Abstract:Background: The milk fat globule membrane (MFGM) is a thin film that exists within the milk emulsion, suspended on the surface of milk fat globules, and comprises a diverse array of bioactive components. Recent advancements in MFGM research have sparked a growing interest in its biological characteristics and health-related functions. Thorough exploration and utilization of MFGM as a significant bioactive constituent in milk emulsion can profoundly impact human health in a positive manner. Scope and approach: This review comprehensively examines the current progress in understanding the structure, composition, physicochemical properties, methods of separation and purification, and biological activity of MFGM. Additionally, it underscores the vast potential of MFGM in the development of additives and drug delivery systems, with a particular focus on harnessing the surface activity and stability of proteins and phospholipids present on the MFGM for the production of natural emulsifiers and drug encapsulation materials. Key findings and conclusions: MFGM harbors numerous active substances that possess diverse physiological functions, including the promotion of digestion, maintenance of the intestinal mucosal barrier, and facilitation of nerve development. Typically employed as a dietary supplement in infant formula, MFGM's exceptional surface activity has propelled its advancement toward becoming a natural emulsifier or encapsulation material. This surface activity is primarily derived from the amphiphilicity of polar lipids and the stability exhibited by highly glycosylated proteins.

Abstract:Dairy products have become more common in people's daily diets in recent years, and numerous useful components derived from milk are widely employed in the food industry. Milk fat globule membrane (MFGM) is a kind of film that encases milk fat globules, and has been shown to have a high nutritional value. In this work, the protein, lipid, carbohydrate, and other components of MFGM are discussed, and also common separation, preparation, and analysis technologies, physicochemical properties, and functional features of MFGM are reviewed, to provide some guidance for the development and utilization of MFGM.

Abstract:Fat is distributed in the form of tiny, spherical droplets or globules, stabilized in the form of an emulsion. The membrane surrounding fat globules in milk, called the milk fat globule membrane (MFGM). The MFGM is characterized by a complex three-layer mixture of proteins, phospholipids and glycoproteins. Variety of factors can make a inlfuence to MFGM, e.g. physiological condition of cows, the stage of lactation, harvest, processing treatments, etc. Furthermore, both MFGM proteins and MFGM lipids have been shown to possess some unique functional properties and health beneifts. This review summarized the current knowledge of MFGM, focusing on its composition, structure, inlfuence factors and nutritional aspects. Future perspectives were also mentioned.

Abstract:Milk fat globule membrane (MFGM) is a three-layer membrane-like structure encasing natural milk fat globules (MFGs). MFGM holds promise as a nutritional supplement because of the numerous physiological functions of its constituent protein. This review summarizes and compares the differences in MFGM protein composition across various species, including bovines, goats, camels, mares, and donkeys, and different lactation periods, such as colostrum and mature milk, as assessed by techniques such as proteomics and mass spectrometry. We also discuss the health benefits of MFGM proteins throughout life. MFGM proteins promote intestinal development, neurodevelopment, and glucose and lipid metabolism by upregulating tight junction protein expression, brain function-related genes, and glucose and fatty acid biosynthesis processes. We focus on the mechanisms underlying these beneficial effects of MFGM proteins. MFGM proteins activate key substances in in signaling pathways, such as the phosphatidylinositol 3-kinase/protein kinase B, mitogen-activated protein kinase, and myosin light chain kinase signaling pathways. Overall, the consumption of MFGM proteins plays an essential role in conferring health benefits, some of which are important throughout the mammalian life cycle.

Abstract:In this research,milk fat globule membrane (MFGM) was separated from five different species raw milk (cow,goat,buffalo,yak and camel).Compositions of MFGM from five species milk,include protein,amino acid,lipid,polysaccharide,microelement and ash,were separated and analyzed.Comparison results showed that the differences of compositions in five MFGMs were significant (p＜0.05).Basic data and theory basis were provided in this study for the further research on MFGM poteomics and functional features.

Abstract:Background Currently, the omics techniques have unveiled the complexity of milk fat globule membrane (MFGM) composition which determines its multiple biofunctions and unique interfacial behavior. MFGM has been recommended for supplementation in dairy products and the physiological and interfacial functions of its natural and processed forms have attracted significant attention. Scope and approach This review discussed the inherent association among compositions, biofunctions, interfacial behavior, and digestion properties of natural and processed MFGM. The advanced knowledge of MFGM compositions including polar lipids and proteins is reviewed, and the association of the compositions with their biofunctions and the size distributions of milk fat globules (MFGs) are discussed. Then the discrepancies of natural and processed MFGs in interfacial properties and behaviors are summarized, highlighting the regulatory functions of MFGM interfacial behavior in lipid digestion. Key findings and conclusions The intricate compositions of MFGM correspond to its multiple biological activities, and the synergistic effects among individual substances deserve attention. The composition characteristics of native MFGM also determine its interfacial behaviors (lateral phase segregation, charge heterogeneity, morphology, and mechanical property) and further affect the fate of MFGs in gastrointestinal tract. Dairy processing operations including homogenization, thermal treatment, and spray drying cause the decrease of size, loss of natural structures, and changes of compositions of MFGM, impairing its bioactivates and altering the digestion patterns of lipids. The MFGM-based food ingredients deserve further development in production and applications, and future research should focus on the preservation of bioactivities and techo-functions of natural MFGM.

Abstract:The composition and physical-chemical properties of the milk fat globule membrane (MFGM) is a subject that has gained increased interest in the field of food colloids, mainly because the nutritional and technological value of the MFGM. In fact, related changes in integrity and structure during milk processing pose a huge challenge as far as efforts directed to isolate the components of the fat globule membrane. MFGM characteristics and potential utilization are subjects of dissension. Thus, the effects of processing and the colloidal interactions that exist with other milk constituents need to be better understood in order to exploit milk fat and MFGM, their functionality as colloids as well as those of their components. These are the main subjects of this review, which also reports on the results of recent inquiries into MFGM structure and colloidal behavior.

Abstract:Milk fat globule membrane (MFGM) is a complex trilayer structure present in mammalian milk and is mainly composed of phospholipids and proteins (>90%). Many studies revealed MFGM has positive effects on the immune system, brain development, and cognitive function of infants. Probiotics are live microorganisms that have been found to improve mental health and insulin sensitivity, regulate immunity, and prevent allergies. Probiotics are unstable and prone to degradation by environmental, processing, and storage conditions. In this review, the processes used for encapsulation of probiotics particularly the potential of MFGM and its constituents as encapsulating materials for probiotics are described. This study analyzes the importance of MFGM in encapsulating bioactive substances and emphasizes the interaction with probiotics and the gut as well as its resistance to adverse environmental factors in the digestive system when used as a probiotic embedding material. MFGM can enhance the gastric acid resistance and bile resistance of probiotics, mainly manifested in the survival rate of probiotics. Due to the role of digestion, MFGM-coated probiotics can be released in the intestine, and due to the biocompatibility of the membrane, it can promote the binding of probiotics to intestinal epithelial cells, and promote the colonization of some probiotics in the intestine.

Abstract:Breast milk is the safest and most perfect natural food for infant growth and development.As one of the most important components in breast milk,the milk fat globule membrane(MFGM)is a 3-layer membrane structure surrounding milk fat globules(MFG).This unique structure not only maintains the stability of milk but also plays an important role in the digestive and metabolic processes of infants.In this article,we introduce the reader to the composition and structural specificity of MFGM,review the sequential digestion of MFGM depending on several enzymes in the mouth,stomach and intestine of healthy infants,and elaborate on the interaction mechanism between MFGM and various enzymes,in order to provide a reference for relevant studies.
Preparation of milk fat globule membrane ingredients enriched in polar lipids: Composition characterization and digestive properties

Abstract:In this study, milk fat globule membrane (MFGM) ingredients enriched in polar lipids were prepared using membrane filtration, including microfiltration, diafiltration, and ultrafiltration from butter serum powder. Polar lipids (phospholipids, sterols, and gangliosides) in prepared MFGM ingredients were analyzed by 31P nuclear magnetic resonance spectroscopy, GC-MS, and ultra-performance liquid chromatography (UPLC)-MS/MS, respectively. The lipolysis degree and microstructure of MFGM ingredient and soybean lecithin (SL) emulsions during in vitro digestion were also analyzed. Microfiltration showed higher concentration efficiency than ultrafiltration, which increased by 2.16% and 2.73% in phospholipids, respectively. Moreover, diafiltration concentrated more polar lipids (6.39% of phospholipids) than microfiltration. Milk fat globule membrane ingredients had high levels of sphingomyelin (1.27%-1.36%) and ratio of GD3 to GM3 is 9.25- to 9.88-fold. The different lipolysis behaviors between MFGM ingredient emulsions and SL emulsions were correlated with their different polar lipid compositions. Phospholipids from both MFGM ingredients and SL could help maintain the initial structure during the gastric digestion. These results could provide a scientific basis for developing high-polar-lipids food, particularly infant formulas and special functional foods.
Stabilization of Human Milk Fat Analog Emulsions Using Milk Fat Globule Membrane Material–coated Lipid Droplets: Structural and Physical Properties

Abstract:To mimic the human milk fat globule, milk fat globule membrane (MFGM) was used as an emulsifier to coat the lipid droplets for the preparation of the human milk fat analog emulsions (MFEs). The physical properties of MFEs at different pH and ionic strength and digestion properties were evaluated. The results showed that the encapsulation rate of MFEs was more than 90%, and the zeta potential was -18.53 mV. MFGM fragments were adsorbed at the interface of the lipid droplets and remained stably dispersed in the aqueous phase. Aggregation of MFEs occurred at pH of 3.5-5.3 and NaCl >= 50 mM or CaCl2 >= 5 mM. MFEs behaved as viscoelastic liquids at a pH < 6.7 and aggregated at a pH < 4.6, thus forming a gel with increased viscosity. In addition, the digestive characteristics of MFEs were closer to that of human milk. Lipid droplets aggregated during the gastric digestion stage; however, some parts of the globule structure remained until the end of the intestinal digestion stage. This study provides a better understanding of emulsifying and digestive properties of MFGM, and MFGM can be exploited to develop emulsions containing MFGM-coated lipid droplets for the design of infant food.

Moreover, the diafiltration plays an important role to concentrate more polar lipids (6.39% phospholipid). MFGM ingredients had unique phospholipid of sphingolipid and more GD3 than GM3 in gangliosides, while soybean or egg lecithin had other sterols except for cholesterol. MFGM ingredient-emulsification showed easier lipolysis than soybean lecithin-emulsification, which was highly correlated with their different polar lipid compositions.

Abstract:Research on milk fat globule membrane (MFGM) is gaining traction. The interest is two-fold; on the one hand, it is a unique trilayer structure with specific secretory function. On the other hand, it is the basis for ingredients with the presence of phospho- and sphingolipids and glycoproteins, which are being used as food ingredients with valuable functionality, in particular, for use as a supplement in infant nutrition. This last application is at the center of this Review, which aims to contribute to understanding MFGM’s function in the proper development of immunity, cognition, and intestinal trophism, in addition to other potential effects such as prevention of diseases including cardiovascular disease, impaired bone turnover and inflammation, skin conditions, and infections as well as age-associated cognitive decline and muscle loss. The phospholipid composition of MFGM from bovine milk is quite like human milk and, although there are some differences due to dairy processing, these do not result in a chemical change. The MFGM ingredients, as used to improve the formulation in different clinical studies, have indeed increased the presence of phospholipids, sphingolipids, glycolipids, and glycoproteins with the resulting benefits of different outcomes (especially immune and cognitive outcomes) with no reported adverse effects. Nevertheless, the precise mechanism(s) of action of MFGM remain to be elucidated and further basic investigation is warranted.

Abstract:The milk fat globule membrane (MFGM) is the surrounding membrane of the triacyl glyceride-dense fat globules in milk. The native MFGM is a tri-layer structure formed during the biosynthesis of lipids in the rough endoplasmic reticulum of lactocytes within the mammary gland. It is composed of a phospholipid tri-layer, cholesterol, many proteins, glycosylated proteins, and various lipids (Brink and Lönnerdal, 2020). The 2 main sources of MFGM are cream derived during butter production and whey derived from cheese manufacturing. The use of MFGM ingredients in nutritional products, especially in infant nutrition, has received great attention in recent years. Today, there is a range of commercially available MFGM-enriched ingredients from various suppliers across the globe. The basic components of the tri-layer, phospholipids, and proteins have been shown to exert bioactive effects in various models including in humans. It is not yet known how the differences in composition of such complex ingredients impact the bioactive potential of MFGM within the dairy matrix. The MFGM becomes more complex as a result of processing as it loses its native tri-layer structure and MFGM fragments are formed. This review aims to bring to light the underlying mechanisms for the bioactive effects of MFGM ingredients on human health.
Variation in milk fat globule size and composition: A source of bioactives for human health

Abstract:Milk fat globules (MFGs) are secreted from the mammalian gland and are composed of a triacylglycerol core surrounded by a triple membrane structure, the milk fat globule membrane (MFGM). The MFGM contains complex lipids and proteins reported to have nutritional, immunological, neurological and digestive functions. Human and ruminant milk are shown to share a similar MFG structure but with different size, profile and abundance of protein and polar lipids. This review summarizes the reported data on human, bovine, caprine and ovine MFG composition and concentration of bioactive components in different MFG-size fractions. A comprehensive understanding of compositional variations between milk from different species and MFG size fractions may help promote various milk sources as targeted supplements to improve human development and health. MFG size and MFGM composition are species-specific and affected by lactation, diet and breed (or maternal origin). Purification and enrichment methods for some bioactive proteins and lipids present in the MFGM have yet to be established or are not scaled sufficiently to be used to supplement human diets. To overcome this problem, MFG size selection through fractionation or herd selection may provide a convenient way to pre-enrich the MFG fraction with specific protein and lipid components to fulfill human dietary and health requirements.
A novel strategy to construct stable fat globules with all major milk fat globule membrane proteins to mimic breast milk fat emulsions at the protein level

Abstract:Milk fat globule membrane (MFGM) proteins have several biological functions and maintain the fat globule structure. However, the major MFGM protein compositions in simulated human milk emulsions are different from those in human milk due to the composition loss in the isolation process of MFGM materials. To overcome this limitation, we developed a novel strategy, namely, the solution enriched with MFGM was homogenized with cream separated from the milk rich in large-sized fat globules. The results of physicochemical properties and the interfacial protein coverage of the emulsions showed that the emulsions prepared by the new method had a smaller particle size, higher stability, and more interfacial protein coverage when the ratio of fat to protein was 1:3. In addition, proteome differences in interfacial proteins between the new emulsions and simulated infant formula emulsions were investigated, and the results revealed that the interface of the emulsions prepared by the new method contained all major MFGM proteins and unique GO annotations and KEGG pathways. However, only four MFGM proteins (XO, ADPH, PAS 6/7) were quantified at the interface of the emulsions prepared by the common method. Furthermore, the protein number and the total relative abundance of major MFGM proteins were approximately 2-fold and 475-fold higher at the interface of the emulsions prepared by the new method compared to the common method. Overall, the study modulated the interfacial protein composition of fat globules by screening the sources of lipid and homogenization methods and revealed its potential effect on processing stability and biological properties.
Effects of Various Thermal Treatments on Interfacial Composition and Physical Properties of Bovine Milk Fat Globules.

Abstract:This study aimed to investigate changes of milk fat globules (MFG) and their membranes after thermal treat- ments, and further analyzed the relationship between the stability of MFG and interfacial compositions of milk fat globule membrane (MFGM). We characterized the influence of three kinds of thermal treatments on fat globule interfacial components (including interfacial phospholipids and interfacial protein) and physical prop- erties using phospholipidomics and several microscopy techniques. The results showed that size of MFG increased from 2.96 mu m to 3.59 mu m and zeta-potential decreased from-9.71 mV to-13.23 mV after thermal treatment, suggesting that MFGM was damaged and MFG occurred coalescence. Thermal treatment increased the Young's modulus of MFGM and made membranes more fragile. The abundance of MFGM proteins decreased while casein and beta-lactoglobulin increased after thermal treatment. Results of phospholipidomics showed that 27 phospholipid species could be used to distinguish the samples. Pasteurization reduced mainly SM and PC located in the outer bilayer of MFGM, while ultra-pasteurization reduced not only SM and PC but also PI and PE located in the inner leaflet. Based on correlation analysis, the increase in Young's modulus of MFGM during thermal treatment might be related to changes in chemical components on the membrane, suggesting a potential link between the change of MFGM components and fat globule coalescence behavior.

Abstract:This work aimed to investigate the properties of emulsions prepared with milk fat globule membrane (MFGM) and its components, which are milk fat globule membrane proteins (MFGMPs) and milk fat globule membrane lipids (MFGMLs). The results demonstrated that the particle size of emulsions prepared with MFGM and MFGMPs decreased with increasing concentrations (from 1% to 4%); however, these droplet sizes were smaller than those prepared with MFGMLs of the same concentration. In addition, the emulsion prepared with MFGMLs showed a clear phase separation, as opposed to those from MFGM and MFGMPs. These results indicated that MFGMLs alone could not contribute to the stability of the emulsions. The microscopic images also confirmed that the droplet sizes of emulsions prepared with MFGM and MFGMPs were smaller than those prepared with MFGMLs. Moreover, the emulsions prepared with MFGM and MFGMPs had lower shear stress than those prepared with MFGMLs. The MFGM and MFGMPs were found to be sensitive to pH; thus, their physical stabilities behaved according to their pH. The heat treatment results showed that the emulsions prepared with MFGM were stable over the heat of temperature ranging from 35 degrees C to 85 degrees C, whereas those prepared with MFGMPs were stable to heat of temperature ranging from 35 degrees C to 65 degrees C and became unstable at the temperature of over 65 degrees C.
Structural and physical-chemical properties of milk fat globules fractionated by a series of silicon carbide membranes

Abstract:Driven by the acknowledged health and functional properties of milk fat globules (MFGs), there is a growing interest to develop gentle methodologies for separation of fat from milk. In this study, separation of fat from raw milk and fractionation in streams containing MFGs of different size was achieved using a series of two silicon carbide ceramic membranes. A first step consisting of a 1.4 µm membrane aimed to concentrate the bulk of the fat, i.e. the larger MFGs (D[4,3] ∼ 4 µm) followed by a 0.5 µm fractionation aimed to concentrate the residual milk fat in the permeate, i.e. fraction with the smaller MFGs (D[4,3] ∼ 1.8-2.4 µm. The fat separation performance showed a yield of 92 % for the 1.4 µm membrane and 97 % for the 0.5 µm membrane. Both fat enriched retentates showed, by the confocal laser scanning microscopy, intact MFGs with limited damage in the MFG membrane. The fatty acid profile analysis and SAXS showed minor differences in fat acid composition and the crystallization behavior was related to differences in the fat content. The 0.5 µm permeate containing the smallest MFGs however showed larger aggregates and a trinomial particle size distribution, due to probably pore pressure induced coalescences. The series of silicon carbide membranes showed potential to concentrate some of MFGM proteins such as Periodic Schiff base 3/4 and cluster of differentiation 36 especially in the 0.5 µm retentates. A shift in casein to whey protein ratio from 80:20 (milk) to 50:50 was obtained in the final 0.5 µm permeate, which opens new opportunities for product development.
