Serum-free Culture of Primary Retinal Neurons of Newborn SD Rat in Vitro

Abstract:Objective To establish a culture system for primary retinal neurons culture(PRC) with serum-free medium,and provide a basis for further study on retinal neural cells in vitro.Methods The retina of Sprague-Dawley rats at postnatal 3 days to 5 days were dissected into cell suspension with trypsin digestion.Dissociated cells were seeded in NeurobasalTM-A medium supplemented with B27,and the growth regularity of cells in vitro was observed under the phase contrast microscope.At 5 days after cultivation,the retinal cells were initially identified by immunocytochemical method with anti-MAP-2,anti-rhodopsin monoclonal antibody and anti-GFAP monoclonal antibody,and the percentage of rod photoreceptor was counted in each 10-field high power view under light microscope.Results Retinal neural cells grew very well in serum-free medium,some cells possessed axons and some axons connected in networks.Approximately 95.6% of these cells were retinal neural cells by immunocytochemical identification in 5-day-culture,among which the percent of rod photoreceptor cells was about 38.6%,and simultaneously the growth of glial cell was well inhibited.Conclusion Purified cultures of rat retinal neurons are obtained using serum-free medium,which offered useful models for the generation of enriched photoreceptor populations in vitro.
