Abstract:Density functional theory calculations and experimental studies have been carried out on the intramolecular Pauson−Khand-Type reaction mediated by a PdCl2-thiourea catalyst, which proceeds under mild reaction conditions and provides a useful alternative to traditional Pauson−Khand reactions. The classical mechanism of the Pauson−Khand reaction involving the alkyne/alkene C−C bond formation as the key step has been found to be energetically unfavorable and is not in line with the experimental observations. A novel reaction mechanism has been proposed for the reaction. The first step involves the cis-halometalation of the alkyne, followed by sequential alkene and carbonyl insertion. The rate-determining fourth step is an intramolecular C−Cl oxidative addition, leading to a PdIV intermediate. A C−C bond formation by reductive elimination completes the reaction. The mechanism is in agreement with the key experimental observations including (1) the need of a chloride for catalytic activity and the absence of catalysis with Pd(OAc)2 alone; (2) the rate acceleration by the addition of LiCl; both with PdCl2 and Pd(OAc)2 catalysts; and (3) the preferred formation of the trans diastereomer in substituted cases. The cis halometalation and the formation and stability of the PdIV intermediate is studied in detail and provides general insights into these novel steps.

Abstract:A series of novel carbohydrate-based iminophosphinite ligands have been synthesized for the first time and successfully applied in Pd-catalyzed asymmetric allylic alkylation. The substituent effects on the catalytic reaction were also investigated, and 92% ee was achieved when the p-nitro-substituted ligand 10d was used.

Abstract:A novel C-2 symmetrical and sterically bulky thiourea ligand 1 has been successfully applied to Heck, Suzuki and Suzuki-type carbonylative coupling reactions under aerobic conditions. Since the metal-sulfur bond in the thiourea complexes is stronger than the metal-phosphorus bond of typical phosphine complexes, thiourea ligands generally do not easily dissociate from the metal center under catalytic conditions, which establishes the thiourea 1-based palladium complexes as effective catalysts for the palladium-catalyzed cross-coupling reactions.

Abstract:Which way? Palladium thiourea catalyzed Pauson–Khand reaction of norbornene with substituted alkynoates to afford selectively either α-carboxylated adducts or β-carboxylated adducts with the reverse regioselectivity has been demonstrated for the first time. Abstract:C-H activation and functionalization are on the forefront of modern synthetic chemistry. Imagine if any C-H bond of a molecule could be converted to a C-X bond, where X is the target functionality-this would alter the synthetic blueprints for complex target molecules since it would provide novel disconnections in retrosynthetic analysis. Collaborations between many experimental and computational groups have led to rapid developments of new C-H functionalization methods. Our groups represent an example of this; we were brought together as part of the NSF-supported Center for Selective C-H Functionalization. Many examples of experimental-computational synergy for selective Pd(II)-catalyzed C-H activation of aryl and alkyl groups are described in this Account. We describe computations by the Houk group made in response to experimental stimuli by the Yu group. The first section discusses the experimental and computational investigations of oxazoline-directed stereoselective Pd(II)-catalyzed C(sp3)-H bond activation that occurs through the concerted metalation-deprotonation (CMD) pathway involving a monomeric Pd(II) complex. The second section involves two types of bidentate ligands, mono-N-protected amino acid (MPAA) and acetyl-protected aminoethyl quinoline (APAQ) ligands that promote the C-H activation reactions with the ligand as the internal base. In the MPAA-assisted remote C-H bond activation, the basic dianionic amidate ligand participates in the deprotonation of a specific C-H bond. This mechanism accounts for the improved reactivity and selectivity in C-H activation reactions with MPAA ligands. The chiral APAQ ligands enable asymmetric palladium insertion into prochiral C-H bonds on a single methylene carbon center. The dianionic amidate of the APAQ ligand acts as an intramolecular base to deprotonate the methylene C-H asymmetrically and facilitate chiral Pd-C bond formation. The origins of the dramatic differences of five-membered (relatively inactive) and six-membered (highly active) chelation in β-methylene C(sp3)-H activation reactions by a Pd(II) catalyst were explained with density functional theory (DFT) calculations. This is mainly due to the steric repulsions between the ArF group of the substrate and the quinoline group of the ligand. The steric repulsion between the ArF group of the substrate and the quinoline group of the APAQ ligand destabilizes the five-membered chelate transition structure, increasing the energy of the transition state. The third section discusses a mechanism involving a Pd-Ag heterodimeric complex intermediate in the template-directed, Pd(II)-catalyzed remote meta functionalization of toluene derivatives and benzoic acid derivatives. The nitrile directing group of the template coordinates with Ag while the Pd is placed adjacent to the meta-C-H bond in the transition state, leading to the observed high meta selectivity. The selective activation of remote meta-C-H bonds at various distances can be achieved by tuning the template. The dual role of AgOAc as both an oxidant and part of the heteronuclear active species in the mechanism involving PdAg(OAc)3 was determined by DFT calculations and is in accord with literature information about complexes. For the systems discussed in these three sections, the similarity is that they all proceed via the CMD mechanism. The differences lie in the proton acceptors and the active Pd species. Common CMD involves a monomeric Pd mechanism with acetate as the proton acceptor. Both MPAA and APAQ ligands react via monomeric Pd mechanisms with a ligand moiety (the amidate oxygen) as the proton acceptor. Nitrile-containing template-mediated meta-C-H activations proceed via a Pd-Ag heterodimeric mechanism, still with acetate as the proton acceptor. The interaction between our two groups, experts in experiment and computation, and the discoveries made possible by that interplay are highlighted in this Account.

Abstract:[GRAPHICS]A novel thiourea-based C-2-symmetric ligand was synthesized, and its application in the palladium-catalyzed Heck and Suzuki coupling reactions of arenediazonium salts was evaluated, The reactions, which were performed at room temperature, without added base, and under aerobic conditions, produced product in 4 h with good yield. Abstract:A series of six disubstituted diacylthioureas 1,3‐ and 1,4‐C6H4[C(O)NHC(S)NR1R2]2 (L1‐6) were synthesized with varied Rn substituents (R1 = R2 = Et for L1‐2; R1 = R2 = Bn for L3‐4; R1 = iPr and R2 = Ph for L5 (1) and L6 (2)). Treatment of Ln with Cu(I) halide precursors CuX(PPh3)3 (X = Cl, Br, I) produced the discrete binuclear adducts Ln[CuX(PPh3)2]2 (3‐11 and 14‐16; n = 1‐3 and 6) by binding to Cu(I) centres via the monodentate‐S mode. In contrast, L4 and L5 yielded only the chloride products of binuclear L4[CuCl(PPh3)2]2 (12) and mononuclear L5CuCl(PPh3)2 (13), respectively, with one S arm in the latter remaining dangling, while bromide or iodide analogues were not available for L4 and L5, possibly due to the steric hindrance imposed by larger halide anions or bulky isopropyl substituents. The reactions of Ln with nitrate [Cu(NO3)(PPh3)2]led to the double‐deprotonation of ligand protons to generate dianions Ln' (1,3‐/1,4‐C6H4[C(O)NC(S)NR1R2]22‐) and led to the consequent formation of binuclear diacylthioureato Cu(I) complexes Ln'[Cu(PPh3)2]2 (n=1 (17), 2 (18), 4 (19)) via the κ‐O,S‐bidentate mode. The obtained ligands and complexes were spectroscopically and structurally characterized. These Cu(I) products (3‐19) were experimentally used as catalysts for the oxidation of 1‐phenylethanol.

Abstract:We report herein the synthesis of novel chiral S,N-heterobidentate thiourea-oxazoline ligands and their application to palladium-catalyzed enantioselective bis(methoxycarbonylation)s of terminal olefins under mild conditions. Copper salts were found to play multiple roles in this reaction. Substituted 2-phenylsuccinates were obtained in >90% yield and up to 84% cc under optimized conditions.
Mechanistic Origin of Ligand-Controlled Regioselectivity in Pd-catalyzed C-H Activation/arylation of Thiophenes.

Abstract:The use of ligands to control regioselectivity in transition-metal-catalyzed C-H activation/functionalization is a highly desirable but challenging task. Recently, Itami et al. reported an important finding relating to Pd-catalyzed ligand-controlled α/β-selective C-H arylation of thiophenes. Specifically, the use of the 2,2'-bipyridyl ligand resulted in α-arylation, whereas the use of the bulky fluorinated phosphine ligand P[OCH(CF(3))(2)](3) resulted in β-arylation. Understanding of this surprising ligand-controlled α/β-selectivity could provide important insights into the development of more efficient catalyst systems for selective C-H arylation, and so we carried out a detailed computational study on the problem with use of density functional theory methods. Three mechanistic possibilities--S(E)Ar and migration, metalation/deprotonation, and Heck-type arylation mechanisms--were examined. The results showed that the S(E)Ar and migration mechanism might not be plausible, because the key Wheland intermediates could not be obtained. On the other hand, our study indicated that the metalation/deprotonation and Heck-type arylation mechanisms were both involved in Itami's reactions. In the metalation/deprotonation pathway the α-selective product (C5-product) was preferred, whereas in the Heck-type arylation mechanism the β-selective product (C4-product) was favored. The ligands played crucial roles in tuning the relative barriers of the two different pathways. In the 2,2'-bipyridyl-assisted system, the metalation/deprotonation pathway was energetically advantageous, leading to α-selectivity. In the P[OCH(CF(3))(2)](3)-assisted system, on the other hand, the Heck-type arylation mechanism was kinetically favored, leading to β-selectivity. An interesting finding was that P[OCH(CF(3))(2)](3) could produce a C-H···O hydrogen bond in the catalyst system, which was crucial for stabilization of the Heck-type transition state. In comparison, this C-H···O hydrogen bond was absent with the other phosphine ligands [i.e., P(OMe)(3), PPh(3), PCy(3)] and these phosphine ligands therefore favored the metalation/deprotonation pathway leading to α-selectivity. Furthermore, in this study we have provided theoretical evidence showing that the Heck-type arylation reaction could proceed through an anti-β-hydride elimination process.

Abstract:In this work, we present an effective strategy to enhance the reactivity and durability of molecular organometallic catalysts by constructing into porous frameworks, as demonstrated by triphenylphosphine (PPh 3 ). Such PPh 3 moieties accessible via the porous structures could be partially metalated by Pd species to generate a highly efficient and recyclable heterogeneous catalyst for Suzuki coupling of aryl chlorides. Site isolation in the rigid framework stabilizes the catalytically active monophosphine‐ligated complex against deactivation into less active assemblies of bisphosphine‐Pd. Meanwhile, the densely populated free ligands inhibit the catalytically active sites from decomposition, thereby highlighting the beneficial effects of such a platform. The tunability of porous polymer synthesis thus gives great promise to impart numerous organometallic catalysts constituted by readily available ligands with unique reactivity, which are not trivially achievable with traditional systems.

Abstract:Pd-Catalyzed ligand-directed C-H functionalization has become one of the most important synthetic methods in organic chemistry. A theorecical study with the B3PW91 density functional theory method is carried out on Pd-catalyzed ligand-directed C-H acetoxylation. The thermodynamic properties in substrates binding step and C-H activation step are calculated. The data indicate that the selectivity of competition reactions between substrates with different directed group is determined by the substrates binding step. A larger complex stability constant will facilitate the formation of the corresponding product. However, the C-H activation step is unrelated to the reaction selectivity. Furthermore, no correlation is found between the reaction selectivities and the gas-phase basicities, Mulliken charges or HOMO energies of the directed groups.
