Abstract:Hexafluoroisopropanol (HFIP) has been extensively applied in organic synthesis as a versatile reaction media. However, the application of HFIP as an catalyst in organic synthesis has been rarely studied. Presented here, is the example of HFIP-catalyzed Mannich-type reaction between quinoxalin-2(1H)-ones and difluoroenoxysilanes. The developed protocol tolerates various substituents under mild reaction conditions, providing a divergent approach to the construction of difluoro-substituted 3,4-dihydroquinoxalin-2(1H)-ones and monofluoroquinoxalin-2(1H)-ones in 51-92% yields.

Abstract:The three-component reaction between an isoquinoline, a dialkyl acetylenedicarboxylate and 2,2,2-trifluoro-1-phenylethanone involving a 1,4-dipolar cycloaddition cascade approach is developed using a continuous-flow microreactor. The reaction conditions (solvent, flow rate and temperature) are optimized leading to the synthesis 2-(trifluoromethyl)-2H-[1,3]oxazino[2,3-a]isoquinoline derivatives in 79-83% isolated yields, whilst eliminating the formation of 11bH-pyrido[2,1-a]isoquinoline-12,3,4-tetracarboxylate by-products. The continuous-flow method is also applied to similar reactions using aromatic aldehydes as the carbonyl component.

Abstract:Iodine was used to catalyze the hetero-Diels-Aider reaction of pentafluorobenzylidineaniline (C6F5CH=NAr 1) with enol ethers to afford the corresponding tetrahydroquinolines derivatives as a mixture of cis/trans stereoisomers in moderate yields. These products could also be prepared by onepot, three-component reaction of pentafluorophenylaldehyde, anilines, and enol ethers under the same reaction condition. Mild and neutral reaction conditions, facile experimental procedure, and low price of iodine should make this method attractive for practical synthesis of many fluorinated tetrahydroquinoline derivatives. Abstract:A highly regioselective intramolecular oxy-fluorination of alkenyl oximes was achieved. This new transformation represents an efficient method for the preparation of monofluoromethyl-substituted isoxazolines. The synthetic application of the oxy-fluorination product was demonstrated by a one-step synthesis of monofluoromethyl-substituted β-hydroxyl ketone derivatives.

Abstract:Infrared irradiation promoted the synthesis of substituted 5-oxo-1, 2, 3, 4, 5, 6, 7, 8-octahydroquinoline derivatives. The condensations of 5, 5-dimethyl-1, 3-cyclohexane-dione, 1, 3-diaryl-2-propen-1-one and ammonium acetate in the absence of solvent under infrared irradiation for 3 h gave the title compounds in 67.6%∼86.5% yields.

Abstract:Herein, we report the development of a FSO 2 radical initiated tandem cyclization reaction under visible light photoredox catalysis, which allows for a facile access to FSO 2 -functionalized oxindoles from N -arylacrylamides. This reaction features mild reaction conditions, readily available starting materials, and SuFExable products. The introduction of sulfonyl fluoride groups into oxindoles could be of interest for related study in the context of discovering new bioactive molecules. Abstract:An electrochemical cascade radical cyclization of 2‐aryl‐N‐acryloyl indoles with sodium trifluoromethanesulfinate as a coupling partner has been explored, which afforded the unexpected bis‐trifluoromethylated indole[2,1‐a]isoquinolines as products. Experimental results and DFT calculations disclose that this reaction involves trifluoromethylation‐triggered cyclization and the second trifluoromethylation as the key steps. This strategy does not need any transition‐metal catalysts or oxidants with a readily available trifluoromethylating reagent enabling facile synthesis of bis‐trifluoromethylated indole‐fused tricycles.

Abstract:A metal-free electrochemical oxidative difluoroethylation of 2-arylbenzimidazoles was accomplished, which provided an efficient strategy for the synthesis of MeCF2-containing benzo[4,5]imidazo[2,1-a]-isoquinolin-6(5H)-ones. In addition, the method also enabled the efficient construction of various difluoroethylated indolo[2,1-a]isoquinolin-6(5H)-ones. Notably, this electrochemical synthesis protocol proceeded well under mild conditions without metal catalysts or exogenous additives/oxidants added.

Abstract:A decarboxylative/dehydrofluorinative formal [3 + 2] cycloaddition aromatization of isoquinolinium N-ylides with difluoroenoxysilanes has been developed. This methodology provides a facile and straightforward synthetic pathway to afford highly functionalized fluorinated pyrrolo[2,1-a]isoquinolines in good to excellent yields under mild conditions. Moreover, gram-scale and synthetic derivatization experiments for the late-stage functionalization of drug molecules have also been demonstrated.

Abstract:AbstractThe interest in indole dearomatization, which serves as a useful tool in the total synthesis of related alkaloid natural products, has recently been renewed with the intention of developing new methods efficient in both yield and stereoselective control. Here, we report an enzymatic approach for the oxidative dearomatization of indoles in the asymmetric synthesis of a variety of furoindolines with a vicinal quaternary carbon stereogenic center. This approach depends on the activity of a flavin‐dependent monooxygenase, TsrE, which is involved in the biosynthesis of bicyclic thiopeptide antibiotic thiostrepton. TsrE catalyzes 2,3‐epoxidation and subsequent epoxide opening in a highly enantioselective manner during the conversion of 2‐methyl‐indole‐3‐acetic acid or 2‐methyl‐tryptophol to furoindoline, with up to >99 % conversion and >99 % ee under mild reaction conditions. Complementing current chemical methods for oxidative indole dearomatization, the TsrE activity‐based approach enriches the toolbox in the asymmetric synthesis of products possessing a furoindoline skeleton.
