Promoter Methylation of Death-Associated Protein Kinase Gene and Calgranulin B Mrna As Indicators of Therapeutic Effect in Barrett′s Esophagus

Abstract:Objectives To evaluate the therapeutic effect of endoscopic mucosal resection with ligator on Barrett′s esophagus by detecting promoter methylation of death-associated protein kinase gene and calgranulin B mRNA in esophageal mucosa. Methods Fifty-seven patients with Barrett′s esophagus received endoscopic mucosal resection with ligator. Endoscopic finding, pathologic diagnosis, promoter methylation of DAPK gene, calgranulin B mRNA and fecal calgranulin B were documented before and after the treatment, respectively. Promoter methylation of DAPK gene was detected by methylation-specific PCR. Calgranulin B mRNA was observed by real-time PCR. Fecal calgranulin B protein was determined by enzyme-linked immunosorbent assay. Results There were significant differences in promoter methylation of DAPK gene and calgranulin B mRNA before and after treatment. As for fecal calgranulin B protein, no change occurred. No correlation was revealed between mucosal calgranulin B mRNA and fecal calgranulin B protein. Conclusions Endoscopic mucosal resection with ligator is a safe and effective treatment for Barrett′s esophagus. Promoter methylation of DAPK gene and calgranulin B mRNA in esophageal mucosa reflect the therapeutic effect on Barrett′s esophagus.
