Abstract:Catestatin (CST) was first named in 1997 for its catecholamine-inhibitory activity. It was discovered as a potent inhibitor of catecholamine secretion and as a regulator of histamine release. Accumulating evidence shows that CST is involved with cardiovascular diseases; however, whether CST is a protective factor for these conditions and the mechanisms by which such actions may be mediated are not well understood. In this article, we review recent basic research and clinical trials in the study of CST and summarize the association of CST with cardiovascular diseases. We review data obtained from MedLine via PubMed and from our own investigations.

Abstract:Catestatin is a multifunctional peptide that is involved in the regulation of the cardiovascular and immune systems as well as metabolic homeostatis. It mitigates detrimental, excessive activity of the sympathetic nervous system by inhibiting catecholamine secretion. Based on in vitro and in vivo studies, catestatin was shown to reduce adipose tissue, inhibit inflammatory response, prevent macrophage-driven atherosclerosis, and regulate cytokine production and release. Clinical studies indicate that catestatin may influence the processes leading to hypertension, affect the course of coronary artery diseases and heart failure. This review presents up-to-date research on catestatin with a particular emphasis on cardiovascular diseases based on a literature search.

Abstract:Background: Cardiovascular disease remains one of the leading causes of death globally. Myocardial ischemia and infarction, in particular, frequently cause disturbances in cardiac electrical activity that can trigger ventricular arrhythmias. We aimed to investigate whether catestatin, an endogenous catecholamine-inhibiting peptide, ameliorates myocardial ischemia-induced ventricular arrhythmias in rats and the underlying ionic mechanisms. Methods and results: Adult male Sprague-Dawley rats were randomly divided into control and catestatin groups. Ventricular arrhythmias were induced by ligation of the left anterior descending coronary artery and electrical stimulation. Action potential, transient outward potassium current, delayed rectifier potassium current, inward rectifying potassium current, and L-type calcium current (ICa-L) of rat ventricular myocytes were recorded using a patch-clamp technique. Catestatin notably reduced ventricular arrhythmia caused by myocardial ischemia/reperfusion and electrical stimulation of rats. In ventricular myocytes, catestatin markedly shortened the action potential duration of ventricular myocytes, which was counteracted by potassium channel antagonists TEACl and 4-AP, and ICa-L current channel agonist Bay K8644. In addition, catestatin significantly increased transient outward potassium current, delayed rectifier potassium current, and inward rectifying potassium current density in a concentration-dependent manner. Catestatin accelerated the activation and decelerated the inactivation of the transient outward potassium current channel. Furthermore, catestatin decreased ICa-L current density in a concentration-dependent manner. Catestatin also accelerated the inactivation of the ICa-L channel and slowed down the recovery of ICa-L from inactivation. Conclusions: Catestatin enhances the activity of transient outward potassium current, delayed rectifier potassium current, and inward rectifying potassium current, while suppressing the ICa-L in ventricular myocytes, leading to shortened action potential duration and ultimately reducing the ventricular arrhythmia in rats.

Abstract:BACKGROUND:Catestatin, a chromogranin A-derived peptide, is a potent antagonist of nicotine-evoked catecholamine release. We know that catecholamine plays an important role in cardiovascular remodeling induced by hypertension, therefore we hypothesized that catestatin would affect target-organ structure during hypertension.METHODS:Twelve spontaneously hypertensive rats (SHRs) were randomized to SHR control group and catestatin group, the normal control group was comprised of six healthy Wistar-Kyoto rats of the same age. Tail-cuff blood pressure and pulse rate were obtained at weeks 1, 4 and 8. At the end of the eight-week period, the heart, abdominal aorta and left kidney were excised and weighed, VG staining was done and the intima-media thickness of vessels and the collagen volume fraction were assessed by an image acquisition and analysis system. The proliferating cell nuclear antigen (PCNA) was observed by immunohistochemistry, and real time reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the mRNA levels of proliferative genes including cyclin A, ki67 and PCNA in the abdominal aorta.RESULTS:All the parameters in SHR observed in the present study increased significantly compared to Wistar Kyoto rats (P < 0.01). With intervention with catestatin, the systolic blood pressure decreased slightly but it was not significantly different from the SHR control, the cardiac mass index and left ventricular mass index both decreased significant ly, the collagen volume fraction decreased by nearly 30% in the heart, by 25% in vessels and by 10% in the kidney, and the intima-media thickness and expression of proliferative genes, including cyclin A, ki67 and PCNA, in the abdominal aorta also decreased significant ly.CONCLUSIONS:The present study indicated that catestatin could ameliorate proliferating changes of heart, kidney and vessels during hypertension, especially to the deposition of interstitial collagen. Blood pressure was not the main factor to mediate this effect, which suggested that catestatin could become a novel protective factor for hypertensive target organs.

Abstract:Objective Catestatin is a newly discovered endogenous neuropeptide with potent catecholamine release-inhibitory activity.The purpose of the present study was to evaluate the possible pathophysiologic role of catestatin through investigation of plasma catestatin levels in patients with congestive heart failure(CHF).Methods Plasma catestatin was measured with enzyme-linked immunosorbent assay and norepinephine was measured with high performance liquid chromatography in 45 patients with CHF and 30 controls.Results Plasma catestatin(0.88±0.30 vs 0.41±0.14 ng/ml) and norepinephrine(421.9±61.4 vs 210.2±55.4 pg/ml) levels were significantly higher in patients with CHF than those in controls(P<0.01).Both neuroendocrine factors levels decreased after treatment,but still remained significantly higher than those in controls(P<0.01).However,in CHF patients after treatment,the catestatin/norepinephrine ratio was significantly increased(2.39±0.70 vs 1.95±0.64,P<0.01).Plasma catestatin levels were similar among patients with different NYHA classes,as well as between patients with normal and reduced ejection fraction.Plasma catestatin level was correlated positively with NTproBNP level(r=0.38,P<0.05),while no correlation was found between catestatin and left ventricular ejection fraction,hsCRP or cTNI.The area under ROC curve of catestatin in CHF for diagnosis was 0.94.Conclusion Plasma catestatin levels in patients with CHF were significantly elevated and positively correlated with NTproBNP.Catestatin might be used as a new biomarker in CHF.

Abstract:Background: Catestatin is an active neuropeptide derived from chromogranin A, which can suppress catecholamine to release. Since catecholamine plays an important role in cardiovascular remodeling secondary to hypertension, we therefore hypothesized that catestatin would affect structural changes of hypertensive target organs. Methods: Spontaneous hypertensive rats were used to observe the effect of catestatin. After 8-week-intervention, removing the heart, abdominal aorta and left kidney, calculating mass index and conducting VG staining. The intima-media thickness of vessel and collagen volume fraction were measured by image acquisition and analysis system, and the expressions of proliferative genes in abdominal aorta were detected by Real Time RT-PCR. Results: The cardiac mass index, left ventricular mass index, kidney mass index, intima-media thickness, collagen volume fraction of acquired tissues and proliferative genes of abdominal aorta all increased notable in spontaneous hypertensive rats, comparing with Wistar Kyoto rats. With intervention of catestatin, the cardiac mass index and left ventricular mass index both decreased significantly, the collagen volume fraction decreased nearly by 30% in heart, by 25% in vessel and by 10% in kidney, samely the intima-media thickness and expression of proliferative genes. Conclusions: The present study indicated that catestatin could ameliorates proliferative changes in cardiac, renal and vascular in hypertension, especially to interstitial collagen deposition, which suggest that catestatin maybe a novel protective factor for hypertensive target organs.
Catestatin reverses the hypertrophic effects of norepinephrine in H9c2 cardiac myoblasts by modulating the adrenergic signaling

Abstract:Catestatin (CST) is a catecholamine release-inhibitory peptide secreted from the adrenergic neurons and the adrenal glands. It regulates the cardiovascular functions and it is associated with cardiovascular diseases. Though its mechanisms of actions are not known, there are evidences of cross-talk between the adrenergic and CST signaling. We hypothesized that CST moderates the adrenergic overdrive and studied its effects on norepinephrine-mediated hypertrophic responses in H9c2 cardiac myoblasts. CST alone regulated the expression of a number of fetal genes that are induced during hypertrophy. When cells were pre-treated CST, it blunted the modulation of those genes by norepinephrine. Norepinephrine (2 µM) treatment also increased cell size and enhanced the level of Troponin T in the sarcomere. These effects were attenuated by the treatment with CST. CST attenuated the immediate generation of ROS and the increase in glutathione peroxidase activity induced by norepinephrine treatment. Expression of fosB and AP-1 promoter-reporter constructs was used as the endpoint readout for the interaction between the CST and adrenergic signals at the gene level. It showed that CST largely attenuates the stimulatory effects of norepinephrine and other mitogenic signals through the modulation of the gene regulatory modules in a characteristic manner. Depending upon the dose, the signaling by CST appears to be disparate, and at 10-25 nM doses, it primarily moderated the signaling by the β1/2-adrenoceptors. This study, for the first time, provides insights into the modulation of adrenergic signaling in the heart by CST.
Dramatic Changes in Catestatin Are Associated with Hemodynamics in Acute Myocardial Infarction.

Abstract:Acute myocardial infarction (AMI) is characterized by complex neuroendocrine activation. To investigate catestatin profiles, serial catestatin levels were determined by enzyme-linked immunosorbent assay in the first week after AMI in 50 patients. Catestatin levels reduced at admission and negatively correlated with heart rates; it increased significantly on the third day but remained decreased at 1 week and positively with blood pressure. In a subgroup of 20 patients admitted within 4 h after onset, circulating catestatin correlated inversely with norepinephrine. Catestatin might be involved in the course of AMI and act as a tool in monitoring the progression of AMI.

Abstract:Aims Endogenous catecholamine release‐inhibitory peptide catestatin has been associated with heart failure (HF). This subgroup analysis of our cohort of HF compared the different effects of catestatin as a predictor for cardiac outcomes in patients with HF with reduced (HFrEF), mildly reduced (HFmrEF) or preserved (HFpEF) ejection fraction. Methods Plasma catestatin was measured in the HF patient cohort of 228 cases with a whole spectrum of ejection fraction. The cardiac deaths were analysed according to prespecified subgroups. Results Over a median follow‐up of 52.5 months, the association between plasma catestatin and cardiac death was different in patients with HFrEF, HFmrEF or HFpEF [hazard ratio (HR) 1.53, 95% confidence interval (CI) 0.99–2.37 and HR 2.73, 95% CI 1.56–4.75, respectively; interaction P = 0.022]. Patients with HFmrEF/HFpEF were older and more likely to be female, with non‐ischaemic cardiomyopathy and atrial fibrillation but lower levels of plasma B‐type natriuretic peptide (BNP). Similar adverse cardiac events occurred in patients with HFmrEF/HFpEF as in HFrEF. Plasma catestatin was a better predictor for cardiovascular death in the HFmrEF/HFpEF patients [area under the receiver operating characteristic curve (AUC) = 0.72, 95% CI 0.45–0.74] than in the HFrEF patients (AUC = 0.59, 95% CI 0.587–0.849). The optimal cut point of plasma catestatin level of 0.86 ng/mL predicted a 2.80‐fold elevated risk for cardiac death in HFmrEF/HFpEF. Conclusions Elevated plasma catestatin might be a more sensitive predictor for cardiac outcome in patients with HFmrEF/HFpEF than in HFrEF.

Abstract:Catestatin was discovered as a potent inhibitor of catecholamine secretion and plays important roles in the cardiovascular system. Our previous study demonstrates a close relationship between catestatin levels and prognosis of ST-elevation myocardial infarction (STEMI). Using the same population, the goal of this study is to investigate the ability of catestatin to predict left ventricular (LV) remodeling in STEMI patients. 72 patients and 30 controls were included. Catestatin was sampled after admission to the emergency room (ER), at day3 (D3), and day7 (D7) after STEMI. Echocardiography was performed at D3 and after 65 months for evaluation of LVEDD, EF, IVS, LVPW, E, A, E’, E/A, and E/E’. The changes of these parameters from D3 to 65 months were used to reflect the changes of ventricular structure and function. We found that plasma catestatin levels at D3 were highly correlated with the changes of LVEDD, EF, E, A, E’, E/A, as well as E/E’. Patients with higher catestatin levels developed worse ventricular function during the follow-up period. Single-point catestatin was effective to predict LVEDD change. And concurrently increasing catestatin and NT-proBNP levels predicted the highest risk of LV remodeling. This study suggests an important prognostic information of catestatin on LV remodeling.

Abstract:Background Catestatin has been reported as a pleiotropic cardioprotective peptide. Heart failure with preserved ejection fraction (HFpEF) was considered a heterogeneous syndrome with a complex cause. We sought to investigate the role of catestatin in HFpEF and diastolic dysfunction. METHODS AND RESULTS Administration of recombinant catestatin (1.5 mg/kg/d) improved diastolic dysfunction and left ventricular chamber stiffness in transverse aortic constriction mice with deoxycorticosterone acetate pellet implantation, as reflected by Doppler tissue imaging and pressure‐volume loop catheter. Less cardiac hypertrophy and myocardial fibrosis was observed, and transcriptomic analysis revealed downregulation of mitochondrial electron transport chain components after catestatin treatment. Catestatin reversed mitochondrial structural and respiratory chain component abnormality, decreased mitochondrial proton leak, and reactive oxygen species generation in myocardium. Excessive oxidative stress induced by Ru360 abolished catestatin treatment effects on HFpEF‐like cardiomyocytes in vitro, indicating the beneficial role of catestatin in HFpEF as a mitochondrial ETC modulator. The serum concentration of catestatin was tested among 81 patients with HFpEF and 76 non–heart failure controls. Compared with control subjects, serum catestatin concentration was higher in patients with HFpEF and positively correlated with E velocity to mitral annular e′ velocity ratio, indicating a feedback compensation role of catestatin in HFpEF. Conclusions Catestatin protects against diastolic dysfunction in HFpEF through attenuating mitochondrial electron transport chain–derived reactive oxygen species generation. Serum catestatin concentration is elevated in patients with HFpEF, probably as a relatively insufficient but self‐compensatory mechanism.

Abstract:Catestatin (CST) was first discovered as a potent non-competitive and reversible inhibitor of catecholamine secretion. Recent reports on plasma CST level in heart diseases suggested a cardioprotective role for this peptide. Given that cardiac remodeling is the dominant pathologic process in cardiac dysfunction, we propose that CST participates in the regulation of concern pathways and contributes to the inhibition of cardiac remodeling. In this minireview, the potential mechanism of cardiac remodeling involving CST will be discussed from three aspects: hypertrophy, fibrosis, and apoptosis.

Abstract:Background and aims: The neuropeptide catestatin (CST) is an endogenous nicotinic cholinergic antagonist that acts as pleiotropic cardiac protective hormone. This study investigated the association between CST and coronary artery disease (CAD) and the underlying mechanisms. Methods and results: The serum concentration of CST among 224 CAD patients and 204 healthy controls was compared, and its association with atherosclerosis severity in 921 CAD patients was further analyzed. Compared to healthy subjects, serum CST concentration was lower in patients with CAD [ 1.14 (1.05-1.24) ng/mL vs. 2.15 (1.92-2.39) ng/mL, p < 0.001] and was inversely correlated with disease severity (r=-0.208, p < 0.001). In cultured endothelial cells, CST suppressed TNF-alpha-elicited expression of inflammatory cytokines and adhesion molecules by activating angiotensin-converting enzyme-2 (ACE2). Administration of CST reduced leukocyte-endothelium interactions in vitro and in vivo, and attenuated the development of atherosclerotic in ApoE(-/-) mice fed a high-fat diet. These protective effects by CST were blocked by an ACE2 inhibitor. Conclusions: Serum CST concentration is lower in CAD patients and is inversely associated with the severity of atherosclerosis. CST acts as a novel anti-atherogenic peptide that inhibits inflammatory response and EC-leukocyte interactions via an ACE2-dependent mechanism.
Correlation Of Plasma Catestatin Level And The Prognosis Of Patients With Acute Myocardial Infarction

Abstract:Catestatin is a peptide which is a potent inhibitor of catecholamine secretion and played essential functions in the cardiovascular system. Previous research found that dramatic changes of catestatin were associated with hemodynamics in acute myocardial infarction (AMI) during the first week after the AMI symptoms onset, but whether catestatin is also involved in the pathophysiological progression after AMI and then a predictor for outcomes is not clear. The aim of this study is to determine the correlation of plasma catestatin levels at different time points and the prognosis of AMI. 100 participants recruited were all patients with AMI, all of who received successful primary percutaneous coronary intervention (PCI) within 12h from the AMI symptom onset in our center; the concentrations of plasma catestatin were evaluated from blood samples of those 100 participants. Subsequent 65 months' follow-up was performed after discharging to evaluate cardiac adverse events and the association between catestatin levels and prognosis of AMI was examined. We confirmed the dramatic change of catestatin concentrations in the first week of AMI, and the levels of catestatin on D3 were much higher in adverse events group than those in non-adverse events group (p<0.0001), but the ratio of D7/D3 was significantly lower. In addition, the Kaplan-Meier analysis showed that the groups in which the levels on D3 were higher (p<0.0001) and the ratios of D7/D3 were lower (p<0.0001), patients trended to be more susceptive to adverse events after AMI. Furthermore, according to the analysis, we surmised catestatin level on D3 as an appropriate predictor for outcomes in patients with AMI with good specificity as well as sensitivity. All of the evidence confirmed that catestatin plays an important role in the progress of AMI, and may act as a promising target for prognostic prediction.

Abstract:Background Catestatin plays an important role in the adjustment of blood pressure and cardiac function. We investigated levels of plasma catestatin in essential hypertension and the relationship between catestatin and left ventricular hypertrophy. Methods Plasma was collected from 136 patients with essential hypertension and 61 healthy controls. Plasma catestatin was measured by enzyme-linked immunosorbent assay (ELISA). Plasma norepinephrine was measured by high-performance liquid chromatography. All patients underwent echocardiography, measurement of fasting blood glucose, body mass index (BMI) and lipid levels. Results Plasma levels of catestatin and norepinephrine were significantly higher in patients with essential hypertension than in normal controls (both P<0.01). The ratio of catestatin to norepinephrine was significantly lower in patients with essential hypertension than in normal controls (P<0.01). In patients with essential hypertension, plasma norepinephrine level was significantly higher in patients with than without left ventricular hypertrophy (P<0.01). Plasma catestatin level was lower, but not significantly, in patients with than without left ventricular hypertrophy. The ratio of catestatin to norepinephrine was significantly lower in patients with than without left ventricular hypertrophy (P<0.01). Conclusion Plasma catestatin is elevated in patients with essential hypertension. The ratio of catestatin to norepinephrine was lower in patients with left ventricular hypertrophy. Catestatin might participate in the development of hypertension and left ventricular hypertrophy.

Abstract:Aims: We have recently identified a functional pool of intracellular β 1 adrenergic receptors (β 1 ARs) crucial for cardiac function. However, the systematic signaling network of intracellular β 1 ARs remains elusive. Here, we aim to uncover the integrative control of cardiac function, compartmentalized β 1 AR signaling, and potential interface between two critical modulators of catecholamine homeostasis, MAO-A and OCT3. Methods and Results: Cardiac deletion of MAO-A (MAO-A-CKO) elevated myocardium catecholamines, cAMP levels and cardiac ejection fraction by disrupting intracellular catecholamine degradation. Deletion and pharmacological inhibition of MAO-A both enhanced cardiac adrenergic response, with elevated ejection fraction and heart rate. Inhibiting OCT3 by corticosterone prevented the impacts of MAO-A inhibition on cardiac function. Using subcellular anchored Förster resonance energy transfer (FRET) biosensors, we then determined the regulation of compartmentalized β 1 AR/PKA signaling at the sarcoplasmic reticulum (SR) and plasma membrane (PM) microdomains. We also examined local PKA-substrates phosphorylation and myocyte excitation-contraction coupling. MAO-A deletion and inhibitor (MAOi) enhanced local PKA activity at the SR, phosphorylation of phospholamban, Ca 2+ cycling, and contractile response. Whereas overexpression of MAO-A impairs SR-localized PKA activity and phospholamban phosphorylation. In contrast, these modifying of MAO-A did not affect PKA activity at PM and L-type Ca 2+ channel phosphorylation. Inhibiting OCT3 blocked blocking catecholamine transport and diminished the efficacy of MAO-A inhibitor (MAO-A) and MAO-A-CKO to regulate SR-localized β 1 AR signaling. In parallel, OCT3 inhibition and deletion prevented the MAO-A-dependent enhancement of Ca 2+ cycling and cardiac contractile function. Conclusion: Together, we show that MAO-A and OCT3 act in a concert to fine-tune the intracellular catecholamines, activation of SR-localized β 1 AR-PKA signaling, and cardiac fight-or-flight response. Further, we reveal a drug contraindication between anti-inflammatory corticosterone and anti-depressant MAOi in modulating adrenergic regulation in the heart, providing novel perspectives of those drugs in cardiac implications.

Abstract:Catestatin (CST) is a catecholamine secretion inhibiting peptide as non-competitive inhibitor of nicotinic acetylcholine receptor. CST play a protective role in cardiac ischemia/reperfusion (I/R) but the molecular mechanism remains unclear. Cardiomyocytes endogenously produced CST and its expression was reduced after I/R. CST pretreatment decreased apoptosis especially endoplasmic reticulum (ER) stress response during I/R. The protection of CST was confirmed in H9c2 cardiomyoblasts under Anoxia/reoxygenation (A/R). In contrast, siRNA-mediated knockdown of CST exaggerated ER stress induced apoptosis. The protective effects of CST were blocked by extracellular signal-regulated kinases 1/2 (ERK1/2) inhibitor PD90895 and phosphoinositide 3-kinase (PI3 K) inhibitor wortmannin. CST also increased ERK1/2 and protein kinase B (Akt) phosphorylation and which was blocked by atropine and selective type 2 muscarinic acetylcholine (M2) receptor, but not type 1 muscarinic acetylcholine (M1) receptor antagonist. Receptor binding assay revealed that CST competitively bound to the M2 receptor with a 50% inhibitory concentration of 25.7 nM. Accordingly, CST inhibited cellular cAMP stimulated by isoproterenol or forskolin, and which was blocked by selective M2 receptor antagonist. Our findings revealed that CST binds to M2 receptor, then activates ERK1/2 and PI3 K/Akt pathway to inhibit ER stress-induced cell apoptosis resulting in attenuation cardiac I/R injury.
Prognostic Differences of Catestatin among Young and Elderly Patients with Acute Myocardial Infarction

Abstract:BACKGROUND: Previous studies have reported inconsistent findings regarding the association between catestatin and outcomes of acute myocardial infarction (AMI). This study aims to investigate the prognostic value of catestatin for long-term outcomes in patients with AMI. METHODS: One hundred and sixty-five patients with AMI were enrolled in this series. The plasma catestatin levels at baseline and clinical data were collected. All patients were followed up for four years to investigate whether there were major adverse cardiovascular events (MACEs), including cardiovascular death, recurrent AMI, rehospitalization for heart failure, and revascularization. RESULTS: There were 24 patients who had MACEs during the follow-up period. The MACEs group had significantly lower plasma catestatin levels (0.74??0.49 ng/mL vs. 1.10??0.79 ng/mL, P=0.033) and were older (59.0??11.4 years old vs. 53.2??12.8 years old, P=0.036). The rate of MACEs was significantly higher in the elderly group ( _60 years old) than in the young group (<60 years old) (23.8% [15/63] vs. 8.8% [9/102], P=0.008). The catestatin level was significantly lower in the MACEs group than that in the non-MACEs group (0.76??0.50 ng/mL vs. 1.31??0.77 ng/mL, P=0.012), and catestatin was significantly associated with MACEs (Kaplan Meier, P=0.007) among the elderly group, but not in the young group (Kaplan Meier, P=0.893). In the Cox proportional hazards regression, high catestatin was one of the independent factors for predicting MACEs after adjustment for other risk factors (hazard ratio 0.19, 95% confidence interval 0.06-0.62, P=0.006) among elderly patients. CONCLUSIONS: Elderly AMI patients with lower plasma catestatin levels are more likely to develop MACEs. Catestatin may be a novel marker for the long-term prognosis of AMI, especially in elderly patients.
