Abstract:Purpose To investigate the distribution patterns of microlymphatic vessels and microvessels in colorectal carcinomas and their relationship with tumor metastasis and prognosis. Methods Immunohistochemical staining of Podoplanin and CD34 were used to detect microlymphatic density (MLD) and microvascular density (MVD) in 227 colorectal cancers. Results The MLD in the invasive front (19.64±7.21) was significantly higher than that in the central region (5.64±2.87) (P0.05). Similarly, the MVD in the invasive front (40.93±11.71) was much higher than that in the tumor center (21.07±8.38) (P0.05). Higher MLD in the invasive front was significantly correlated with tumor invasive depth, lymphatic/vascular invasion, lymphatic node metastasis and distant metastasis (P0.05). Higher MLD and MVD in the invasive front was associated with adverse prognosis of colorectal carcinoma (P0.05). However, both were not independent prognosticators in colorectal cancers. Conclusions Microlymphatic vessels and microvessels are predominantly distributed in the invasive front of colorectal carcinoma. High MLD and MVD at this region may be of significance in predicting poor prognosis in patients with colorectal cancers.

Abstract:Objective To investigate the relationship between microvessel density (MVD) and Vascalar endothelial growth factor(VEGF), clinicpathological characteristics and the prognosis in primary breast cancer patients. Methods MVD and VEGF in 47 cases of breast cancer were studied by immunohistochemistry on formalin-fixed, paraffin-embedded tumor section. Correlations between MVD, VEGF, clinicpathological factors and prognosis were statistically analyzed. Results ①The mean of MVD was 43.13±15.40, range 21~89, median 42 in all patients. VEGF expression was observed in 28 of 47(59.6%). ②MVD and VEGF were statistically significantly higher in lymph node positive, and relapsed or metastatic group than in lymph node negative and disease free survival group (P<0.01), whereas no correlations with menopausal status, histologic grade, estrogen receptor and progesterone receptor (P>0.05). ③The MVD of the VEGF positive cases in breast cancer was significantly higher than that of the VEGF negative case (P<0.001). ④Kaplan-Meier survival analysis indicated that both overall survival (OS) and disease free survival (DFS) statistically significantly between low-MVD and high-MVD group, or between VEGF expression negative and VEGF expression positive, respectively. Conclusion MVD and VEGF were relationship with lymph node positive number, relapsed or metastatic status, DFS and OS. It could be an independent prognostic factor in primary breast cancer.
Evaluation of vascular endothelial growth factor and tumor microvessel density in prognoses of patients with breast cancer: A follow-up of 1-10 years

Abstract:Aim: To observe the expression of vascular endothelial growth factor(VEGF) and the distribution of microvessel density(MVD) in breast carcinoma and benign breast disease, discuss the relationship of MVD and VEGF with the tumor size(T), histological grade, the level of estrogen receptor and axillary lymphatic metastasis, and relapse and metastasis, and evaluate the prognostic value of MVD and VEGF. Methods: The pathological sections were stained by immunohistochemical staining using antibody of vascular endothelium for factor VIII antigen and multiclone antibody of anti-VEGF respectively. Under the microscope, the stained degree of VEGF were observed, and the contribution of microvessel counting was calculated. The prognoses of patients were followed up. Results: In the breast cancer and benign breast disease, the positive rates of VEGF expression were 51.35% and 16.13% respectively(χ 2=11.21, P <0.005), and the mean levels of MVD were (30.8±19.4) and (16.09±7.9) every 200 times sight respectively(t=4.47, P <0.01). MVD was associated with tumor size(t=2.0, P <0.05), histological grade (F = 3.48, P <0.05), axillary lymphatic metastasis(χ2=4.24, P < 0.05), and relapse and metastasis(t=2.84, P <0.05), and VEGF was related to axillary lymphatic metastasis(χ2=5.62, P <0.05) and relapse and metastasis(χ2=5.13, P <0.05). The relapse-free survival rate was significantly associated with VEGF(χ 2=5.12, P <0.05) and MVD (χ2= 8.752, P <0.05). Moreover, there was no obvious correlation between MVD and VEGF. Conclusion: The distribution of MVD and the postive rate of VEGF expression in breast carcinoma were significantly higher than those of benign breast disease. VEGF and MVD of breast carcinoma, related to the prognoses of patients, can be regarded as a prognostic indicator for the patients with breast cancer. VEGF is not the only factor influencing tumor angiogenesis.

Abstract:BACKGROUND & OBJECTIVEThis study was designed to investigate the vascular endothelial growth factor (VEGF) and the microvessel density(MVD) in node-negative breast carcinoma, and to discuss the relationship of MVD and VEGF to the tumor size, histologic grade, and relapse and metastasis. Meanwhile, the prognostic value of MVD and VEGF was estimated.METHODSMVD and VEGF were evaluated after immunohistochemical staining using antibody of vascular endothelium for factor VIII antigen and VEGF in 62 patients with node-negative breast carcinoma respectively. The patients were followed up.RESULTSVEGF and MVD were all associated with histological grade and relapse (P < 0.05), but no association with ER and tumorsize; the relapse-free survivals (RFS) and overall survivals of patients with high MVD or high VEGF expression were significantly lower than that of the patients with low MVD or low VEGF expression (P < 0.01). Moreover, the difference of survivals in the patients with high MVD and high VEGF expression were more obvious(P < 0.005). Meanwhile, there was association between MVD and VEGF (P < 0.01).CONCLUSIONSThe node-negative patients with high VEGF expression or high MVD would have poor prognosis. VEGF and MVD could be significant prognostic indicators for the patients with node-negative breast carcinoma. It has better prognostic value to connected VEGF and MVD. VEGF was an important factor in tumor angiogenesis.

Abstract:Objective To investigate the relationship between vascular endothelial growth factor (VEGF) and microvascular density (MVD) with lymph node metastasis in breast carcinoma (BC). Methods Expression of VEGF protein and VEGF mRNA and MVD in 47 cases of breast carcinoma were studied by immunohistochemistry methods and reverse-transcription polymerase chain reaction (RT-PCR) technique. Results VEGF mRNA expression in BC tumor tissues were higher than in adjacent nomal tissues (55.30% vs 17.02%). VEGF mRNA expression in BC lymph node metastasis showed a tendency to relation. There was a close correlation between VEGF and MVD with lymph node metastasis ( P 0. 05). MVD positive rate in BC tumor tissues were higher than in adjacent nomal tissues ( P 0. 05) . Conclusion VEGF is closely related to angiogenesis of BC. The increase of VEGF and MVD may promote lymph node metastasis of BC.

Abstract:Objective: To detect the expression of VEGF and MVD count in invasive ductal carcinoma of breast to clarify the association of VEGF expression and MVD count with the clinicopathologic features. Methods: The expressions of VEGF, ER, PR, C-erbB-2 and MVD count in 88 cases of invasive ductal carcinoma of breast were examined by immunohistochemistry staining ( SP- method). Results: Sixty- two out of the eighty- eight specimens of breast carcinoma ( 70.45%) showed positive expression of VEGF. The positive rate of VEGF in cases with lymph node metastasis was higher than that without lymph node metastasis ( P < 0.05). The positive rate of VEGF in stage IIb- III was higher than that in stage I- IIa ( P < 0.05). The positive rate of VEGF in C- erbB- 2 positive group was higher than that in C- erbB- 2 negative group ( P < 0.05). Higher expression of VEGF was observed in cases with higher tissue differentiation degree ( P < 0.05). Also, significant higher MVD count was observed in cases with higher tissue differentiation degree ( P < 0.01). The MVD count increased significantly with the increase of the expression of VEGF ( P < 0.01). Conclusion: The result of this study suggested that in invasive ductal carcinoma of breast, angiogenesis and metastasis were mediated mainly by VEGF. The expression of VEGF and MVD might be reference predictors for the biological behavior of breast carcinoma. The antiangiogenic therapy which used VEGF as a target would become a new method to treat patients who were C- erbB- 2 positive in the future.

Abstract:BACKGROUND & OBJECTIVE:TNM staging system is used widely to predict prognosis of non-small cell lung carcinoma (NSCLC) patients, but patients with the same stage may have very different survivals; better prognostic index is needed. Angiogenesis is considered to be essential for tumor development, progression, and metastasis, but the prognostic impacts of vascular endothelial growth factor (VEGF) and microvessel density (MVD) in NSCLC is controversial. This study was to evaluate the prognostic value of VEGF and MVD in NSCLC.METHODS:VEGF and MVD in 214 specimens of stageI-II NSCLC (20 in stage IA, 137 in stage IB, and 57 in stage IIB) were detected by tissue chip and SP immunohistochemistry. No patient underwent postoperative antitumor treatment.RESULTS:VEGF expression didn't relate to gender, age, blood type, pathologic type, and TNM stage (P0.05). MVD correlated with age and pathologic type (P0.05), but did not relate to gender, blood type, and TNM stage (P0.05). The mean value of MVD was 65.8+/-5.2 in VEGF-low patients, and 67.5+/-2.5 in VEGF-high patients (P0.05). The 5-year survival rate was significantly lower in MVD-high patients than in MVD-low patients (34.5% vs. 60.0%, P=0.013). Furthermore, multivariate Cox regression analysis showed that MVD (P=0.000) was an independent prognostic factor of NSCLC.CONCLUSION:High MVD closely relates to poor prognosis of stageI-II NSCLC.
Prognostic Significance of Vascular Endothelial Growth Factor Expression and Microvessel Density in Carcinoma of Ampulla of Vater.

Abstract:BACKGROUND/AIMS:To investigate whether the expression of vascular endothelial growth factor (VEGF) and microvessel density (MVD) are of prognostic significance in ampullary carcinoma.METHODOLOGY:Twenty-two resected tumor specimens from patients with ampullary carcinoma were immunohistochemically stained for VEGF and CD34 (surrogate for vessels) by streptavidin-peroxidase method.RESULTS:Expression of VEGF in tumor tissue was found in 50% of patients. The mean MVD for entire group was 26.4 +/- 12.8. A significantly higher MVD was observed in the tumors with positive VEGF expression (35.0 +/- 9.6) compared with that of negative VEGF expression (17.7 +/- 9.3) (p<0.01). The expression of VEGF and MVD were closely related lymph node status and tumor TNM stage. The positive expression rate of VEGF and the average MVD in patients with lymph node metastases were 85.7% and 33.1 +/- 10.8 respectively, which were significantly higher than those in patients without lymph node metastases (33.3% and 22.8 +/- 11.8 respectively) (p<0.05). The positive expression rate of VEGF and the average MVD in patients with stage III and were 75% and 36.3 +/- 8.4 respectively, which were significantly higher than those in patients with stage I (25% and 18.4 +/- 10.1 respectively) or stage II (50% and 23.8 +/- 13.4 respectively) (p<0.05). The Kaplan-Meier survival curves showed that the 3-year survival rate for patients with positive VEGF expression or a high MVD (9.1% and 10% respectively) were lower than those in patients with negative VEGF expression or a low MVD (63.64% and 58.33% respectively) (p<0.05).CONCLUSIONS:VEGF is positively correlated with MVD in ampullary carcinoma. VEGF and angiogenesis may play an important role in lymph node metastasis and progression of ampullary carcinoma. VEGF and MVD appear to be important prognostic predictor in patients with ampullary carcinoma.

Abstract:Objective To investigate the relationship of vascular endothelial growth factor (VEGF) and tumor angiogenesis to the clinical pathological parameters of breast carcinoma. Methods S-P immunohistochemical methods were used to detect the expression of VEGF protein and micro-vessel density (MVD) in 46 specimens of breast carcinomas. Results The results showed that VEGF protein located in cytoplasm and CD34 in cytoplasm of vascular endothelial cell. The positive rates of VEGF protein in breast carcinoma were 76.09%, and the expression of CD34 was positive in all 46 samples. Both VEGF expression and MVD value were positively correlated with the histological grade (P0.05) and lymph node metastasis (P0.01), but not related to the histological type (P0.05). The level of MVD was significantly higher in VEGF positive group than in VEGF negative one (t=2.220,P0.05). Conclusion VEGF might be highly related to angiogenesis of breast carcinoma and promote growth, invasion and metastasis of tumor. The expression of VEGF or MVD might be one of the reference predictors for the biological behavior of breast carcinoma.

Abstract:OBJECTIVE:To study the relationship between the microvascular density(MVD) and the prognosis in laryngeal cancer.METHOD:Microvessel quantitation in 31 cases of laryngeal cancer was done with factor VIII-related antibody and SABC method.RESULT:The quantitation of microvessels in laryngeal cancer were negative of histologic grade and type, but were significantly different (P < 0.05) between metastasis (10.23 +/- 0.78) and without metastasis(6.38 +/- 0.33) two years after operation.CONCLUSION:The angiogenesis of laryngeal cancer is correlated with the lymph node metastasis and prognosis.

Abstract:The aim of this study was to investigate the role of ultrasonographic microvascular imaging in the evaluation of prognosis of patients with invasive breast cancer treated by adjuvant therapies. A total of 121 patients with invasive breast cancer underwent ultrasonographic contrast-enhanced imaging, vascular endothelial growth factor (VEGF) staining, and microvessel density (MVD) counts. The parameters of microvascular imaging and the expression of VEGF and MVD in primary breast cancer were calculated. The correlation between these factors and the overall and progression-free survival rate were analyzed using the Kaplan-Meier method. Among 121 cases, the positive VEGF cases were 75 and negative ones were 46. The cut point of 52.3 was calculated by the regressive curve for MVD counts. The data showed the mean intensity (MI) was positively associated with both the MVD counts (r = .51, p < .001) and VEGF expression (r = .35, p < .001). For the prognosis of patients, high VEGF expression and MVD counts were associated with reduced progressive and survival times (PFS, p = .032 and p = .034; OS, p = .041 and p = .038, respectively). The correlation between parameters of microvascular imaging, VEGF expressive status, and the MVD counts were established. The cut point of mean intensity (MI = 40) was used to investigate as an independent predictor for PFS (p = .021) and OS (p = .025), respectively, due to a strong correlation between MVD counts and VEGF expression in patients with invasive breast cancer. The microvascular imaging could be a visual and helpful tool to predict the prognosis of patients with invasive breast cancer treated by adjuvant therapies.

Abstract:Objective:To analyse the expression of vascular endothelial growth factor(VEGF)and microvessel density(MVD) in pancreatic carcinoma for evaluating the effect of angiogenesis in the development of this tumor.Methods:Collection of 43 ca- ses of pancreatic carcinoma sample and 43 normal pancreatic tissues as control,using immunohistochemistry(IHC)to detect the expression of VEGF and MVD.Results:The positive rate of VEGF was 76.7% in pancreatic carcinoma and 53.5% in nor- mal pancreatic tissue,while MVD was 50.01±26.33 in pancreatic carcinoma and 12.66±5.76 in normal pancreatic tissue. These two parameters was significantly higher in tumor group than in normal group(P0.05).In pancreatic carcinoma, VEGF and MVD in the group with lymph node metastasis was significantly higher than in the group without lymph node metas- tasis,as well as in the group with advanced TNM staging VS early staging(P0.05).VEGF expression in pancreatic carcino- ma was found to be consistently correlated with MVD(P0.05).In the pancreatic carcinoma,survival time in the group with higher expression of VEGF or MVD was significantly shorter than the group with lower expression of VEGF or MVD(P(0.05).MVI)was the unique factor which can impact survival time(P0.05).Conclusion:(1)VEGF plays an important role in the angiogenesis,tumor growth and metastasis of pancreatic carcinoma.(2)VEGF and MVD,due to the consistent correlation of the both.could he considered as an ideal indicator of angiogenesis for the pancreatic carcinoma.(3)VEGF and MVD can be used as predictors for the biological behavior and the prognosis of the pancreatic carcinoma.

Abstract:Objective To explore the relationship between the microvessel density (MVD)with tumor invasion and metastasis in patients with esophageal carcinoma.Methods The surgical specimens of 48 patients with esophageal squamous cell carcinoma were collected,SP immunohistochemical method was used to mark CD34,then MVD in tumor tissues were detected.Results The MVD in patients with esophageal carcinoma wasn't related to age,sex,volume of tumor and invasive depth ( P 0 05),but MVD was related to the tumor differential grades( P 0 05) and the lymph node metastasis( P 0 0001).Conclusion MVD can be used as an important biology index to indicate the malignant degree and prognosis indicator of esophageal carcinoma.

Abstract:Objective To investigate the expression of vascular endothelial growth factor (VEGF) in colorectal cancers and its relation with the microvessel density (MVD).Methods MVD and VEGF were immunohistochemical studies by SP in formalin fixed paraffin embedded specimens of 52 colorectal cancers,10 colorectal adenomas,8 normal colorectal mucosas.Results (1)MVD of colorectal cancers (20 73±10 16)were significantly higher than colorectal adenomas(8 53±2 33) and normal colorectal mucosas (3 50±0 82), P 0 01;colorectal adenomas were also significantly higher than normal colorectal mucosas( P 0 05).The MVD in colorectal cancers significantly correlated with depth of invasion,lymph node metastasis,and distant metastasis,but the relation to the histological types and tumor size was not significant.The MVD of Duke stage D,C were significantly higher than A,B( P 0 01).(2)The expression positive rate of VEGF in colorectal cancers (65 4%,34/52) were significantly higher than colorectal adenomas,normal colorectal mucosas (30%,3/10,12 5%,1/8), P 0 01.The VEGF expression significantly correlated with the depth of invasion ( P 0 05),lymph node metastasis and distant metastasis,but the relation to histological types and tumor size was not significant.With the advancing of Duke stage,the VEGF expression increased accordingly,and Duke D,C were significantly higher than Duke B,A ( P 0 01,or P 0 05).(3)A close correlation was present between MVD with VEGF(γ=0 534, P 0 05).Conclusion MVD and VEGF expression were directly correlated with the biological behaviors of colorectal cancers.VEGF plays a central role in tumor associated angiogenesis of colorectal cancers.

Abstract:Objective To study the angiogenesis in breast carcinoma and its correlation to expression of vascular endothelial growth factor (VEGF) and microvessel density (MVD) and prognosis.Methods Immunohistochemistry method was used to detect the expression of VEGF and MVD in the cancerous and corresponding paracancerous tissues in 109 cases of breast carcinoma.Results The positive rate of VEGF expression in breast carcinoma tissue was 93.57%,higher than that of the neighboring noncancerous tissues (42.67%),P0.05.In addition,MVD and VEGF had a closely positive correlation between tumor size and histological grade (P0.01).Conclusion VEGF is an important angiogentic factor in the breast cancinoma.VEGF and MVD are closely correlated with lymph node metastasis or tumor recurrence.

Abstract:Objective: To investigate the relationship of vascular endothelial growth factor (VEGF) and angiogenesis with the progression of colorectal carcinoma. Methods: VEGF protein expression and microvascular density in 63 cancer tissues, 10 neighboring non-cancerous and 10 normal tissues were examined by SABC immunohistochemical technique. Results: The positive rate of VEGF expression in colorectal carcinoma tissues was higher than that of neighboring non-cancerous tissues (P<0.05), MVD was significantly higher in VEGF-positive colorectal carcinoma than in VEGF-negative colorectal carcinoma (P<0.05), but VEGF and MVD were not related to histological grade (P>0.05). MVD and VEGF were positively correlated with the depth of invasion, Dukes stage, lymph node metastasis and blood metastasis of tumor (P<0.05). The prognosis of patients with positive VEGF and high MVD was significantly poorer than that of those with negative VEGF and low MVD (P< 0.05). Conclusion: VEGF is closely related to angiogenesis of colorectal carcinoma and promotes growth, invasion and metastasis of colorectal carcinoma. MVD and VEGF can be used as useful markers which guide the choice and better use of therapy, for the stage and prognosis in colorectal cancer.

The relationship between preoperative mammographic signs and expression of VEGF and MVD was analyzed. [Results] The expression of VEGF had positive correlation to their corresponding MVD values (r=0.691, P0.01). VEGF and MVD value were related to the tumor size, speculation sign, cluster of calcification, enlarged ressels and the axillary lymphnode metastasis (P0.05), but were not related to crass skin, retracted nipple (P0.05). [Conclusion] The mammographic signs of breast cancer are closely related to expression of VEGF and MVD. This mammographic signs can predict biological behaviour of breast cancer and maybe evidence for early diagnosis and estimation prognosis of breast cancer.
Vascular Endothelial Growth Factor and Microvascular Density in Esophageal and Gastric Carcinomas

Abstract:AIM: To observe the relationship between the expression of vascular endothelial growth factor (VEGF), microvascular density (MVD) and the pathological characteristics of esophageal and gastric carcinomas.METHODS: S-P immunohistochemical staining was used to investigate the expression of VEGF in all the specimens. The antibody against factor VIII-related antigen was used to display vascular endothelial cells, and MVD was examined by counting the factor VIII-positive vascular endothelial cells.RESULTS: The positive rates of VEGF expression in esophageal carcinoma and gastric carcinoma were 81.36 % and 67.5 % respectively, and the MVD averaged 41.81+/-8.44 and 34.36+/-9.67 respectively, which were higher than those in benign diseases. The expression of VEGF and MVD were closely correlated with the degree of differentiation, lymphatic metastasis, but not related to depth of cancer invasion. In early stage gastric carcinoma, the rate of expression of VEGI and MVD was lower than that in progressive gastric carcinomas.CONCLUSION: The expression of VEGF is correlated with tumor angiogenesis, and VEGF plays an important role in new blood vessels formation, the expression of VEGF and MVD play an important role in tumor growth and metastasis. MVD and the expression of VEGF may be two important indexes for patients' prognosis.
Relationship Between the VEGF and Pathological Factor As Well As Prognostic Significance in Breast Cancer

Abstract:Objective: To investigate the expression of vascular endothelial growth factor (VEGF) in breast cancer and to evaluate the prognostic significance of VEGF.Methods: A total of 109 patients were selected to this study from the years l985 to l986. VEGF and microvessel density(MVD) were analyzed by immunohistochemistry in primary lesions in these patients. Resu1ts: High expression group of VEGF and low expression group of VEGF had significant difference in lymph node involvement(52.2% and 23.5%, P0.05) and histologic grade. There was a trend of increase of expression level of VEGF compared with decrease of 5-years 10-years 15-years survival but which hadn't significant difference.VEGF expression is not connected with tumor size,menopause and estrogen receptor. Conclusion: VEGF expression is significantly related to the lymph node involvement and histologic grade and may be used as one of the prognostic indicators in breast cancer.

Abstract:Objective To investigate the correlation between the microvessel density(MVD) and expression level of vascular endothelial growth factor(VEGF) and biological behavior of non-small cell lung cancer(NSCLC) and its prognosis. Methods MVD and VEGF expression in 56 cases of NSCLC and 16 cases of nomal lung tissues was tested with immunohistochemistry methods. Results (1)significant differences in MVD and VEGF expression were found between NSCLC and nomal lung tissues (P0.05),lung cancer MVD increased with the increase of the VEGF expression,they had positive relation.(2)MVD and VEGF expression in lung adenocarcinoma was higher than that in squamosumcarcinoma (P0.05),they increased with the lymphnode metastasis and advancing of TNM Stage (P0.05).(3)Between the survival time in the group less than 2 years and the group more than 2 years, both MVD and VEGF expression had significant difference(P0.05).Conclusion MVD and VEGF are closely related to the occurrence,development and metastasis of NSCLC, they can be used as a major judgement parameter for biological behavior and prognosis of NSCLC.
