Comparative Study on the Anti-nociceptive Effects of Radix Angelicae Dahuricae Decoction and Its Lyophilized Powder

Abstract:Objective To compare the anti-nociceptive effects of two dosage forms of Radix Angelicae dahuricae(AD),the decoction(ADd)and the lyophilized powder(ADp),on peripheral tissue injury caused persistent spontaneous pain and hyperalagesia.Methods Two models for persistent pain test induced by bee venom(BV)and formalin were used to observe and compare the anti-nociceptive effects.Results (1)Compared with the control group treated with normal saline,ADd and ADp in different concentrations(0.1 g/ml,0.4 g/ml,and 2.0 g/ml,10 mL/kg),could significantly reduce the times of spontaneous paw flinching reflex(SPFR)induced by BV,showing insignificant difference between the two dosage forms;those of 0.4 g/ml also showed significant suppression on formalin induced SPFR;(2)ADd of the latter two dosages administered before stimulation could effectively inhibit the primary heat hyperalagesia induced by BV; but the inhibition to primary mechanical hyperalagesia only showed by ADd of 2.0 g/ml;(3)the anti-nociceptive effect of ADd to BV induced SPFR and primary heat hyperalgesia could not be reversed by naloxone,a non-selective opioid receptor antagonist.Conclusion Both dosage forms of AD have anti-nociceptive effect on the persistent spontaneous pain primary heat/mechanical hyperalagesia indaced by peripheral tissue danage in dose-dependent manner,but the analgesia of AD is not mediated by endogenous opioid receptor.
