Abstract:A palladium-catalyzed direct 2-alkylation reaction of free N-H indoles has been developed. This reaction relies on a norbornene-mediated cascade C-H activation process at the indole ring, which features high regioselectivity and excellent functional group, tolerance. The reaction represents the first example for a generally applicable, direct C-H alkylation of indole at the 2-position.

Abstract:A palladium-catalyzed direct 2-alkylation reaction of free N-H indoles was developed based on a norbornene-mediated regioselective cascade C-H activation. The detailed reaction mechanism was investigated by NMR spectroscopic analyses, characterization of the key intermediate, deuterium labeling experiments, and kinetic studies. The results indicate that a catalytic cycle operates, in which an N-norbornene type palladacycle is formed as the key intermediate. Oxidative addition of alkyl bromide to the Pd(II) center in this intermediate is the rate-determining step of the reaction. The synthetic utility of this indole 2-alkylation method was demonstrated by its application in natural product total synthesis. A new and general strategy to synthesize Aspidosperma alkaloids was established employing the indole 2-alkylation reaction as the key step, and two structurally different Aspidosperma alkaloids, aspidospermidine and goniomitine, were synthesized in concise routes.

Abstract:The apparent and the real: What looks like a Friedel-Crafts alkylation reaction of electron-deficient pyrroles is actually a Pd(II)-catalyzed, norbornene-mediated C-H activation reaction, in which the alkylation of the pyrrole core occurs by reductive elimination. As well as ethyl-1H-pyrrole-2-carboxylate, several other 2,3-disubstituted pyrroles underwent the selective C5 alkylation in good yield.

Abstract:An effective and practical strategy has been established for the direct and site-selective arylation of indoles at the C4 and C5 positions with the aid of a readily accessible, cheap, and removable pivaloyl directing group at the C3 position. This transformation shows good functional-group tolerance and could serve as a powerful synthetic tool for the synthesis of medicinally relevant compounds. This method and those developed in previous research together enable the regiocontrolled direct arylation of indole at each C-H bond without prefunctionalization of the reactive sites.

Abstract:Recent advances in transition metal-catalyzed selective C-H functionalization of indoles have garnered tremendous attention. Great efforts have been devoted to C2 and C3 arylation because of the inherent reactivity of the pyrrole ring. Until recently, elegant methods have been developed to enable selective direct arylation on the benzenoid moiety at C4, C5, C6, and C7. This review highlights the contributions made in benzenoid direct arylation of indoles and presents their potential in organic synthesis.

Abstract:The indole scaffold will continue to play a vital role in the future of drug discovery and agrochemical development. Regioselective direct arylation of indoles on the benzenoid moiety is a challenging task due to the inherent reactivity of the C2 and C3 positions. Here, we have developed an effective strategy for the regioselective direct arylation of indoles at the C7 position with (hetero)aryl bromides by the rational design of a directing group. The key to the high selectivity and reactivity of this method is the appropriate selection of a class of directing groups, N-PR 2 (R = t Bu and c Hex), that are easily removed in the presence of the Wilkinson’s catalyst. Using the present method as a key step, formal synthesis of marine alkaloid dictyodendrin B has also been demonstrated.

Abstract:A Rh(I)‐catalyzed chelation‐assisted C2‐selective C−H decarbonylative alkylation of indoles with readily available, cheap, safe and structurally diverse alkyl carboxylic acids or anhydrides has been developed. A wide variety of primary and secondary alkyl carboxylic acids and differently substituted indoles are compatible with this transformation, allowing facile synthesis of various C2‐alkylated indoles with high efficiency and broad tolerance of diverse functional groups. The reaction proceeds in the absence of any external oxidant, and the presence of readily installable and removable N‐pyrimidyl directing group is critical for catalysis. The process is convenient and scalable, and avoids the use of a dried solvent and an inert atmosphere. Moreover, selective C7‐alkylation and C2, C7‐dialkylation have also been achieved by slightly modifying the reaction conditions.

This protocol targets α-alkylfurans, achieving moderate to good yields under very practical reaction conditions. Abstract:With alanine as a transient directing group, Pd-catalyzed regioselective alkynylation at the indole C4-position was successfully established in a good yield. The total synthesis of the PAF antagonist demonstrated the synthetic utility of this protocol. The regioselectivity was explicitly proven by the prepared C4-selective palladacycle intermediate in the catalytic process and the DFT calculation of the energy barriers of C4- and C2-site-selective C-H activation of indole.

Abstract:Selective C-H bond alkynylation toward modular access to material and pharmaceutical molecules is of great desire in modern organic synthesis. Reported herein is Ir(III)-catalyzed regioselective C-H alkynylation of ketones and esters, which is generally applicable for the rapid construction of molecular complexity. This protocol provides a complementary process for conventional alkyne synthesis. Further functionalization of carbonyl-derived material molecules and pharmaceuticals demonstrates the potential synthetic utility of this methodology.

The method has the advantages of wide functional group tolerance, high selectivity, broad range of substrates and good yields.

Abstract:A novel palladium catalyzed direct ortho-arylation of N-phenacylpyridinium bromide was developed. The amazing N-phenacyl group regioselectively activates the C-H bond of pyridine and automatically departs from the arylated products. A kinetic isotope effect study proved that the reaction went through a C-H-bond activation pathway and 2,6-diphenylpyridine was produced stepwise from 2-phenylpyridine.

Abstract:<p>A new Rh(III)-catalyzed direct C-H alkenylation of 2,3-unsubstitued indoles with alkenyl borates through C-H activation followed by transmetallation was described. This protocol provides an efficient method for the synthesis of terminal C2-alkenylindoles under mild conditions, and also shows broad substrate scope and high functional group tolerance with respect to both components. Abstract:Methods for the enantioselective direct oxygenation of internal alkenes have provided chemists with versatile and powerful toolboxes for the synthesis of optically pure alcohols, one of the most privileged structural motifs. Regioselectivity, however, remains a formidable challenge in the functionalization of internal alkenes. Here we report a palladium-catalysed highly regio- and enantioselective remote 1, n -dioxygenation ( n ≥ 4) of internal alkenes with engineered pyridine-oxazoline (Pyox) ligands. The reactions proceed efficiently and exhibit a broad substrate scope with excellent regio- and enantioselectivity, affording optically pure 1, n -diol acetates as the key synthons for important bioactive molecules. Experimental studies and density functional theory calculations provide evidence that the regioselectivity is governed by the reactivity disparity of two allylic C–H bonds, where the oxypalladation is reversible and the first palladium migration step proves to be the regioselectivity-determining step, enabled by the modified phenyl-substituted Pyox ligands.

Abstract:Which way? Palladium thiourea catalyzed Pauson–Khand reaction of norbornene with substituted alkynoates to afford selectively either α-carboxylated adducts or β-carboxylated adducts with the reverse regioselectivity has been demonstrated for the first time. Control of the regioselectivity in this transformation is governed by the addition of LiCl. A mechanistic interpretation to account for this observed regioselectivity is presented.

Abstract:Indole represents a privileged scaffold extensively used in drug design due to its promise to improve both binding affinity and specificity receptors. Direct functionalization of indole framework is an efficient route to achieve structurally diverse indole derivatives. Herein, highly regio-selective C3-alkylation of indoles via hydrogen borrowing methodology (HBM) has been established using alcohols as the alkylation reagents by palladium catalysis. This method enables an efficient synthesis of C3-alkylated indoles, demonstrating high group adaptability to both indole and alcohol substrates (20 examples, up to 94% yield).

Abstract:The chosen one! A novel Pd-catalyzed direct and selective C3-acetoxylation of indole derivatives has been accomplished (see scheme). This selective CH activation reaction was implemented without the assistance of directing groups and took place under mild conditions. The kinetic study revealed that the reaction was zero-order with respect to the oxidant and first-order with respect to the indole derivatives.
