Abstract:Aim: To quantitatively analyze the nitric okide (NO) and Ca2+ in apoptosis of esophageal carcinoma cells induced by arsenic trioxide (As2O3).Methods: The cell line SHEEC1, a malignant esophageal epithelial cell induced by HPV in synergy with TPA in our laboratory, was cultured in a serum-free medium and treated with As2O3. Before and after administration of As2O3, NO production in cultured medium was detected quantitatively using the Griess Colorimetric method. Intracellular Ca2+ was labeled using the fluorescent dye Fluo3-AM and detected under confocal laser scanning microscope (CLSM), which was able to acquire data in real-time enabling Ca2+ dynamics of individual cells in vitro. The apoptotic cells were examined under electron microscopy.Results: Intracellular concentration of Ca2+ increased from 1.00 units to 1.09-1.38 units of fluorescent intensity at As2O3 treatment and NO products subsequently released from As2O3-treated cells increased from 0.98-1.00 X 10(-2) mol.L-1 up to 1.48-1.52 x 10(-2) mol.L-1 and maintained in a high level contineously. Finally apoptosis of cells occurred, chromatin being agglutinated, cells shrunk, nuclei became round and mitochondria swelled.Conclusion: Ca2+ and NO increased with cell damage and apoptosis in cells treated by As2O3. The Ca2+ is an initial messenger to the apoptotic pathway. To investigate Ca2+ and NO will be a new direction for studying the apoptotic signaling messenger of the esophageal carcinoma cells induced by As2O3.

Abstract:Objective To explore the relationship between apoptosis indeced by arsenic trioxide(As 2O 3) and the transcription factor NF-κB activity in K562 cells.Methods K562 cells were cultured with As 2O 3 in different concentrations Western blot was used to analyse the expression of NF-κB in nuclei and IκB-α in cytoplasm of K562 cells Apoptosis and degradation of IκB-α protein were also observed by flow cytomerty.Results After exposure to As 2O 3 , K562 cells were induced to apoptosis The level of IκB-α was downregulated after As 2O 3 stimulation, while NF-κB was upregulated Flow cytometry also confirmed the changes of IκB-α Conclusion As 2O 3 can induce apoptosis of K562 cells, and this effect involves the degradation of IκB-α and the activation of NF-κB

Abstract:Aim: To investigate the effects of inhaled nitric oxide (NO) on pulmonary inflammation, apoptosis, peroxidation and protein nitration in a rat model of acute lung injury (ALI) induced by meconium. Methods: Twenty-four healthy male Sprague-Dawley rats were randomly devided into 3 groups ( n =8): meconium-induced ALI with intratracheal instillation of 1 mL/kg saline (Mec/saline group), continuous inhalation of NO at 20 μL/L. (Mec/iNO), and the control group (control). Electromicroscopic examination was used to determine the extent of epithelial apoptosis. TUNEL was used to detect DNA fragmentation in pulmonary apoptotic cells, expressed as the apoptosis index (AI). Western blotting was used to detect pulmonary inducible NO synthase (iNOS) expression. RT-PCR was used to detect interleukin (IL)-1β mRNA expression. Cell count inbronchoalveolar lavage (BAL), myeloperoxidase (MPO) activity, as well as malondialdehyde (MDA) and nitrotyrosine formation, the markers of toxic NO-superoxide pathway in rat lung parenchyma specimens, were also examined. Results: Expression of iNOS protein and IL-1β mRNA were increased significantly in the Mec/saline group (both P <0.01) compared with the control group. BAL cell count, MPO activity, lung injury score, pulmonary AI, MDA level and nitrotyrosine formation were also increased significantly (all P <0.01). The meconium-induced iNOS protein and IL-1β mRNA expression were inhibited significantly by NO inhalation when compared with the Mec/saline group (both P <0.01). BAL cell count, MPO activity and lung injury score were also decreased significantly ( P <0.01 or P <0.05). However, there were no statistical differences in MDA level, nitrotyrosine formation or pulmonary AI between the Mec/saline and Mec/iNO groups. Electromicroscopic examination revealed a significant degree of epithelial apoptosis in both the Mec/saline and Mec/iNO groups. Conclusions: Early continuous inhalation of NO 20 μL/L may protect the lungs from inflammatory injury, but does not decrease epithelial apoptosis or lung nitrotyrosine formation. Inhalation of NO alone is not associated with a detectable increase in oxidant stress.

Abstract:In order to explore the early apoptotic signal messengers and to search the apoptotic pathway, the morphological and functional changes of mitochondria were examined, and nitric oxide (NO) and calcium ions (Ca2+) were measured in the course of apoptosis in esophageal carcinoma cells induced by As2O3. The esophageal carcinoma cell line SHEEC1, established by HPV in synergy with TPA in our laboratory, were cultured with serum-free medium in a culture flask, 24-well plate and small culture chambers, and added with As2O3 at 1, 3, 5 mumol/l. After 0, 2, 4, 8, 12, 24 h of drug adding the NO were measured from extracellular cultured medium and the SHEEC1 cells were collected from flasks for electron microscopic examination. Fluorescent intensity (FI) of rhodamine 123 (Rho123) labeled cells was detected using laser confocal scanning, microscope (LCSM) for evaluation of mitochondrial membrane potential. Intracellular Ca2+ of cells in small culture chambers labeled with Fluo-3 dye were measured using LCSM over time. After adding As2O3 SHEEC1 cells revealed characteristic morphological and functional changes of mitochondria such as hyperplasia, swelling and disruption, accompanying decrease of transmembrane potential (FI of Rho123 decreased). The Ca2+ level increased at once after adding As,03 and the NO concentration increased step by step till 24 h, then apoptotic morphology of cells occurred. The results suggest that by inducement of As2O3, increasing Ca2+ and NO, the apoptotic signal messengers, will initiate the mitochondria-dependent apoptotic pathway.

Abstract:OBJECTIVE:To observe the apoptosis induced by exogenous NO in Tca8113 cells and to investigate the possible mechanism.METHODS:SNP as NO donor was used to treat the tongue squamous cell carcinoma Tca8113 cells. Cytotoxic and apoptotic effects of NO on Tca8113 cells were examined by using MTT assay, acridine orange (AO) staining, Wright-Giemsa staining, agarose gel electrophoresis and flow cytometry. Western blot was performed for investigating the apoptotic mechanism.RESULTS:NO had a remarkable proliferation inhibiting effect on Tca8113 cells. After being exposed to exogenous NO, Tca8113 cells showed series of apoptotic morphological changes such as cell shrinkage, nuclear condensation; and also showed DNA fragmentation, G2/M phase arrest as well as upregulation of the tumor suppressor P53 protein.CONCLUSION:Exogenous NO has a proliferation inhibition and apoptosis induction effect on Tca8113 cells in a concentration and time-dependent manner, P53 protein may be involved in the apoptosis induced by NO.
Study on the Technology of Nitric Oxide (no) Detection in Vitro and in Vivo

Abstract:To reveal the intracellular distribution of nitric oxide (NO) in single cell and to detect NO quantitatively in animal models, various methods are developed. A confocal microscope and 3D visualization technique were applied to visualize NO concentration in arbitrary directions and in different sections of cells. It was shown that NO distributions in cultured neurons of rat hipopocampus were not homogeneous. A kind of highly sensitive and selective NO electrode was utilized to measure the NO concentration in SD rat hippocampus, simultaneously with blood pressure and ECG recording. By the administration of N-G-nitro-L-arginine methylester (L-NAME), the changes of NO level in the hippocampus in vivo were detected. It was shown that the NO level decreased greatly when NOS was inhibited by L-NAME with significant increase in blood pressure. The developed methods may be helpful to investigate the physiological mechanism of NO and its role in pathology.

Abstract:To explore the relationship between the reactive oxygen species (ROS) and apoptosis in esophageal carcinoma cells (SHEE85) induced by arsenic trioxide (As2O3), we focused on changes of apoptosis, ROS, and antioxidants. Apoptosis of SHEE85 was confirmed by means of DNA fragmentation stained by Hoechst 33342, Sub-G(1) cells scored by flow cytometry and ultrastructure of cells by electron microscopy. To evaluate the level of ROS, the chemiluminescent method was used for measuring the production of superoxide anion (O-2(-.)). Lipid peroxide (malondialdehyde, MDA), superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were measured respectively by the photometry method. In the cells treated with As2O3 at a concentration of 5.0 mumol/l for 2-24 h, the content of cellular O-2(-.) and MDA was increased, but SOD and GSH-Px were significantly lower in the process of apoptosis in SHEE85. As2O3 at concentration of 0.5 mumol/l did not cause cell apoptosis but promoted cell proliferation. These results suggest that As2O3 at a high dosage (5 mumol/l) causes cell apoptosis and at a low dosage (0.5 mumol/l) causes cell proliferation. The essential mechanisms of cell apoptosis induced by As2O3 may be related to the increase of ROS and decrease of anti-oxidation. ROS and antioxidants participate in the apoptotic pathway of esophageal carcinoma cells.
Nitric Oxide Induces Oral Squamous Cell Carcinoma Cells Apoptosis with P53 Accumulation.

Abstract:Nitric oxide has been reported to have cytotoxic effects in several tumor cells. The objective of this study was to investigate the effects of exogenous nitric oxide on apopotosis in oral squamous cell carcinoma cells and to reveal its possible mechanism. Tca8113 cells were cultured with various concentrations of nitric oxide that were released from sodium nitroprusside (SNP). Nitrite/nitrate levels in the culture supernatant were determined using a commercial available nitric oxide kit. Cellular proliferation was determined by MTT assay. Apoptosis was detected by flow cytometry. Expression of inducible nitric oxide synthase (iNOS) was determined by immunocytochemistry. p53 expression was assessed by Western blot. SNP can release nitric oxide into the culture medium in a dose-dependent manner. Nitric oxide remarkably inhibits proliferation in a dose and time-dependent manners and lead to apoptosis of the Tca8113 cell. The p53 expression was elevated accompanying by the increased apoptotic cells. No difference of iNOS was found whether or not the cells were treated with SNP. Exogenous nitric oxide had an inhibitory effect on Tca8113 cells proliferation in a dose and time-dependent manners and possibly via p53 dependent apoptosis pathway. Exogenous nitric oxide had no significant effect on cellular iNOS protein.

Abstract:The processes of apoptosis and inflammatory responses, which are defensive strategies utilized by cells to confront external substances, can give rise to diverse diseases when prolonged or disrupted, such as cancer, Alzheimer's disease, and Parkinson's disease. Here we engineered a live-cell imaging fluorescent probe for nitric oxide (NO) based on naphthalimide and o-phenylenediamine, enabling the sensitive detection of NO in cancer cells and thereby live-monitoring the doxorubicin-induced apoptosis and lipopolysaccharide-triggered inflammation reactions. Importantly, we found that the level of released NO can sensitively indicate the early stages of both cellular inflammatory responses and apoptotic processes. It suggested that cellular NO in fact behaves as a new class of signaling molecule for inflammatory responses and apoptosis processes, providing a potent tool for live-monitoring cellular physiological reactions.

Abstract:Ezrin is a key protein in membrane-cytoskeleton interaction. Expression of ezrin in actin-rich cell surfaces may play a role in the modulation of cell shape and adhesion. The aim of this study was to detect ezrin, actin and cytoskeleton and to explore their relationship in the apoptosis of esophageal epithelial cells (SBEE) induced by arsenic trioxide (As2O3). The SHEE is an immortalized human fetal esophageal epithelial cell line, and the cells were treated by administering 5, 10 and 20 mumol of As2O3. The proliferation and apoptosis of SHEE cells were examined by flow cytometry with propidium iodide staining. Ezrin expression was detected by immunocytochemistry and Western blotting. Actin filament was stained by FITC-labeled phalloidin and detected quantitatively by fluorescent microscopy. Cell morphology and microfilaments were examined by electron microscopy. The results revealed that As2O3 induced an inhibition of proliferation and the promotion of apoptosis in SHEE cells. The ezrin, actin and cytoskeleton were decreased after As2O3 treatment and the cellular morphology of apoptosis developed. Our results suggested that the morphological changes of arsenic-induced apoptosis of human esophageal epithelial cells were initiated by ezrin and actin-cytoskeletal aberrance.
Isoalantolactone induces apoptosis through reactive oxygen species-dependent upregulation of death receptor 5 in human esophageal cancer cells.

Abstract:Esophageal cancer is the eighth most prevalent cancer and has high mortality in our society. Isoalantolactone, extracted from Inula helenium L, has shown potent anticancer effects on a variety of cancers. However, its effect on human esophageal cancer, and the underlying molecular mechanism, remain to be investigated. In the present study, we demonstrated that isoalantolactone induced apoptosis in esophageal cancer cells. Treatment with isoalantolactone activated caspases-3, -7, and -10, and upregulated death receptor (DR)5. Furthermore, DR5 knockdown partially reversed the effect of isoalantolactone. These results indicated the extrinsic apoptosis was induced by isoalantolactone. In addition, intracellular reactive oxygen species (ROS) were significantly elevated after treatment with isoalantolactone. N-Acetylcysteine, an ROS scavenger, blocked both the apoptosis and decreased cell viability caused by isoalantolactone. In vivo, significant suppression of tumor growth by isoalantolactone was observed in an ECA109 cell xenograft mouse model. Isoalantolactone showed no obvious adverse effects on mouse weight and histology of heart, liver, spleen, lung, and kidney. In conclusion, our results revealed that isoalantolactone induced apoptosis through the extrinsic pathway via upregulation of DR5 and elevation of ROS in human esophageal cancer cells. Isoalantolactone, therefore, could be a potential candidate in developing anticancer agents for esophageal cancer patients.
Real-time Detection of Nitric Oxide in Cultured Rat Aorta Endothelial Cells Induced by Shear Stress

Abstract:To establish a stable and real-time method to detect the production of intracellular nitric oxide (NO) of endothelial cells under different shear stresses. After the cultured endothelial cells were loaded with DAF-FM, the relative NO production was determined by fluorescence intensity, which was detected using Zeiss Axioskop 2 fluorescence microscope and ICCD-camera. The fluorescence of DAF-FM can reflect NO production. Shear stress can induce a dose-dependent increase of NO production, and the increase can be totally inhibited by L-NAME and partly inhibited by Ca2+-free buffer. The method can be used to detect the change of NO production in real time, and it can also be used to study the mechanism related to NO increase induced by shear stress.
Direct Acupuncture of Nitric Oxide by an Electrochemical Microsensor with High Time-Space Resolution

Abstract:Measurement of signal molecule is critically important for understanding living systems. Nitric oxide (NO) is a key redox signal molecule that shows diverse roles in virtually all life forms. However, probing into NO's activities is challenging as NO has restricted lifetime (<10 s) and limited diffusion distance (usually <200 mu m). So, for the direct acupuncture of NO within the time-space resolution, an electrochemical microsensor has been designed and fabricated in this work. Fabrication of the microsensor is achieved by (1) selective assembly of an electrocatalytic transducer, (2) attaching the transducer on carbon fiber electrode, and (3) covered it with a screen layer to reduce signal interference. The fabricated microsensor exhibits high sensitivity (LOD, 13.5 pM), wide detection range (100 pM-5 mu M), and good selectivity. Moreover, studies have revealed that the availability of the sensor for efficient detection of NO is due to the formation of a specific DNA/porphyrin hybrid structure that has synergetic effects on NO electrocatalysis. Therefore, NO release by cells and tissues can be directly and precisely traced, in which we have obtained the release pattern of NO by different cancer cell lines, and have known its dynamics in tumor microenvironment. The fabricated electrocatalytic microsensor may provide a unique and useful tool for the direct assay of NO with high time-space resolution, which promisingly gives a technical solution for the bioassay of NO in living systems.

Abstract:High levels of intracellular reactive oxygen species (ROS) in cells is recognized as one of the major causes of cancer cell apoptosis and has been developed into a promising therapeutic strategy for cancer therapy. However, whether apoptosis associated biophysical properties of cancer cells are related to intracellular ROS functions is still unclear. Here, for the first time, we determined the changes of biophysical properties associated with the ROS-mediated oesophageal cancer KYSE-150 cell apoptosis using high resolution atomic force microscopy (AFM). Oridonin was proved to induce ROS-mediated KYSE-150 cell apoptosis in a dose dependent manner, which could be reversed by N-acetylcysteine (NAC) pretreatment. Based on AFM imaging, the morphological damage and ultrastructural changes of KYSE-150 cells were found to be closely associated with ROS-mediated oridonin-induced KYSE-150 cell apoptosis. The changes of cell stiffness determined by AFM force measurement also demonstrated ROS-dependent changes in oridonin induced KYSE-150 cell apoptosis. Our findings not only provided new insights into the anticancer effects of oridonin, but also highlighted the use of AFM as a qualitative and quantitative nanotool to detect ROS-mediated cancer cell apoptosis based on cell biophysical properties, providing novel information of the roles of ROS in cancer cell apoptosis at nanoscale.
Arsenic Nanoparticles Trigger Apoptosis via Anoikis Induction in OECM-1 Cells

Abstract:Arsenic compounds have been used as therapeutic alternatives for several diseases including cancer. In the following work, we obtained arsenic nanoparticles (AsNPs) produced by an anaerobic bacterium from the Salar de Ascotán, in northern Chile, and evaluated their effects on the human oral squamous carcinoma cell line OECM-1. Resazurin reduction assays were carried out on these cells using 1–100 μM of AsNPs, finding a concentration-dependent reduction in cell viability that was not observed for the non-tumoral gastric mucosa-derived cell line GES-1. To establish if these effects were associated with apoptosis induction, markers like Bcl2, Bax, and cleaved caspase 3 were analyzed via Western blot, executor caspases 3/7 via luminometry, and DNA fragmentation was analyzed by TUNEL assay, using 100 μM cisplatin as a positive control. OECM-1 cells treated with AsNPs showed an induction of both extrinsic and intrinsic apoptotic pathways, which can be explained by a significant decrease in P-Akt/Akt and P-ERK/ERK relative protein ratios, and an increase in both PTEN and p53 mRNA levels and Bit-1 relative protein levels. These results suggest a prospective mechanism of action for AsNPs that involves a potential interaction with extracellular matrix (ECM) components that reduces cell attachment and subsequently triggers anoikis, an anchorage-dependent type of apoptosis.

Abstract:With two-photon and confocal laser scanning microscopy in combination with fluorescent probes Hoechst 33342, 2',7'-dichlorofluorescin diacetate (DCFH-DA) and Fluo 3-AM, we simultaneously observed arsenic trioxide (As(2)O(3))-induced changes in nuclear morphology, reactive oxygen species (ROS) and intracellular calcium concentration (Ca(2+))(i) within human skin squamous carcinoma cells (Colo-16 cells). Our results indicated that As(2)O(3) induced (Ca(2+))(i) elevation and ROS production within Colo-16 cells, and both (Ca(2+))(i) elevation and ROS production were involved in the apoptosis of Colo-16 cells. These results suggested that two-photon and confocal laser scanning microscopy might provide a real-time, sensitive and noninvasive method for simultaneously multi-parameter observation of As(2)O(3)-induced apoptosis at the single cell level.
808 Nm-Light-Excited Upconversion Nanoprobe Based On Lret For The Ratiometric Detection Of Nitric Oxide In Living Cancer Cells

Abstract:NO (nitric oxide) has dual functions in cancer, promoting carcinogenesis in low concentrations and inducing tumor cell apoptosis at high concentrations. The dual-edged-sword functions of NO make it particularly appealing to develop a sensitive and specific chemical probe for its detection. However, most NO sensors suffer from poor Stokes shifts and are limited by ultraviolet (UV) or visible light excitation, which render it difficult to avoid the intrinsic background signal. In this study, an 808 nm laser-excited Nd3+-sensitized upconversion nanoprobe based on LRET (luminescence resonance energy-transfer) for NO detection was constructed for the first time. This probe was composed of Nd3+-sensitized core-shell upconversion nanoparticles (540 nm and 660 nm emission) as the energy donor and RhBs as the acceptor. In the presence of NO, RhBs was converted into Rhodamine B and its strong absorption subsequently quenched the 540 nm fluorescence of UCNPs, while the emission at 660 nm remained constant. The ratiometric detection of the fluorescence at 540 nm, as compared to 660 nm, can precisely respond to the difference in NO levels with a detection limit of 0.21 mu M. Importantly, compared with conventional UCNPs excited at 980 nm, the 808 nm light excitation led to lower water absorption and deeper tissue penetration, thus avoiding overheating and allowing for long-term biological imaging. This strategy has been perfectly applied to detecting the NO levels in living cells to differentiate the tumor cells from normal cells based on varied intracellular NO concentration. Further, the nanosystem realized the real-time monitoring of NO during treatment with NO donor drugs, which could inspire the future application of this probe to guide NO therapy.
Label-free fluorometric assay for cytochrome c in apoptotic cells based on near infrared Ag2S quantum dots

Abstract:As an important biomarker, cytochrome c (Cyt c) plays a crucial role in mitochondrial electron transport chain and cell apoptosis. Herein, a label-free near-infrared (NIR) Ag2S quantum dots (QDs)-based fluorescent strategy was constructed for the sensitive and selective detection of Cyt c. In this system, Cyt c was hydrolyzed by trypsin, and the resulting heme-peptide fragment exhibited peroxidase-like activity for catalytic decomposition of H2O2 into hydroxyl radical (·OH). The presence of caffeic acid in this system resulted into the formation of caffeic acid-quinone due to the strong oxidizing ability of ·OH. The production of caffeic acid-quinone led to the fluorescence quenching of Ag2S QDs through electron transfer mechanism. Based on the cascade reaction, we successfully developed a label-free approach to detect Cyt c using Ag2S QDs as nanoprobes. Under the optimized conditions, the fluorescence intensity of Ag2S QDs was linearly relative to the concentration of Cyt c over the range from 2.0 to 150 nM with a detection limit of 1.7 nM. In addition, this strategy was successfully applied for quantitative detection of Cyt c in cell lysates of H2O2 or etoposide (anticancer drug)-induced apoptotic cells, providing great potential application for cell-based oxidation pressure determination and screening of anticancer drugs.
Macrophage-derived nitric oxide induces apoptosis of rat hepatoma cells in vivo

Abstract:Although nitric oxide (NO) has been postulated to play important roles in host defense mechanisms against tumor cells, a direct evidence supporting this hypothesis is lacking. To obtain molecular insights into the antitumor action of NO, its metabolism and effect on ascites hepatoma (AH-130) cells were investigated in tumor-bearing rats. Kinetic analysis revealed that substantial amounts of nitrite and nitrate, metabolites of NO, appeared in plasma and ascites of AH-130-inoculated rats. Western blot analysis revealed that a large number of macrophages that expressed inducible type of NO synthase (iNOS) appeared in cancerous ascites, particularly during 1 to 2 weeks after inoculation of AH-130 cells. When NO generation by peritoneal macrophages increased, a significant fraction of AH-130 in ascites fluid underwent apoptosis as judged from the fragmentation of their nuclear DNA. Kinetic analysis revealed that NO strongly inhibited mitochondrial electron transport and changed calcium status in AH-130 cells, particularly under low oxygen tensions such as in cancerous ascites. Intraperitoneal injection of NO donor strongly enhanced DNA fragmentation of AH-130 cells. Antimycin A, a specific inhibitor for mitochondrial electron transport, also induced DNA fragmentation of AH-130 cells by a mechanism that was inhibited by adding ascorbate and tetramethyl-p-phenylene diamine (TMPD) as electron donors. These results indicate that NO derived from peritoneal macrophages inhibits mitochondrial electron transport and disturbs calcium homeostasis in ascites hepatoma AH-130 cells, thereby inducing their apoptosis in vivo.
