Abstract:Background: Soy isoflavones, such as genistein and daidzein, are bioflavonoids found in soy products that are able to interact with various hormones such as estrogen. Epidemiological studies reveal a proper level of isoflavones in diet can prevent many diseases like cancers or diabetes. Therefore, it is important to study the biotransformation and xenobiotic metabolism of soy isoflavones. Methods: A systematic review of published studies was carried out to investigate the characterization of isoflavones and their metabolites, sample pretreatment and quantitative analysis of isoflavones, and the influence of soy isoflavones on drug and xenobiotic metabolism. Results: Aglycones with weak estrogen-like activities are the biologically active forms of the soy isoflavones in mammals. The most recent advances including extraction, purification and detection of isoflavones in soybean and soy products are discussed. The effects of soy isoflavones on drug and xenobiotic metabolism involve in regulation of phase I cytochrome P450 (CYPs) enzyme and phase I detoxifying enzymes expression and activity. At the molecular level, soy isoflavones have proved capable of estrogenic/antiestrogenic with tissue-selective, anti-cancer, antiobesity, anti-oxidation, and tyrosine kinase inhibition activities. Conclusion: This review summarized different aspects of soy isoflavones and their molecular mechanisms of pharmacological action on xenobiotic, which demonstrated that soy isoflavones can decrease the incidence of many diseases and benefit for human health. However, since the lack of clinical research for evaluation of the proper dosage of intake of soy isoflavones in diet or adjunctive therapy, there is a need for further studies on the selection of doses, biomedical applications and adverse effects of isoflavones for human health.

Abstract:Soybeans and their food products exist in the market in various forms, ranging from crude oils and bean meals to nutritious products (e.g. soy milk powers). With the availability of technologies for mass production of soy products and for enrichment of soy components (e.g. phospholipids, saponins, isoflavones, oligosaccharides and edible fiber), the nutritional values of soy products have been enhanced remarkably, offering the potential for functional food development. Among different bioactive components in soybeans, one important component is isoflavones, which have been widely exploited for health implications. While there are studies supporting the health benefits of isoflavones, concerns on adverse effects have been raised in the literature. The objective of this article is to review the recent understanding of the biological activities, adverse effects, and use of isoflavones in functional food development.

Abstract:Soybeans are rich in proteins and lipids and have become a staple part of the human diet. Besides their nutritional excellence, they have also been shown to contain various functional components, including isoflavones, and have consequently received increasing attention as a functional food item. Isoflavones are structurally similar to 17-β-estradiol and bind to estrogen receptors (ERα and ERβ). The estrogenic activity of isoflavones ranges from a hundredth to a thousandth of that of estrogen itself. Isoflavones play a role in regulating the effects of estrogen in the human body, depending on the situation. Thus, when estrogen is insufficient, isoflavones perform the functions of estrogen, and when estrogen is excessive, isoflavones block the estrogen receptors to which estrogen binds, thus acting as an estrogen antagonist. In particular, estrogen antagonistic activity is important in the breast, endometrium, and prostate, and such antagonistic activity suppresses cancer occurrence. Genistein, an isoflavone, has cancer-suppressing effects on estrogen receptor-positive (ER+) cancers, including breast cancer. It suppresses the function of enzymes such as tyrosine protein kinase, mitogen-activated kinase, and DNA polymerase II, thus inhibiting cell proliferation and inducing apoptosis. Genistein is the most biologically active and potent isoflavone candidate for cancer prevention. Furthermore, among the various physiological functions of isoflavones, they are best known for their antioxidant activities. S-Equol, a metabolite of genistein and daidzein, has strong antioxidative effects; however, the ability to metabolize daidzein into S-equol varies based on racial and individual differences. The antioxidant activity of isoflavones may be effective in preventing dementia by inhibiting the phosphorylation of Alzheimer’s-related tau proteins. Genistein also reduces allergic responses by limiting the expression of mast cell IgE receptors, which are involved in allergic responses. In addition, they have been known to prevent and treat various diseases, including cardiovascular diseases, metabolic syndromes, osteoporosis, diabetes, brain-related diseases, high blood pressure, hyperlipidemia, obesity, and inflammation. Further, it also has positive effects on menstrual irregularity in non-menopausal women and relieving menopausal symptoms in middle-aged women. Recently, soybean consumption has shown steep increasing trend in Western countries where the intake was previously only 1/20–1/50 of that in Asian countries. In this review, Ihave dealt with the latest research trends that have shown substantial interest in the biological efficacy of isoflavones in humans and plants, and their related mechanisms.

Abstract:To investigate the effects of a synthetic soybean isoflavone (ISF) on growth performance, meat quality, and antioxidation in male broilers, 1,500 birds that were 42 d old were allotted to 5 treatments with 6 replicates per treatment (50 birds per replicate). Birds were fed diets supplemented with 0, 10, 20, 40, or 80 mg of ISF/kg, respectively, for a period of 3 wk ad libitum. The results showed that dietary supplementations with 10 or 20 mg of ISF/kg increased weight gain by 13.6 and 16.2% (P < 0.01) and elevated feed intake by 7.37% (P < 0.05) and 11.2% (P < 0.01), respectively. Addition of 10 mg of ISF/kg decreased feed:gain by 5.5% (P < 0.05). Supplementation with 40 mg of ISF/kg in the diet slightly increased water-holding capacity by 17.24% (P < 0.1), and the addition of 20 or 40 mg/kg of ISF significantly increased the pH value of meat (P < 0.01), although adding 40 or 80 mg of ISF/kg increased the lightness of meat color (P < 0.05). Malondialdehyde production was slightly reduced in plasma of 20 mg of ISF/kg supplemented chickens (P < 0.1) and significantly decreased in breast muscles of 20, 40, or 80 mg of ISF/kg supplemented chickens (P < 0.01). The addition of 40 or 80 mg of ISF/kg significantly increased total antioxidant capability (P < 0.01) and slightly elevated total superoxide dismutase activity (P < 0.1) in plasma of chickens. The dose of 80 mg of ISF/kg slightly improved catalase activity in plasma (P < 0.06). In breast muscle, treatment of birds with 40 or 80 mg of ISF/kg caused an increase of total superoxide dismutase activity by 25.36% (P < 0.05) or 63.93% (P < 0.01). Catalase activity significantly increased by 70.61% by the supplemental ISF at the 40-mg level (P < 0.05). Also, 10, 20, or 40 mg doses of ISF/kg decreased lactic acid production (P < 0.05). The results of this study indicate that dietary ISF could improve growth performance and meat quality by decreasing lipid peroxidation and improving antioxidative status in male broilers.

Abstract:Soy and soy-based foods are considered healthy, particularly in many Asia-Pacific countries, where soy products have long been consumed. Soy and soy-related products have been found to help prevent the occurrence of cardiovascular diseases and certain types of cancer, such as breast and prostate cancer. These products can also have antioxidative effects that alleviate hot flashes during menopause and bone loss. These biological and therapeutic functions are primarily due to the isoflavones derived from soy, whose structure is similar to the structure of 17-β-oestradiol. Despite the many health benefits for humans and animals, the application of isoflavones remains controversial because of their anti-oestrogenic properties. We focused on general information regarding isoflavones, as well as their structure, function, and application. We summarized evidence showing that dietary or supplemental isoflavones exert protective effects on the health of humans and animals. Based on the literature, we conclude that soy foods and isoflavones may be effective and safe; however, more high-quality trials are needed to fully substantiate their potential use.

Abstract:Isoflavones, rich in soybean, are currently receiving much attention because of their potential role in preventing and treating cancer and other human chronic diseases. The present review provides an overview of the recent results in this research field. Data from epidemiological reports and laboratories have shown that isoflavones have multi-biological and pharmacological effects in animals and humans. These include estrogenic and antiestrogenic effects, cell signalling conduction, as well as cell growth and death. Based on these properties, soy protein and isoflavones have been associated with reduced incidences of breast and prostate cancers, cardiovascular diseases or osteoporosis, and exhibit some other favorable effects. The mechanism through which isoflavones may exert the above-mentioned functions are not only based on the estrogenic properties of isoflavones, but also on their role as protein tyrosine kinase inhibitors, as regulators of gene transcription, modulators of transcription factors, antioxidants, as well as by altering some enzyme activities. However, to draw a clear conclusion regarding the clinical use of isoflavones further investigation would be required, although only a few effects of short- or long-term use of soy proteins are known in humans.

Abstract:Background: Isoflavones are naturally occurring flavonoids, commonly found in the food consumed for centuries in the East-Asian population, characterized by a structure able to exert nonsteroidal estrogen-like activity on human cells. They have attracted researcher interest all around the word, following the results obtained in epidemiological and clinical studies. The involvement of isoflavones and their metabolites in various biological processes suggests that they can influence several metabolic pathways and can influence the gene expression at epigenetic level, involving effects that probably are due to early life exposure. They show positive health effects on several diseases, especially in the prevention of coronary heart and neurological diseases, hormone-related cancers, osteoporosis, and postmenopausal symptoms. Methods: We have performed a critical evaluation of available literature trough a structured search of bibliographic databases about isoflavones health promoting properties, risk assessment and mechanisms of action. In addition, we supplied useful information on their biochemical properties, sources and bioavailability. Results: Although these molecules have been the subjects of numerous researches, their role for the wellness of the human organism remains controversial. Moreover, there are substantial inconsistencies between the results obtained by epidemiologic studies conducted on Eastern population, which found high health promoting properties, and Western clinical trials, which found much less positive effects. Conclusion: Further epidemiologic studies and well-designed prospective human studies are to determine the beneficial effects of isoflavones exposure, as well as establishing its safe therapeutic.

Abstract:Isoflavones are phytoestrogens with potent estrogenic activity; genistein, daidzein and glycitein are the most active isoflavones found in soy beans. Phytoestrogens have similarity in structure with the human female hormone 17-β-estradiol, which can bind to both alpha and beta estrogen receptors, and mimic the action of estrogens on target organs, thereby exerting many health benefits when used in some hormone-dependent diseases. Numerous clinical studies claim benefits of genistein and daidzein in chemoprevention of breast and prostate cancer, cardiovascular disease and osteoporosis as well as in relieving postmenopausal symptoms. The ability of isoflavones to prevent cancer and other chronic diseases largely depends on pharmacokinetic properties of these compounds, in particular absorption and distribution to the target tissue. The chemical form in which isoflavones occur is important because it influences their bioavailability and, therefore, their biological activity. Glucose-conjugated isoflavones are highly polar, water-soluble compounds. They are hardly absorbed by the intestinal epithelium and have weaker biological activities than the corresponding aglycone. Different microbial families of colon can transform glycosylated isoflavones into aglycones. Clinical studies show important differences between the aglycone and conjugated forms of genistein and daidzein. The evaluation of isoflavone metabolism and bioavailability is crucial to understanding their biological effects. Lipid-based formulations such as drug incorporation into oils, emulsions and self-microemulsifying formulations have been introduced to increase bioavailability. Complexation with cyclodextrin also represent a valid method to improve the physicochemical characteristics of these substances in order to be absorbed and distributed to target tissues. We review and discuss pharmacokinetic issues that critically influence the biological activity of isoflavones.

A review on the effect of soy bioactive components on growth and health outcomes in pigs and broiler chickens

Abstract:While soy products have long been included in animal diets for their macronutrient fractions, more recent work has focused on the immunomodulatory potential of bioactive components of this feedstuff. This comprehensive review aims to identify the current state of knowledge on minor soy fractions and their impact on the health and growth of pigs and broiler chickens to better direct future research. A total of 7,683 publications were screened, yet only 151 were included in the review after exclusion criteria were applied, with the majority (n = 87) of these studies conducted in pigs. In both species, antinutritional factors and carbohydrates, like stachyose and raffinose, were the most frequently studied categories of bioactive components. For both categories, most publications were evaluating ways to decrease the prevalence of the examined components in soy products, especially when fed at earlier ages. Overall, most studies evaluated the effect of the bioactive component on performance-related outcomes (n = 137), followed by microbial analysis (n = 38) and intestinal structure and integrity measures (n = 37). As they were analyzed in the majority of publications, antinutritional factors were also the most frequently investigated category in relation to each specific outcome. This trend did not hold true for microbiota- or antioxidant-associated outcomes, which were most often studied with carbohydrates or polyphenols, respectively. Changes to the host microbiota have the potential to modulate the immune system, feed intake, and social behaviors through the microbiota-gut-brain axis, though few publications measured behavior and brain characteristics as an outcome. Other identified gaps in research included the study of soy saponins, as most research focused on saponins derived from other plants, the study of phytosterols outside of their role in cardiovascular or reproductive outcomes, and the general examination of bioactive peptides. Overall, given soy's popularity as a current constituent of animal feed, additional research into these bioactive components may serve to define the value of soy products through their potential ability to support the productivity, health, and well-being of animals.

Abstract:Scientific advancements in recent years have shed new light on the relationship between diet and human health. Nutrients play an important role in the prevention of many civilization diseases, such as osteoporosis, type II diabetes, hypercholesterolemia, and cardiovascular diseases. The biological activity of natural plant components allows their use in the treatment of various diseases, especially civilization diseases, to be speculated. Special attention is paid to phenolic compounds that have numerous health-promoting properties. Isoflavones, phenolic compounds, are commonly found in legumes, especially in soybeans. Their structural similarity to 17-β-estradiol (E2), the main female sex hormone, allows them to induce estrogenic and antiestrogenic effects by binding to estrogen receptors, and their consumption has been associated with a decreased risk of hormone-related cancers. In addition, numerous epidemiological studies and related meta-analyses suggest that soy consumption may be associated with a lower incidence of certain diseases. However, there are some doubts about the potential effects on health, such as the effectiveness of cardiovascular risk reduction or breast cancer-promoting properties. The purpose of this review is to present the current knowledge on the potential effects of soy isoflavone consumption with regard to civilization diseases.

Effects of Soybean Isoflavone on Reproductive Development in SD Rats

Abstract:OBJECTIVE:To assess the effects of soybean isoflavone on reproductive development in SD rats.METHODS:Forty 4 week-old SD rats (20 males and 20 females) were randomly divided into a control group and a soy isoflavone (SI) group according to their body mass, and were fed with normal diet or normal diet added with soy isoflavone (100 mg/kg BW), respectively. The body weight, daily food intake, estrous cycle, fertility index, pregnant index, gestation length, number of pups, blood hormone level, the weight of reproduction organ and the quality of sperm of the parental generation and the sex ratio, anogenital distance, vagina opening time, prepuce separation time of F1 generation were recorded. The data were analyzed by two independent t test, and the data of pups in each litter were analyzed as a unit.RESULTS:Compared with the control group, the body weight and food utilization of male and female rats in SI group were lower before copulation (P < 0.05). The total number of sperm of parental males was higher (P < 0.05). Serum luteinizing hormone (LH) level of parental male was lower (P < 0.05), serum luteinizing hormone (LH), prolactin (PRL) and progesterone (P) of parental female were lower in SI group (P < 0.05). There were no significant differences of other indices such as estrous cycle, mating index, fertility index between two groups.CONCLUSION:SI has certain effect on the bodyweight of parental males, food utilization of females, and the total number of sperm and the level of some hormones.
Effects of Soy Isoflavones on Lymphocyte Activation and Natural Killer Cells Activity in Vitro of Rats(in English)

Abstract:AIM: To study the effects of soy isoflavones on immunocytes of rats in vitro. METHODS: Peripheral blood mononuclear cells were obtained from Wistar rats and were cocultured with ConA and various concentrations of genistein or soy daidzein (daidzein) for 68 h. The proliferation of lymphocytes stimulated by ConA in vitro was determined by tetrazolium assay (MTT). In another experiment, isolated peripheral blood mononuclear cells and YAC-1 cells were used as effector cells and target cells, respectively. Natural killer cells (NK) activity was evaluated by lactic dehydrogenase (LDH) assay. RESULTS: Daidzein significantly increased ConA-induced lymphocytes proliferation in a concentration-dependent manner (0.01-1 μmol*L-1) in comparison with ConA-induced vehicle control(P<0.01). In addition, 1 μmol*L-1 daidzein significantly enhanced NK activity(P<0.01). There was no significant effect on the proliferation of lymphocytes and NK activity cocultured with genistein within the same dosing regimens. CONCLUSION: Daidzein, rather than genistein, is able to enhance the immunological function in vitro, which suggests a novel approach to understanding the action mechanisms of daidzein by which soy foods may contribute to observed cancer prevention.

Abstract:Fifteen Holstein cows in milk mataphase were used in a completely randomized design to determine the effects of Daidzein and Genistein on incretion parameters.The cows were randomly divided into three groups,which included 200 mg Daidzein/d group,200 mg Genistein/d group and control in 60 ds.Results showed that INS,T3,T4 were not significantly different among groups(P0.05),but GH,PRL and E2 in Daidzein and Genistein groups were significantly improved compared to control(P0.05).

Abstract:Despite recommendations to increase plant food consumption for public and planetary health and the role that soy foods can play in plant-predominant diets, controversies around the effects of soy foods, especially soy isoflavones, are a barrier to their intake. Given their cardioprotective effects and ability to alleviate menopausal symptoms, addressing these concerns is particularly relevant to women. This systematic review and meta-analysis of randomized controlled trials aimed to determine the effect of soy isoflavones on measures of estrogenicity in postmenopausal women. MEDLINE, Embase, and Cochrane Library were searched through August 2024 for randomized trials ≥3-mo investigating soy isoflavones compared with non-isoflavone controls in postmenopausal women. Outcomes included endometrial thickness (ET), vaginal maturation index (VMI), follicle-stimulating hormone (FSH), and estradiol. Independent authors extracted data and assessed risk of bias. Grading of Recommendations, Assessment, Development and Evaluation was used to assess certainty of evidence. We included 40 trials (52 trial comparisons, n = 3285) assessing the effect of a median reported dose of 75 mg/d of soy isoflavones in substitution for non-isoflavone controls over a median of 24 wk. Soy isoflavones had no statistically significant effect on any measure of estrogenicity; ET [mean difference, -0.22 mm (95% confidence interval, -0.45, 0.01 mm), PMD = 0.059], VMI [2.31 (-2.14, 6.75), PMD = 0.310], FSH [-0.02 IU/L (-2.39, 2.35 IU/L), PMD = 0.987], and estradiol [1.61 pmol/L (-1.17, 4.38 pmol/L), PMD = 0.256]. The certainty of evidence was high to moderate for all outcomes. Current evidence suggests that soy isoflavones do not exhibit estrogenic effects compared with non-isoflavone controls on 4 measures of estrogenicity in postmenopausal women. This synthesis supports that soy isoflavones likely act as selective estrogen receptor modulators, differing clinically from the hormone estrogen. Addressing public health concerns may promote soy foods as high-quality plant protein sources with low environmental impact and cost, particularly benefiting postmenopausal women and aligning with sustainable dietary patterns and guidelines. Abstract:Isoflavones are secondary metabolites that represent the most abundant category of plant polyphenols. Dietary soy, kudzu, and red clover contain primarily genistein, daidzein, glycitein, puerarin, formononetin, and biochanin A. The structural similarity of these compounds to β-estradiol has demonstrated protection against age-related and hormone-dependent diseases in both genders. Demonstrative shreds of evidence confirmed the fundamental health benefits of the consumption of these isoflavones. These relevant activities are complex and largely driven by the source, active ingredients, dose, and administration period of the bioactive compounds. However, the preclinical and clinical studies of these compounds are greatly variable, controversial, and still with no consensus due to the non-standardized research protocols. In addition, absorption, distribution, metabolism, and excretion studies, and the safety profile of isoflavones have been far limited. This highlights a major gap in understanding the potentially critical role of these isoflavones as prospective replacement therapy. Our general review exclusively focuses attention on the crucial role of isoflavones derived from these plant materials and critically highlights their bioavailability, possible anticancer, antiaging potentials, and microbiome modulation. Despite their fundamental health benefits, plant isoflavones reveal prospective therapeutic effects that worth further standardized analysis.

Effect of Soy Isoflavones(SI)on Anti-oxidative Enzyme and Lipid Peroxide in Rat

Abstract:Objective To study the effect of soy isoflavones on some kinds of anti-oxidative enzyme and lever of lipid peroxide in rat. Method Six groups male Wistar rats weighting(160±20)g were studied:5 groups were fed a diet with high cholesterol,high fat and high cholic acid salt for 20 weeks.4 of these groups were fed varying levels of isoflavone 30 mg/(kg,bw),90 mg/(kg,bw)and 270 mg/(kg,bw),one group(EC)was fed estrogen 0.25 mg/(kg,bw),because of the estrogen effect of isoflavone and one group was fed high fat and cholesterol and cholic acid salt only as a AS model.One group fed normal diet(without soybean)as a normal control.After 20 week,rats were killed and we detect the lever of anti-oxidative enzyme and lipid peroxide in the serum and tissue of liver of rats. Results The level of lipid peroxide(LPO)is decreased and the level of some kinds of anti-oxidative enzyme is increased,such as superoxide dismutase(SOD)and GSH-PX. Conclusions The results suggest dietary isoflavones can regulate the lever of anti-oxidative enzyme and lipid peroxide in body and has the effect of inhibiting the formation of AS induced by high fat and cholesterol diets.
