Abstract:A novel cDNA of phospholipid hydroperoxide glutathione peroxidase (PHGPx), which encodes a functional protein capable of complementing the yeast PHGHX-deletion mutant, was recently discovered in radish (Raphanus sativus) and designated as RsPHGPx [Yang X-D, Li W-J, Liu J-Y (2005) Biochim Biophys Acta 1728:199–205]. Sequence alignment suggested that RsPHGPx contains a targeting peptide required for transport to mitochondria, but the experimental evidence for the exact intracellular distribution of RsPHGPx remains to be elucidated. To uncover the cellular localization of plant PHGPx, we first investigated RsPHGPx’s intracellular distribution. Western blot analysis of subcellular fractions using the RsPHGPx antiserum clearly indicated the distribution of RsPHGPx in the radish mitochondrial fraction. Furthermore, a construct expressing the RsPHGPx precursor tagged with green fluorescent protein was introduced into tobacco and yeast cells, and the fusion protein was transported into both mitochondria, indicating that RsPHGPx was indeed localized in mitochondria. To explore the biochemical functions of this enzyme, we tested the enzymatic activity of the recombinant RsPHGPx protein. It displayed GSH-dependent peroxidase activity and exhibited the largest affinity to and the highest catalytic efficiency on phosphatidylcholine hydroperoxide, suggesting that phospholipid hydroperoxide is probably the optimum substrate for RsPHGPx. Furthermore, RsPHGPx showed a much higher V max value, by two orders of magnitude, than those of all other known plant PHGPxs. Taken together, these results showed evidence for the first time of mitochondrial localization and higher activity of PHGPx in plants and provided a framework for continued studies on the physiological functions of RsPHGPx.

Abstract:A novel antioxidant peptide EDEQKFWGK from porcine plasma hydrolysate (PPH) was separated by chromatography, HPLC, and identified by LC-MS/MS. Results showed that EDEQKFWGK had better antioxidant ability (Hydroxyl RAS 32.19%, ABTS RAS 92.93% and DPPH RAS 26.76%) compared with glutathione (30.11%, 82.01%, 26.44%) due to the presence of hydrophobic, aromatic acids (F, W) and acidic amino acids (E, D), decreasing ROS by providing hydrogen atom and chelating metal ions. Furthermore, the antioxidant properties of synthetic EDEQKFWGK still significant despite in vitro digestion because of the production of smaller active peptide. Additionally, it could increase SOD, CAT, GSH-Px to resist oxidative damage in HepG2 cells by inhibiting ROS (O2- , OH·), forming complexes to prevent OH· from destroying DNA and binding to ARE to promote antioxidase expression. Thereby, the novel peptide EDEQKFWGK from porcine plasma had much stable antioxidant properties and hade great potential in formulating functional foods. PRACTICAL APPLICATIONS: This research isolated a novel antioxidant peptide. Moreover, the antioxidant effects of peptide were confirmed under the in vitro digestion model and oxidative damage HepG2 cells model. The results showed the antioxidant peptide could play better effect after digestion and protect the cells from oxidative damage. These data could expand the sequence data of antioxidant peptides and promote the high-value utilization of PPH.

Abstract:A full-length cDNA encoding putative phospholipid hydroperoxide glutathione peroxidase (PHGPx) was cloned from Raphanus sativus. The cDNA, designated RsPHGPx, includes an open reading frame which encodes 197 amino acid residues. The alignment of amino acid sequences showed that RsPHGPx had the highest sequence homology to plant PHGPx and contained an N-terminal extension characteristic of a mitoctiondrial targeting peptide. Northerri blot analysis indicated that RsPHGPy was constitutively and ubiquitously expressed during radish development, and its expression was differently regulated by various stress conditions. The expression of RsPHGPx in a yeast PHGPx-deletion mutant significantly rescued the mutant sensitivity to oxidation-sensitive linolenic acid, just as the yeast PHGPx3 gene did. Abstract:Glutathione (Grx) is a glutathione-dependent oxidoreductase belonging to the thioredoxin (Trx) superfamily. It maintains redox homeostasis in organisms and plays a significant role in resisting oxidative stress. Nevertheless, Grx-related sequence information and functional validation are greatly inadequate in crustaceans. In this study, the Grx3 of Penaeus monodon (PmGrx3) was cloned. The full-length cDNA of PmGrx3 was 1546 bp, which consists of 113-bp 5′-UTR, 458-bp 3′-UTR, and 975-bp ORF, encoding a 324 amino acid putative protein. The putative PmGrx3 protein was 36.04 kDa with a theoretical isoelectric point of 4.98. Sequence alignment indicated that PmGrx3 had the highest amino acid sequence homology with the Grx3 of Penaeus vannamei at 96.57% and was clustered with the crustaceans. Quantitative real-time PCR (qRT-PCR) analysis revealed that PmGrx3 was detected in all tissues examined, with the highest expression in the muscle, stomach, and testis, as well as the lowest expression in abdominal nerve. In addition, PmGrx3 was expressed during the developmental period and was the most highly expressed in the zygote. Both ammonia-N stress and bacterial infection caused different degrees of upregulation or inhibition of the PmGrx3 expression in the hepatopancreas and gills. The present study indicates that PmGrx3 may be a key oxidoreductase in the defense system of shrimp against environmental stress and pathogen infection.

Abstract:Overexpression of phospholipid hydroperoxide glutathione peroxidase (PHGPx) genes has been reported to play an important role in protecting host cells from oxidative injury in several model systems. A radish phospholipid hydroperoxide glutathione peroxidase (RsPHGPx) known to have high catalytic activity was applied to mouse 3T3 fibroblasts to determine the protective effects of PHGPx against oxidative injury triggered by hydroperoxides such as hydrogen peroxide (H(2)O(2)), tert-butyl hydroperoxide (t-BHP) and phosphatidylcholine hydroperoxide (PCOOH). We observed that preincubation of cells with RsPHGPx significantly increased cell viability, reduced levels of malondialdehyde (MDA), inhibited generation of reactive oxygen species (ROS), and maintained natural cell shapes after treatment with H(2)O(2), t-BHP or PCOOH, indicating that the exogenous RsPHGPx can act as an effective hydroperoxide-scavenger and may also protect target cells from oxidative damage. These results suggest the possibility for use of RsPHGPx as a therapeutic protectant.
Identification of a New Antioxidant Peptide from Porcine Plasma by in Vitro Digestion and Its Cytoprotective Effect on H2O2 Induced HepG2 Model

Abstract:Antioxidant peptide, referred as Trp-Gly-Pro-Gly-Val-Glu (WGPGVE), was isolated and identified from porcine plasma which was hydrolysised 5 h by alkaline protease. The sequence of WGPGVE had hydrophobic amino acids G, V, aromatic amino acids W, and acidic amino acid E, as well as its values of hydroxyl, ABTS, DPPH radicals scavenging rates and iron chelating rate were improved by 30.15%, 10.05%, 38.69%, 51.40% respectively, compared to glutathione (29.32%, 81.06%, 25.43%, 17.82%). Moreover, under in vitro digestion model, it exhibited stable antioxidation (50.15%, 92.49%, 24.29%, 35.35%) after the hydrolysis effect of pepsin and trypsin. Additionally, WGPGVE could protect HepG2 cells against H2O2 by promoting the expression of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) and decreasing reactive oxygen species (ROS), malondialdehyde (MDA) contents. Thus, the new sequence antioxidant peptide could be potentially used in pharmaceuticals or functional foods and promote the application of porcine plasma in food.

Abstract:A novel radish RsPHGPx cDNA, which encodes a functional phospholipid hydroperoxide glutathione peroxidase (PHGPx) protein, was identified in the previous work. In the study genomic organization and the upstream regulatory sequence analysis of this gene was presented. Southern blot analysis showed that RsPHGPx gene existed in radish genome in manner of single copy. Moreover, a 3.3 kb genomic DNA fragment of RsPHGPx gene was isolated by combination of common PCR and genome-walking method. Sequence analysis on this genomic fragment demonstrated that RsPHGPx gene consists of seven exons separated by six introns, and suggested that a short 5'-flanking sequence immediately before the exon I should be the putative RsPHGPx promoter region, which is proceeded by the upstream neighboring biotin synthase gene. Cis-acting elements search showed that the putative promoter contains elements responsive to hormones (eg. E-Box and W-Box), abiotic stresses (eg. MYB and MYC binding sites), and light (Box H and I-Box), etc. Northern blot analysis indicated that the expression of RsPHGPx was subjected to up-regulation of chilling and down-regulation of ABA and successive illumination (in etiolated seedlings), implying the regulatory roles of some predicted elements. However the up-regulation effect of herbicide paraquat, which can induce oxidative stress, suggested the presence of some unknown elements in the promoter region. This is the first report on gene structure and upstream regulatory sequence analysis in reported plant PHGPx genes, which will be a prerequisite to understand regulatory mechanism of PHGPx gene expression in plants.

Abstract:The radish phospholipid hydroperoxide glutathione peroxidase (RsPHGPx) fusion proteins expressed in Escherichia coli with high efficiency were purified and the antioxidant effects of RsPHGPx together with glutathione on hydroperoxide-mediated injury in mouse NIH3T3 fibroblasts were investigated.The cell viability measurement confirmed that 10μg/ml RsPHGPx,when added to the fibroblasts separately,can only slightly increase the cell viability.However,when combained with glutathione,the antioxidant effects increase dramatically.MDA assay suggests that RsPHGPx can scaveng the lipid peroxides,the results are accordance with the MTT results.ROS flourescence investigations and DAPI experiments show that RsPHGPx can increase the protection effect via inhibiting ROS genreation and this protection help maintain the normal nuclear and membrane conditions of NIH3T3 fibroblasts.The combain experiment of RsPHGPx and glutathione provides experimental evidences for the future production of RsPHGPx.

Abstract:Phospholipid hydroperoxide glutathione peroxidase (PHGPx) is an unique antioxidant enzyme that directly reduces lipid hydroperoxides in biomembranes. In the present work, the entire encoding region for Oryza sativa PHGPx was expressed in Escherichia coli M15, and the purified fusion protein showed a single band with 21.0 kD and pI = 8.5 on SDS- and IFE-PAGE, respectively. Judging from CD and fluorescence spectroscopy, this protein is considered to have a well-ordered structure with 12.2% alpha-helix, 30.7% beta-sheet, 18.5% gamma-turn, and 38.5% random coil. The optimum pH and temperature of the enzyme activity were pH 9.3 and 27 degrees C. The enzyme exhibited the highest affinity and catalytical efficiency to phospholipid hydroperoxide employing GSH or Trx as electron donor. Moreover, the protein displayed higher GSH-dependent activity towards t-Butyl-OOH and H(2)O(2). These results show that OsPHGPx is an enzyme with broad specificity for hydroperoxide substrates and yielded significant insight into the physicochemical properties and the dynamics of OsPHGPx.
Micropreparation of a native PHGPx protein from radish seedlings by immunoaffinity chromatography

Abstract:Radish phospholipid hydroperoxide glutathione peroxidase (RsPHGPx) Was identified as a mitochondrion-targeting PHGPx in previous work. To determine its cleavage site of the targeting peptide, the immunoaffinity chromatography (IAC) purification approach was carried out to isolate the native RsPHGPx protein. Polyclonal antibodies directed against recombinant RsPHGPx were raised in rabbit. Monospecific anti-RsPHGPx antibodies were isolated by means of affinity chromatography using the recombinant RsPHGPx as affinity ligand, and employed in assembling an IAC column. A single-step, highly specific and easy-to-use protocol was developed for purification of the active RsPHGPx protein through the assembled IAC column. Using this approach, a specific protein of the expected molecular size was obtained from the mitochondrial fraction of radish seedlings. Western blot analysis showed that it could be specifically recognized by anfi-RsPHGPx antibodies, and an enzyme activity assay indicated that it exhibited significant PHGPx activity, suggesting that the purified protein should be the desired native RsPHGPx. These results will lead to clarification of the targeting peptide and the active mature protein of RsPHGPx and will be helpful to further probe the intracellular localization mechanism and biological function of this plant PHGPx.

Abstract:The entire encoding region for Momordica charantia phospholipid hydroperoxide glutathione peroxidase (McPHGPx) was cloned into pET-28a(+) vector and expressed in Escherichia coli BL21(DE3). The purified recombinant McPHGPx displayed GSH-dependent peroxidase activity towards phospholipid hydroperoxide, H 2 O 2 , and tert -butyl hydroperoxide and had the highest affinity with and catalytic efficiency for phospholipid hydroperoxide. The optimum temperature of the enzyme activity ranged from 40 to 50°C, thus it is a thermostable enzyme compared to other PHGPx enzymes. Furthermore, McPHGPx expression in Saccharomyces cerevisiae PHGPx-deletion mutant rescued the susceptibilities to the oxidation-sensitive polyunsaturated fatty acid (linolenic acid), indicating its PHGPx complementation function in yeast. These results have well documented that McPHGPx functions as a PHGPx in vitro and in vivo and will be beneficial for further functional studies on plant PHGPx enzymes.

Abstract:The distribution of plant phospholipid hydroperoxide glutathione peroxidase (PHGPx) genes was investigated by screening conducted using the PCR and southern blotting methods in ten important plant species. The results demonstrated the presence of PCR fragments and southern hybridization positive bands produced from the DNA samples of radish, wrinkled giant hyssop, aloe, tomato, bitter gourd, wheat, rape and garlic suggesting that the PHGPx gene may exist in each of these eight plant species with 1 or 2 copies in their genomes. These results provided basic information for cloning these genes.

Abstract:A cDNA encoding putative phospholipid hydroperoxide glutathione peroxidase (PHGPX) was isolated from rice using rapid amplification of cDNA ends. This cDNA, designated ricPHGPX, includes an open reading frame encoding a protein of 169 amino acids which shares about 60% and 50% amino acid sequence identity with plant and mammalian PHGPXs, respectively. The gene is expressed at a relative high level in flag leaves and the expression can be markedly induced by oxidative stress, suggesting that the product of the gene plays a key role in defense against oxidative damage in rice.
Optimized Expression and Identification of the Radish Phospholipid Hydroperoxide Glutathione Peroxidase in Pichia Pastoris

Abstract:The expression of RsPHGPx gene in Pichia pastoris was investigated.The RsPHGPx gene inserted into secretory vector pPIC9K was transformed into Pichia pastoris strain GS115.The singlecopy recombinant strains were screened by G418.In addition,the induction conditions were optimized to get the highest expression of the target protein (the optimum:1%methonal,pH6.0,28℃).Finally,it was shown that the recombinant RsPHGPx could be secreted into the culture supernatant to a level of 102mg/L after 60 hours of induction.The fractional ammonium sulfate precipitation,desalination,and gel chromatography were used to purify protein and more than 90% purity of RsPHGPx was obtained.The bioactivity of RsPHGPx was high-dependent redox-active of GSH and reached its max secreted volume at 60h,the specific activity is 4.2μmol/min·mg.The research has laid the foundation for gaining and exploiting a large amount of RsPHGPx.

Abstract:An N-terminal 32 amino acids radish PHGPx gene(Δ32RsPHGPx) was cloned into expression vector pGEX-6P-1 and transformed into E.coli strain BL21(DE3).Δ32RsPHGPx for crystallization was prepared via Glutathione SepharoseTM affinity,cation-exchange and gel filtration chromatographies.The concentration of it was 10 g/L and the purity was over 95%.Δ32RsPHGPx showed obvious PHGPx activity towards lipid hydroperoxides.In addition,a tertiary structure model of Δ32RsPHGPx displayed the thioredoxin fold.
Expression, purification and crystal growing conditions of recombinant oryza sativa PHGPx

Abstract:Phospholipid hydroperoxide glutathione peroxidase (PHGPx) is a unique antioxidant enzyme that directly reduces lipid hydroperoxides in biomembranes. It plays potential important roles in oxidative stress response. Oryza sativaPHGPx gene was cloned into expression vector pGEX-6P-1 and transformed into E. coli strain BL21 (DE3). OsPHGPx for crystals was prepared with employing Glutathione Sepharose (TM) affinity, cation-exchange and gel filtration chromatography. The purity of the purified OsPHGPx was over 95%. OsPHGPx showed an obvious PHGPx activity towards lipid hydroperoxides. MALDI-TOF analysis shows that the exact molecular weight of OsPHGPx was 19275. 568, which was in accordance with the theoretical molecular weight. The microcrystals of OsPHGPx were obtained under several conditions. In addition, a tertiary structure model of the OsPHGPx generated from http://swissmodel. expasv. org/ displayed the thioredoxin fold.

Abstract:The radish phospholipid hydroperoxide glutathione peroxidase (RsPHGPx) and its mutant (mRsPHGPx) fusion proteins expressed in Escherichia coli BL21 (DE3) with high efficiency were purified with Glutathione Sepharose 4B affinity chromatography and cleaved on-column with PreScission protease, and both the recombinant proteins without tags were obtained at a purity more than 90%. The enzymatic measurement confirmed that the activity of mRsPHGPx was far less than that of RsPHGPx. Effects of RsPHGPx and mRsPHGPx on restoration of B16 melanoma cells irradiated with uhroviolet were investigated. The results showed that the active RsPHGPx could significantly increase the cell viability and reduce the lipid peroxidation of cell membranes, while mRsPHGPx with very low activity had no effects, suggesting RsPHGPx could inhibit the ultroviolet-mediated injury in cell membranes mainly by rapidly scavenging various lipid peroxides.
Glutathione peroxidase gene BnGPX1 cloning from rapeseed and expression under abiotic stress

Abstract:A glutathione peroxidases gene,designated as BnGPX1(GenBank assession number HM130680),was a key enzyme protecting plants against oxidative damage generated by reactive oxygen species(ROS) under abiotic stresses.It was cloned from rapeseed cultivar Ningyou 16(Brassica napus L.) by RT-PCR(reverse transcription PCR).The open reading frame(ORF) of BnGPX1 was 711 bp,encoding 236 amino acids with isoelectric point(pI) of 9.37 and molecular mass of 26.10 kD.BnGPX1 had 3 characteristic domains of plant GPXs and 3 conserved cysteine residues.Its genomic DNA(gDNA) fragment(GenBank assession number HM130681) was isolated by PCR.The sequence of gDNA demonstrated 6 exons separated by 5 introns.All introns were spliced following the consensus sequence with GT and AG at 5' and 3' ends respectively.BnGPX1 construction was similar to the Arabidopsis AtGPX1 gene.Expression patterns of BnGPX1 in rapeseed tissues under different abiotic stresses were obtained by semi-quantitative RT-PCR.Results showed that BnGPX1 was constitutively and ubiquitously expressed at high levels in stems,leaves,silliques and flowers,but at lower level in roots.The expression was increased under salt,drought and high temperature condition.It did not respond to cold stress.

Abstract:More than 20 sequences of phospholipid hydroperoxide glutathione peroxidase (PHGPX) from a sequence database were analyzed. The analyses show that the primary structures of most PHGPX proteins have three highly conserved regions forming a catalytic center and have more than 50% amino acid sequence identity in common. However, two PHGPXs from bovine and swine with the same function have very low similarity with typical PHGPXs and do not have the three highly conserved regions. Thus, the PHGPX proteins are divided into two types: those with the three highly conserved regions, designated as PHGPX-I, and the others as PHGPX-II. In general, type I proteins are composed of ca.170 amino acid residues; a few of them have an extra signal peptide sequence at the N-terminal of the protein. The composition of plant and animal PHGPX amino acids is very different, with most plant PHGPXs being weak acidic, while most animal ones are alkaline. Another specific conservative motif is also found in plant PHGPX proteins. System evolution analysis shows that ortholog and paralog evolution models both exist in PHGPXs, with the plant PHGPX and the animal PHGPX diverging exclusively into two branches in PHGPX-I. The information revealed by the evolution tree agrees with the general species evolution process from low to advanced and from simple to complicated.
Selenium-containing 15-Mer Peptides with High Glutathione Peroxidase-like Activity

Abstract:Glutathione peroxidase (GPX) is one of the most crucial antioxidant enzymes in a variety of organisms. Here we described a new strategy for generating a novel GPX mimic by combination of a phage-displayed random 15-mer peptide library followed by computer-aided rational design and chemical mutation. The novel GPX mimic is a homodimer consisting of a 15-mer selenopeptide with an appropriate catalytic center, a specific binding site for substrates, and high catalytic efficiency. Its steady state kinetics was also studied, and the values of k(cat)/K(m)(GSH) and k(cat)/ K(mH(2)O(2)) were found to be similar to that of native GPX and the highest among the existing GPX mimics. Moreover, the novel GPX mimic was confirmed to have a strong antioxidant ability to inhibit lipid peroxidation by measuring the content of malondialdehyde, cell viability, and lactate dehydrogenase activity. Importantly, the novel GPX mimic can penetrate into the cell membrane because of its small molecular size. These characteristics endue the novel mimic with potential perspective for pharmaceutical applications.
