Abstract:Objective To investigate the serum levels and clinical significance of osteopontin (OPN) and disintegrin and metalloprotease 8 (ADAMS) in patients with lung cancer.Methods Twenty four patients with small cell lung cancer (SCLC) (SCLC group),60 patients with non-SCLC (NSCLC) (NSCLC group) and 20 healthy volunteers (control group) were detected the serum levels of OPN and ADAM8 by ELISA.The relationship of OPN and ADAM8 with pathological characteristics of NSCLC was analyzed.The efficacy of serum OPN and ADAM8 for diagnosing SCLC and NSCLC was analyzed by ROC curve.Results The expressions of OPN and ADAM8 were significantly higher in NSCLC group ((36.86±11.00) μg/L,(2 695.99±568.86) ng/L) and SCLC group ((35.47±12.01) μg/L,(2 428.82± 415.42) ng/L) than those in control group ((25.45±10.71) μg/L,(1 737.29±594.36) ng/L) (P＜0.05),and there were no significant differences between NSCLC group and SCLC group (P＞0.05).OPN was positively correlated with ADAM8 in NSCCLC group (r=0.440,P=0.001),and there was no linear correlation between OPN and ADAM8 in SCLCgroup and control group (r=0.180,P=0.468;r=0.110,P=0.725).The levels of OPN and ADAM8 were significantly higher in the patients with pathological stage Ⅲ + Ⅳ,lymph node metastasis and distal metastasis than those in the patients with pathological stage Ⅰ + Ⅱ,with no lymph node metastasis or distal metastasis (P＜0.05).When the optimal cut-off value of OPN was 26.73 μg/L,the AUC for diagnosing NSCLC was 0.779 (95％ CI:0.618-0.940,P=0.002),the sensitivity was 86.7％,and the specificity was 66.7％.When the cut-off value of ADAM8 was 1 673.76 ng/L,theAUC for diagnosing NSCLC was 0.869 (95％CI:0.752-0.987,P＜0.001),the sensitivity was 100.0％,and the specificity was 66.7％.When the optimal cut-off value of OPN was 24.50 νg/L,the AUC for diagnosing SCLC was 0.745 (95 ％ CI:0.559-0.932,P=0.025),the sensitivity was 88.9％,and the specificity was 58.3％.When the cut-off value of ADAM8 was 1 783.50 ng/L,the AUC for diagnosing SCLC was 0.806 (95％CI:0.627-0.984,P=0.005),the sensitivity was 94.4％,and the specificity was 66.7％.Conclusion Both serum OPN and ADAM8 have certain values to the diagnosis of NSCLC and SCLC,and their levels are correlated with the clinicopathological characteristics of NSCLC,therefore they can be used as molecular indexes for disease monitoring and prognosis evaluation of NSCLC.
