Abstract:A new series of dimers of de(N-methyl) erythromycin derivatives, joined by a -COCH2- or -CH2CH2- linker, was synthesized and their in vitro antiproliferative activity against three human cancer cell lines was evaluated. Structure–activity relationships indicated that transformation of the linker -COCH2- into -CH2CH2- led to an increase in the potency of the compound; removal of cladinose and conversion of the carbonyl group at the C9 position to 9(S)-hydroxyl- or 9(S)-amino-containing groups resulted in a marked decrease in activity. Among the compounds synthesized, compounds 3d and 3e, both had C9 oxime groups and showed strong inhibitory activity against the three cell lines. Flow cytometry studies on HT-1080 cells indicated that the mechanism underlying the dimers' antiproliferative activity involved inducing cell cycle arrest in the G0/G1 phase. It was discovered that the percentage of HT-1080 cells arrested depended on the concentration of the erythromycin derivative dimer and on the length of time of treatment.

Abstract:A series of new emodin derivatives 6 similar to 14 containing nitrogen atoms have been synthesized. Their structures were established by H-1 NMR, IR, MS spectra and elemental analyses. The cytotoxicity bioassays of these new emodin derivatives were carried out using the standard MTT method, and most of emodin derivatives showed potent cytotoxicity. The mixture of positional isomers 7a and 7b demonstrated the strongest cytotoxicity activity against the cell line of KB and MCF-7.

Abstract:Twenty-one new diaryl ether substituted piperazine derivatives were synthesized and their structures were confirmed by H-1 NMR, ESI-MS and FIRMS data. The preliminary biological activity tests showed that some of the title compounds had good activities. Compound 4i exhibited a potent cytotoxicity against the Ecal09 and CNE cell lines with IC50 Values of 7.13 and 4.54 mu mol.L-1, respectively, which showed the similar activities to the positive control, cisplatin. Compound 5d also possessed an efficient cytotoxicity aganist CNE cell lines with an IC50 value of 8.49 mu mol.L-1.
