Etiological Diagnosis of Atypical Acute Conjunctivitis with Polymerase Chain Reaction

Abstract:OBJECTIVE To discuss the clinical significance of polymerase chain reaction(PCR)for etiological diagnosis of atypical acute conjunctivitis(AAC).METHODS44 eyes from 44 patients with AAC and 22 eyes from 22 healthy persons were chosen,in which the more severe one of two eyes were taken.All DNA were extracted from secreta of conjunctival sac in subjects and divided into two parts,and the PCR amplifications were separately carried out with bacteria primer and adenovirus primer to observe the results with gel imaging analysis.Meanwhile,bacterial culture of the patients with AAC is performed.RESULTS520 bp stripes were amplified in 12eyes(27%)in AAC group,and 1 004 bp stripes in 11eyes(25%),and both 520 bp and 1 004 bp stripes were amplified at the same time in 3eyes(7%),and 520 bp stripes with no1 004 bp stripes were amplified in 1eye(5%)in normal group.Positive results were shown in 17eyes(39%)with bacterial culture with no statistical significance as compared with the PCR test results.CONCLUSIONPCR can be used in the rapid etiological diagnosis of AAC.
