microscopy revealed some pathologic changes including a close proximity of MCs,nerve terminals and blood vessels,proliferation and thickness of blood vessels and apoptosis of cells.Conclusion Neurogenic inflammation is one of the mechanisms of visceral hypersensitivity of rats exposed to CUS and MC may involve in it.
