Pharmacokinetics and Bioavail Ability Studies of Colistin Sulfate in Pigs

Abstract:Pharmacokinetics and bioavailability of colistin were studied in 5 healthy pigs following single intravenous(2 5 mg/kg b.w.)and intramuscular (2 5 mg/kg b.w.and 5 0 mg/kg b.w.)by clinder plate assay of microbiological method.The colistin sulfate concentration time data were analysed with 3P97 program.Injection was separately administered into muscle(IM) with the dose of 2 5 and 5 0 mg/kg body weight,whose Cmax were 3 73±0 28 and 6 40±0 18μg/ml;Tmax were 32 22±1 51 min and 34 08±1 76min;t 1/2β were 255 99±13 65 min and 264 08±28 57 min;injection was administered into aural vein(IV) with the dose of 2 5 mg/kg body weight,whose t 1/2β was 251 28±12 53 min.Examples of the experimentally determined serum concentration of colistin sulfate generated two exponential model with first order absorption.The mean absolutely bioavailability of 2 5 and 5 0 mg/kg b.w.(IM)were 98 30% and 88 54%.
